Characteristic of Progeny in Pepper Transformants (고추 유전자변형체 후대 생육특성 검정)

  • For the resistance test for Phytophthora blight of $T_1$ and $T_2$ transformants in pepper, Phytophthora blight fungus was inoculated to seedlings of the $T_1$ and $T_2$ transformants by concentration (density: zoospore $10^3/ml$). Occurrence rate of blight at 5days after inoculation was 4.0 % in T1-1 line and 10.0% in $T_1-2$ line, and its rate for 12 days after inoculation was 52.0% in $T_1-1$ line, 64.0% in $T_1-2$ line, respectively. Therefore, the lower occurrence rate to blight was enable to select resistant transformants in the some inoculation density (zoospore $10^3/ml$), meanwhile 'Kumtap' and 'Subicho' were 100% in highest occurrence rate to blight. For field test, in which blight was commonly occurred, of the Youngyang Pepper Experiment Station, the acquired transformant resisting to blight was similar to characteristics of domestic varieties, 'Subic ho' for fruit shape, but there are some differences in growth, days to flowering, fruit characteristics. Occurrence of blight in $T_2-1-6$, and $T_2-4-9$ lines was smaller approxmately 30% than commercial varieties, 'Kumtap', although occurrence of blight in field was showed higher difference among tested lines. In this study, we concluded that the transformants showing blight resistance selected from habitual field could be fixed at every generation, and the developed transformation system was also considered to develop transformants in pepper.

Phylogenetic study of the fern genus Hypodematium (Hypodematiaceae), focusing on Korean native taxa (한국산 금털고사리속의 계통분류학적 연구)

  • In Korea, Hypodematium glanduloso-pilosum was formerly known as the only Korean native species in the genus. Recently, however, we reported one unrecorded taxon, H. squamuloso-pilosum Ching, which was found on rocks at a limestone mountain in Yeongwol, along with one new taxon, H. angustifolium in Okcheon. Traditionally, Hypodematium taxa are often distinguished from each other by vegetative characters such as pinnatifid lamina, glandular hairs, and narrower or linear lanceolate scales on stipe bases. H. glanduloso-pilosum, distributed widely throughout the country except on Jejudo Island in Korea, exhibiting variations in leaf segregation, indusia positions, hair distributions and size. The high variation in the morphological characters in the widely distributed taxon has caused problems delimitating three native species from each other. To evaluate the phylogenetic relationships among H. glanduloso-pilosum and taxa related to Hypodematium (all Korean native taxa), we carried out morphological and molecular analyses (cpDNA rbcL and psbA-trnH) of populations of the genus Hypodematium in Korea. Although H. glanduloso-pilosum exhibits high variations in some morphological characters, the species is characterized by stipes and indusia with densely multicellular hairs and rod-shaped glandular hairs or hairs and lanceolate or oblong lanceolate scales in rhizomes and stipe bases distinguished from those of other Korean native taxa (H. squamuloso-pilosum and H. angustifolium). In the analyses of cpDNA data, three Korean native taxa are placed in the same clade, i.e., in the glanduloso-pilosum group. Moreover, our analyses propose that H. squamuloso-pilosum (China and Korea), H. angustifolium (Korea), and H. fordii (China and Japan) share the same glanduloso-pilosum clade with H. glandulosopilosum (China, Korea, and Japan).

The Influence of Students' Perception of Tutor's roles on Deep Learning, Achievement, and Course Evaluation in Online Gifted Education Program (온라인 영재교육 프로그램에서 중학생의 튜터 역할에 대한 인식이 심층학습, 학업성취, 수업평가에 미치는 영향)

  • This study investigated the relationships among middle school students' perceptions on the roles of online tutor, their deep learning, achievement, and overall evaluation of learning experiences in the context of inquiry based online gifted mathematics and science learning. For this purpose, 249 middle school students who took online course were surveyed about their perceptions on the degree to which their tutor performed the roles as an online tutor. The students were also asked about the activities which indicate deep learning approaches and overall course experiences such as the level of satisfaction, understanding and engagement in the course. The regression analyses were conducted to examine the relationships of students' perceptions on the roles of online tutor, deep learning, achievement, and overall course experiences. The results first showed that the roles of online tutor which affects students' deep learning approach such as high-order learning, integrative learning, reflective learning were the role as a subject matter and evaluation expert. Among the sub variables of deep learning approach the variable that was related to students' overall achievement was the use of high-order learning strategy. Second, the achievement in inquiry task was related to the role of tutor as a guide of learning process and method. Third, students' overall course evaluations such as the level of satisfaction, understanding and engagement were not related to any role of tutor.

Studies On the Major Components and Antioxidative Properties of Whole Fruit Powder and Juice Prepared from Premature Mandarin Orange (미숙 온주밀감을 이용한 통 밀감 분말과 착즙액의 성분함량 및 항산화성에 관한 연구)

  • Major components and antioxidative properties of whole fruit powders and juices prepared from premature mandarin, Citrus unshu, were investigated. Dietary fiber, flavonoid, and organic acid contents greatly increased in samples with lower degree of fruit maturity (August) than higher ones (October), and in whole fruit powders than extracted juices, whereas free sugar contents, especially sucrose content, increased in fruits with higher degree of maturity than lower ones. Antioxidative properties were not affected by degree of fruit maturity, whereas total polyphenol content and electron-donating abilities were higher in extracted juices than whole fruit powders. Nitrite-scavenging abilities of all samples were similar except at pH 6.0.

Compressive Creep Behavior of Rice Starch Gels (쌀 전분 젤의 creep 특성)

  • The creep behavior of gels made with $30{\sim}45%$ gelatinized rice starch was measured over a wide range of temperature. Compressive creep curves of rice starch gels conformed to a six element mechanical model consisting of one Hookean, two Voigt and one Newtonian component. The creep compliance of gels decreased with increasing starch concentrations. Among viscoelastic constants of the mechanical model, elastic modulus was mainly influenced by the change of starch concentrations. The concentration-invariant compliance curve was obtained by reduction to 38% using reduction parameter $a_{c}$. The creep compliance curves of 45% starch gels increased with temperature, which indicated that rice starch gels became softer and less rigid with increasing temperature. When the compliance at $20^{\circ}C$ was set as a reference curve, creep compliance data for 45% gels at various temperature could be superimposed as a continuous smooth curve. The apparent activation energies of 45% rice starch gels calculated by the modified WLF equation were not intrinsic, but decreased as temperature increased.

Quality Characteristics of Ogapiju Prepared by Different Raw Materials (원료 첨가를 달리한 오가피주의 품질 특성)

  • Quality characteristics of ogapiju prepared by adding different raw materials such as Acanthopanax(0, 0.1, 0.25, 0.5%), Ganoderma lucidum(0, 0.05%), pine needle (0, 0.05%), and red ginseng(0, 0.05%) were evaluated by chemical analyses and sensory evaluation. Total organic acid contents of samples containing ogapi and other medicinal herbs were higher than that of control group. Major free amino acid was histidine. Contents of total amino acid and free sugar were highest in sample C (Acanthopanax 0.1, G. lucidum 0.05, pine needle 0.50, red ginseng 0.05%). In each sample 31-49 volatile components were identified by solid-phase microextraction method, and 42 components were detected by sniff-test using GC-olfactometry. Ogapiju showed higher content of ${\alpha}-copaene$ than control group ${\alpha}-Pinene$, camphene, ${\beta}-pinene$, sabinene, ${\alpha}-terpinene,\;{\gamma}-terpinene$, p-cymene, terpinolene, ${\alpha}-thujone,\;{\beta}-thujone,\;{\alpha}-terpineol$, carvone, and ${\beta}-ionone$ were not identified in control group. Volatile composition of ogapiju was characterized by higher amount of terpenoids. Green and herbaceous note was stronger in ogapiju than control group. Sensory evaluation indicated that good taste and palatability were highest in sample C.

Changes in Moisture Content and Quality of Chewing Gum during Storage (저장중 츄잉껌의 수분함량과 품질변화)

  • The changes in sensory and mechanical texture of chewing gum during storage at various relative humidity were studied to define the quality index for the prediction of shelf-life. The initial moisture content of chewing gum was 2.57% (dry basis). The BET monolayer value at $a_{w}$ 0.19 was calculated to be 1.57% (dry basis). The sensory scores of chewing gum were closely correlated with moisture content and instrumental texture parameters with 0.1% significant level. Therefore the quality of stored chewing gum was directly related with moisture content above BET monolayer. The products became organoleptically acceptable in the range of moisture content $2.17{\sim}3.16%(dry basis)$. This range of moisture content ie equivalent to the ranges of instrumental parameter, fracture force$0.8{\sim}1.8{\times}10^{7}$, fracture modulus $1.1{\sim}2.4{\times}10^{8}$, puncture force $0.5{\sim}1.1{\times}10^{7}[dyne/cm^{2}]$ and brittleness $0.7{\sim}1.4{\times}10^{8}[dyne/cm^{3}]$, respectively.

Non-thermal Pasteurization of Carrot Juice by High Voltage Pulsed Electric Fields with Exponential Decay Pulse (고전압 Exponential Decay Pulse를 이용한 당근주스의 비열(非熱) 살균)

  • Carrot juice inoculated with $2\;{\times}\;10^8\;cfu/mL$ of Escherichia coli was treated with pulsed electric fields(PEF) for the purpose of a development of new cold pasteurization processes. Inactivation of E. coli in carrot juice increased with increase in intensity of the electric field strength and treatment time. The cells were suspended at concentration of ca. $2\;{\times}\;10^8$ cells per ml. A reduction of 4D was obtained at 40 kv/cm and 256 exponential decay pulses at room temperature. Critical electric field strength(Ec) and treatment time(tc) needed for inactivation of E. coli were 11.74 kV/cm and $3.6\;{\mu}s$ at room temperature, respectively. The combination of PEF and thermal treatment inactivated E. coli more effectively. The reductions of up to 5.5D were observed when the carrot juice was treated with PEF of 22.5 kV/cm and $205\;{\mu}s$ at $50^{\circ}C$. PFF treatment did not effect in color, pH, $^{\circ}Brix$, titratable acidity and ${\alpha}-,\;{\beta}-carotene$ contents of carrot juice.

Studies on Optimum Cooking Conditions for Commercial Continuous Rice Cooker (대량연속취반장치에서 최적 취반조건 연구)

  • The optimum cooking conditions for large scale continuous gas cooker were studied with three varieties of rice. Optimum soaking time for the cooker was estimated to be 30 min at cooking temperature above $20{\circ}C$, while 60 min at cooking temperature below $10{\circ}C$. The ratio of water-to-milled rice giving the best eating quality was founded to be $1.41{\sim}1.48$, and corresponding moisture content of cooked rice ranged from 60 to 62% for different rice varieties. The optimum ratio of water to milled rice. (Chucheong) decreased from 1.50 to 1.37 with increase in the amount of milled rice from 4.0 to 6.5 kg in a cooking vessel. However, actual water uptake by rice (g water absorbed per g rice) on cooking was nearly constant value of 1.13, which means that evaporated water during cooking increases with decreasing the amount of rice in a cooking vessel.

Effect of Sucrose and Gluten on Glass Transition, Gelatinization, and Retrogradation of Wheat Starch (밀전분의 유리전이와 호화 및 노화에 대한 sucrose와 글루텐의 영향)

  • Differential scanning calorimetry (DSC) was used to study effects of sucrose and gluten on wheat starch glass transition, gelatinization, and retrogradation. Glass transition temperature ($T_{g}$) of wheat starch decreased as the ratio of sucrose or gluten to starch increased. Both peak temperature ($T_{G}$) and enthalpy values of gelatinization endotherm increased or decreased with increasing ratio of sucrose or gluten, respectively. Wheat starch gel with no sucrose and gluten recrystallized up to 4 weeks of storage at $4^{\circ}C$, whereas those with sucrose and gluten completed recrystallization within 1 week. Both wheat starch gels with no sucrose and gluten, and those with sucrose and gluten at storage temperature of $32^{\circ}C$ recrystallized up to 4 weeks, with wheat starch-sucrose-gluten (1 : 0.5 : 0.12) system, which had highest ratios of gluten and sucrose to starch, showing lowest recrystallization. Nucleation and propagation rates of starch gel recrystallization based on polymer crystallization principles can be converted into peak width (${\delta}T$) and peak temperature ($T_{R}$) of retrogradative endotherm by DSC, because higher nucleation rate at storage temperature of $4^{\circ}C$ close to $T_{g}$ showed higher ${\delta}T$, whereas higher propagation rate at $32^{\circ}C$ (close to $T_{G}$) had higher $T_{R}$.
