Biochemical and Cytological Changes of Pseudomonas sp. DJ-12 Cells in Response to Catechol Treatment (Catechol 처리에 의한 Pseudomonas sp. DJ-12의 생화학 및 세포학적 변화)

  • Aromatic hydrocarbons which are not easily degraded by microorganisms can be accumulated in the conlaminated environment for a long lime, producing toxic effects on wild lives and humans. However, the sublethal concentrations of the chemicals induce the synthesis of stress-shock proteins in the cells and increase the adaptability of the organisms to the environmental stresses. In this study, therefore, the cells of Psezido~nonus sp. DJ- 12 treated with catechol at various concentrations were inveshgated for their survival, biodegtadability of catechol, production of stress-shock proteins, and cytological changes. The organisms were capable of degrading catechol at the range of 0.5 to 1.0 mM concentration wilhin 6 hours incubation, but they were killed by $10^2$-10$^3$ celllinl at 3 mM or higel- concentration without any catechol degradation. These cells treated with catechol begm lo produce DnaK and GroEL at 1 mM and 0.5 mM. respectively. Pseudumonas sp. DJ-12 treated with 10 mM catechol for I hour exhihiled some punctuated pores on the cell wall and contortion of the rod shape. The cells treated with he sublethal concentration of catechol showed the increased tolerance for suvival when exposed to the lethal concentration, and such tolerant effects were functioned crossly among benzoate, 4-chlorobenzoate, 'and catechol.

Characteristics of Adhesive Disks in Parthenocissus tricuspidata during Attachment (착생에 따른 담쟁이덩굴 흡착근의 부착 특성)

  • Parthenocissus tricuspidata is an epiphyte that lacks a main axial stem, but develops adhesive disks along the stem for climbing support. In this study, scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) were utilized to examine the brick wall surface and the adhesive disks of P. tricuspidata that attached to the surface successfully. The study was mainly focused the outermost layers of both structures before and after adhesion to find out whether there has been some structural and/or physical interactions between the two. The adhesive disks adhered firmly to the brick wall by secreting adhesive materials that help them for a tight attachment to the surface. The rough wall surface appeared facilitating better attachment of the adhesive disks by infiltrating the materials into those spaces leading to some degree of interactions at the interface. EDS analysis on the outermost layers of the adhesive disks that were separated from the substrates was also consistent with the SEM data on the interaction between the adhesive disks and the substrate surface. EDS analysis of the brick wall surface and the adhesive disks demonstrated similar elements of O, Si, Fe, Al, K, Mg, and Na in their components.

Immuno-stimulating Activities of Mannose-rich Polysaccharides Isolated from Korean Black Raspberry Wine (복분자주에서 분리한 Mannan 다당의 면역증진 활성)

  • Polysaccharides isolated from Korean black raspberry wine were examined for their chemical properties and immuno-modulating activities. The molecular mass of RB-1b-I, the main polysaccharide in black raspberry wine, was estimated as 180 kDa and it contained a significant proportion of mannose (76.8%) and 4 different minor component sugars such as galactose (15.8%), arabinose (3.8%), glucose (2.6%) and rhamnose (1.2%). This indicated that RB-1b-I was mainly present as a mannan, which had originated from the cell walls of fermenting yeasts. On the other hand, RB-1b-I induced high levels of macrophage activation as well as mitogenicity regarding murine splenocytes in vitro. The intravenous administration of RB-1b-I significantly augmented NK cytotoxicity against YAC-1 tumor cells. RB-1b-I also showed potent anti-complementary activity in a dose-dependent manner via both alternative and classical pathways. Results indicated that Korean black raspberry wine contains peculiar polysaccharides which provide beneficial immuno-stimulating activities for human health.

Analysis of Off-flavor Compounds from Over-extracted Coffee (과추출에 의한 커피의 이취성분 분석)

  • To verify the volatile compounds contributing to coffee off-flavor, medium roasted Colombian, Sumatra, Ethiopian and light and dark roasted Colombian bean coffees were sequentially and studiously over extracted. Eight espresso samples and eight filter-dripped samples were analyzed by gas chromatography/mass spectrometry with the solidphase microextraction method and evaluated by sensory tests. In total, 67 compounds were detected, and desirable aroma decreased sharply, while off-flavors increased rapidly after 30 mL of espresso and 200 mL of drip coffee. Percent peak area of 4-ethylguaiacol revealed a linear increase during extraction and was highly correlated with increasing off-flavor, suggesting that it could be an indicator of over extraction. Considering the odor activity value, guaiacol and 4- vinylguaiacol were also contributory compounds to off-flavors.

Developmental Patterns of Glandular Trichomes in Leaves of Vitex negundo (좀목형 엽육 표피조직의 분비모 발달 양상)

  • Vitex negundo is an aromatic plant which releases a unique scent due to the presence of essential oil stored presumably within glandular trichomes. The focus of this research was to study developmental patterns of glandular trichomes in Vitex negundo leaves using electron microscopy. There are two types of glandular trichomes which develop on the leaf epidermis of Vitex negundo, peltate glandular type (PT) and capitate glandular type (CT). Structural features differ significantly depending on size and density, formation of secretory cavity, plastid, etc during developmental stages. In young leaves, undifferentiated PTs are densely distributed in the upper epidermis, but are not externally exposed in the lower epidermis because they are covered by non-glandular simple trichomes. Upon leaf development, PTs and CTs show clear structural differentiation in the upper and lower epidermis. PTs are composed of up to eight head cells (ca. 35~40 ${\mu}m$) and one stalk cell (ca. 5 ${\mu}m$), while CTs are composed of four head cells (ca. 10~15 ${\mu}m$) and 1~2 stalk cells (ca. 10 ${\mu}m$). Although secretory cavities develop on the secretory head cells, their size, structure, and formation proceed very differently depending on trichome type. In early development of PT, a large cavity with numerous secretory vesicles form rapidly from the head cells. In CT, however, only a small secretory cavity is formed, slowly relative to PT, without secretory vesicles. The PTs are considered to play an important role in releasing the aromatic components of Vitex negundo.

Improvement of Insulation System for LNG Storage Tank Base Slab (LNG 저장탱크 바닥판 단열 시스템 개선)

  • Liquefied natural gas(LNG) is natural gas that has been converted temporarily to liquid form for ease of storage and transport it. Natural gas is the worlds cleanest burning fossil fuel and it has emerged as the environmentally preferred fuel of choice. In Korea, the demand of this has been increased since the first import from the Indonesia in 1986. LNG takes up about 1/600th the volume of natural gas in the gaseous state by cooling it to approximately $-162^{\circ}C(-260^{\circ}F)$. The reduction in volume therefore makes it much more cost efficient to transport and store it. Modern LNG storage tanks are typically the full containment type, which is a double-wall construction with reinforced concrete outer wall and a high-nickel steel inner tank, with extremely efficient insulation between the walls. The insulation will be installed to LNG outer tank for the isolation of cryogenic temperature. The insulation will be installed in the base slab, wall and at the roof. According to the insulation's arrangement, the different aspects of temperature transmission is shown around the outer tank. As the result of the thermal & stress analysis, by the installing cellular glass underneath the perlite concrete, the temperature difference is greatly reduced between the ambient temperature and inside of concrete wall, also reducing section force according to temperature load.

Evaluation of Design Characteristics in the Reinforced Railroad Subgrade Through the Sensitivity Analysis (민감도 분석을 통한 철도보강노반 설계 특성 평가)

  • By changing from ballasted track to concrete slab track, new type railroad subgrade is strongly required to satisfy strict regulations for displacement limitations of concrete slab track. In this study, sensitivity analysis was performed to assess the design characteristics of new type reinforced railroad subgrade, which could minimize residual settlement after track construction and maintain its function as a permanent railway roadbed under large cyclic load. With developed design program, the safety analysis (circular slip failure, overturning, and sliding) and the evaluation of internal forces developed in structural members (wall and reinforcement) were performed according to vertical installation spacing and stiffness of short and long geotextile reinforcement. Based on this study, we could evaluate the applicabilities of 0.4 H short geogrid length with 0.4 m vertical installation spacing of geotextile as reinforcement and what the ground conditions are for the reinforced railroad subgrade. And also, we could grasp design characteristics of the reinforced railroad subgrade, such as the importance of connecting structure between wall and reinforcement, boundary conditions allowing displacement at wall ends to minimize maximum bending moment of wall.

A Model for Evaluating the Radioactive Contamination in the Urban Environment (도시환경에서 방사성물질의 오염평가 모델개발)

  • A model for evaluating radioactive contamination in the urban environment, named METRO-K, was developed as a basic step for accident consequence analysis in case of an accidental release. The three kind of radionuclides $(^{137}Cs,\;^{106}Ru,\;^{131}I)$ and the different chemical forms of iodine (particulate, organic and elemental forms) are considered in the model. The radioactive concentrations are evaluated for the five types of surface (roof, paved road, wall, lawn/soil, tree) as a function of time. Using the model, the contaminative impacts of the surfaces were intensively investigated with respect to with and without precipitation during the measurement periods of radionuclides in air. In addition, a practical application study was conducted using $^{137}Cs$ concentration in air and precipitation measured in an European country at the Chernobyl accident. As a result precipitation was an influential factor in surface contamination. The degree of contamination was strongly dependent on the types of radionuclide and surface. Precipitation was more influential in contamination of $^{137}Cs$ than that of $^{131}I$ (elemental form).

Study on Indoor Wireless Environment of mmWave WLAN Communication (초고주파 근거리 통신의 실내 무선 환경 연구)

  • Recently, as the demand for transmission of ultra-high quality media data such as UHD, AR, and VR increases, various technologies for this have been actively developed and IEEE 802.11ad standard have been commercialized. In this paper, a test bed is constructed to analyze the indoor wireless environment using the IEEE 802.11ad standard based on mmWave, and the experimental results of various indoor wireless environments are introduced and analyzed. We compared the data from the module by data transmission, such as signal to noise ratio(SNR) and throughput. And we measured the beam pattern and width of the module and compared the effects on the indoor environment of the corridor and the office. This shows that the signal reflection of the wall shows higher SNR values and is more suitable to use for indoor than outdoor. It is confirmed that the loss when not in line of sight(LoS) is not enough to compensate the wall reflected signal. As a result, it is judged to be suitable for the indoor use of the mmWave LAN and can be usefully used for further experiments.

Homology Modeling and Characterization of Oligoalginate Lyase from the Alginolytic Marine Bacterium Sphingomonas sp. Strain MJ-3 (알긴산을 분해하는 해양미생물인 Sphingomonas sp. MJ-3 균주의 올리고알긴산 분해효소의 상동성 모델링 및 특성연구)

  • Alginates are found in marine brown seaweeds and in extracellular biofilms secreted by some bacteria. Previously, we reported an oligoalginate lyase from Sphingomonas sp. MJ-3 (MJ3-Oal) that had an exolytic activity and protein sequence homology with endolytic polymannuronate (polyM) lyase in the N-terminal region. In this study, the MJ3-Oal was tested for both exolytic and endolytic activity by homology modeling using the crystal structure of Alg17c from Saccharophagus degradans 2-40T. The tyrosine residue at the $426^{th}$ position, which possibly formed a hydrogen bond with the substrate, was mutated to phenylalanine. The FPLC profiles showed that MJ3-Oal degraded alginate quickly to monomers as a final product through the oligmers, whereas the Tyr426Phe mutant showed only exolytic alginate lyase activity. $^1H$-NMR spectra also showed that MJ3-Oal degraded the endoglycosidic bond of polyM and polyMG (polymannuronate-guluronate) blocks. These results indicate that oligoalginate lyase from Sphingomonas sp. MJ-3 probably catalyzes the degradation of both exo- and endo-glycosidic bonds of alginate.
