Measurement and Within-tree Distribution of Larval Entrance and Adult Emergence Holes of Japanese Pine Sawyer, Monochamus alternatus(Coleoptera: Cerambycidae) (솔수염하늘소의 유충침입공과 성충탈출공의 측정과 소나무 내 분포)

  • Larval entrance and adult emergence holes of Japanese pine sawyer (Monochamus alternatus), primary vector of pinewood nematode (Bursaphelenchus xylophilus), were measured in dead pine logs from 1999 to 2002. Their distributions within pine log were also analyzed. More numbers of entrance and emergence holes were distributed on crown than trunk part as 56.2 and 27.7 holes/m$^2$, respectively Higher proportions of entrance (27.5%) and emergence holes (22.4%) were distributed on the log with 8 to 10 cm diameter; the larger or the smaller logs had fewer holes. Surface area of entrance hole was 65.8 $\textrm{mm}^2$ and diameter of emergence holes was 7.0 mm in average. Average depth of pupal chamber was 24.8 mm from surface to the bottom of the chamber, and its volume ranged from 200 to 2.000 ㎣ Average distance between entrance and emergence holes on bark surface was 3.3 cm. Gallery length from the beginning of entrance hole to the end of emergence hole was 46.2 mm.

  • The Luciola lateralis larva took 5.3 days from climbing on the land to the pupal cocoon formation. It took 6.6 days for a larva to eventually transform to a pupa after building a pupal cocoon. The size of pupal cocoon was 10.1 mm in length, 4.7 mm in width and its wall thickness was 1.3 mm. The mean pupal period was 10.5 days. The adult stayed 6.8 days in the pupal cocoon before escaping the cocoon. The peak adult emergence appeared around 9 p.m., and decreased after 10 p.m. The optimal soil temperature for emergence was $23.4^{\circ}C$. The female adult of the natural population (Nat-type) lived shorter, laid fewer eggs, and the oviposition frequency was fewer than that of the Lab-type individuals. However, a few individuals from the natural population laid 200-400 eggs. The less number of oviposition in the natural population may be due to the fact that the female adults might lay eggs before the collection for the experiment.

Gnawing and Escaping Behaviors of Monochamus alternatus (Coleoptera: Cerambycidae) in a Confined Environment: Suggesting a Bioassay Method of Netting for Adult Escape Prevention (인위적 구속환경에서 솔수염하늘소의 쏠기와 탈출행동: 성충탈출 방지용 그물망의 생물검정법 제안)

  • The Japanese pine sawyer, Monochamus alternatus Hope, is a representative vector of the pine wood nematode, Bursaphelenchus xylophilus, which causes wilting symptoms in pine trees. A control method using a net has been introduced, which is an alternative method to the fumigation for the control of dead pine trees by pine wilt disease. This study was carried out to investigate the factors that induce gnawing and escaping behaviors of M. alternatus. The behaviors were examined after M. alternatus adult was placed in a confined space at different temperatures. M. alternatus adults could escape through mesh net torn by gnawing when they were confined in a space of 30 mm or less in diameter. The success rate of escape was high at 20 to $30^{\circ}C$, and no adults escaped at $15^{\circ}C$. The enticement of M. alternatus adults by food didn't affect the success rate of escape. In the case of not being confined in a narrow space, the escaping hole could not be formed because the gnawing was not concentrated on one part. M. alternatus moved its body in a narrow space using the tarsus of middle and hind legs, and made an escape hole by concentrically gnawing the obstacle on the front side with mandible, and showed a behavior of getting out while supporting the body by supporting the front legs. The present results will be able to use as an important basic information for evaluating the performance of mesh net which confines M. alternatus adults and suggested by alternative method to fumigation technology.

  • Japanese pine sawyer, Monochamus alternatus, is a primary vector of pinewood nematode, Bursaphelenchus xylophilus in Korea. Emergence characteristics of the adults were studied in Jinju, Korea using dead pine logs in which the larvae of the sawyer overwintered from 1999 to 2002. Emergence data showed that the adults began to emerge out of the logs in sunny place from May the 15th in average of the four years. Cumulative emergence ratio (CER) reached at 50% at mid-June, and the emergence terminated by early or late July depending on years. In shady place, however,50% CER was reached 17 days later than in sunny place. The 50% CER of males reached 1 to 4 days earlier than that of females depending on years. Of the adults emerged from the logs collected from February to April in 2001, 97.6% was univoltine, and the rest was biennial which emerged from May to July of the next year. Female and male adults emerged throughout 24 hours; 32.3% of the total adults emerged from 8 to 12 a.m. It took 68.0 seconds for the adults to escape from pine logs.

  • This study investigated the life history of the plum sawfly Monocellicampa pruni Wei, 1998 through field observations and laboratory experiments. M. pruni is a univoltine, complete metamorphosis species with five instars at the larval stage. Adults emerge from the soil around mid-March when Japanese plum trees are in their flowering period. The adults deposit one (rarely two) egg(s) under the epidermis layer of the calyx. After hatching, the larva soon burrows into the fruitlet, consumes the endocarp and remains inside the fruit within its four-time moulting transitions before reaching full growth. At maturity (in May), the larva leaves the infested fruitlet through an exit hole and burrows into the soil at a depth of 2 - 11 cm for spinning its cocoon and hibernates until the next spring as the larval form. Under laboratory conditions (T = 20℃; RH = 40 - 60%), the male lived for a slightly shorter period than the female, 6.03 ± 0.40 and 7.55 ± 0.45 days, respectively. A female could produce 30.29 ± 4.50 eggs in her lifespan. In the field, the duration of the egg was approximately 10 - 11 days and that of the larva was approximately 31 - 34 days.

Proteomic Study for Low Molecular Weight Peptides in the Mealworm Tenebrio molitor (갈색거저리 유래 저분자단백질체의 분석)

  • In this study, we examined low molecular weight peptides using proteomics in order to identify their original proteins, derive their peptides, and determine the functions of the proteins in Tenebrio molitor, the mealworm (larvae, pupae, or adult) from which the peptides were extracted. Fifty-four proteins were finally identified through an analysis of proteome to derive the analyzed peptides. The proteins that induced low molecular weight peptides were identified to be the most abundant in adults only, and the next highest were derived from a group containing both adults and larva. However, other groups, including pupa, were detected to have a lower frequency of peptides. As a result of orthologous classification of the detected proteins, the general function prediction was only investigated at the highest frequency among the examined proteins. Proteins related to chromatin structure and dynamics were detected by their higher frequency among functional classes. The next highest frequency was shown by proteins related to amino acid transport and metabolism and carbohydrate transport and metabolism. Therefore, it is assumed that proteins correlated with chromatin, amino acid, and carbohydrate metabolisms are easily induced into low molecular weight peptides, and that their peptides could play a role as bioactive substances.

Efficient Mass-rearing Method of Mythimna loreyi (Lepidoptera: Noctuidea) using Artificial Diets (인공사료를 이용한 뒷흰가는줄무늬방나방(Mythimna loreyi) (나비목: 밤나방과)의 효율적인 대량 사육 방법)

  • A lot of individuals of Mythimna loreyi have been attracted to the sex-pheromone traps of Spodoptera frugiperda during recent few years. However, there is no information about this pest. Thus, an efficient mass-rearing method of M. loreyi is demanded to study this pest. In this study, we compared the effects of artificial diets and rearing methods on its larval development and oviposition to suggest an efficient mass-rearing method of M. loreyi. Between S. frugipera and Agrotis ipsilon artificial diets, A. ipsilon diet showed more rapid larval development with higher survivorship, and decreased pupa weights and oviposition numbers compared to S. frugipera diet. Moreover, a grouping rearing caused more rapid larva development, decreased pupa weight and survivorship compared to an individual rearing. Therefore, for mass-rearing of M. loreyi, it is considered efficient to rear the newly emerged larvae in groups using A. ipsilon artificial diet and then rearing them individually after second or third larval stadium.

Anti-obesity Effect of Ethanol Extracts from Silkworm (Bombyx mori) Pupae Powder Fermented with Cordyceps militaris in the Primary Adipocytes and High Fat Diet-induced Obesity Model Mice (일차 지방세포와 고지방식이로 유발한 비만모델동물에서 Cordyceps militaris로 발효시킨 누에 (Bombyx mori) 가루 에탄올 추출물의 항비만 효과)

  • Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.
