Germination and Antioxidant Activity of Korean Oak Pollen Treated with Medicinal Mushrooms (약용버섯을 이용한 도토리화분의 세포 발아 및 항산화 활성)

  • In this study, we measured antioxidant activity as DPPH radical scavenging and the total polyphenol content of pulverized and lyophilized oak pollens inoculated with fungi to confirm the husk removal effect. The total polyphenol content of oak pollen was highest in lyophilized pollen medium inoculated with Armillaria mellea, and was lowest in pollen inoculated with Lentinula edodes. Total polyphenol content of the lyophilized pollen was higher than that of the refined pollen and the pulverized pollen in oak pollen germinated with A. mellea. The total polyphenol content of the lyophilized oak pollen germinated with A. mellea was 1.4-fold higher than that extracted with water. Measurement of antioxidant activity using the DPPH (2, 2 diphenyl-1-picrylhydrazyl) free radical scavenging method showed that the lyophilized oak pollen germinated with A. mellea had the highest and that germinated with L. edodes was lowest in antioxidant activities. The lyophilized oak pollen germinated with A. mellea was 2 to 4 times higher than that extracted with water in the antioxidant activity of DPPH free radical scavenging. Many germinated cells were formed around pore of acorn pollen inoculated with L. edodes, while those were formed at the end of hyphae derived from oak pollen inoculated with A. mellea.

Genetic Diversity of Mulberry Dwarf Phytoplasma(MD) by SSCP Technique (SSCP기법에 의한 뽕나무오갈병 파이토플라스의 유전적 다형성 분석)

  • Phytoplasmas were detected consistently in 42 mulberry cultivars showing dwarf disease using DNA analysis by amplification with phytoplasma universal primer pairs P1/P7 (about 1.8 kb and R16F2n/R2 (about 1.2 kb). The point mutation from 42 cultivars of mulberry tree was detected by single-strand conformation polymorphism (SSCP) analysis. The SSCP profiles were clearly observed from all of cultivars in 8% polyacrylamide gel, electrophoresizing for and running 8-15 hrs. at 150V, $10^{\circ}C$. The MD and JWB phytoplasma PCR products was mixed and electrophoresis was performed to detect their polymorphism. In this results, the SSCP profiles of all bands of MD and JWB were analyzed on single lane and were distinct in their each of band patterns. The SSCP analysis was possible to detect of 1.8 kb and 1.2 kb nucleotide size and near close band patterns were distinct by mix of two samples. Previously, it was only possible to detect of point mutation under 600 bp nucleotide sequence by SSCP analysis but this modification of SSCP technique was possible to detect clearly SSCP band patterns of about 1.8 kb and 1.2 kb nucleotides.

Cudrania tricuspidata Root Extract as Whitening and Antiwrinkle Cosmetic Agent (꾸지뽕나무 뿌리 열수추출물로부터 미백 및 주름개선 화장품 소재의 개발)

  • Water extracts of root, leaf, branch and fruit of Cudrania tricuspidata were tested to use as natural antiwrinkle and whitening cosmetic ingredient. Root extract indicated 86.5 mg/g polyphenol concentration and 55.5 mg/g flavonoid concentration. From MTT test, root, branch and fruit extract did not show cell toxicity up to $1,000{\mu}g/mL$, however, leaf extract displayed significant cell toxicity in concentrations higher than $200{\mu}g/mL$. Cudrania tricuspidata root extracts displayed excellent 81% tyrosinase inhibition (whitening effect), 58% elastase inhibition (antiwrinkle effect) at $500{\mu}g/mL$ and showed minor UVB absorbance. When solution and lotion formulation were made including Cudrania tricuspidata root extract, they are stable at $25^{\circ}C$ for 21 days, but showed significant deviation in viscosity and particle size at $47^{\circ}C$. From the study, Cudrania tricuspidata root extract demonstrated good potential for natural whitening and antiwrinkle cosmetic agent.

Isolation of Non-toxic Bacillus thuringiensis Strains from the Dead Larvae of Apriona germari and Aphodius apicalis (뽕나무하늘소(Apriona germari) 및 왕똥풍뎅이 (Aphodius apicalis) 사충으로부터 무독성 Bacillus thuringiensis의 분리)

  • Four strains of Bacillus thuringiensis were isolated froin the dead larvae of mulberry longicorn beetle (Apriong germari) and dung beetle (Aphodius apicalis). One nf four B. thuringiensis isolates turned out to be subspecies darinstadiensis but the remains were not identified using 33 B. thuringiensts flgellar ( H ) antibodies. Furthermore. bioassays of spore-parasporal inclusion protein mixture conducted against third instar larvae of A. gerrntrri or A. apicalis, second instar larvae of Bombyx mori, and third instar larvae of Cu1ex pipiens pullens showed that the isolates were non-toxic. To further confirm, four isolates were characterized and analysed by SDS-PAGE and agarose gel electrophoresis. The results revealed that parasporal protein and plasmid DNA patterns of four isolates are different from those of darmstadiensis and 20 known non-toxic B. thuringiensis strains, suggesting that the four isolates are novel non-toxic B. thuringiensis.

Evaluation of Fruit Characteristics According to Mulberry Breeding Lines for Fruit Production (오디 생산을 위한 뽕나무 계통별 과실 특성 평가)

  • We evaluated fruity characteristics to select the desirable mulberry varieties for the production of fruit. Tested items were total yield, weight of single fruit, soluble solids and fruit size including length, width, and petiole length. 'Susungppong', 'Cheongnosang', 'Suwonnosang' and 'Jeolgokchosaeng (Chungbuk)' were showed high yield for three years. In weight of single fruit, 'Jeolgokchosaeng (Chungbuk)' was 4.5 g. 4 mulberry varieties were showed move than 17 Brix $\%$ in soluble solids. From that results, eight suitable varieties such as 'Jeolgokchosaeng (Chungbuk)', 'Dangsang 7', 'Jangsosang', 'Susungppong', 'Suwonnosang', 'Palcheongsipyung', 'Kangsun', and 'Jukcheonchosaeng' were selected for the production of fruit.

  • The incidence of mulberry dwarf disease was surveyed on seventy two cocoon producing areas throughout the country from 1983 to 1984. There was a low tendency of incidence in Gyeonggi, Gangwon and Chungbuk, the northern parts of 37 degrees north latitude. However, in Chungnam and Gyeongnam, was a relatively high incidence observed. The most severe incidence areas of mulberry dwarf disease were found in Jeonnam, Jeonbuk and Gyeongbuk.

  • Ethanol 75% extracts of Morus alba Linne(Ma) and Sophora flavescens AITON(Sf) were fractionated by chloroform($CHCl_{3}$), ethyl acetate(EtOAc), butanol(BuOH) and water. Each fractionate was added to tryptic soy broth varying in extract levels and 5 strains of Listeria monocytogenes(Lm) were inoculated in them for comparing growth inhibition at $32^{\circ}C$. The $CHCl_{3}$ and EtOAc fraction of Ma were superior in growth inhibition of 5 strains but only $CHCl_{3}$ fraction of Sf showed comparatively higher inhibition to Lm ATCC 19113. Lm ATCC 19111 and 19113 were totally inhibited by 500 ppm of EtOAc fraction and Lm ATCC 19112, 19114 and 15313 were also absolutely inhibited by 1000 ppm of $CHCl_{3}$ fraction of Ma 75% ethanol extract.
