Characteristics of mulberry cultivar Shimheung (Morus alba L.) for mulberry fruit production (오디 생산용 뽕나무 품종 '심흥'의 특성)

  • We bred a mulberry cultivar named Shimheungppong, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at three places(Suwon, Wanju, and Jinju) for six years from 2004. This is tetraploidy variety belonging to (Morus alba L.) selected among mulberry genetic resources preserved in Suwon. Shimheungppong was high yielding cultivar in fruit productivity by 5% compared to control cultivar 'Chungilppong(Morus alba L.)' for four years. Although Shimheungppong was a little lower in sugar content of mulberry fruits, it was bigger than that of Chungilppong. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

Physiological Activities of Extracts from Different Parts of Cudrania tricuspidata (꾸지뽕나무 부위별 추출물의 생리활성 탐색)

  • The physiological activities of extracts from the leaf, stem, and fruit of Cudrania tricuspidata were investigated. The electron-donating ability (EDA) of the 70% (v/v) ethanolic extract of stem was 90.20%; this was the highest value of all the extracts tested and higher than the L-ascorbate solutions. The total polyphenol contents were the highest in the leaf extracts under all extraction conditions. Especially, 70% (v/v) methanolic extract of leaf contained the highest total polyphenol content of 224.48 mg%. SOD-like activity showed the highest activity in water extract of leaf at 64.53%. Tyrosinase-inhibitory activities were the most effective in all extracts of fruit. ACE inhibitory activities were the highest in water extract of fruit. Nitrite-scavenging abilities under acidic conditions (pH 1.2 and pH 3.0) were the most effective in all the extracts. The results of this study will be useful for understanding the physiological activities of Cudrania tricuspidata extracts.

  • This study was planned to find out the effects of several food-plants on the pupal weight and the number of eggs in the ovary of the fall webworm, Hyphantria cunea $D_{RURY}$. The larvae were separately reared on different food-plants in the Petri dish. Four to six day old pupae were used for the study of the pupal weight. The number of eggs in the ovary was counted under the binocular dissecting microscope. The loaves of the food-plants were sampled on Juiy 16, and analyzed by Kjeldahl method for the determination of total nitrogen content. The results obtained in this study were as follows; (1) The pupal weights were highly different according to the species of plants and sexes. The greatest pupal weight found in the pupae reared on the mulberry and the least one in the pupae reared on the apple tree. The weight female pupae was greater than that of male pupae in every case. (2) The number of eggs in the ovary was positively correlated to the pupal weight. (3) The nitrogen content of the food-plants used in this experiment was varied with the species of the plants. However, the nitrogen content of the food-plants was not significantly correlated to the pupal weight and the number eggs in the ovary.

Aspects of Popcorn Disease Occurrence on Mulberry Fruits in Korea (국내 오디균핵병의 발생 양상)

  • Occurrence of popcorn disease on mulberry fruits was surveyed in several locations in Korea from 2005 to 2007. The diseased fruits turned grayish white and changed to hard and black sclerotia during overwintering after falling onto the ground. Apothecia produced from overwintered sclerotia in the fields of mulberry trees were observed in early May. Two types of apothecia were produced from the sclerotia, which were cup-shaped or club-shaped. The fungus with cup-shaped apothecia was identified as Ciboria shiraiana, and that with club-shaped apothecia as Scleromitrula shiraiana. C. shiraiana occurred at the ratio of about 6 vs. 4 in the fields. Occurrence of the disease differed remarkably depending on years, locations, and varieties of mulberry trees investigated. Field survey of the disease incidence showed that Baekhahyeongsang, Dahojosaeng, Hasusang, a native variety and Sawonppong 22 were resistant to the disease, Cheongilppong, Daeseongppong, Guksang 20 and Turkey-D were moderately susceptible, and Cateneo, Moreji and Sanggwa were highly susceptible. Disease incidence in the fields mulched with nonwoven fabrics or plastic films was much lower than that in the non-mulched fields. Disease incidence in the fields mulched with rice straws was similar to that in the non-mulched fields.

Component(mineral) difference of Phellinus linteus on cultured to timber (기주식물에 따른 목질진흙버섯의 일반성분(무기물) 함량변화 I)

  • We investigated mineral difference from mushroom Phellinus linteus and Phellinus baumii on mulberry, oak and elm. ICP test, The major mineral components were K, Ca, and Fe, while Mg, Na, P, Cr, Mn, Cu and Zn were determined as micro mineral components. In case of P.linetus and P.baumii the component of K was shown as 699.0mg/100g and 630mg/100g, in oak, 340.7mg/100g and 314.6mg/100g in elm, and 311.6mg/100g and 311.6mg/100g in mulberry respectively. In Ca, mulberry fruiting body were shown about 2 times highest and expecially in case of Na was oak fruiting body were shown as 10 times highest as against the others sample groups. In micro mineral components, Zn component was shown very diffeence about sample groups. We concluded that the highest mineral component of K was shown in mushroom cultivated on oak of both P.linetus and P.baumii. As a results, mineral component has showed very difference on cultivated to timber. According to the results of mineral component we concluded that Phellinus linteus and Phellinus baumii has mineral component different depending on timber.

Production of Novel Flavonoids in Cell Cultures of Cudrania tricuspidata (꾸지뽕나무 (Cudrania tricuspidata)세포배양에 의한 신규 Flavonoids 생산)

  • To produce novel bioactive flavonoids, Gericudranin A and Gericudranin B, a cell culture system of Cudrania tricuspidata including callus induction and optimization of culture conditions was established. Friable calli were efficiently induced from the hypocotyl segments of seedlings on B5 medium supplemented with 1.0 mg/L NAA, 0.1 mg/L kinetin and 3% sucrose. Several factors were optimized for the Gericudranin production and the cell growth in suspension cultures. Low level of basal salt medium (1/8 MS), 1.0 mg/L IAA and 0.1 mg/L zeatin, and high level of sucrose (5%) were effective for the production of Gericudranins, whereas WPM with 1.0 mg/L NAA, 0.1 mg/L zeatin, and 5% sucrose were more effective for the cell growth. When cells were cultured on MS liquid medium supplemented with 1.0 mg/L IBA, about 2200 $\mu\textrm{g}$/g dry wt of Gericudranin A could be produced. The level might be about 10 times of the native inner bark. About 2350 $\mu\textrm{g}$/g dry wt of Gericudranin B was also produced on MS liquid medium with 5% sucrose, 1.0 mg/L NAA, 0.1 mg/L kinetin. The content was estimated about 3 times of the level of native inner bark.

Effect of Vinyl Mulching on Growth of Various Sizes of Mulberry Saplings in a Dense Planting (밀식 뽕밭 조성시의 묘목규격별 비닐 피복효과)

  • This experiment was carried out to develope a new planting system from the economical point of view, using the various sizes of mulberry saplings ranged from 3mm to 7mm (3, 4, 5 and 7mm) in diameter of branches. They were planted densely (2,285 trees/10a) in vinyl film-and non-mulching system. The results are summarized as follows. 1. There are no differences in growth and yield of the planting year between 6mm and 7mm saplings. 2. The vinyl mulching affords an abundant growth of the saplings due to retaining of soil temperature and moisture. The dense planting with vinyl film mulching gave rise to 26% and 7% increase of leaf yield, using the saplings sized of 7mm and 5mm, respectively. 3. On the other hand, it was evident that the use of 6mm saplings in non-mulching and 5mm ones in mulching redused possibly the sapling cost. It was also noteworthy an additional profit took places in the yield increase, the cost reduction of saplings and weeding by using the 7mm and 5mm saplings in mulching system. It amounted to the increase of ₩33,800 and ₩24,520, respectively over the use of 7mm saplings in non-mulching system.

Structure Analysis of Water-soluble Polysaccharides Extracted from The Unripe Fruit of Cudrania tricuspidata (꾸지뽕나무 열매에서 추출한 수용성 다당류의 구조분석)

  • The unripe fruit of cudrania tricuspidata was extracted with 50% ethanol. The crude water-soluble extracts were separated by liquid-liquid separation with n-hexane, ethyl acetate and butanol followed by precipitation with ethanol, and then the water-soluble polysaccharide (F1) was isolated by the fractionation through gel permeation chromatography using preparative PLaquagel-OH column with water. The structure was characterized by monosaccharide composition with HPAEC-PAD, methylation analysis with GC-MS, FT-IR and HPLC. According to the data, F1 was com posed of glucose (22.84 mM), galactose (13.75 mM), arabinose (45.87 mM), xylose (7.49 mM). It was revealed which uronic acid and acetyl group were not attached in F1. And it is constituted of 1-linked arabinose, 1,4-linked arabinose, 1,3-linked glucose, 1,4-linked galactose, 1,4-linked glucose, 1,3,6-linked galactose, 1,3,6-linked glucose and the ratio was showed 1.1 : 1.0 : 4.9 : 7.5 : 3.0 : 3.1 : 1.4 : 1.5.

Tyrosinase Inhibitory Xanthones from Cudrania tricuspidata (꾸지뽕나무에서 분리한 xanthones의 tyrosinase 저해제 연구)

  • The methanolic roots bark extract of Cudrania tricuspidata (Carr.) Bureau was chromatographed, which yielded three xanthones 1-3 by tyrosinase inhibitory activity-guided fractionation. The structures were fully characterized by analysis of physical and spectral data. Among them, furano prenylxanthone 3, never reported as tyrosinase inhibitor, showed potent activity with $IC_{50}$ value of $16.5{\mu}M$, and appeared to inhibit the polyphenol oxidase activity of tyrosinase in an uncompetitive inhibitor($K_i=1.6{\mu}m$) when L-tyrosine was used as a substrate. Moreover, potent inhibitor furano prenylxanthone 3 had an extended lag time of 310 sec at $20{\mu}M$, while lag time of kojic acid as positive control was prolonged with 350 sec at the same concentration.

Phenotypic and Genotypic Correlations between Some Characters of Mulberry Trees (상수 각형질 상호간의 표현형상관과 유전상관)

  • The experimental studies were intended to clarify the effects of leaf yield calculations, and also aimed at estimating the correlations between some characters for the selection of desirable stocks of mulberry trees. The analysis of covariances was the variance components procedures in a replicated trial for the varieties, and also it was used to obtain the genotypic, phenotypic and environmental correlations between the eight characters-branch length, node number, branch diameter, branch number per stock, total branch weight, old branch weight, new shoot and leaf weight, and leaf weight, etc. The results obtained are summarized as follows: Phenotypic, genotypic and environmental covariances are shown in Table 1 and phenotypic correlations, genotypic correlations and environmental correlations are shown in Table 2. Genotypic correlations were slightly higher than the corresponding phenotypic correlations between the characters in the varieties, but the variation in values due to the change of environment appeared between the characters. Genotypic correlations between the leaf yield of mulberry trees and other characters indicated that high leaf yield was genetically correlated with six characters namely branch length, node number, branch number per stock, total branch weight, old branch weight and new shoot weight, etc. It was also observed that lower correlations were appeared between the leaf yield and branch diameter, and branch diameter and other all characters studied in both genotypic and phenotypic correlation coefficients.
