Ulcer Healing Effects of Vitamin E on Chronic Gastric Ulcer Induced by Alcohol in Young Adult Rats (알코올로 유도한 만성위궤양 흰쥐 모델에서 비타민 E 보충이 위궤양 치유에 미치는 영향)

  • This study was carried out to examine the effects of vitamin E on chronic gastric ulcer induced by alcohol treatment in rats. Chronic gastric ulcer model was established by oral administration of 70% ethanol at one time and supply of 15% ethanol for additional 7 days. Male Sprague-Dawley rats, approximately 200 g, were fasted for 24 hours and orally gavaged with 1 mL of 70% ethanol for the induction of acute ulcer. A supply of 15% ethanol dissolved in distilled water for 7 days were followed to maintain chronic gastric ulcer. Acute ulcer group was sacrificed at 3 hours after oral administration of 1 mL of 70% ethanol. Chronic groups were divided into three groups according to vitamin E levels; low-vitamin E (LVE, 0 mg/mL oil/day), normalvitamin E (NVE, 1 mg/mL oil/day) and high-vitamin E (HVE, 10 mg/mL oil/day). These groups were fed vitamin E free diets which were made of vitamin E free vitamin mix followed AIN-93M pattern for 7 days. Histological findings of congestion, hemorrhage and necrosis in gastric tissue were shown severely in acute ulcer group and LVE group of chronic ulcer groups. The concentration of gastrin in serum was significantly higher in LVE group. The content of histamine in stomach was lower in acute ulcer group but there was no significant difference among the chronic groups regardless of vitamin E levels. Content of malondialdehyde (MDA) in gastric tissue was higher in HVE group and activities of antioxidant enzyme, glutathione peroxidase (GPx) and catalase, were lower in HVE group. Myeloperoxidase (MPO) activities as a marker of neutrophils infiltration was significantly higher in LVE group. These results suggested that vitamin E supplementation has positive effects on healing of alcohol-induced chronic gastric ulcer through alleviation of gastric tissue injuries and reduction of the MPO activity in gastric tissue and gastrin in serum.

  • 식생활의 서구화로 제빵 산업이 발달하고, 빵을 주식으로 하는 인구가 늘고 있다 또한 기존의 재료보다는 기능성이 첨가된 부재료를 활용한 건강 지향적인 식품의 수요가 증가하고 있는 추세이다. 최근 밀배아의 용도 다양화를 위하여 팔은 연구가 진행되고 있으며, 상업적으로 이용되고 있다. 소맥의 제분과정에서 부산물로 얻어지는 밀배에는 식물 중에서 가장 많은 천연 비타민 E 복합체가 이상적인 상태로 함유되어 있다. 밀배아에 들어 있는 비타민 E는 천연 비타민 E 복합체와 합성 비타민 E로 구분되어진다. 천연비타민E는 8가지 이성체를 가진 자연 비타민 E를 말하며, 4종류의($\alpha$$\beta$${\gamma}$$\delta$)의 토코페롤과 4종의 토코페롤의 복합체이다. 비타민 E는 사람을 포함한 고등동물에 필수적인 영양소로 식물성기름, 곡류, 배아 등에 많이 들어 있으며, 여러 이성질체중 $\alpha$-토코페롤이 체내에서 가장 생물학적 활성이 큰 것으로 알려졌으며, 생체막조직에서 인지질의 천연항산화제 역할을 하여 체내대사를 정상으로 유지하는데 큰 기능을 하는 것으로 보고되고 있다. 우리나라에서 식용으로 소비되는 밀은 거의가 수입에 의존하고 있으며, 밀배아는 거의 전량이 가축의 사료용으로 사용되고 있고 근래에 와서는 배아유, 배아떡, 복합 조미료, 쿠키, 배아를 첨가한 국수 등에 이용되고 있다. 따라서 본 실험에서는 식빵제조시 밀배아 분말을 0, 2, 4, 6, 8, 10%씩 첨가하여 기능성 식빵을 제조하였을 때의 반죽의 물성, 호화특성을 살펴보고 빵으로 제조하였을 때의 식빵의 품질 특성으로서 1차 발효 손실률, 굽기 손실률, 부피, 질감, 노화도, 관능적 특성 및 mould-free shelf life를 측정하였다.te, ferric citrate가 유제품에 사용하기에 적합한 것으로 나타났다. 생이용성을 평가하기 위하여 우유에서 low molecular weight components(ILC)를 분리하고 철분과 복합체를 형성시킨 다음, 철분 결핍된 쥐의 소장에서 loop을 형성시켜 ILC-철분 복합체를 injection하여 철분 흡수도를 조사하였다. Ferrous lactate 100ppm에서 약 25.6%흡수되었고 ferric citrate 100ppm은 24.7%, ferrous sulfate는 19.7%흡수되었다. ILC를 첨가하지 않은 100ppm 철분염 용액은 ferrous sulfate를 제외하고는 흡수도가 감소되었다. 철분 결핍된 쥐에게 gavage 방법에 의하여 철분강화우유를 투여하였을 때 철분 25ppm 시료에서는 ferrous sulfate가 12.5%로 가장 높았고 ferrous lactate는 8.1%, ferric citrate는 6.5% 흡수되었다. 철분 100ppm수준에서는 흡수율이 낮아져 ferrous sulfate는 25ppm 시료보다 절반이하 수준이었다. Ferric citrate는 차이가 거의 없었으며 ferrous lactate는 70%수준이었다. 이상의 결과에서 철분강화우유에 사용하기 적합한 철분염은 ferrous lactate, ferric citrate였는데 특히 ferrous lactate는 제품의 이화학적 품질, 생이용성 측면 모두에서 가장 좋은 것으로 나타났다.다 높았으며, 1회당 평균 8.1$\pm$5.1개의 난포란을 회수하였다. investigation can be separated into sampling and analytical uncertainties, it can be used as a criterion where the resources for the inves

Effect of Vitamin E Treatments on The Humoral and Cellular Immune Responses in Mice. - Animal experiment for nursing care of vitamin E-deficient patients- (비타민 E 투여가 마우스의 체액성 및 세포성 면역반응에 미치는 영향 -비타민 E 결핍환자의 간호중재 개발을 위한 동물실험 -)

  • Vitamin E, which has its advocates in the treatment of diabetes mellitus. autoimmune disease, cancer and peripheral vascular and thromboembolic disease, has now been alleged to have a powerful antioxident effect and to affect various biological activities such as fertility factor, inhibition of human platelet aggregation and stabilization of biological membranes. The present study was designed to test whether vitamin I(alpha-tocopherol) can : (1) enhance the hemagglutinin response to sheep red blood cells (SRBC), (2) modulate Arthus and delayed type hypersensitivity(DTH) to SRBC and contact hypersensitivity to dinitrofluorobenzene (DNFB). (3) enhance the mitogenic response of murine splenocyte, (4) decrease the recovery of Cryptococcus neoformans from brain, lung, liver, spleen and kidney of infected mice and (5) have an inhibitory or enhancing effect on the induction of active systemic anaphylaxis(ASA) induced by chicken-gamma globulin (CGG) in mice. Mice were given either intramuscular injections of 0.3ml (300mg) of vitamin I before immunization or were infection for 10 consecutive days or were given by vitamin I esophageal intubation, 0.1ml(100mg), for 20 days before sacrifice for the mitogenic response experiments. It was found that vitamin E treated mice showed a significant enhancement in hemagglutinin response, Arthus reaction and DTH to SRBC and contact hypersensitivity to DNFB. There was no significant difference in the mitogenic response to phytohemagglutinin(PHA), but the response to concanavalin A(ConA) or pokeweed mitogem(PWM) was increased in vitamin E-treated mice. Interestingly, the vitamin E administration before C. neoformans infection decreased significantly the recovery of C. neoformans from brain lung, liver, spleen and kidney of the infected mice as compared with that of the control mice, strongly suggesting that vitamin E pretreatment may increase the resistance of mice to the fungal infection. Unexpectedly, vitamin E administration enhanced the production of CGG -induced ASA. Taken together, it can be concluded that vitamin I administration may in-crease the humoral and cellular immune response and resistance. to C. neoformans infection, but enhance the induction of ASA to CGG. Further studies are necessary to clarify the underlying mechanism accounting for these effects.

  • Effects of dietary fatty acids and vitamin E on antioxidant system were studied in rat liver and serum. Sources of dietary fat (10 wt%) were safflower oil (SO) poor in $\omega$3 fatty acid and mixed oil (MO) with computer-adjusted fatty acid ratios (AA/DHA=1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/l.5/1) with (ME) and without (MO) vitamin E (500 mg/kg diet). Rats were fed the three kinds of diet from 3~4 wks prior to the conception. At the age of 3 and 9 wks of the second generation rat, antioxidant vitamins and glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities were measured in the liver and serum. The concentrations of $\beta$-carotene were lower in ME than in MO and SO in the liver at the age of 3 wks. It seemed that vitamin E has an inhibitory action on the uptake of $\beta$-carotene or acts as a preferred antioxidant to $\beta$-carotene. The concentrations of lycopene were lower in SO than in MO in the liver at the age of 3 wks. The concentrations of cryptoxanthin showed no significant changes within groups. The activities of GSH-Px tended to increase in ME compared to MO and the ratios of SOD/GSH-Px tended to decrease in ME compared to MO in the liver at the age of 3 weeks. The activities of antioxidant enzyme at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidant enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period.

Equine Motor Neuron Disease in a Jeju Pony (제주 조랑말에서의 말운동신경세포질환 일례)

  • Equine motor neuron disease (EMND) is a spontaneous neurologic disorder of horses, which results from the degeneration of motor neurons in the spinal cord and brain stem. An 8-year-old Jeju pony gelding presented with weight loss, muscle tremors, frequent recumbence, low head carriage, sweating, and standing with four limbs close together. The gelding has been on the same stable and limited access to pasture for several years. The gelding has been fed with dried hays and commercial concentrated feeds. Laboratory test revealed very low serum vitamin E level (0.14 ${\mu}g/mL$; reference range > 1.5 ${\mu}g/mL$), mildly elevated creatine kinase (402 IU/L; reference range 119-287 IU/L) and aspartate aminotransferase (876 IU/L; reference range 226-336 IU/L). Oral glucose absorption test showed decreased glucose absorption. Histopathologic examination using a biopsy specimen from sacrocaudalis dorsalis medialis muscle revealed atrophic and hypertrophic muscle fiber, centralization of nucleus, degenerating and necrotic muscle fibers. Taken together, the gelding was diagnosed as EMND. After oral vitamin E administrated for 5 weeks, the gelding showed normally improved stance, decreased periods of recumbency, improved head carriage and weight gain except consistent tremors. This is the first report that successfully treated EMND case occurred in a Jeju pony in Korea.

Assessment of selenium and zinc status in female collegiate athletes (대학 여자 운동선수의 셀레늄 및 아연 영양상태)

  • Purpose: This study was aimed to assess selenium and zinc status in female collegiate athletes and their relationship with dietary intake. Methods: Female collegiate athletic groups of judo and aerobics, and healthy sedentary collegiate females were recruited for this study and their serum selenium and zinc contents were measured by the neutron activation analysis (NAA) method. In addition, the dietary intake of subjects was measured using the two days 24-hour recall method. Results: Serum selenium in judo athletes was $10.7{\mu}g/dl$, which was significantly lower than that of aerobic athletes ($12.2{\mu}g/dl$), but not different from that of the sedentary group ($11.4{\mu}g/dl$). Additionally, serum zinc levels were $96.1{\mu}g/dl$ and $90.2{\mu}g/dl$ in aerobic and judo athletes, respectively, which did not differ significantly. Moreover, dietary selenium and zinc intake of the athletic groups did not differ significantly from that of the sedentary female group. Overall, 33.3% of the serum selenium concentration variation was explained by the intake of vitamin E, selenium and saturated fatty acids, while 14.7% of the serum zinc level variation was explained by the intake of saturated fatty acids. The strongest dietary indicator for serum selenium and zinc levels was saturated fatty acids intake. Conclusion: Judo athletes appear to have lower selenium status than aerobic athletes, suggesting different body selenium status according to sport type. To maintain body selenium and zinc levels, the dietary intake of saturated fatty acids should be decreased.
