Effects on Growth and Yield of Korean Malting Barley Cultivars by Soil-borne Bymovirus Infection (맥류의 토양 전염성 Bymovirus의 감염이 국내 육성 맥주보리의 생육 및 수량에 미치는 영향)

  • Viral diseases, especially Barley yellow mosaic virus (BaYMV) and Barley mild mosaic virus (BaMMV) have been most serious in barley fields. In this study, we investigated the effect of different level of resistance to viral diseases on the plant growth and yield in malting barley. In diagnosis of virus infection, BaYMV and BaMMV were detected in 'Doosan 29' (susceptible), however, 'Jinyangbori' (moderate susceptible) and 'Hopumbori' (moderate) was infected by only BaYMV. Plant height was restrained about 8~29% in overwintered plant regeneration stage depending on the resistant of each cultivar. The culm length damaged also to 9~12% by BaYMV infection. The tiller numbers reduced to 10~14% in overwintering season, however, the head numbers in harvest season more decreased to 26~33%. Heading date was delayed to 3~3 days by the infection. In examination of yield components, 1,000 kernel weight and $\ell$ weight reduced according to culrivar's resistant degrees to 4.0~6.4% and 1.0~4.2%, respectively. The yield of abortive grain was doubled in BaYMV infection comparing to non-infested field. Three varieties tested in the non-infected field over two years were not significantly different for yield potential with ranges of 509kg~632kg/10a. However, significant yield reduction was observed in 'Saessalbori' and 'Baegdong' with ranges of 77~177kg/10a as compared to 'Hopumbori' (467 kg/10a) when tested in the virus-infected field. Yield potentials of these cultivars reduced by 26~43%, respectively, in the virus-infected field as compared to those in the non-infected field.

Breeding of the Scab-Resistant Pear Cultivar 'Greensis' (배 검은별무늬병 저항성 품종 '그린시스' 육성)

  • To develop scab-resistant pear (Pyrus spp.) varieties with fruits that are as crisp and juicy as Asian pears, a cross was made between 'Whangkeumbae' and 'Bartlett' varieties (P. pyrifolia ${\times}$ P. communis) at the Pear Research Institute of the National Institute of Horticultural & Herbal Science, Rural Development Administration, in 1994. Among the 285 seedlings, 'Greensis' was first selected in 2006 for its good eating quality and named in 2012 after regional adaptation tests in nine regions and ten experimental plots from 2007 to 2012. The tree showed a vigorous growth habit and semi-spreading characteristics, like 'Whangkeumbae'. The optimum fruit harvest date was also around Sept. 26 and fruit was round in shape and green in skin color at maturity. Average fruit weight was 470g, and the soluble solids content was $12.4^{\circ}Brix$. The flesh was very crisp and juicy, and had good eating quality. Its' leaf size was similar with 'Bartlett' and smaller than 'Whangkeumbae'. The average of full bloom date of 'Greensis' was determined as Apr. 26, which was six days later than 'Whangkeumbae' and similar with 'Bartlett'. S genotypes of 'Greensis' were identified as $S_4S_e$ by S-allele PCR product sequencing analysis. It seems that the $S_4$ allele was inherited from 'Whangkeumbae' and the Se allele from 'Bartlett'. 'Greensis' displayed strong resistance to scab disease caused by Venturia nashicola, similar to European pear cultivars like 'Beurre Hardy' and, 'Conference'. 'Greensis' was also highly resistant to black leaf spot (Alternaria kikuchiana) in the field

Identification of Quantitative Trait Loci Associated with Anthracnose Resistance in Chili Pepper (Capsicum spp.) (고추 탄저병 저항성 관련 양적형질 유전자좌 분석)

  • Pepper ($Capsicum$ spp.) anthracnose caused by $Colletotrichum$ $acutatum$ is a destructive disease susceptible to areas where chili peppers are grown. $Capsicum$ $baccatum$ var. $pendulum$ (Cbp) is resistant to anthracnose and has actively been used for interspecific hybridization for the introgression of resistance gene(s) into cultivated chili peppers. The goals of this study were to determine the inheritance of resistance to anthracnose within $Capsicum$ $baccatum$ and to map quantitative trait loci (QTLs) for the anthracnose resistance. A genetic mapping population consisting of 126 $F_2$ plants derived from a cross between $Capsicum$ $baccatum$ var. $pendulum$ (resistant) and $Capsicum$ $baccatum$ 'Golden-aji' (susceptible) was used for linkage mapping. The linkage map was constructed with 52 SSRs, 175 AFLPs, and 100 SRAPs covering 1,896cM, with an average interval marker distance of 4.0cM. Based on this map, the number, location, and effect of QTLs for anthracnose resistance were studied using plants inoculated in the laboratory and field. A total of 19 quantitative trait loci (2 major QTLs and 16 minor QTLs) were detected. Two QTLs ($An8.1$, $An9.1$) showed 16.4% phenotypic variations for anthracnose resistance after wounding inoculation. In addition, five minor QTL loci ($An7.3$, $An7.4$, $An4.1$, $An3.1$, $An3.2$) showed a total of 60.73% phenotypic variations of anthracnose resistance in the field test. Several significant QTLs were also detected and their reproducibility was confirmed under different inoculation conditions. These QTLs are now being confirmed with different breeding populations. Markers tightly linked to the QTLs that are reliable under different environmental conditions will help to determine the success of marker-assisted selection for anthracnose -resistant breeding programs in chili pepper.

Evaluation of Scab Resistance and Effect of Photosynthetic Rates on Fruit Characteristics among Elite Pear Seedlings (배 우량계통의 검은별무늬병 저항성 평가 및 광합성률이 과실 품질에 미치는 영향 구명)

  • BACKGROUND: The scab, which is caused by Venturia nashicola, gives serious damages to pear trees. 'Niitaka' accounts for 82% of areas in pear cultivation. However 'Niitaka' is a scab susceptible cultivar. So, most of Korean farmers who growing pear trees have suffered by economic losses with the scab. In this research, we evaluated the scab resistance among elite pear seedlings to clarify genetics about the scab resistance. And we analyzed photosynthetic features with these seedlings to develop suitable cultivar which is advantageous for producing quality fruits during the growth and development of plants. METHODS AND RESULTS: We measured the rates of scab incidence among seedlings in a field experiment condition and an in-vitro test. An in-vitro test has been done with field experiment-based results. We made plant materials by grafting branches of each seedlings with 'Kongbae' rootstocks. And they had been grown for one month. Then, scab conidia suspension is sprayed to seedlings and sustained for 40 days under the controlled environment. As the results, 6 seedlings displayed lower incidence rates than other seedlings and 'Niitaka'. We also measured instant photosynthetic rates of each seedlings to determine the correlation between photosynthetic rates and fruit characteristics. However, it seemed that there is no correlation between them. CONCLUSION(S): Among the seedlings, 6 seedlings displayed the higher resistance to scab than other seedlings and 'Niitaka'. This characteristics is considered to be come from the gene expression of European pear. And we found that photosynthetic rate in trees rarely does not influence the fruit characteristics. It is considered to be affected by cultivar's own characteristics.

Incidence of Rice stripe virus during 2002 to 2004 in Korea and Chemical Control of Small Brown Plant Hopper (2002-2004년의 벼줄무늬잎마름병 발생상황 및 약제처리에 의한 애멸구의 화학적 방제)

  • Incidence of rice stripe disease, caused by Rice stripe tenuivirus (RSV), was surveyed during 2002 to 2004. The incidence area and ratio of diseased fields were decreased gradually during those period. Rate of diseased plants were 45.8%, 45.0% and 43.7% in the susceptible cultivars Chucheong, Saechucheong and Ilpum, respectively. However, the rate was 4.4% in resistant cultivar Hwaseong. In addition, breakdown rate was also significantly high in the susceptible cultivars in Chucheong, Saechucheong and Ilpum showing 33.6, 33.2 and 31.9%, respectively. In Hwaseong, the breakdown rate was 0.8%. Collection efficiency was compared between two insect vector collection methods. Insect-sucking machine method was much more efficient than sweeping net method in collecting small brown plant hopper (SBPH). According to the survey of the population density of the insect vector during 2002 to 2004, the national average population density was gradually decreased year by year, 3.6, 2.3, and 1.3%, respectively. This result was significantly related with the decrease of the incidence of rice stripe disease. Control efficiency of rice stripe disease by treating several seedling box and water surface with insecticides against SBPH resulted that imidacloprid GR, Fipronil FG, Clothianidin+Probenazole GR and Thiamethoxam GR showed over 80% of control efficiency at 28 days after treatment at the early stage in nursery.

A New Early Maturing Blackish Purple Pigmented Glutinous Rice Variety, 'Josaengheugchal' (조생 흑자색 찰벼 품종 '조생흑찰')

  • 'Josaengheugchal', a new blackish purple pigmented glutinous japonica rice cultivar, was developed by the rice breeding team of Department of Functional Crop, NICS, RDA in 2004. This cultivar was derived from a cross between 'Tohoku 149' as black glutinous source and 'Sx 864' as purple colored rice in 1992 and 1993 winter season, and selected by pedigree breeding method until $F_6$ generation. As a result, a promising line, YR15907-6-8-1-5, was advanced and designated as the name of 'Milyang 194' in 2001. The local adaptability test of 'Milyang 194' was carried out at seven locations from 2002 to 2004 and it was named as 'Josaengheugchal'. 'Josaengheugchal' is an early maturing cultivar and has 71 cm culm height. It has higher anthocyanian content compared with 'Heugnambyeo'. It is moderately resistant to leaf blast but susceptible to other disease and insect pests. The yield potential of 'Josaengheugchal' in brown rice was about 4.21 MT/ha at ordinary fertilizer level in local adaptability test. This cultivar would be adaptable to the plain paddy field of middle, Honam, and Yeomgnam in Korea under ordinary and double cropping system.

A Wide Region of Tropical Asia Adaptable Japonica Rice 'Asemi' (아시아 광지역 적응성 자포니카 벼 '아세미')

  • 'Asemi' is a rice variety derived from a cross between 'Jinmibyeo' which has translucent milled rice and medium maturity and 'Cheolwon46', an elite line with high yield and early maturity by the rice breeding team at NICS, RDA in 2013. The heading date of 'Asemi' is August 1, six days earlier than the check variety 'Hwaseong'. It has 82 cm culm length and 109 spikelets per panicle. 'Asemi' is resistant to blast disease, stripe virus and tungro virus, but susceptible to other viruses and planthoppers. The milled rice of this variety exhibits translucent, clear non-glutinous endosperm and short grain shape. It has protein content (6.7%) higher than 'Hwaseong', and amylose content (19.5%) similar to 'Hwaseong'. The milled rice recovery rate of 'Asemi' is similar to that of 'Hwaseong'. However, the head rice rate of 'Asemi' is higher than that of 'Hwaseong'. Milled rice yield of 'Asemi' is 5.23 MT/ha in ordinary cultivation. ' Asemi' could be adaptable to the wide region of tropical Asia (Registration No. 5639).

Growth Environments and Management Strategies for Pinus densiflora Village Groves in Western Gangwon Province (강원도 영서지역 소나무 마을숲의 생장환경과 관리방안)

  • The purpose of this study was to survey structures and growth conditions of Pinus densiflora village groves, and to establish management strategies for their desirable growth and conservation. Twelve village groves were selected in western Gangwon province for the study. The age of the study groves ranged from 50 to 200 years. Average dbh (diameter at breast height) and density of trees for each study grove were 27~52cm and 0.5~9.3 trees/$100m^2$, respectively. Soil environments were favorable to Pinus densiflora growth in the majority of the study groves, but 2 study groves with sandy soils showed considerably poor nutrient contents. Low tree vitality was found in some of the study groves due to poor conditions of root growth from soil fill and trampling. There were detachment of cambial tissue and damage of stem cavity at 6 study groves, which were caused by artificial injury, careless pruning, and frost damage. Light disease damage by Rhizosphaera kalkhoffii and phomopsis blight were found at 6 study groves. Light pest damage by Thecodiplosis japonensis was also found at 6 study groves, but the pest damage at 2 study groves was relatively considerable. Thus, major factors limiting normal growth of Pinus densiflora village groves were infertility, soil fill and trampling, stem damage, and disease and pest. Desirable management strategies were explored to solve growth-related problems and to conserve the study groves. The management strategies included fertilization of organic matter and lime, removal of soil fill, soil plowing and graveling, wood-trail installation or woodchip mulching, supply of wood fences and protective frames, surgical operation for damaged stems, vitality enhancement, and trunk injection to improve growth environments or control stem damage and disease/pest.

  • Mycelial growth of Colletotrichun dematium and C. gloeosporioides isolated from anthracnose of Lycium chinense was favorable on V-8 Juice Agar media and Oatmeal Agar media of the four different media tested. Sporulation of C. dematium was favorable on PDA media and V-8 Juice Agar media and that of C. gloeosporioides was favorable on all media tested. The optimum temperature for mycelial growth of C. gloeosporioides and C. dematium was $25^{\circ}C$ and $28^{\circ}C$, respectively and sporulation of the fungi was stimulated under alternating NUV light and darkness. Of the seven nitrogen sources tested, gelatin was the most favorable for mycelial growth of C. gloeosporioides and glycin and gelatin were favorable for that of C. dematium. Of the eleven carbon sources tested, dextrin and sorbitol were favorable for mycelial growth of C. gloeosporioides and dextrin was the most favorable for that of C. dematium. Among the eleven fugicides tested, Benomyl, Folpet, Dithianon, Carbendazim and S-3308L were found superior for the inhibition of mycelial growth and spore germination of the fungi. They were, also, found superior for the control of anthracnose disease of L. chinense when they were applied to the plants in the field.

Evaluation of resistance to Pierce's disease among grapevine cultivars by using the culture filtrates produced from Xylella fastidiosa (Xylella fastidiosa의 배양여액을 이용한 포도나무 피어스병 품종 저항성 검정)

  • This study investigated whether culture filtrates produced by Xylella fastidiosa can be used to determine the varietal susceptibility to Pierce's disease in grapevines (Vitis spp.) as a substitute for pathogen inoculation or field screening. A bioassay of grape leaves with culture filtrates from the pathogen showed that their phytotoxicities were active and host-selective. Ethyl acetate extracts from them also showed toxicities and host selectivity in both bunches of grapes and muscadine grapes. The sensitive range of plants to the culture filtrates and their ethyl acetate extracts was consistent with the host range of the Pierce's disease pathogen. Susceptible cultivars are sensitive to even highly diluted culture filtrates, while resistant cultivars were not affected even at their original culture filtrates. Susceptible cultivars were more sensitive to the undiluted culture filtrate than were highly diluted culture filtrates, and the younger leaves were the most sensitive to the culture filtrates in grapes. Although some European grape cultivars showed moderately susceptibility in this study, the determination of varietal resistance to Pierce's disease by the treatment of culture filtrates of pathogens could provide valuable information for the preliminary selection of genetic resources and seedlings from hybridization in a disease resistant grape breeding program.
