New Records of Six Species of the Tetraodontidae (Pisces: Tetraodontiformes) from Korea (한국산 참복과 (복어목) 어류 6 미기록종)

  • Six species of the family Tetraodontidae collected from coastal waters of Korea were described with photographs, all of which are unreported previously from Korea. They are Lagocephalus inermis (Temminck et Schlegel), Takifugu alboplumbeus (Richardson), T. pseudommus (Chu), T. reticularis (Tien, Cheng et Wang), Arothron hispidus (Linnaeus) and A. nigropunctatus (Bloch et Schneider).

Isolation and Identification of Tetrodotoxin-producing Marine Microorganism from Pufferfish (복어로부터 복어독(Tetrodotoxin) 생성능이 있는 해양 미생물의 분리 및 동정)

  • A novel marine microorganism, Vibrio sp. YE-101, was isolated from pufferfish and investigated for its ability to synthesize tetrodotoxin (TTX). Various strains isolated from the intestine of pufferfish were grown on TCBS agar plate, and then cultured on Ocean Research Institute (ORI) medium supplemented with 3% NaCl at 23$^{\circ}C$ for 3days. The cells were harvested, disrupted, fractionated by Bio-Gel P-2 column chromatography and then TTX-producing strain, Vibrio sp. YE-101, was identified using mouse bioassay. The isolated TTX from Vibrio sp. YE-101 was also analyzed and identified by HPLC and gas chromatography-mass spectrometer (GC-MS). The mass fragmentation of trimethylsilyl derivatives of C9-base of TTX from Vibrio sp. YE-101 was interpreted and the pattern of fragmentation was same with that of authentic standard. The purfied TTX was also positive to the mouse bioassay, which clearly represents that Vibrio sp. YE-101 can synthesize TTX.

Tetrodotoxin in a Pufferfish, Fugu xanthopterus (Korean Name, Ggachibog) (까치복(Fugu xanthopterus)의 독성)

  • A total of 24 specimens of the pufferfish, Fugu xanthopterus, purchased at a fixhmarket in Pusan, korea were examined for toxicity using the assay method of tetrodotoxin (TTX). Also, the toxins isolated from the puffer liver were partially purified and analyzed for their chemical composition by instrumental behaviors. On the whole, when the level of toxicity in each organ was analyzed compared to that of liver, they were 100 % of the lover, 92 % for the intestine, 75% for the skin, 17% for the muscle, 785 for the testis, 87% for the ovary, and 71% for bile. The highest and average scores of toxicity for the liver were 917 and $231{\pm}51MU/g$ liver, respectively. The toxins of the puffer gave four peaks in HPLC whose retention times (10, 20, 22 and 25 min) were close to those of TDA, TTX, 4-epi-TTX, and and -TTX, respectively.

  • The purpose of this study is to make a correlation analysis between mechanical characteristics and sensory characteristics of puffer fish fillet under storage conditions and to find out those quality characteristics. With the storage time, VBN increased while color, smell and texture values decreased. As the appearance, the fish showed excellent quality stored at $4^{\circ}C$ for 8-12 hours, at $12^{\circ}C$ for 4-8 hours and at $20^{\circ}C$ for 4 hours. When the L value increased, color preference in sensory results decreased in correlation physicochemical properties, mechanical properties and sensory properties of puffer fish fillet. However, mechanical properties and sensory evaluation showed a different aspect. When the higher temperature, redness(a-value) decreased. Regardless of storage temperature, yellowness (b-value) increased. In the results of the hardness, the value was higher, the sensory showed the better texture.

  • The pufferfish Takifugu vermicularis radiatus (Gukmeri-bog) was examined for toxicity. Thirty-one specimens, which had obtained from creeks of Inchon and Ansan in Spring and Autumn of 1992 and 1993, Korea, were assayed for anatomical distribution of toxicity by mouse bioassay method. Ovary, testis, liver and muscle showed very strong toxicity, and intestine and skin did moderate toxicity. Their average toxicities were moderately toxic in all. Moreover, the toxicities of liver and intestine were stronger in Spring than Autumn. Judging from the results, T. vermicularis radiatus is considered to be very harmful fish. Consequently, it is necessary to take a proper preventive step against food poisoning by pufferlish ingestion.

  • The present study was conducted for the data of toxicity and taste components of the pufferfish, Sphoeroides annulayus (bull's eye fuller), transported from Mexico. All other parts including muscle and skin were nontoxic ranging below $10\;{\mu/g$ except gonad, The amounts of IMP and ADP were $5.6\;{\mu}mol/g\;and\;2.7\;{\mu}mol/g$, and the ratio to the total ATP and its related compounds was $41.1{\%}$. The great portion of free amino acids in the muscle of the puffer was occupied by L-glycine, L-alanine, L-anserine, L-threonine and L-valine. Their amounts were $233.5 mg/100 g, 169.0 mg/100 g, 149.1 mg/100 g, 135.7 mg/100 g and 132.3 mg/100 g$. Their concentration ratio to total free amino acids were $14.28{\%},\;10.33{\%},\;9.12{\%},\;8.30{\%}\;and\;8.09{\%}$, respectively. The content was $50.12{\%}$ of the total free amino acids. In addition, the amounts of taurine and L-histidine were 119.3mg/100 g and 14,7 mg/100 g.

The Study on the Analysis Method of Tetrodotoxin in Puffer Fish (복어 중 테트로도톡신 분석법에 관한 연구)

  • The current standard for testing tetrodotoxin (TTX) in foodstuffs is the mouse bioassay (MBA) in Korea as in many other countries. However, this test suffers from potential ethical concerns over the use of live animals. In addition, the mouse bioassay does not test for a specific toxin thus a sample resulting in mouse incapacitation would need further confirmatory testing to determine the exact source toxin (e.g., TTX, STX, brevotoxin, etc.). Furthermore, though the time of death is proportional to toxicity in this assay, the dynamic range for this proportional relationship is small thus many samples must be diluted and new mice be injected to yield a result that falls within the quantitative dynamic range. Therefore, in recent years, there have been many efforts in this field to develop alternative assays. High performance liquid chromatography (HPLC) coupled with mass spectrometry (MS) has been emerged as one of the most promising options. A LC-MS-MS method involves solid-phase extraction (SPE) and followed by analysis using an electrospray in the positive ionization mode and multiple reactions monitoring (MRM). To adopt LC-MS-MS method as alternative standard for testing TTX, we performed a validation study for the quantification of TTX in puffer fish. This LC-MS-MS method showed good sensitivity as limits of detection (LOD) of $0.03{\sim}0.08{\mu}g/g$ and limits of quantification (LOQ) of $0.10{\sim}0.25{\mu}g/g$. The linearity ($r^2$) of tetrodotoxin were 0.9986~0.9997, the recovery were 80.9~103.0% and the relative standard deviations (RSD) were 4.3~13.0%. The correlation coefficient between the mouse bioassay and LC/MS/MS method was higher than 0.95.

  • 제주도 해산어류의 양식은 대부분 육상 양식수조를 이용하여 사육하고 있다. 좋은 입지적 조건과 연중수온 변동폭이 적고 오염원으로 부터 차단된 지하해수 등을 이용하여 안정적인 생산을 하고 있으나, 최근에 들어 단일 어종에만 치우친 넙치의 생산에 대한 우려로 일부 업체에서는 경제성이 높은 능성어류나 복어류에 대한 연구로 품종 다양화를 모색하고 있다. 국내의 자주복 양식은 일부 양식장에서 일본으로부터 수정란을 도입하여 종묘생산과 양성을 실시하고 있다. (중략)
