Effects of YK-209 Mulberry Leaves on Antioxidative Defense System of Liver in Streptozotocin-Induced Diabetic Rats (YK-209 뽕잎이 Streptozotocin 유발 당뇨쥐 간조직의 항산화계에 미치는 영향)

  • The purpose of this study was investigated the effects of YK-209 mulberry leaves on antioxidative defense system of liver in diabetic rats induced with streptozotocin (STZ). Male Sprague-Dawley rats weighing 100$\pm$10 g were randomly assigned to one normal and four STZ-induced diabetic groups; YK-209 mulberry leaves free diet (DM group),0.1% YK-209 mulberry leaves diet (DM-0.1Y group),0.2% YK-209 mulberry leaves diet (DM-0.2Y group) and 0.4% YK-209 mulberry leaves diet (DM-0.4Y group). Diabetes was induced by intravenous Injection of 55 mg/kg body weight of STZ in sodium citrate buffer (pH 4.3) via tail vein after 4 weeks feeding of experimental diets. Rats were sacrificed at the 9th day of diabetic states. Liver weight in all four diabetic groups were higher than normal group, but YK-209 mulberry supplementation groups were lower than DM group. Hepatic superoxide dismutase (SOD) activity was significantly decreased in all diabetic groups, compared with normal group. Hepatic glutathione peroxidase (GSHpx) activity was 7.3% decreased in DM group, compared with normal group, but those of DM-0.1Y and DM-0.2Y groups were maintained the normal level. The hepatic thiobarbituric acid reactive substances was markedly increased by 144% in DM group, compared with normal group, but those of DM-0. 1Y, DM-0.2Y groups were maintained the normal level. The contents of lipofuscin in liver were increased by 100% in DM group compared with normal group, but those of DM-0. 1Y, DM-0.2Y and DM-0.4Y groups were decreased to 42% 43% and 44%, respectively, compared with DM group. The hepatic superoxide radical (0$^2$-) contents in DM group were increased to 81%, compared with normal group, but those of DM-0.1Y and DM-0.4Y groups were similar to those of normal group. The present result indicate that YK-209 mulberry leaves regarded to suppress lipid peroxidation as an free radical scavenger system by the inhibition of oxidative stress.

Physiological Activities of Mulberry Leaf and Fruit Extracts with Different Extraction Conditions (용매에 따른 뽕잎과 오디의 생리활성 효과)

  • Physiological activities in mulberry leaf and fruit were examined. Electron-donating ability (EDA), tyrosinase activity, SOD-like action (SOD), angiotensin I-converting enzyme-(ACE) inhibitory activity,and nitrite-scavenging ability of mulberry leaf and fruitextracted with water, with 50% (v/v) or with 100% ethanol, were measured. The EDA of mulberry leaf and fruit extracted with water or 50% (v/v) ethanol were greater (by 65.72-81.30%) than that with the 100% ethanol extract, whereas the activities of both former extracts were lower than those with 1.0% and 0.1% (both w/v) L-ascorbate solutions. The SOD-like activities of water, 50% (v/v) and 100% ethanol extracts of all samples were 24.13.26.80% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Nitrite-scavenging activity at pH 1.2 was observed in all extracts. The results further our understanding of the physiological activities of mulberry leaf and fruit extracts.

Influence of Late Summer Fertilizer on the Yield and Cold Damage of Mulberry in Kangwon Province (강원도 지방의 뽕밭에서 철늦은 여름 덧거름이 수량 및 가지끝마름에 미치는 영향)

  • Cold damage of mulberry in Kangweon Province is generally higher that of other provinces. Farmers suspected cold damage caused from late summer fertilizer. To understand the effect of late summer fertilizer. To understand the effect of late summer fertilizer, two additional summer fertilizer times, August 10 and 20, were compared with conventional time(June, 20). Mulberry yield increased by 10~30% in 1m middle cut, and 10~17% in leaf picking method in August 10 treatment, compared with conventional. Additional summer fertilizer did not increase cold damage rate of branches. Leaf yield in leaf picking was higher by 6~29% that that of 1m middle cut.

  • An optimal manufacturing process was established for the mass-production of mulberry leaves tea. The produced tea contained abundant dietary fibers, $\beta$-carotenes, and calcium. Also the polyphenol content and antioxidant activity of mulberry leaves tea were higher than those of simply dried mulberry leaves. These results imply that this tea manufacturing process is effective to utilize the useful components of mulberry leaves.

Effect of Mulberry Leaf Tea for the Removal on Cd and Pb in drinking water (뽕잎차에 의한 음용수중 Cd과 Pb의 제거효과)

  • To investigate the detoxificative effect of tea, five kinds of tea(mulberry, anaerobic treated mulberry, green, barly tea and corm tea) were selected and determined their dotoxication activities for Cd and Pb in drinking water. The effect of tea on the removal of Cd and Pb were increased proportionally to the contents of teas. Anaerobic treated mulberry leaf tea showed stronger detoxication activity than the others. In drinking water contaminated with Cd, the removal effect of Cd was high 27% by anaerobic treated mulberry leaf tea and 14% by mulberry leaf tea as compared to green tea. Also, in drinking water with Pb, the removal effect of anaerobic treated mulberry leaf tea was the best among five kinds of tea. Pretreatment method on the removal effect of Cd was better than post-treatment method in the treatment method.

  • The investigation of 218 residual pesticides for 19 types of tea (persimmon leaf tea, chrysanthemum tea, green tea, lavender tea, rosemary tea, dandelion leaf tea, puer tea, mulberry leaf tea, hydrangea leaf tea, jasmine tea, nuomixiang tea, buckwheat tea, mugwort tea, lotus leaf tea, oolong tea, longjing tea, rose tea, tiehkwanyin tea and huoguo tea) obtained from markets in Ansan and Suwon was carried out to assess the risk for residual pesticides in tea. The detection rate was 23.1 % (19 samples of total 65 tea samples) and the detected pesticides were 15 pesticides, such as bifenthrin, bromopropylate, chlorpyrifos, cyhalothrin, cypermethrin, chlorfenapyr, dicofol, endosulfan, fenpropathrin, fludioxonil, fenvalerate, iprobenfos, isoprothiolane, tetradifon and triazophos. The range of concentrations for the detected residual pesticides was 0.01 to 1.24 mg/kg which showed below their maximum residue limits (MRL), but the residual concentration of bifenthrin in a puer tea showed above the legal limit of 0.3 mg/kg. The result of risk assessment of residual pesticides for the detected 15 samples showed that EDI (estimated daily intake) of the pesticides detected ranged 0.0001~0.0844% of their ADI (acceptable daily intake).

Hypoglycemic Effects of Pills Made of Mulberry Leaves and Silkworm Powder in Streptozotocin-Induced Diabetic Rats (뽕잎과 누에가루 혼합환의 Streptozotocin유발 당뇨쥐에서의 혈당강하 효과)

  • This study was carried out to examine the effects of pills made of mulberry leaves and silkworm powder on lowering blood glucose level. Experimental animals were Sprague-Dawley male rat weighing 100$\pm$10 g and pills were supplemented with 0.4% (4 g/kg) diet. Experimental groups were assigned to diabetic group (DM group) and pill supplemented groups. Pill supplemented groups were classified 100% mulberry leaves (M group), mixing 25% silkworm powder to mulberry leaves (25SM group), mixing 50% silkworm powder to mulberry leaves (50SM group), mixing 70% silkworm powder to mulberry leaves (75SM group) and 100% silkworm powder (100S group). Experimental diets and water fed ad libitum, and streptozotocin was injected to induce diabetic state after 3rd weeks and sacrificed on the 9th day. The contents of 1-deoxynojirimycin(DNJ) were increased with adding the silkworm powder. The contents of GABA and rutin were increased with adding the mulberry leaves. In vitro, intestinal mucosa $\alpha$-glucosidase inhibitory activities were significantly increased in pills which mixed with silkworm powder by 50%. Blood glucose levels were high in groups which mixed with silkworm powder by 50% compared to DM group. Intestinal mucosa maltase activity in proximal part was significantly reduced in pill supplemented group compared to DM group and pill supplemented groups were no significant difference. Enzyme activity in middle part was no significant difference in experimental groups. Enzyme activity in distal part was decreased in pill supplemented groups, especially in 50SM, 75SM and 100S groups were significantly reduced compared to DM group. Sucrase and lactase activities in pill supplemented groups were significantly reduced at proximal part, and there was no significant difference in middle and distal parts. In conclusion, pills made of mulberry leaves and silkworm powder increased the $\alpha$-glucosidase inhibitory activity in vitro and reduced the blood glucose levels by controlling the disaccharidase activities of intestinal proximal part in STZ-induced diabetic rat. The synergistic effect was the highest when mulberry leaves was mixed with silkworm powder by the ratio of 50 : 50.

Quality and Antioxidative Characteristics of Cudrania tricuspidata Leaves Tea (꾸지뽕잎차의 품질 및 항산화 특성에 관한 연구)

  • To obtain basic da1a on the preparation of Cudrania tricuspidata leaves tea, the quality and anti-oxidative characteristics of dried raw leaves (RT), pan-fired leaves tea (PT) and fermented leaves tea (FT) were investigated. General characteristics of RT, PT and FT, respectively, were: moisture content 18.47, 6.23 and 8.50%; crude protein content 17.77, 20.46 and 19.13%; and carbohydrate content 54.42, 62.52 and 61.96%. The crude lipid and ash contents were in the range 0.05 - 0.07% and 9.27 -10.74% respectively; the water soluble solid content was in the order FT > PT > RT and ranged from 23.10 - 37.38%; there were no significant differences in the total polyphenol content (815.24 - 835.16 mg%). Although $L^*$ values of PT (20.94) and FT (20.85) were lower than those of RT (34.71), the $a^*$ value in PT and the $b^*$ value in FT were highest. In all ethanol extracts the reducing power, electron-donating ability and superoxide dismutase (SOD)-like activity increased in a concentration-dependent manner. Furthermore, the activity in FT was higher than in PT or RT. The total free amino-acid content was higher in FT (1429.93 mg%) than RT (1108.94 mg%) or PT (833.13 mg%). The major amino acids were L-asparagine and L-valine in RT, L-cysteine and L-glutamic acid in PT and L-proline in FT. In a sensory evaluation of PT and FT, bitter and astringent tastes were decreased relative to RT, while sweet and savory tastes, flavor, color and overall acceptability were increased. These results indicate that FT bas a higher antioxidant effect and free-amino-acid content than PT, while the sensory quality of FT is similar to that of PT.

Effect of Mulberry Leaf Sprayed with Rutin and GABA on Silkworm Growth (Rutin 및 GABA 첨식이 누에 성장에 미치는 영향)

  • This experiment was carried out to investigate the feeding effect of rutin (quercetin-3-o-${\beta}$-rutinoside) and ${\gamma}$-aminobutyric acid(GAB) sprayed on mulberry leaves on silkworm growth. Amount of ingest, digest and feces showed no difference statistically except rutin. Rutin made 6% increase of amount of digest, compared to the control. However, rutin or GABA decreased maximaly silkworm weight by 28% during the experimental period. GABA showed body weight loss by 15%, rutin by 7% respectively, compared to the control. Rutin and GABA extended three days larval period and extended one day mounting period and decreased 15.4% and 91% in number of mounted silkworm compared with the control, respectively.

Anti-bacteria effect of green tea, mulberry, and mate leaves extracts on S. mutans (녹차잎, 뽕잎, 마테잎 추출물이 S. mutans에 미치는 항균효과)

  • This study aims to figure out how green tea leaves, mulberry leaves, and mate leaves that are often taken around in drinking tea influence on S. mutans known as dental caries causative bacteria. After vaccinating 1% of S. mutans on the extract added badge and incubating 6 hours and 10 hours in $37^{\circ}C$, it calculated absorbance and bacterial colony number. Extract of green tea leaves and mate leaves were added with 0, 0.5, 1.0, 2.0, and 4.0% of concentration. As a result of growth suppressive effect of S. mutans, it may be observed that colony number significantly decreases in higher concentration. When it calculated bacterial colony number with adding 2% extract after 10 hours, 99.0% for green tea leaves, 97.1% for mulberry leaves, and 89.6% for mate leaves appeared.
