Adaptive $CO_2$Fixation Nitrate assimilation of Portulace oleracea in Zoysia japonica Community (잔디군락에 출현하는 쇠비름의 $CO_2$고정과 질소동화)

  • $CO_2$와 질소동화작용에 있어서의 잎과 줄기간의 세포간 관계가 잔디군락의 자연 환경 상태하에서 자라는 Portulaca oleracea 에서 조사되었다. 기공이 줄기에서는 관찰되지 않는 반면 잎에서는 $1cm^2$당 3,275개가 나타났다. 어린 쇠비름 잎의 기공의 조헝운 범에누누 높고 낮에는 낮다. 최고 활성을 갖는 마디의 순서는 위에서 아래로 떨어짐에 따라 변화한다.P.oleracea 의 줄기조직은 CMA의 최고 활성을 나타내었으나 잎에서는 CMA의 활성이 나타나지 않았다. 빠른 산의 성화는 새벽에 원줄기에서 보여지나 줄기는 서서히 증가하는 것 밖에 관찰되지 않는다. RuPB carboxy-lase의 활성은 거의 관찰 되지 않는다. 특히 잎새에서의 PEP carboxylase의 활성은 이른 아침에 줄기에서는 낮고 잎에서는 매우 높음을 보여준다. 이런 결과는 새벽에 열려진 기공을 통과할$CO_2$가 PEP carboxylase에 의해 잎에서 동화되고 $C_4$생산물이 줄기로 이동함을 나타낸다. Nitrate 축적과 nitrate reductase, nitrite reductase, glutamine symithetase, gluamate symthase, glutamade dehydrogenase의 농도는 잎보다 줄기에서 더 높았고 밤보다 낮에 더 높다. 이것은 뿌리를 통해 흡수된 nitrate의 상당한 양이 줄기에서 동화되고 줄기 조직을 통해 잎으로 이동되어 거기에서 감소된다.

Effects of Dessication, Sucrose and Salt Stress on the Regeneration of Portulaca oleracea Cultured Cells (건조, 염분 및 탕의 처리가 쇠비름(Portulaca oleracea L.) 배양세포의 재분화에 미치는 영향)

  • The optimal level of growth regulator for callus initiation stem explants was BAP 0.1mg/L combined with 2,4-D 1.0 mg/L in Murashige and Skoog (MS) medium supplemented with 30g/L sucrose and 10g/L agar, and that for cell growth was BAP 0.1+2,4-D 0.5 mg/L in MS liquid medium. The regeneration frequency of P. oleracea cells was significantly increased by subjecting the cells to dessication for 1and 2 h up to 83%, respectively, as compared with untreated control showing 61%. Cell viability and survival rate was inhibited by pretreatment of 0.6% NaCl for 2 days, while regeneration ability was not affected by the treatment. Pretreatment of 100g/L sucrose for 2 days markedly stimulated the regeneration of cells up to 81%. These results suggest that in addition to physiological changes, water stress resulted from dessication and high concentration of sucrose and NaCl is closely related to the regeneration of P. oleracea cultured cells.

  • Volatile terpenes responsible for allelopathic activity in four weed species were identified using gas chromatography (GC) and GC-mass spectrometry. Artemisia asiatica Nakai contained 26 volatile terpenes, consisting of 16 monoterpenes and 10 sesquiterpenes, whereas there were only four terpenes in Trifolium repens L. and three terpenes each in Portulaca oleracea L. and Capsella bursa-pastoris (L,) Medik. Suspected allelochemics were ${\alpha}$-phellandrene, 1,8-cineole, limonene, ${\alpha}$-pinene, borneol, selinene, and caryophyllene in A. asiatica, ${\alpha}$-pinene and ${\beta}$-caryophyllene in T. repens, and ${\alpha}$-pinene in C. bursapastoris. No these compounds were found in P. oleracea.

  • Investigation on weed flora in Lycium chinense upland fields was conducted to understand the current status of weed flora and establish the control measures. Investigation was conducted twice, May and June on 2015. From this investigation, 91 species of 30 families including 27 exotics were identified into 36 species of annuals, 28 species of biennials and 27 species of perennials. Dominance was the highest with Portulaca loeracea followed by Digitaria ciliaris, Stellaria media, Oxalis corniculata, Acalypha australis, Chenopodium album, Cyperus iria etc. in order. Chenopodium album was the highest in importance analysis and the followings were in order of Conyza canadensis, Conyza bonariensis, Senecio vulgaris, Amaranthus lividus, Sonchus asper etc.

  • To establish the system of in vitro plant regeneration, the different explants (stem with axillary bud and stem without axillary bud) of Hylotelephium ussuriense were cultured on the Murashige and Skoog's medium containing 6-benzylaminopurine (BA) and indolebutyric acid (IBA). The adventitious shoot induction was more effective in the stem with axillary bud explants than the stem without axillary bud explants, and was the best on MS medium containing 3.0 mg/L BA and 0.01 mg/L IBA. Frequency of plantlet growth was not significantly treated on MS and sucrose. Total chlorophyll contents under ventilation treatment were higher than those in control (non-ventilation). This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Weed Flora of Ulreung Island in Korea (울릉도(鬱陵島)에 발생(發生)하는 잡초종(雜草種))

  • About 140 weed species belonging to 42 families occurring in summer were observed in Ulreung island. The Compositae was the most wildely occurring family covering 27 weed species, followed by 21 species in Graminae, 9 in Polygonaceae, 7 in Leguminosae, and 7 in Labiatae etc. In terms of the lands classified, about 60 species in 26 families were observed in the cultivated and the medical crops grown areas, respectively, and 116 species in 40 families occurred in the non-cultivated land like the vicinity of the cultivated area and 94 species in 34 families in the valley. No. of species and families were much greater in the non-cutivated land than those of the cultivated one. The most dominant weed species in both the cultivated and its vicinity in Ulreung island were Digitaria sanguinalis, followed by Portulaca oleracea, Polygonum hydropiper, Equisetum arvensis, Artemisia princeps, Commetina communis, Setaria viridis in order. Community analysis was done by the method of Toyohara in two cultivated lands such as the general crop land and the medical herb crop grown land, and two non-cultivated areas such as the vicinity of the cultivated land and valley. The cultivated land consisted of the communites of P. hydropiper and Cyperus amuricus, including Bidens tripartita in P. hydropiper community. The medical crop grown land composed of three major communities like P. hydropiper, Amaranthus mangostanus and Sonchus asper in which A. mangostanus and S. media were presented in the ecoton, indicating community being transiting. In the non-cultivated areas like the vicinity of crop land, the communities of Erigeron annuus, P. oleracea, and Oxalis corniculata were dominant, and Dystaenia takeshimana was included in the community of E. annuus and Stellan'a aquatica in P. oleracea community. In the valley, Ranunculus quelpaertensis community was existed in between the communities of Sonchus asper and Plantago asiatica.

  • This study was carried to know factors affecting emergence of major upland weeds in soil in order to get basic information on weed control methods. Firthy eight percent of weed seeds were distributed within soil surface to 10cm in soil and 2% of weed seeds were observed in 40~50cm soil layer in field. As planting depth was deeper, emergence of weeds became poor. However Capsella bursa-pastoris can emerge at soil surface. Amaranthus retroflexus, Amaranthus lividus, Porturaca oleracea, Chenopodium album, Solanum nigrum upto 3cm, Eleusine indica, Echinochloa crus-galli, Setaria viridis, Digitaria sanguinalis upto 7cm. Emergence of weeds was very poor in very acid soil but good in soil pH 5.5~6.0. However emergence of weed was not affected by pH 3.5~4.0 or above. Emergence of E. indica, C. bursa-pastoris, A. retroflexus, A. lividus, C. album, E. crus-galli, Solanum nigrum, and S. viridis was good in loam soil and P. oleranea, D. sanguinalis in sandy loam soil. Emergence of weed seeds was not affected by fertilization.

Effect of Split Nitrogen Application Times on Turf Vegetation of Creeping Bentgrass (질소 분시횟수가 Creeping Bentgrass 잔디초지의 식생에 미치는 영향)

  • This study was conducted from March 21 to July 8 in 2004 at JeJu Island to investigate the influences of split nitrogen application times on creeping bentgrass vegetation. Nitrogen rate was 20 kg/10a and it was applied from once to 5 times. The result obtained were summarized as follows; plant height was getting longer as nitrogen was split applied from once to 5 times. So it was longest at 5 times split nitrogen application, but it was no significance from 4 to 5 times. Root length, Minolta SPAD-502 chlorophyll meter reading value, leave and root weight n[e directly proportional plant height response. Degree of land cover Iud density of creeping bentgrass increased as nitrogen was split applied from once to 5 times. But degree of land cover md density of reed decreased. The number of reed species on decreased as nitrogen was split applied from once to 5 times. Then ranking of the dominant weeds were Portulaca oleracea Polygonum hydropiper and Stellaria media (at once split time nitrogen application), Portulaca oleracea, Polygonum hydropiper and Digitaria adscendens (at 2 split times nitrogen application), Digitaria adscendens and Portulaca oleracea Polygonum hydropiper, (at 3 split times nitrogen application), Portulaca oleracea, Poa annua and Polygonum hydropiper (at 4 md 5 split times nitrogen application). These results indicate that the optimum frequency of split N applications is four times for growth of creeping bentgrass in volcanic ash soils of Jeju island.

Anthracnose of Amaranthus mangostanus Caused by Glomerella cingulata in Korea (Glomerella cingulata에 의한 비름 탄저병)

  • Symptoms similar to anthracnose were observed on Amaranthus mangostanus in Sancheon-gun, Gyeongnam province, where the plants were autogenously formed community. The symptoms were appeared in stem and spread, eventually whole plants died. Mycelial colony of the isolate was whitish gray to dark gray on potato dextrose agar. Conidia were single celled, colorless, cylindrical and measured as $10.5{\sim}21.7{\times}3.8{\sim}6.0{\mu}m$. Appressoria were dark brown, ovate to obovate and sized as $5.6{\sim}13.7{\times}4.6{\sim}11.4{\mu}m$. Perithecia were brown to black in color and shaped as globose to obpyriform and sized as $79.7{\sim}286.7{\mu}m$. Asci had eight ascospores and sized as $47.7{\sim}89.7{\times}8.1{\sim}13.3{\mu}m$. Ascospores were slightly curved at the center cylindrical, fusiform and measured $9.3{\sim}20.3{\times}4.6{\sim}6.3{\mu}m$. Optimum temperature for growth was $30^{\circ}C$. On the basis of morphological characteristics and pathogenicity test to host plants, the fungus was identified as Glomerella cingulata. This is the first report on the Anthracnose of Amaranthus mangostanus caused by Glomerella cingulata in Korea.
