Studies on Proper Medium for Somatic Embryogenesis in Suspension Culture of Rehmania glutinosa and Encapsulation of Somatic Embryos (지황의 현탁배양에서 체세포배 형성에 관여하는 요인분석과 체세포배의 Encapsulation)

  • This study was conducted to find the factors affecting somatic embryogenesis in suspension culture of Rehmania glutinosa and investigate the possibility of artificial seed production by encapsulation of somatic embryos. Linsmeier-Skoog medium was appeared as proper for somatic embryogenesis. Sucrose with $3{\sim}5%$ as carbon sources was good for somatic embryogenesis, and both ammonium and nitrate nitrogen were necesary for normal somatic embryo production. BA with NAA or kinetin with NAA were better than the use of cytokinin alone for both somatic embryogenesis and numbers of somatic embryos. $AgNO_3$ as protectant for vitrification of seedlings in vitro culture had no harmful effect on somatic embryos. Sphericity of encapsulated seeds was good at 3% gel of sodium alginate but germination was better at 2.5% sodium alginate level. Artificial seeds were germinated and developed normal shoots and roots under in vitro condition.

Effect of Abscisic Acid, Kinds and Concentrations of Osmoticum on Somatic Embryo Induction, Germination and Plantlet Regeneration in Larix kaempferi (ABA 및 삼투압제 종류 및 농도에 따른 낙엽송 (Larix kaempferi) 체세포배 유도, 발아 및 식물체 재분화 효과)

  • This study was conducted to examine effects of concentrations of abscisic acid (ABA) or /kinds of osmotica on induction of somatic embryos (SEs), germination and plantlet regeneration in Japanese larch (Larix kaempferi). In comparison of duration of culture, concentrations of ABA and osmoticum, the highest induction number (191/g tissue) of the SE was showed in $60{\mu}M$ ABA+0.2 M sucrose for 4 weeks culture. However, the lowest number (3.5~23.5) of SEs was induced from $4{\mu}M$ ABA+0.1 M sucrose, regardless of culture duration for SEs induction. In comparison of germination efficiency of SEs, the highest induction frequencies of cotyledon (90.9%), hypocotyl (95.8%) and root (96.5%), respectively, were obtained from the SEs that cultured from the treatment of $60{\mu}M$ ABA+0.2 M sucrose with 5 weeks culture. In contrast, the lowest germination response was showed in SEs that induced from the treatment of $4{\mu}M$ ABA+0.1 M sucrose. In comparison of effect of different kinds/concentrations of osmotica for germination and plantlet regeneration, the best response was obtained from the treatment of 0.2 M sucrose with induction of cotyledon (98.3%), hypocotyl (78.4%), root (57.5%) and plantlet regeneration (54.8%), respectively.

In Vitro Propagation of Persimmon(Diospyros kaki) by Embryo Culture (감나무의 배 배양법에 의한 기내 증식)

  • The embryos(6-8mm) isolated from seeds of Diospyros kaki were cultured on Murashlge-Skoog(MS), Woody Plant Medium(WPM), Campbell Durzen(CD), Lictvay's Medium(LM), Kao-Michaluk(KM), Nitsch, White, Heller, Wolter-Skoog(WS) media. The results showed that MS and WPM media were most suitable to the development of embryos into plantlets with length of $5.4{\pm}1.2 cm$ and 5-6 leaves. However, when LM and KM media were used, the addition of 1 to $2{\mu} moles/\ell GA_3$ was required for the germination of the embryos. Superoxide dismutase (SOD) activities, one of the changing factors in leaves according to physiological status displayed to be exceptionally significant in the leaves of plantlets germinated from seeds in potting sand soil contrary to those of cultured embryos specially around germination period.

Production of Artificial Seeds by Alginate-encapsulation of Rice Somatic Embryos (벼의 수화겔 인공종자 생산)

  • Somatic embryos derived from cell suspension cultures of rice were singly alginate-encapsulated to be used as artificial seeds. When placed on half strength MS solid medium,73% of the encapsulated somatic embryos were capable of germination Encapsulation per se did not affect the germination frequency of embryos. When incubated by wrapping with moistured non-sterile filter paper, 60% of the encapsulated somatic embryos germinated. However encapsulated zygotic embryos without endosperm showed a high germination frequency regardless of the sterility of the incubation conditions. The results suggest that a greater susceptibility of somatic embryos to contaminants is attributed to lower germination frequency of encapsulated somatic embryos in non-sterile conditions.

Optimum Chilling Terms for Germination of the Dehisced Ginseng(Panax ginseng C. A. Meyer) Seed (개갑 인삼종자의 발아 적정 저온감응기간)

  • Experiments were conducted to study the optimum chilling period for breaking physiological dormancy of dehisced ginseng(Panax ginseng C. A. Meyer) seed. Embryo of ginseng seed is too small to be noticed with naked eyed on harvesting time. Embryo grew to half size of endosperm after seeds were stratified for 3 months. It takes 6 months for this embryo to reach the size enough for germination. And it grew faster indoors than outdoors. Dehisced ginseng seed with full-size embryo did not germinate at room temperature and required chilling treatment for 75days in outdoors and 90 days in cold chamber. While seed receiving sufficient chilling treatment were left to be in room temperature, the chilling effect decreased remarkably.

키토산 처리 콩나물의 성장특성

  • 키토산처리가 콩나물의 성장특성에 미치는 영향을 조사하기 위하여 콩에 0.05% 키토산(493kDa, 금호화성, 한국)을 함유하는 0.05% 초산 용액 4배량을 가하여 8시간 침지한 후 2$0^{\circ}C$에서 2시간 간격으로 5일간 주수하면서 발아율, 수율, 배축과 자엽의 중량변화, 수분함량, 배축의 길이와 두께 등을 조사하였다. 발아율은 키토산처리구(88-94%)가 대조구(69-85%) 보다 높았다. 5일간 재배한 콩나물의 생체중량은 처리구(70.28 g/10 g-pot)가 대조구(63.86 g/10 g-pot) 보다 18% 높았다. 재배중의 수분함량은 대조구(59.53-73.33%) 보다 처리구(64.66-84.45%)가 5.13-11.12%가 높았다. 재배중의 자엽의 중량은 대조구(25.06-27.21 g/10 g-pot)가 처리구(23.42-25.55 g/10 g-pot) 보다 높았으며 배축의 중량은 대조구(9.27-36.25 g/10 g-pot)보다 처리구(15.19-44.73 g/10 g-pot)가 높았다. 재배중 배축의 길이와 두께는 대조구(6.51-19.87 cm)보다 처리구(7.67-22.38 cm)가 길었으며, 두께는 대조구(2.08-2.12 mm)가 처리구(203-2.08 mm)보다 약간 두꺼웠다. 이상의 결과 키토산처리는 콩나물의 발아율을 높이며, 콩나물의 수분함량이 높고, 배축의 길이가 기나 두께는 다소 가는 특성을 나타내었다.

Cotyledon Structure and Germinability of Somatic Embryos Formed from Inflorescence Explants of Cnidium officinale M. (천궁 (Cnidium officinale M.)의 화기절편으로부터 형성된 체세포배의 자엽구조와 발아)

  • Calli were induced from the explants of infloresence, petiole and leaf blade of Cnidium officinale on MS medium with 2.4-D, while embryogenic callus was induced only from inflorescence explants. Somatic embryos of 78 per explant were formed during subculture of the explants on medium without 2.4-D after culture on medium with 2 mg/L 2.4-D. Cotyledonary variation, cup-shaped cotyledon of 49% and other abnormal cotyledons of 13.5 % was observed on the somatic embryos. However this variation could be overcomed by the addition of activated charcoal onto culture medium. Somatic embryos at cotyledonary stage germinated on MS basal medium but the germination rate was very poor, blow 50 %. Somatic embryos on the medium with activated charcoal showed improved germinaton.

Effects of Heat Treatment and Selected Medicinal Plant Extracts on GABA Content after Germination (열처리와 한약재 추출물이 발아현미의 GABA 함량에 미치는 영향)

  • The purposes of this work were to study germination conditions on $\gamma$-aminobutyric acid (GABA) contents and to develop a simple and effective technique for the production of germinated brown rice with an enhanced GABA levels. The samples were subjected to heat treatments at 40 and $45^{\circ}C$ before the soaking step. Moreover, four medicinal plant extracts including Schizandra chinensis, Dimocarpus longan Lour, Angelica dahurica, and Gastrodia elata Blume were used as soaking and germinating media. GABA levels were enhanced in the germinated brown rice compared to the non-germinated brown rice and the highest GABA contents were observed in heat treatment at $35^{\circ}C$. The brown rice soaked in Gastrodia elata Blume extract showed the highest GABA contents compared to the control sample. These results demonstrate that the GABA levels during germination could be significantly enhanced by the heat treatment and the treatment of medicinal plant extracts in the soaking step.

Germination and Antioxidant Activity of Korean Oak Pollen Treated with Medicinal Mushrooms (약용버섯을 이용한 도토리화분의 세포 발아 및 항산화 활성)

  • In this study, we measured antioxidant activity as DPPH radical scavenging and the total polyphenol content of pulverized and lyophilized oak pollens inoculated with fungi to confirm the husk removal effect. The total polyphenol content of oak pollen was highest in lyophilized pollen medium inoculated with Armillaria mellea, and was lowest in pollen inoculated with Lentinula edodes. Total polyphenol content of the lyophilized pollen was higher than that of the refined pollen and the pulverized pollen in oak pollen germinated with A. mellea. The total polyphenol content of the lyophilized oak pollen germinated with A. mellea was 1.4-fold higher than that extracted with water. Measurement of antioxidant activity using the DPPH (2, 2 diphenyl-1-picrylhydrazyl) free radical scavenging method showed that the lyophilized oak pollen germinated with A. mellea had the highest and that germinated with L. edodes was lowest in antioxidant activities. The lyophilized oak pollen germinated with A. mellea was 2 to 4 times higher than that extracted with water in the antioxidant activity of DPPH free radical scavenging. Many germinated cells were formed around pore of acorn pollen inoculated with L. edodes, while those were formed at the end of hyphae derived from oak pollen inoculated with A. mellea.

Effect of Plant Growth Regulators on Somatic Embryogenesis from Cotyledon of Herbaceous Peony (Paeonia lactiflora Pall.) (芍藥(Paeonia lactiflora Pall.)의 子葉組織 培養시 식물생장조절제가 體細胞胚發생에 미치는 影響)

  • This experiments were carried out to determine the optimum concentrations of plant growth regulators for the direct embryogenesis from the cotyledon culture of Paeonia lactiflora Pall. Zygotic embryos rescued from true seeds were developed abnormally in the medium containing ABA. But somatic embryogenesis from cotyledons of abnormal seedlings was induced more effectively. Also the somatic embryogenesis from cotyledons was promoted in the medium containing ABA. The frequency of embryogenesis was maximum(59.9%) from the cotyledons cultured on MS medium containing 0.5 mg/L ABA on which the frequency of somatic embryos with two cotyledons was 22.6%.
