Culture of Bovine Embryos Derivedfrom IVM/IVF into Blastocysts in Defined Simple Media with Different Concentrations of $\textrm{CO}_2$and $\textrm{O}_2$ ($\textrm{CO}_2$$\textrm{O}_2$의 농도가 소 체외주정란의 체외발육에 미치는 영향)

  • 소 체외수정란의 체외 발육에 $CO_2$$O_2$의 농도가 미치는 영향에 대하여 두가지 배양액과 서로 다른 Co-culture 체계를 이용하여 검토하였다. 체외 수정후 40~44시간에 회수간 2~8 세포기 수정란을 여러가지 gas조건하의 다른 배양조건에서 무작위로 옮겨 실험을 수행하였다. 5% $CO_2$$O_2$,10% $O_2$및 20% $O_2$조건에서 배양을 실시한 결과, 상실배가 이상 발육된 체외 발육성적은 각각 22.1% 15.0% 및 21.1%였다.(p<0.05). 한편 배양액에 따른 헤외 배양성적은 KOSM배양액(19.1%)의 경우가 Menezo's B2 배양액(13.7%)에서 배양한 경우보다 더 높은 성적을 얻었다.(p>0.05,Table 1). 2~8세포기 수정란을 5% $CO_2$또는 10% $CO_2$와 5%, 10% 및 20%의 $O_2$조건 하에서 체외 배양을 실시한 경우 각각 15% 와 8%의 체외 발육 성적을 얻었고(P>0.05), $O_2$조건의 경우 10% $O_2$(17%)와 20% $O_2$(20%)의 배양조건을 이 5% $O_2$(26%)보다 낮은 성적을 나타냈다. (P<0.05), (Table2). 체외 수정 후 얻은 2~8세포기 수정란을 단순 배양액 과 두개의 다른 공동 배양체계를 이용하여 5% $CO_2$와 5% 및 20% $O_2$의 배양 조건하에서 체외 배양을 실시한 결과 상실배와 배반포기 까지 발육된 체외 발육 성적은 배양액과 공동배양 체계에 따른 차이는 인정되지 않았다(p>0.05). 그러나 $O_2$의 농도는 소 체외 수정란의 체외 발육성적에 영향을 미치는 것으로 나타났다.(Table 3). 본실험은 $CO_2$$O_2$의 농도가 소 체외수정란의 체외 배양할때 배양체계에 따라 다른 영향을 미치는 것으로 나타났다.

Multiple Shoot Formation of Gentiana axillariflora Leveille by in Vitro Culture (큰용담의 기내증식에서 multiple shoot의 유기)

  • This study was aimed to proliferate Gentiana axillariflora Leveille which is one of the important medicinal and ornamental plants, by establishment of multiple shoot formation and embryogenesis through tissue culture technique. Callus was formed on MS (Murashige and Skoog) medium supplemented with 2,4-D, CPA, but not formed with BAP. The addition of 2,4-D 2 mg/ l into the medium was effective for callus formation and the rate of callus formation was about 90%. Somatic embryos were obtained on MS medium for two months. When callus was cultured on MS medium with combination treatment of 2,4-D 0.5 mg/ l and BAP 0.5 mg/ l, the number of embryo formed was better than that of other single or combination treatments and the total numbers of embryo a were 18.8 (number of total embryo/number of explants incubated = 753/40) at mean. Callus induction from stem and node explants was increased by addition of TDZ 2 mg/ l in the presence of 2,4-D 2 mg/ l, respectively. The best result about the differentiation of shoots was obtained on MS medium added BAP 2 mg/ l from node culture. Multiple shoots from shoot apex were induced on MS medium containing BAP 1 mg/ I and TDZ 1 mg/ l , BAP 2 mg/ l and TDZ 1 mg/ l. The number of multiple shoots per one explant was above seventy plants. It was the most effective regeneration system for rapid multiplication of Gentiana axillariflora Leveille.

Citric Acid Production Using Encapsulated Aspergillus niger (Aspergillus niger 고정화 캡슐을 이용한 구연산 생산 특성)

  • The encapsulatpd A. niger grew up inside the capsule and mycelia penetrated through the pore of the capsule membrane. The mycelia on the capsule wall became loose when the carbon source and oxygen were deficient in the medium. On the contrary, the production rate increased and mycelia made a lump tightly when the carbon source and oxygen were sufficient. Namely, number of proper capsule of unit volume in the medium was existed. The phenomenon which was swelled of capsule membrane in cultivation could prevented by adding CaCl2 into the medium. According to the time adding CaCl2 into the medium, the production rate of citric acid was influenced. In case of adding CaCl2 into the medium at 7th day cultivation, the production yield of citric acid was increased about 40 percent higher than that of adding CaCl2 initially. The production yield of citric acid using encapsulated A. niger of flask culture was influenced with oxygen supply. The production yield of citric acid ($\Delta$p/$\Delta$s) of the flask culture was increased 3.88 time by using T-flask instead of parafilm sealed flask. Therefore, the productivity and consumption rate concerning production which was taken carbon source were increased when oxygen supply was sufficient. The production of citric acid using encapsulated A. niger was increased average 30 percent higher than that of bead in between 6th and 13th day cultivation.

배양액이 마우스 핵이식 재구축배의 초기발생에 미치는 영향

  • 본 실험은 Piezo-미세조작기(PrimeTech Ltd., Japan)를 사용하여 마우스 핵이식 후 재구축배를 CZB와 KSOM 두가지 배양액을 사용하여 체외배양성적을 비교 검토하였다. MII의 미수정란은 성숙한 4~5주령 B6D2Fl에 hCG 주사 후 14시간째에 과적 방법을 통해 난관의 팽대부로 부터 회수하였고, metaphase II chromosome-spindle complex와 최소량의 세포질을 내경이 10$\mu\textrm{m}$인 피펫으로 흡입하여 탈핵하였다. 핵이식에 사용된 난구세포(8-l0$\mu\textrm{m}$)는 3시간동안 12% PVP 에처리 하여 piezo-미세조작기를 이용하여 세포질에 세포의 핵을 직접 미세주입 하였다. 핵이식 후 생존한 재구축배는 2시간동안 배양한 후 10mM SrC1$_2$와 5$\mu\textrm{g}$/$m\ell$의 cytochalasin B가 첨가된 $Ca^{2+}$-free CZB에서 6시간 활성화 처리하였다, 활성화 처리 후 위전핵이 관찰된 재구축란을 CZB 와 KSOM 배지에서 배양하면서 발달률을 비교하였고, 상실배 및 배반포배로 발달한 재구축배를 day 3 대리모에 이식하였다. 표 1에서 보는 바와 같이 재구축배의 2-cell로의 발달률에 있어서 KSOM이 CZB에 비하여 유의적으로 높게 나타났으며(P<0.05), 또한 4-cell과 상실배/배반 포배로의 발달률에 있어서도 KSOM이 CZB에 비하여 유의적으로 높은 발달률을 나타내었다(P<0.01). 또한 KSOM 배지에서 배양된 상실배/배반포배를 대리모에 이식한 경우에 11.5 d.p.c에 생존한 태아가 관찰되었다. 이상의 결과로 핵이식 재구축배의 활성화 처리 후의 발생에는 KSOM 배지가 CZB 배지에 비하여 유효함을 확인 할 수 있었다.그와 같은 배양 기술을 이용하여 외래유전자를 도입한 일련의 결과에 관하여 보고 하고자한다., 이것은 세포내 유전자가 transfection되지 않은 세포도 neo selection에서 선발된다는 것을 제시하고 있다. 따라서 체세포를 이용한 형질전환동물 생산을 위해서는 세포내 유전자 도입과 선발 과정에서 나타난 colony에 대하여 보다 엄격한 screen을 하는 것이 필요한 것으로 생각된다.로 우점하였다. 여름철 식물플랑크톤 대발생에 영향은 수온과 직산염이 중요하였으나, 부유물질 크게 기여하지 못하였다.애를 확인하고 지도 관점을 파악하는 것을 포함한다. 그러나 본 논문은 역사발생적 수학 학습-지도 원리의 실제적인 적용에 관하여는 기초적인 연구에 지나지 않기 때문에, 역사발생적 원리를 학교수학에 실제적으로 적용하기 위해서는 각각의 내용에 대한 철저한 역사적 분석을 바탕으로 하는 후속 연구가 필요하다./TEX>구성교육${\lrcorner}$이 조선총독부의 관리하에서 실행되었다는 것을, 당시의 사범학교를 중심으로 한 교육조직을 기술한 문헌에 의해 규명시켰다.nd of letter design which represents -natural objects and was popular at the time of Yukjo Dynasty, and there are some documents of that period left both in Japan and Korea. Effects of FSH and LH on Maturation of Bovine Preantral Follicle (소 Preantral Follicle 성숙에 미치는 FSH와 LH의 영향)

  • The present study was conducted to develop an in vitro culture system that would support bovine follicle growth from preantral to antral stage, oocyte maturation, fertilization, and embryonic development. Bovine preantral follicles (150$\pm$1.2 ${\mu}{\textrm}{m}$) surrounded by theca cell were isolated ezymatically and mechanically from ovarian cortical slides in Leibovitz L-l5 medium containing 1 mg/$m\ell$ collagenase and 0.2 mg/$m\ell$ DNase I and cultured for 25 days in the presence of different concentrations of bovine FSH and LH in $\alpha$MEM medium. The survival and growth rates of follicles cultured in the presence of FSH (10~150 ng/$m\ell$) were significantly higher than those of control group (P < 0.001), but no significant differences were observed in survival and growth rates of follicles between the LH treatment groups (1~125 ng/$m\ell$) and the control. The survival (40%) and growth (244 $\pm$ 0.5 $\mu\textrm{g}$) of follicles cultured with FSH (90 ng/$m\ell$) and LH (25 ng/$m\ell$) were higher than those of control (25%, 160 $\pm$1.0 $\mu\textrm{g}$). Finally, 50% percent of healthy antral follicles were obtained, and almost 60% of them has complete meiotic division with 1st polar body (18.1%) and 10.0% have developed to the cleaved embryo and blastocyst stage. These results suggest that bovine preantral follicle with intact theca cell can grow to the antral stage using these culture conditions, and that oocytes from in vitro-matured bovine preantral follicle may acquire meiotic competence and can undergo fertilization and development.

  • Cell lines of Ginkgo biloba were derived from different plant parts and from ten varieties spanning various geographic locations. They had various properties of growth and product formation. More than three flavonol glycosides were present in low concentration in callus and suspension cultures. Cell growth and biosynthesis of flavonol glycosides were found to be affected by medium composition. Culture conditions which influenced cell growth and product formation were also examined. Light stimulated the flavonol glycosides biosynthesis and ten times higher flavonol glycosides content was obtained as compared with the result without light.

  • To elucidate the optimal conditions of mycelial growth of Pholiota adiposa, the effect of a kinds of media, incubation temperature and shaking culture were investigated. Mushroom complete medium was selected as superior media for mycelial growth in this fungus. The optimal temperature for mycelial growth of this fungus shows at 25℃. Shaking culture could be obtain the mycelia in two or three times than the stationary culture.

Effect of EGF and IGF-I on in vitro Maturation of Porcine Oocytes and Development of Porcine IVM/IVF Embryos (EGF와 IGF-I의 첨가배양이 돼지 미성숙 난포란의 체외성숙과 배발달에 미치는 영향)

  • The present study was carried out to examine the effect of EGF and IGF-I in vitro maturation (IVM) of porcine oocytes and development of porcine IVM/IVF embryos. The results were summarized as follows : 1. The rates of nuclear maturation, penetrated oocytes, pronuclear formation, polyspermic oocytes and mean numbers of the penetrated sperm were not different in NCSU-23 maturation medium with 0, 1, 5 and 10 ng/ml EGF and IGF-I (P>0.05). 2. The rates of blastocyst formation at day 7 after in vitro fertilization in 0, 1, 5 and 10 ng/ml EGF groups were $11.2{\pm}1.5%$, $15.0{\pm}8.3%$, $16.8{\pm}2.8%$ and $21.4{\pm}2.0%$, also 0, 1, 5 and 10 ng/ml IGF-I groups were $11.2{\pm}1.5%$, $15.0{\pm}8.3%$, $16.8{\pm}2.8%$ and $21.4{\pm}2.0%$, respectively. In the total cells case, EGF groups were $22.8{\pm}3.7$, $25.7{\pm}5.5$, $26.0{\pm}4.2$ and $35.1{\pm}4.7$, also IGF-I groups were $21.5{\pm}3.7$, $25.2{\pm}2.8$, $26.2{\pm}2.9$ and $33.2{\pm}3.6$, respectively. Both 10 ng/ml EGF group and 10 ng/ml IGF-I group were significantly higher than those of other treatment groups (P<0.05). 3. The rates of blastocyst formation at day 7 in the NCSU23 culture medium of porcine IVF-produced embryos with 0, 1, 5, and 10 ng/ml EGF groups were $14.0{\pm}1.7%$, $16.2{\pm}1.4%$, $16.9{\pm}1.2%$ and $23.1{\pm}1.6%$, also 0, 1, 5, 10 ng/ml IGF-I groups were $13.6{\pm}1.7$, $15.7{\pm}4.5$, $16.0{\pm}0.2$ and $25.0{\pm}0.8$, respectively. And in the total cells case, EGF grups were $21.8{\pm}2.9$, $25.2{\pm}2.8$, $39.7{\pm}2.7$ and $46.2{\pm}3.6$, also IGF-I groups were $20.7{\pm}2.9$, $26.2{\pm}2.9$, $24.6{\pm}2.4$ and $46.1{\pm}3.5$, respectively. Both 10 ng/ml EGF group and 10 ng/ml IGF-I group were significantly higher than those of any other treatment groups (P<0.05). In conclusion, these results suggested that the addition of 10 ng/ml EGF and IGF-I were effective on the blastocyst formation and total cells of blastocysts.

Plant Regeneration via Secondary Somatic Embryogenesis and Acclimatization in Panax ginseng (장뇌삼의 2차 체세포배 발생을 통한 식물체 유도 및 순화)

  • This study was conducted to establish the optimal condition for plant regeneration and acclimatization from somatic embryos of Panax ginseng. Cotyledon segments of Panax ginseng produced primary and secondary somatic embryos when cultured on MS and WPM media supplemented with 7% sucrose. To induce plantlet conversion, cotyledonary somatic embryos were cultured on WPM solid medium with $GA_3$ at various concentrations (1~30 mg/L) for 4 weeks. Plantlets were transferred to 1/2 WPM solid medium with $GA_3$ at various concentrations (0~5 mg/L) and 0.5% activated charcoal for shoot and root elongations. Elongated plantlets further developed into well-developed leaf and root system on 1/3 SH medium with 0.5% activated charcoal under ventilation condition for 5 months. The highest survival rate to soil was 75% when plantlets were regenerated on 1/3 SH medium without sucrose under ventilation condition.

Liquid Culture of Basidiomycetes on Natural Media (천연물을 이용한 담자균의 균사체 배양)

  • This study was designed to develop functional drink from jujube extract through a simple submerged culture of three basidiomycetes species. The optimum Brix and pH of the jujube extract for culturing the Ganoderma lucidum appeared to be 5 Brix and pH 4. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Coriolus versicolor appeared to be 5 Brix and pH 5. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Phellinus igniarius appeared to be 3 Brix and pH 5. For the maximum mycelial production eighteen days of culture period was required for Phellinus igniarius. The antitumor activity of the polysaccharides extracted from the fermented drinks was demonstrated through the tumor cell line experiments. The $IC_{50}$ values of the jujube drinks fermented with Ganoderma lucidum and Phellinus igniarius against stomach cancer cell line appeared to be one fourth that of the jujube drink which was not fermented with basidiomycetes.
