Study on the Natural Spat Collection of the Pen Shell, Atrina pectinata (키조개, Atrina pectinata의 자연채묘연구)

  • In this experiment of natural Atrina pectinata spat collection, the number of the pen shell spats settled on a collector were 4.2, 6.8, and 2.7 in the first, second, and third year, respectively. The highest number of pen shell spat settled on the collector was observed in August, and the shell length of juvenile pen shells grew up to 42.5 mm six months after setting. The relation between shell length (SL) and days (D) after setting could be expressed as; SL = 0.3854D + 0.8423 ($r^2$ = 0.9764). The spats developed the best in depth of 10-20 m. Spat collectors in mesh size of $1{\times}2mm$ mm were better for spats to survive than the ones in $1{\times}1mm$. The most efficient material as substrate for the settlement of pen shell spat was a polyethylene monofilament in the spat collectors.

  • The extracellular matrix (ECM) is a key factor affecting cell growth and adhesion to the culture surface, and it is also important for maintaining the innate characteristics of cells. Here, we describe the effects of the ECM on cardiomyocyte (HL-1 cell line) growth, viability, phenotype, and contractile ability. Five different ECM materials were investigated to analyze their effects on the cell growth. The physical morphology of the ECM-coated surfaces was scanned with an atomic force microscope (AFM), and the attachment, growth, proliferation, viability, and phenotype of the cells were analyzed using fluorescence immunostaining and an inverted phase contrast microscope.

Dietary Value of Benthic Diatoms for Newly Settled Abalone, Haliotis discus hannai Ino (참전복 치패에 대한 부착성 규조류의 먹이효율)

  • Growth trends of 10 selected species of benthic diatoms, considered essential dietary requirement of the newly settled abalone were monitored. Navicula sp. (B-38), N. incerta and Caloneis schroderi, grew faster than the other tested diatoms. 16 and 32 % abalones fed on Raphoneis sp. and Phaeodactylum settled, respectively; less abalones souled, when fed on Navicula sp., Hantzxchia marina or Nitzschia sp. In the first experiment, survival of the settled abalone was the highest (63 %) and lowest (31 %) for those fed on Rhaphoneis sp. and Navicular sp. respectively. However, in the second and third series of esperiments, abalones fed on Rhaphoneis sp. and Navicula sp. showed the highest (67, 49 %) and lowest (35, 18 %) survival. C. schroderi proved to be the best diet, as the shell length of those fed on the diatoms was 83 ${\mu}$m, as against about 36 ${\mu}$m of those abalones, receiving H. marina or Nitzschia sp., diatoms of the lowest dietary value.

Intermediate Culture of the Spat of Arkshell, Scapharca broughtonii in Summer (피조개, Scapharca broughtonii 부착치패의 하계 중간양성)

  • Arkshell, Scapharca broughtonii spats were placed in natural environmental condition for 30 days from July 28 to August 27, 2004, in order to estabilish intermediate culture technique. Growth and survival of the spats in different intermediate culture areas with various culture methods were measured. Water temperature of studied area ranged from 24.1 to 28.5$^{\circ}C$, salinity was 15.4 to 33.3 psu, dissolved oxygen was 3.92 to 12.6 mg/l. Scapharca broughtonii spats cultured in Yeosu developed the best, 10.15 ${\pm}$ 1.12 mm in average shell length, and the highest survival was recorded as 77% in Namhae. Shell lengths of the Scapharca broughtonii spats cultured in the water depths of 2, 5, and 10 m were 7.14 ${\pm}$ 1.14 mm, 6.98 ${\pm}$ 1.74 mm and 6.27 ${\pm}$ 1.33 mm, and the survivals showed 75.5%, 77.0% and 76.5%, respectively. When 1 mm, 2 mm, and 3 mm-sized spats were cultured for 30 days in water depth of 5 m, the shell length increased to 6.73 ${\pm}$ 1.46 mm, 6.98 ${\pm}$ 1.74 mm and 7.04 ${\pm}$ 1.19 mm, and survivals were 67.0%, 77.0% and 58.5%, respectively. The shell lengths of spat cultured for 30 days in mesh sizes of 1 ${\times}$ 1 mm, 2 ${\times}$ 2 mm and 3 ${\times}$ 3 mm, 5 m below the surface were 8.14 ${\pm}$1.23 mm, 8.26 ${\pm}$ 1.19 mm and 8.78 ${\pm}$ 1.16 mm, and survivals were 41.5%, 43.0% and 44.5%, respectively.

  • 부착 생물에 관한 연구는 선박, 해수 이용시설, 수산 양식업 둥의 방제대책과 관련하여 부착 생물의 종류, 부착 량, 성장도 및 계절적 변화 둥의 연구가 중요한 문제하고 생각한다. 특히 양식시설이 밀집한 진해만 해역의 굴 양식 시설에 부착하는 대부분의 생물들이 Ascidicea, Crustacea, Cirripedia, Bivalvia, Bryozoa, Polychaeta, marine algae 등이며, 이들 가운데 피해가 심한 다모환충류에 관하여 분류학적인 연구를 시도하였던 바 수하연의 양식 굴 사이에서 많은 양의 갯지렁이류를 얻었기에 보고 한다. 수하연에 서식하는 갯지렁이류는 제2부착성(Kazihara, 1964)의 갯지렁이류로서 왕관 모양의 큰 아가미 혹은 몸앞 부분에 실모양의 감각 촉수를 가지고 굴에 피해를 입히는 짧은수술갯지렁이 Loimia medusa, 미끈꽃갯지렁이 Myxicola infundibulum와 제1부착성으로서 굴 패각, 양식 시설 등에 석회질의 집을 형성시켜 피해를 입히는 우산관덮개꽃갯지렁이 Hydroides ezoensis, 둥근구멍환덮개꽃갯지렁이 Dexiospira alveolatus 등의 7과 12속 13종을 보고한다. 이 가운데 새날개갯지렁이 Chaetopterus variopedatus와 둥근구멍관덮개꽃갯지렁이 Dexiospira alveolatus는 한국 미기록종이다.

참전복, Haliotis discus hannai 부착치패의 먹이 선택성과 성장

  • 우리나라에서 건강식품으로 널리 알려져 있는 참전복은 불로장수의 식품으로 널리 애용되고 있을 뿐만 아니라 발암억제 물질인 paolin을 함유하고 있어, 세계적으로 상품가치가 인정되고 있다. 이러한 중요성에도 불구하고 참전복 부착치패의 성장 및 생존율 제고는 아직까지도 해결되어야 할 문제로 남아 있는 실정이다. 참전복 부착치패의 먹이선택성 및 유인성에 관한 기존의 연구를 보면 Viana et al. (1994)은 평균 각장 4.2 cm인 치패를 이용하여 배합사료의 9가지 원료에 대한 유연성을, Harada and Kawasaki (1982)는 각장 2.4 cm 이상인 치패를 대상으로 해조류 3종(미역, 톳, 파래)의 유인효과에 관하여 보고하였다. 그러나 이와 같은 보고들은 전복 종묘생산 현장에서 보다 빠른 시기에 박리하여 해조로 먹이 전환을 하기 위한 노력과는 어느 정도의 거리가 있다고 판단된다. 따라서 본 연구에 있어서는 평균 각장 5.0$\pm$1.0 mm인 치패를 박리하여 Navicula spp., 해조 2종(다시마, 파래) 및 자체 제조한 사료를 공급하여 먹이별 유인효과를 관찰하였다. 조직공학용 전기방사 나노섬유 지지체 제조방법

  • 이 글에서는 나노스케일의 직경을 갖는 섬유를 빠른 생산속도로 제작할 수 있는 전기방사공정(electrospinning process)에 대한 개요와 조직공학용 지지체(tissue engineering scaffold)로의 응용을 위한 제조방법에 대해 소개하고자 한다. 세포의 증식, 분화 등의 생물학적 활동에 기반한 조직공학 및 조직재생 분야에서는 일시적 또는 영구적으로 세포가 부착하여 생장할 수 있는 지지체(scaffold)의 활용이 필수적이다. 세포가 이상적으로 성장할 수 있는 지지체를 제작하기 위해서는 세포의 부착 특성, 화학적/물리적/구조적 성장 환경 등이 고려되어야 한다. 따라서 이상적인 세포 성장 환경을 구현하기 위해 실제 세포 주변의 미세환경(microenvironmenr)조건을 모사하는 연구가 많이 이루어지고 있다. 세포외기질(extracellular matrix)이라고 하는 나노크기의 직경을 갖는 섬유기반의 세포 주변 환경을 모사하는 방법의 하나로 전기방사 공정이 '90년대에 들어 활용되기 시작하였다. 현재까지도 전기방사를 이용하여 제작되는 나노섬유는 공정조건 및 재료를 다양하게 응용하여 조직의 물리 화학적 특성을 잘 반영할 수 있는 장점이 있어 조직공학용 지지체로서 광범위하게 활용되고 있다.

Development of Solid-Gelatin Microcarrier for Large Scale Production of Anchorage-Dependent Animal Gell Lines (부착성 동물세포의 대량배양을 위한 젤라틴 미립담체의 개발)

  • Solid gelatin microcarrier with the size distribution between $100{\mu}{\textrm}{m}$ and $400{\mu}{\textrm}{m}$ was prepared for the attachment and growth experiment for anchorage-dependent animal cell lines, i.e., L 929 and BHK 21. The growth and the maximum cell densities on this gelatin based and polyacrylamide (PAA) microcarriers were compared with those on the commercial dextran based Cytodex 3 microcarrier. Both cell lines showed good comparable attachment and growth patterns on the above three microcarriers. The mouse fibroblast, L 929 showed about the same maximum cell density on PAA, gelatin and Cytodex 3 MC'S BUT BHK 21, the baby hamster kidney cell line, showed the best result on Cytodex 3, which was about $4\times10^6$ cells/ml with the microcarrier concentration of 10 g/1.

Effects of White Sesame Seed Extract and β-Sitosterol on Growth, Migration, and Adhesion of H1299 Human Lung Cancer Cells (흰깨 추출물과 β-Sitosterol이 H1299 폐암세포의 성장, 이동, 부착에 미치는 효과)

  • The current study aimed to investigate effects of ethanol extract of white sesame seed (WSE) as well as a major constituent of white sesame seed, ${\beta}-sitosterol$, on the growth, migration, and adhesion of H1299 human lung cancer cells. Treatment with WSE at concentrations of 150, 300, and $600{\mu}g/mL$ dose-dependently inhibited cell growth (to 51.5~82.6% of control). Treatment with ${\beta}-sitosterol$ at concentrations of 3.125, 6.25, 12.5, and $25{\mu}M$ inhibited cell growth to a greater extent (to 27.5~49.0% of control) than that with WSE (P<0.05). Treatment with WSE (at concentration of $600{\mu}g/mL$) or ${\beta}-sitosterol$ (at concentration of $25{\mu}M$) resulted in increased sub-G1 cell population, indicating their apoptosis-inducing activities. ${\beta}-sitosterol$ was effective in inhibiting both cell migration (to 80.8~86.2% of control at a concentration range of $3.125{\sim}25{\mu}M$) and adhesion (to 21.5~37.4% of control at a concentration range of $6.25{\sim}25{\mu}M$), whereas WSE at a concentration range of $150{\sim}600{\mu}g/mL$ was ineffective. These results indicate that ${\beta}-sitosterol$ is more active than WSE in inhibiting growth, migration, and adhesion of H1299 human lung cancer cells. Further studies are needed to determine if similar effects are reproduced in vivo.
