2000년도 노벨 의학상은 스웨덴의 아비스 칼슨 박사 등 3명 이 수상했다. 그들은 신경전달 물질(neurotransmitter) 중 도파민 (dopamine)과 시냅스(synapse)에 관한 연구로 항 우울제 치료제인 프로작 (prozac)을 개발하여 신경, 정신질환 치료제 개발에 기여한 공로였다. 도파민과 GABA는 신경전달 물질 중의 하나로 도파민은 운동, 정서, 행동, 희노애락 등을 조절하는 것으로 이상 분비될 때 파킨스씨병, 정신분열증, 우울증 등을 유발시킨다. GABA는 억제성 신경전달물질로 이상 분비시 간질 등을 유발시킨다. 도파민과 GABA의 분비는 시냅스 후(postsynapse) 수용체에서 그 기능이 조절된다. 그러나 마약성인 코카인, 헤로인, 몰핀, 암페타민 등 중 독성약물뿐만 아니라 일상 생활에서 흔히 접할 수 있는 흡연, 술 등에 의해서도 그 분비 이상을 초래한다. 따라서 본 논단에서는 최근 뇌신경생물 실험실에서 진행되고 있는 신경전달 물질 중에 도파민 및 GABA 분비에 대 한 연구결과를 바탕으로 소개 하고자 한다.
The enhancing effect of excreted cell factors on the production of somatic embryos from suspension cultures of Daucus carota was studied as a function of factors including molecular size, harvesting time, injection period, and concentration of the extracellular compounds. The production of late-stage embryos was increased up to 1, 500 embryos/ml compared with control cultures when high molecular size and extracellular factors, extracted from newly established embryo culture at early stationary phase, were added at the starting time. The stimulating effect on the production of somatic embryos can be attributed to the presence of high molecular size(> 10 kDa) compounds.

pheromone은 생물의 체내에서 합성되고 체외로 분비되어 동종개체에 작용함으로써 특정의 생리적 변화를 유발하는 물질이다. 생물 중에서는 성장의 어느 단계에서 pheromone과 같은 물질이 관여하고 있다는 것이 알려져 있다. 또한, 단세포생물에서 세포 외로 분비하여 2개의 세포를 서로 접합시키는 물질이 있는데 이러한 물질을 성pheromone이라 부르고 있다. 특히, 효모 버섯, 조류, 섬모충류에서 연구되고 있다. 효모는 성접합형을 달리하는 두 균주가 상호pheromone을 분비, 수용하여 성분화를 행하여가고 있다. 그리고 세균에 있어서도 pheromone 양상의 물질이 세포 외로 분비되고 이 물질에 의하여 세균의 생리적 조건에 영향을 주는 예가 있다. 지금까지 알려져 있는 미생물의 성 pheromone에 대하여 크게 나누어 보면 지용성 성 pheromone과 수용성 성 pheromone으로 대별할 수 있다. 본 총설에서는 세균과 자낭균효모, 담자균효모유래의 성 pheromone 중 peptide(수용성) 성 pheromone에 대해서만 집약하였다.
Vitex negundo is an aromatic plant which releases a unique scent due to the presence of essential oil stored presumably within glandular trichomes. The focus of this research was to study developmental patterns of glandular trichomes in Vitex negundo leaves using electron microscopy. There are two types of glandular trichomes which develop on the leaf epidermis of Vitex negundo, peltate glandular type (PT) and capitate glandular type (CT). Structural features differ significantly depending on size and density, formation of secretory cavity, plastid, etc during developmental stages. In young leaves, undifferentiated PTs are densely distributed in the upper epidermis, but are not externally exposed in the lower epidermis because they are covered by non-glandular simple trichomes. Upon leaf development, PTs and CTs show clear structural differentiation in the upper and lower epidermis. PTs are composed of up to eight head cells (ca. 35~40 ${\mu}m$) and one stalk cell (ca. 5 ${\mu}m$), while CTs are composed of four head cells (ca. 10~15 ${\mu}m$) and 1~2 stalk cells (ca. 10 ${\mu}m$). Although secretory cavities develop on the secretory head cells, their size, structure, and formation proceed very differently depending on trichome type. In early development of PT, a large cavity with numerous secretory vesicles form rapidly from the head cells. In CT, however, only a small secretory cavity is formed, slowly relative to PT, without secretory vesicles. The PTs are considered to play an important role in releasing the aromatic components of Vitex negundo.
The ultrastructure of glandular trichomes in Pelargonium peltatum has been studied with a light microscope, transmission, and scanning electron microscope. Two types of the glands, long-stalked and short-stalked capitate glands, are distinguished with their shape and size of the total glands. Both glands are extreamly abundant in the leaf veins and petioles. These glandular trichomes are consisted of one secretory cell, three stalk cells, and one basal cell. The secretory cells contain a large amount of smooth endoplasmic reticulum. They have also much plastids, vacuoles, Golgi apparati, and mitochondria. High electron-dense deposits are frequently present in vacuoles of secretory cells. It seems to be phenolic compounds which is thought as the major secretory precursors.
동양달팽이(Nesiohelix samarangae)의 자성생식기 중 정자낭(spermatheca)의 미세구조를 투과전자현미경을 이용하여 관찰하였다. 성체의 정자낭의 내강에서는 정자와 이와 관련된 물질들 또는 상피로부터의 분비물질들이 관찰되었다. 정자낭의 내강을 둘러싸고 있는 벽의 상피는 단층원주상피로서 내강과 연한 세포의 원형질막은 미세융모를 가지고 있었으며 섬모는 가지고 있지 않았다. 이 미세융모들의 기부에서는 고배율 하에서 pinocytotic vesicle들이 관찰되었고 때로 세포의 분비물을 세포 밖으로 방출하는 모습도 관찰되었다. 세포들의 상부세포질에는 많은 수의 mitochondria가 있었고, 때로 분비과립과 lysosomd들이 존재하기도 하였다. 이들보다 아래에는 잘 발달한 골지체와 과립소포체(RER)들이 있었다. 이 부위에서 분비과립들이 많이 형성되어 있는 모습이 세포에 따라 관찰되었고, 그 아래에 즉, 각 세포의 하반부 세포질에 상하로 긴 핵이 관찰되었다. 세포의 기부 원형질막은 상부 세포질 내로 깊게 그리고 수없이 많이 주름을 이루고 있어 인접한 세포와의 경계를 구분할 수 없을 정도였다. 기저막의 아래에는 근육다발과 색소세포들이 결합조직 내에서 관찰되었고, 정자낭의 외막은 큰 액포로 인하여 세포질들이 원형질막 주변으로 밀려나 있는 특이한 세포들로 이루어져 있었다. 정자낭의 기능은 정자를 일시 저장하며 정자낭 상피에서 분비되는 소화액들로 정자를 일차적으로 세포외 소화하고 소화되지 않은 물질들을 흡수하여 이차적으로 세포내소화하는 것으로 생각된다.
Neurotransmitter release is regulated by various proteins of the active zone in the presynaptic nerve terminals. Dopamine (DA) is an essential neurotransmitter associated with the pathophysiology of diverse behavioral and mental illness such as schizophrenia and drug addiction. We measured synaptosomal DA release of knockout (KO) mice which lacked major genes related to neurotransmitter release. Synaptosomal DA uptake and release were performed and measured using [$^3H$]-DA and superfusion experiments. 3 of the 17 KO mice exhibited altered DA release compared to their littermate controls. In $Rim1{\alpha}$ KO, [$^3H$]-DA release evoked by membrane depolarization significantly decreased. Both basal (physiological buffer-evoked) and membrane depolarization-evoked DA release significantly decreased in dopaminergic conditional KO of $Rim1{\alpha}{\beta}$. Dopaminergic conditional KO of neurexin3 demonstrated a significant increase of membrane depolarization-evoked DA release. These data explain the similarities and distinctions between DA and other classical neurotransmitters such as glutamate and GABA ($\gamma$-aminobutyric acid) release. In conclusion, $Rim1{\alpha}$ and neurexin3 may be important regulators of presynaptic DA release and related to disorders of the nervous system.
The present study examined the epidermal changes of adhesive disks which occur during attachment in Parthenocissus tricuspidata using scanning and transmission electron microscopy. Several adhesive disks, each covered with a bract, develop from the shoot apical meristem during early development. In the initial stage, the adhesive disks are club-shaped and their upper and lower epidermis are indistinguishable. However, in the actively growing stage, they become spherical and both epidermis are clearly differentiated into the adventitious roots. Prior to wall attachment, the adhesive disks exhibit adaxial convex and abaxial concave shapes, and electron-dense substances are abundant in the vacuoles of epidermal cells. The peripheral area of the adhesive disk is adhered first to the wall surface, while the central area is drawn inward in a vacuum-like state during attachment. As the attachment progresses and the electron-dense substances continue to discharge, the upper and lower epidermis rapidly undergo deterioration and the disks shrink considerably. At this stage, structural changes of the lower epidermis occur much faster than in the upper one. The discharged substance is accumulated on the wall surface, and this aids the attachment of adhesive disks on the wall for long periods. In this manner, the shape and structure of the adhesive disk epidermis change drastically from initial growth to the mature stage. Further, the role of electron-dense substance and shrinkage of the disk during attachment has been discussed in Parthenocissus tricuspidata.
Comparison of lead removal characteristics between two strains, Aureobasidium pullulans and Saccharomyces cerevisiae, and effects of extracellular polymeric substances(EPS) excreted by microorganisms on the removal of lead were investigated. The capacity of lead biosorption to A. pullulans which had EPS was increased as the storage time of the cells increased, due to the increased amounts of excreted EPS. When the EPS were removed from A. pullulans cells, the amounts of adsorbed lead were very small(10% of the cell with EPS). In the case of s. cerevisiae which had no EPS, the lead removal capacity was nearly constant with storage time except early stage, but the spending time to reach an equilibrium state decreased with increasing storage time because of lowering the function of cell membrane. Therefore, it seems that the phenomena of lead biosorption were remarkably affected by the presence of extracellular polymeric substances.
