Effect of Oriental Anti-Stress Agent(Bohyulanshintang) on the Salivary Gland of Rats under Restraint Stress (보혈안신탕(補血安神湯)이 스트레스에 의한 백서 타액선 조직 변화에 미치는 영향에 관한 연구)

  • 일상 생활에서 우리는 스트레스에 항상 노출되어 있으며, 스트레스는 생체의 신경계, 내분비계 및 면역계의 변화를 수반한 항상성의 파괴로 수많은 정신적, 육체적 질병을 야기시킨다. 특히 구강안면영역에서도 다양한 구강점막질환과 구강건조증 등을 발생시킨다. 스트레스를 제거하는 방법으로는 약물요법 및 상담, 명상요법, 종교요법 등 다양한 방법이 제시되고 있는데, 다소의 부작용이 나타나거나 꾸준히 시행하기가 쉽지 않으며 스트레스의 원인을 근본적으로 제거하기가 현실적으로 용이하지 않은 경우가 많아 스트레스에 대한 해결책에 대하여 많은 관심이 집중되고 있다. 이에 본인은 스트레스가 가해졌을 때 백서 악하선에서 관철되며 apoptosis에 대하여 세포보호작용을 하는 clusterin(SGP-2)을 이용하여 구속스트레스를 가하기에 앞서 오랫동안 경험적으로 사용되어 왔고 부작용이 적은 전통약물인 보혈안신탕을 투여하고 스트레스에 의한 타액선의 조직변화를 관찰하여 그 효과를 확인해 보고자하였다. Sprague-Dawley계 응성 백서(200-230g/bw) 33마리를 정상 대조군(3마리), 구속스트레스군(15마리) 및 보혈안신탕 투여 후 구속스트레스군(15마리)으로 나누고 이틀을 각각 구속장치에 구속한 후 0, 1, 3, 5, 7일에 회생시켜 악하선을 적출하였으며, 면역조직화학법 및 Northern Blot을 이용하여 clusterin의 변화를 관찰하였다. 그 결과는 다음과 같다. 1. 구속스트레스군의 악하선 조직에서 clusterin 단백질과 mRNA는 실험 즉일군에서만 미약하게 관찰되었으며 실험 3일과 5일 후에 핵붕괴 및 핵농축 등의 핵변화를 동반한 apoptosis가 관찰 되었다. 2. 보혈안신탕 투여 후 구속스트레스군의 악하선 조직에서 실험 5일군까지 clusterin 이 증가한 후 실험 7일군에서는 감소하였다. 3. 보혈안신탕 투여 후 구속스트레스군의 악하선 조직에서는 apoptosis가 관찰되지 않았다. 4. 보혈안신탕 투여 후 구속스트레스군의 악하선 조직에서 clusterin mRNA가 실험 전군에 걸쳐 미약하게 관찰되었다. 이상의 결과로 타액선 조직은 스트레스 단백질인 clusterin을 생산하여 세포를 보호함으로써 스트레스 상황에 적응하지만, 생리적 적용한계를 넘는 스트레스에 노출될 때는 apoptosis됨이 확인되었다. 그리고 보혈안신탕은 스트레스 상황에서 세포의 생리적 적응력을 높여 세포의 apoptosis를 억제하는 효과를 나타냄이 확인되었다. 따라서 본 연구결과는 구강건조증등의 스트레스성 타액선 질환의 병리기전을 규명하는데 도움이되리라 생각되며, 향후 항스트레스 효과를 가진 보혈안신탕등의 한약재를 임상에 적용함으로써 스트레스로 인한 신체의 병리적 변화를 다소나마 차단할 수 있을 것으로 사료된다.

Effects of N-Methylacetamide on the Viability, Fertility and Hatchability of Cryopreserved Ogye (Korean Native Black Fowl) Semen (N-Methylacetamide 동결보호제가 오계 동결정액의 생존성, 수정 및 부화율에 미치는 영향)

  • The use of methylacetamide (MA) as a cryoprotective agent for freezing Korean Native Black rooster Ogye semen was examined with artificial insemination. The diluted Ogye semen with HS-1 was subjected for 2 step dilution method of cryopreservation in which the final concentration of MA was adjusted to 7.5%. The sperm viability after thawing was reduced from $95.17{\pm}0.93%$ to $55.93{\pm}1.38%$ which was confirmed by live-death analysis based on Fluorescence-Activated Cell Sorting (FACS). The rates of fertilized eggs with fresh or frozen-thawed semen were reduced from $94.98{\pm}3.93%$ to $66.36{\pm}8.43%$ at day 7 with significant difference. However, the hatching rates of experiments at day 21 did not shown difference between $92.64{\pm}2.33%$ and $90.45{\pm}8.05%$ (P<0.05). With these results, the utilization of MA for freezing of Ogye spermatozoa could affect on viability of frozen-thawed semen but not on the fertility of lain eggs and hatchability of fertilized eggs and also provide possible tools of freezing for poultry genetic resource conservation.

Interfacial Reaction between seal and metal interconnect and effets of protecting layer in planar type SOFC stack (평판형 SOFC 스택의 밀봉재와 금속 분리판의 계면반응 및 보호층 효과)

  • 평판형 고체산화물 연료전지 스택의 고온 밀봉 구조에 대하여 설명하고 스택 운전 후 사후 분석을 통하여 밀봉재와 금속 분리판의 계면반응에 대하여 고찰하였다. 대표적인 고온 밀봉재인 Barium-Silicate 계 결정화 유리와 Fe-Cr 계 금속 분리판은 스택의 작동온도인 $700{\sim}850^{\circ}C$ 에서 고온 반응을 통하여 계면에 반응생성물을 형성하는 것이 확인되었다. 이러한 계면반응은 장기 운전시 SOFC 스택 성능 저하의 원인이 되고, 열 싸이클(작동온도${\leftrightarrow}$상온)을 가하면 계면반응 생성물이 delamination 되어 밀봉구조가 파괴되어 수명을 단축시키게 된다. 계면반응은 Fe-Cr 계 금속 분리판의 산화물인 Cr 산화물, Fe 산화물이 밀봉유리 소재와 반응을 일으키는 것이 주요 원인으로 판명되었다. SOFC 스택에서 열 싸이클시 계면반응에 의하여 기밀도가 감소하는 현상이 확인되었으며, 밀봉 구조의 어느 부분에서 계면반응이 진행되는지 관찰하였다. 이러한 계면반응을 막기 위해서는 금속 분리판과 밀봉유리 사이에 계면반응을 억제하는 보호층을 형성하는 방법이 효과적이다. 본 연구에서는 보호층으로서 밀봉유리 및 Fe-Cr 계 금속 분리판과의 계면반응성이 낮고 열팽창 계수가 비슷한 Yttria Stabilized Zirconia 층을 APS(Atmospheric Plasma Spray) 공정을 이용하여 형성하였다. 밀봉유리/YSZ 보호층/금속분리판은 gas-tight 한 밀봉 구조를 형성하였으며, YSZ 보호층은 밀봉유리와 Fe-Cr 계 금속 분리판 소재와 계면반응을 효과적으로 억제하는 것이 확인되었다.

A Study on the Funeral director's working Circumstances and Personal Protective Equipment on Funeral home (안치실 및 염습실 종사자의 주요 감염질병 조사 및 근무기간에 따른 개인위생용품 착용실태에 대한 연구)

  • We are investigated about the state of the working circumstances and infection blocking facility within Metropolitan, Chungcheong and Gyeongsang regions in Korea. The result is as follows. The HAV vaccination rate is 35% and the HBV vaccination rate is 50% among the funeral directors. We expected that the infection risk of funeral directors was very high. Significantly, tuberculosis prevalence rate of funeral director was four times as high as the public. The wear rate of gown, mask, gloves showed relatively high than the others. The wear rate of individual protective equipments has decreased with work experience. The funeral directors have shown that there is difficult to get safe protection from infection. The personal hygiene of funeral directors and the infection blocking equipment should be strengthened institutionally. We are thought to recommend institutional support about infection management, individual protective equipment, vaccination of funeral director and guideline to avoid reversion to previous habitual behavior.

Use of Food-Grade Protective Agents to Improve the Viability of Freeze-Dried Lactic Acid Bacteria (유산균 생존율 향상을 위한 식품첨가물 등급의 동결보호제 탐색)

  • Food-grade protective agents, namely, skim milk, yeast extract, soy powder, and trehalose, were studied for their ability to improve the viability of freeze-dried lactic acid bacteria (LAB), including Weissella cibaria SW1-1, Lactobacillus plantarum A-1, Lactobacillus sakei 2-12 24, and Leuconostoc citreum 3526. The best results were obtained with 10% soy powder; approximately 90% cell viability was observed during the freeze-drying process. Increase in the concentration of soy powder did not cause a proportional increase in the survival rate of LAB. Further, no significant difference was observed when two agents were combined in a 1:1 ratio (p<0.05).

Antioxidant and Neuronal cell protective effects of Stachys sieboldii Miq. according to drying methods (건조방법에 따른 초석잠(Stachys sieboldii Miq.)의 항산화 활성 및 신경세포 보호효과)

  • This study investigated the antioxidant and neuronal cell protective effects of activity of water and 70% ethanol extracts from Stachys sieboldii Miq. according to different drying method(hot air drying and freeze-drying). The total flavonoid and total polyphenol content in water extracts was significantly higher after freeze-drying compared to hot air drying(p<0.05). DPPH and ABTS radical scavenging were increased in a dose-dependent manner. The DPPH and ABTS radical scavenging activity of water extract were significantly higher after free-drying compared to hot air drying(p<0.05). In a cell viability using MTT, the water extract according to hot air drying and freeze-drying of Stachys sieboldii Miq. showed protective effect against H2O2-induced nurotoxicity. The results suggest that the water extracts of Stachys sieboldii Miq. after freeze-drying has antioxidant activities and may be useful for neurodegenerative disorders.

  • The purpose of this study was to understand meaning of a meal among Nursing home elderly and staff. The meaning of a mean was explained to participants through observation and in-depth interviews based on Ethnomethodologic method. The meaning of a meal expressed by the Elderly was applied as personal philosophy on diversity and Caregiver also had become a management work on convenient logic. Elderly has been recognized has as hope of health recovery in important elements such as the air life indispensable to life. In contrast, Caregiver was following their will to live formally without any sense of the meaning of life. they are just extending their life. Meaning of Nursing-home elderly was lighthearted. They eat salt with snack to have massive power and escape from the daily life. However, Caregivers have other control measure. From the above result, In the standard operation of the meal, the manual of the laws of the instructions regarding long-term care insurance for the Aged, must reflect the elderly continued proposed the need to have diversity for the education about understanding and acceptance of the elderly.

Expression of HSP70 Immunoreactivity in EPO Treated Rat Kidney (콩팥에서 Erythropoietin 투여로 인한 HSP70의 발현 변화)

  • Heat shock protein (HSP) 70 functions as a molecular chaperon and reduces stress-induced denaturation and aggregation of intracellular proteins. Erythropoietin (EPO) plays an important role during acute renal failure repair process by rapidly correcting anemia and enhancing renal tubular regeneration. The purpose of this study was to examine the effect of EPO treatment on renal HSP70 expression. Male Sprague-Dawley rats were injected rHUEPO. Kidney were preserved by in vivo perfusion with paraformaldehyde-lysine-periodate (PLP) and processed for immunohistochemistry and electron microscopy. In control kidney, HSP70 was expressed in the cortex, outer medulla and inner medulla. Especially, HSP immunoreactiviy was mainly founded in descending thin limb of outer medulla and inner medullary collecting duct. In EPO treated kidney, HSP70 expression markedly increased in the descending thin limb of outer medulla and newly detected in cortical collecting duct. Electron microscopy showed the presence of HSP immunoreactivity on the intracelluar vesicles and Golgi complex of descending thin limb and cortical collecting duct. These findings suggest that EPO treatment leads to new production of HSP70 in renal tubular cells, and induction of HSP70 by rHuEPO is causally related to protective function.

Analysis of Fatty Acid Composition and Effects of Pumpkin Seed Oil on Human Umbilical Vein Endothelial Cells (호박씨유의 지방산 성분 분석 및 Human Umbilical Vein Endothelial Cell에 미치는 영향 연구)

  • Pumpkin seed oil (PSO) was investigated for its parasite elimination activity and efficacy in treating disorders of the prostate gland and urinary bladder. We confirmed the composition of PSO and identified its ability to improve vessels. Gas chromatography coupled with mass spectrometric (GC-MS) system was used for PSO composition analysis. Cytotoxicity and cell proliferation were confirmed by Cell Counting Kit-8 (CCK-8) assay. Nitric oxide(NO) production was measured with Griess reagent, and intercellular adhesion molecule (ICAM)-1 and vascular cell adhesion molecule (VCAM)-1 mRNA expression levels were measured by reverse transcription polymerase chain reaction (RT-PCR). As a result, PSO revealed the presence of several components such as linoleic acid, oleic acid, palmitic acid and stearic acid. Cytotoxic effects of PSO were not observed, and PSO increased nitric oxide production in human umbilical vein endothelial cells (HUVEC). Additionally, TNF-${\alpha}$-induced cell proliferation and ICAM-1 expression in HUVEC were inhibited by PSO treatment, whereas VCAM-1 expression was not significantly reduced. Taken together, these results show that PSO is worthy of study as a candidate food material for improvement of vascular disease.

  • The appreciation of cultural properties related with what the aspects of society was is mandatory to understand our culture correctly. To make over all cultural properties to our descendants without breakage or damage is our natural duty and national obligation. On the contrary, inhabitants in the preserved area should be restricted and controlled by the cultural property law. The conflict between the inhabitants in the preserved area and the government raises economical problems and damages all the time. Especially it is time to discuss to mediate between them and solve the problems. This feasible study is a proposal to settle them up through renovating abolished school in the preserve area to the museum and a case to preserve the cultual properties as well as habitant's assets in that area.
