Discrimination Method of Brown Planthopper, Nilaparvata lugens(Stal) Nymphs by the Fluorescent Spots between Compound Eyes in Rice Paddies (벼논에서 벼멸구(Nilaparvata lugens Stal)약충 두부의 형광성 반점에 의한 식별법)

  • Several species of planthoppers such as brown planthopper, N. lugens (Stal) (BPH), smaller brown planthopper, Laodelphax striatellus (Fall n) (SBPH), and white-backed planthopper, Sogatella furcifera (Horv th) (WBPH) are distributed in Korean rice paddies but not easy to discriminate them in situ. Accurate discrimination of them is an indispensable process in the forecasting for their outbreak and control. Especially, innovative discrimination method for BPH was required because BPH was one of the most important insect pest of rice. Nymphs and adults of BPHs, SBPHS, and WBPHS, thus, were examined their morphological characteristics in the paddies and laboratory. The nymphs of BPH had different characters from those of SBPH and WBPH. The nymphs of BPH had white fluorescent spot between bottom of compound eye and antenna, while there was no that spot the other two species. The white spot was the brightest at the nymphs just after hatching and getting weaker as the nymph developed. At last the white spot was totally disappeared at the adult stage. This white spot was innovative criterium to discriminate nymphs of BPH, SBPH, and WBPH in rice paddies.

  • A method to control the cigarette beetle. Losioderma serricorne F, m cigarettes by using gamma ray irradiation was evaluated When all stages of cigarette beetle were placed in gelatin capsules and irradiated wlth gamma ray. no eggs survived, but mortalities of pupae, larvae and adults exposed to above 0.8 kGy of gamma ray were only from 0 to 30%. Similar results were also obtained in the control efficacy of the beetle when seuqal kinds ot currently produced ngarettes containing m gelatin capsules were inadhated with gamma ray The cigarette beetle adults and larvae surviving gamma ray inadiatlon were reared on artificial d~etsto show that there were no reproduc. tions was observed in the second generation. The survival periods for each growth stage became shoter with increase gamma ray dosage. The fouirh instar larvae had the longest sulvlvai periods No sign~ficant changes of cigarette flavor and taste were noticed alter gamma ray inadiation. The cigarette packet, metal fotl for packet and cigarette paper by the inad~atlon were not discolored by irradiation.

Feasibility in Differentiation of Resistance of Rice Varieties to Brown Planthopper (Nilaparvata lugens Stal) using Radisoisotope (P-32) Tracer-Technique (P-32를 이용한 벼멸구(Nilaparvata lugens Stal) 저항성 검정법에 관한 연구)

  • It has been documented that the resistance to planthoppers is attributed to the feeding preference of the insects. This might be related to difference in the amount of feeding plant sap between resistant and susceptible hosts. In this aspect, this study was conducted to verify it and to develop an effective screening method for resistance to planthoppers by tracer technique. An effective P-32 labelling of rice seedlings at $2\~3$ leaf stage was dipping the roots in concentration of $0.1{\mu}Ci/ml$ solution for 48 hours. Radioactivity was significantly higher in planthopers fed on susceptible variety for 48 hours as compared to those fed on resistant variety. Radioactivity of adults was higher than that of nymphs and also higher in female than male. The results were highly correlated to that indicated by the feeding preference of the insects and therefore, considered to be valid for a screening technique.

Significance of Semame Seedborne Fungi, with special Reference to Corynespora cassiicola (참깨의 종자전염성 진균과 그 병원성 : Corynespora cussiicola를 중심으로)

  • Alternaria sesami, A. sesamicola, A. tenuis, A. longissima, Cercospora sesami, Cephalosporium sp., Corynespora cassiicola, Fusarium equiseti, F. moniliforme, F. oxysporum, F. semitectum, Macrophomina phaseolina and Myrothecium roridum were detected from 40 seed samples of sesame. A sesami, A. sesamicola, A. tenuis and C. cassiicola were the predominant fungi. Except C. cassiicola, all fungi were almost completly reduced and wiped out the infection by pretreatment with chlorine. Plating components also indicate that C. cassiicola was well-established infections. Seedborne infection of C. cussiicola caused heavy seed rot and seedling mortality. Detailed description has been given on the habit character of C. cassiicola under stereoscopic microscope and the variation in colony character and spore morphology have been taken into account. In inoculation experiments, C. cassiicola produced severe leaf and stem spots and blights on sesame plants resulted in ultimate death of the plants. A. sesami, A. sesamicola A. longissima and C. sesami also produced mild to severe leaf spotting and leaf blight when suspension of their conidia were sprayed on to plants. In soil inoculation experiments, F. oxysporum and M. phaseolina were the most pathogenic causing seed rot and seedling blight.

Incidence of Watermelon Mosaic Virus in Cucurbits (박과 작물에 발생하는 Watermelon Mosaic Virus에 관한 연구)

  • Cucurbits including pumpkin (Cucurbita pepo), gourd (Lagenariaa siceraria), cucumber (Cucumis sativus), melon(Cucumis melo) and watermelon(Cucurbita anguria) were diseased with mosaic symptoms. The causal virus was identified as watermelon mosaic virus(WMV). The WMV was transmitted by Myzus persicae Sulzer, and no seed borne virus was found. The virus caused large local lesions on the inoculated leaves of the Chenopodium amaranticolor and mosaic symptom on the upper leaves of Cucumis melo, Cucumis sativus, Lagenaria siceraria, Cucurbita anguria and Cucurbita pepo. There were no symptoms on the inoculated leaves of the Nicotiana tabacum var. Bright yellow, Nicotiana glutinosa, Vigna unguiculata. Petunia hybrida and Datura stramonium. Thermal inactivation point was $55\~65^{\circ}C$, dilution end point was $10^{-4}\;10^{-5}$ and longevity in vitro of the virus was $7\~8$ days. The virus showed positive reaction against watermelon mosaic virus antiserum in microprecipitin tests. The virus particles were flexuous rods in size of 750 nm.

  • Tropical fire ant, Solenopsis geminata (Fabricius) is distributed over the tropical and temperate areas. It is a polyphagous pest attacking to various plant and causes the pain to the people doing outdoor work. So it is known for a hygienic pest to human being, especially the people doing outdoor activities. It can cause severe pain to human, even with just a bite. Of particular concern is the possibility of its settlement in this country, if it invade to the farmlands and/or forests of Jeju Island and southern part in Korea. It is listed as a regulated pest in Plant Quarantine Act of Korea. This ant is usually imported through a ship and early settled in the port area where it is already distributed. Recently its distribution is rapidly spreading into Southeast Asia and southern part of China. In Korea, it was detected the one time in 1990s, but was detected the ten times from 2000 to 2006 by an imported plant quarantine process. Therefore, we would like to provide some information on the species, including morphology, distribution, host plant, damaged symptom and its general bionomics, which will be useful data for plant quarantine process and control.

Morphology and Biochemcial Characteristics of a Nuclear Polyhedrosis Virus Isolated from the Oriental Tobacco Budworm, Helicoverpa assulta (Guenee) (담배나방 핵다각체병바이러스의 형태 및 생화학적 특성)

  • A nuclear polyhedrosis virus isolated from the oriental tobacco budworm larvae, Helicoverpa assulta (Guenee) was characterized by electron microscopy, SDS-PAGE, restriction endonuclease analysis and cross infectivity. The shape of a polyhedron was $1.0\mu\textrm{m}$ in average with icosahdral outline, and the virus particle was $65nm\times300nm$ in average with rod-shape. The nuclear polyhedrosis virus was contained a single nucleocapsid within a viral envelope embedded in a polyhedron. The polyhedral protein was composed of a single polypeptide with a M.W. of 31 Kd. The genome size of the virus by restriction endonuclease analysis was about 120 Kb. Among several nuclear polyhedrosis viruses, the nuclear polyhedrosis virus from Helicoverpa assulta (HaNPV) and Autographa california nuclear polyhedrosis virus (AcNPV) were infected the oriental tobacco budworm larvae.

  • Experiments were carried out to control white-tip nematode, Aphelenchoides besseyi Christie, during the period just before transplanting. The experimental results were summarized as fellows: 1: From a 12-hr to a 24-hr sled-soaking application into 500 times dilutions of Malathion $80\%$ Ec, Sumithion $50\%$ EC and Lebaycid $50\%$ EC were highly effective than other emulsifierble concentrations of chemicals. 2) The water surface application of PSP-204 $5\%$ G, Diazinon $3\%$ G, Nemagon $20\%$ G and Terracurr-P $5\%$ G were effective. 3: Root·dipping application of Sumithion $50\%$ Ec, Lebaycid $50\%$ Ec and Nemagon $80\%$ Ec were more effective chemicals than others.

  • 1. The number of Rhizoctonia solani, Pythium debaryanum, Fusarium and Trichoderma was detected by the modification of Boosails special plating method when ginseng seed bed was drenched with Captan, Difolatan, Zineb, Maneb and PCNB at weekly intervals. Pythium debaryanum Hesse was described for the first time on Panax ginseng in Korea. 2. The number of Rhizoctonia solani and Pythium debaryanum was decreased gradually as the geason Progressed, whereas that of Fusarium and rrichcderma was increased. 3, The number of Rhixoctonia solani was greatly reduced by PCNB, and soils treated with other fungicides generally showed less Rhizoctonie solani than in the control. The number of Pythium debaryanum was significantly reduced by Zineb, Maneb, followed by Captan and Difolatan. None of the fungicids reduced the number of Fusarium colonies in the fourth week. Effects of the chemicals on Trichoderma were not statistically significant. 4. More fresh weight of the seedling roots was obtained using Difolatan, Maneb and PCNB. Phytotoxicity was noted with Maneb, Zineb and Captan after the third treatment.

Joint Toxic Action of Bifenthrin and Prothiofos Mixture for the Control of Insectivcide-Resistant Diamondback Moth, Plutella xylostella L. (살충제 저항성 배추좀나방 방제를 위한 Bifenthrin과 Prothiofos 혼용의 연합작용)

  • Mixture study of bifenthrin and prothiofos was conducted to control insecticide resistant diamondback moth (DBM), Plutella xylostella, which had shown 581, 18, 19, 11 fold resistance to fenvalerate, cypermethrin, furathiocarb, and prothiofos in Chinju strain, respectively and 38 and 9 fold resistance to fenvalerate and furathiocarb in Seosang strain, respectively. Optimal mixture ratios of bifenthrin and prothiofos was selected against Seosang strain of DBM by leaf dipping method in laboratory and by field test. Maximum co-toxicity coefficient by dipping method was shown at a ratio of 1:50 mixture of bifenthrin 1EC and prothiofos 50EC in active ingredient(a.i.) by 243.2 and then suddenly decreased. The mixed formulation from the dipping test was examined at the cabbage field of Seosang, Hamyang. Although insecticides were reduced to half, control efficacy was similar to farmer's conventional method. In addition, the mixture combination was also more effective against Chinju DBM population which had very high level of resistance to several representative insecticides. Mixture methods of insecticides will be effective countermeasures to the resistance problem of pests.
