Dormancy and Germination Characteristics of Alpine Modest Primrose (Primula modesta var. hannasanensis T.Yamaz.) Seeds (설앵초의 종자 휴면과 발아특성)

  • This research was performed to develop mass propagation method of Alpine modest primrose (Primula modesta var. hannasanensis T.Yamaz.) for improving the conservation and utilization of the species. Seeds were collected on August 2011 in Jeju-city and well-selected seeds were dry-stored at $4{\pm}1.0^{\circ}C$. Seed size ranged $0.44{\pm}0.07{\times}0.61{\pm}0.04mm$, and weight of 1,000 seeds was $51.78{\pm}0.021mg$. Thus the seed was classified as 'dwarf seeds'. As the result of dormancy characteristics, moisture content of freshly matured seeds increased rapidly by water-soaking treatment and seeds did not germinate at $20^{\circ}C$ for 4 weeks under the light condition. Therefore, seeds were estimated to have physiological dormancy. Germination conditions of dry-stored seeds were found to be $20^{\circ}C$ and light condition, and seeds did not germinated under dark condition regardless of all temperature regimes. Therefore, Alpine modest primrose seed was considered as photoblastic type. Percent germination (PG) and germination energy (GE) were greatly improved by soaking the seeds in $GA_3$ and kinetin for 24 hours. Especially, 200~500 mg/L $GA_3$ treatment resulted in the highest PG (95.5%) and GE (98.3%). So, chemical treatment such as $GA_3$ was thought to be a useful method for raising seedling uniformly.

Effect of Water Table Depth in Different Soil Texture on Quality of Barley and Wheat Grain (토성별 지하수위가 밀, 보리의 품질에 미치는 영향)

  • This experiment was performed to characterize the optimum water table level for the grain quality, seed germination and diastic power of barley(var. Olbori) and wheat(var. Grumil). Olbori and Grumil grew in the 550 liter plastic pot that filled with silt loam or sandy loam. During the whole growth period, the underground water level adjusted to be 20, 30, 40, 50 and 70cm. Filled grain ratio and specific gravity were not affected by soil texture and water table. Low level of water table caused the increase of 1,000 grain weight in wheat and barley, but soil texture didn't. Crude protein content tended to be high as the water table level was high, especially in wheat. Change in crude protein content was affected by underground water level more than soil texture. And the affection was slightly higher in sandy loam than silt loam, but the difference was small. The higher level of water table led to the lower crude lipid content in barley and wheat grain. Crude lipid content of both wheat and barley grain grown in sandy loam was higher than those grown in silt loam. As the water table level down, the ash content of barley and wheat grain tend to increase, especially in sandy loam. Wheat flour yield was not affected by soil texture. It was about 65% at 20cm of water level and above 67% at 40cm water level. The seed germination of wheat and barley was more than 95% when the seeds were placed at 2$0^{\circ}C$ for three days. Regardless of soil texture, the lowest germination was seen at 20cm of water table level. And the seed germination rate increased as the underground water level became low. Above 89% of barley grains were germinated within 48 hours except 20cm level of water table in sandy loam. Diastic power of germinated barley was the lowest at 20cm of water table level, and it was almost unchanged below 30cm of water table level. And also it was not affected by soil texture.

Studies on Germination Characteristics and Differential Responses to Herbicides in Setaria Spp. (강아지풀 종류별 종자발아특성과 제초반응 차이)

  • To supply seeds with a good quality as plant materials for herbicide screening and to know whether the germination characteristics could be associated with a differential response to herbicides, germination characteristics and differential responses to herbicides were investigated with 3 species of a genus Setaria ; Yellow foxtail(Setaria glauca P. Beauv, SETGL), Giant foxtail(Setaria faberi Hetrm, SETFA), and Green foxtail(Setaria viridis P. Beauv, SETVI). Degree of dormancy was high in the order of SETGL, SETVI and SETFA. The dormancy of SETGL seed was relatively well removed by room temperature and drying storage, but SETFA and SETVI by low temperature and wetting storage(stratification). For breaking dormancy of SETGL, SETVI and SETFA, it was necessary for being kept under the above storage conditions for at least 2, 4 and 4-5 months, respectively. When the dormancy-breaked seeds were transfered to low temperature($4^{\circ}C$) and drying condition, SETGL showed germination rate of 96% even after 2 month storage. However, SETVI and SETFA showed a decreased germination of 54% and 69%, respectively, with a decreased velocity of germination, indicating that secondary dormancy might be induced. On the other hand, a significant change in germination rate was not observed as the seeds were transfered to room temperature($25^{\circ}C$) and drying condition. The germinability of SETGL seed began to decline from 6th year after storage in room temperature and drying condition. All of 3 species showed relatively high germination rate at alternating temperature of $30^{\circ}C$/$20^{\circ}C$(14hr/10hr) and their germination were not increased by light. All of 3 species exhibited similar responses to cycloxydim, sethoxydim and primisulfuron in greenhouse experiment. In contrast, SETVI and SETFA were relatively susceptible to fenoxapropethyl, SETFA to fluazifop-butyl, SETGL and SETFA to clorimuron-ethyl, and SETGL to EK-2612. The difference in herbicidal response among 3 species was the highest in the treatment of EK-2612. These results suggest that there is no a consistent tendency in responses of 3 species to herbicides which have the same target site. And the relationship between germination characteristics and differential responses to herbicides was not found.

Effect of Harvesting Time on the Quality of Malting Barley (맥주맥(麥酒麥)의 수확시기(收穫時期)가 원맥품질(原麥品質)에 미치는 영향(影響))

  • In order to determine the optimum harvesting time based on grain filling and physico-chemical qualities for malting, three cultivars were harvested at every five days from 30th day after heading date at two experimental sites in the southern part of Korea. Starch accumulation and kernel weight increment were remarkable until 45th day after heading, but negligible after that. Content of ash, crude protein and polyphenolics and ${\alpha}-amylase$ activity decreased with maturation of grains and reached to the lowest value at 45th day after heading date. Germinative energy and capacity were good enough for malting from 40th day after heading date. The optimum harvesting time were estimated at 45th day after heading date in the increment of starch accumulation and kernel weight. At this time matured and immatured kernels were in the ratio 93 : 7. However, it is estimated that early harvesting time was at 40th day after heading date.

Effect of Phosphorous Acid on Control of Phytophthora Blight of Red Pepper (고추 역병에 대한 아인산 (phosphorous acid)의 방제 효과)

  • Control effects of phosphorous acid were investigated on Phytophthora blight of red pepper plants in greenhouse and field. In vitro test, the mycelial growth of Phytophthora capsici was inhibited by the phosphorous acid more than 97% at 1,000 ${\mu}g$ and 10,000 ${\mu}g$ a.i./mL in the liquid and solid culture, respectively. At concentration of 10 ${\mu}g$ a.i./mL of phosphorous acid, in the liquid culture the mycelial growth of P. capsici was inhibited 46.2%, however inhibited only 4.9% on the soild culture. Zoosporangial formation was also inhibited 89.1% by phosphorous acid at 100 ${\mu}g$ a.i./mL. Phosphorous acid affected more zoosporangial formation of P. capsici than its mycelial growth. At the concentrations of 10,000, 1,000 and 100 ${\mu}g$ a.i./mL of phosphorous acid, germination of zoosporangia was inhibited 100, 84.3 and 44.2%, respectively. Mycelial growth and zoosporangial formation of P. capsici were little affected at the concentration of 10 ${\mu}g$ a.i./mL of phosphorous acid. Cermination of zoosporangia was also little affected at this concentration, however growth of the germ tubes was inhibited and the abnormal mycelial growth was observed. Phosphorous acid suppressed the incidence of Phytophthora blight of red pepper plants up to $77.0{\sim}62.0%$, in greenhouse. Phosphorous acid suppressed the incidence of Phytophthora blight of red pepper plants up to 54.0% at the conventional culture in field. Treatments of phosphorous acid increased up to 113% in height, 135% in number of fruit, and 129% in weight of fruit.

Antifungal Activity of Anemarrhena asphodeloides, Coptis japonica and Phellodendron amurense Extracts against Phytophthora Blight (지모, 일황련 및 황백나무 추출액의 항균활성)

  • Antifungal activities of the crude extracts of Anemarrhena asphodeloides. Coptis japonica and Phellodendron amurense were tested against Phvtophthora capsici. and the control effect on red-pepper phytophthora hlight and phytotoxicities of red-pepper were investigated. The results were summarized as follows; Mycelial growth and zoosporangial germination of the red-peppcr phytophthora hlight organism P. capsici were inhihited hy thc crude extracts of plant materials. Methanol extracts or plant materials had hctter antifungal activity than water extracts at hoth a room temperature and a hoiling condition. Antifungal activities of three crude extracts were gradually decreased with prolonged storage period. Red-pepper phytophthora hlight was effectively controlled hy the crude extracts of three plant materials. Of these. the crude extract of C. japonica was marvelously effective. Phytotoxic symptom to red-pepper seedling showed hy water cultural method hut not by pot test. Seed germination and radicle growth of red-pepper were inhihited hy the crude extracts of three plant materials. Phytotoxic symptoms in the leaves and fruits of red-pepper were not ohserved with exogenous foliage application of the three crude extracts.

Growth Characteristics and Adaptability of Introduced Achyranthes bidentata Blume in Korea (도입 우슬의 생육특성 및 국내 적응성)

  • Achyranthes bidentata was recorded to new medicinal crop in a revised 8th edition of the Korean pharmacopoeia. Therefore, A. bidentata began to use for same purpose with Achyranthes japonica which was cultivated since old times in Korea. This study was conducted to investigate the adaptability of A. bidentata in Korea. The germination rate and speed of A. bidentata seeds were higher than those of A. japonica in $15^{\circ}C\;and\;25^{\circ}C$. The growth of A. bidentata was greater than that of A. japonica until the 60 days after transplanting, but growth after that was the opposite. There were remarkable differences between two species in growth characteristics such as flowering date, leaf fall date, plant height, stem color and no. of branches. The flowering and leaf fall date of A. bidentata were earlier than A. japonica by July 7 and September 26. The plant height and number of branches of A.japonica were longer and more than A. bidentata, but the cluster length and no. of flowers per cluster of that were shorter and fewer than this. Number of supporting roots of A. bidentata and A. japonica was 10.7 and 14.6 per plant, respectively. The average yield was not different between two species. The yield of A. bidentata was 166 kg/l0 a in Suwon and 309 kg/10 a in Naju of Korea. The suitable cultivation region of A. bidentata was judged to southern area of Korea.

Dry Matter Distribution during Seedling Development in Soybean (콩의 유묘기간 중 건물중의 분배)

  • To obtain the basic information about the translocation of seed reserves of soybean[Glycine max(L.) Merrill] cultivars, 'Hill', 'Paldalkong' and 'Jangyeobkong' , the dry matter changes of emerging organs during the germination and seedling development were observed at the research farm of College of Natural Resources, Korea University, on May 26, June 5 and June 14. Mean fresh weight, dry weight and moisture content of three soybean cultivars were increased until the V2 stage in all seedling parts except cotyledons. Cotyledon dry weight was continuously decreased and the decreased amount at the V2 stage was about 80% of the cotyledon dry weight at the germination stage. Structural component of cotyledons was 22.2% of the cotyledon dry weight at the germination stage. Metabolic components of cotyledons were markedly decreased until V2 stage, and about 91% of cotyledon metabolic components at germination stage was utilized. However, those of whole seedling were increased after the V1 stage. Therefore, it appeared that role of cotyledons as nutrient supplier for germination and seedling growth was important until the V1 stage especially.

Comparison of Seed Germinating Vigor, Early Germination Characteristics, Germination Speed and Germination Peak Time in New Varieties of The Third Generation of Creeping Bentgrass Under Different Growing Conditions (생육환경에 따른 제3세대 크리핑 벤트그래스 신품종의 종자 발아력, 초기 발아 특성, 발아세 및 발아 피크 기간 비교)

  • Research was initiated to investigate germination characteristics of creeping bentgrass (CB, Agrostis palustris Huds.). Seven varieties of CB were evaluated with different experiments. An alternative environment condition requiring for a CB germination test by International Seed Testing Association (ISTA) was applied in the Experiment I, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$(ISTA conditions). Experiment II was conducted under a room temperature condition of 5 to $25^{\circ}C$(natural conditions). In each experiment, data such as seed germinating vigor, early germination characteristics, germination speed and germination peak time were measured. Significant differences were observed in seed germinating vigor, early germination characteristics, germination speed and germination peak time. Seed germinating vigor was variable with different environments and varieties. It was 61.50 to 98.25% under ISTA conditions and 55.00 to 98.50% under natural conditions. There were considerable variations in early germination characteristics among CB varieties according to different conditions. Early germination characteristics indicated that all varieties were 1 to 4 days faster in germination under ISTA conditions, when compared with natural conditions. The germination speed, measured as days to seed germination of 70% and 90%, was much faster with Penn A-1, Penn A-4 and Penncross under ISTA conditions. But it was even faster with L-93 and Penncross when grown under natural conditions. Differences were also observed in germination peak time with varieties and growing conditions. It was 0.57 to 2.86 days under ISTA conditions and 0.74 to 1.74 days under natural conditions. Regardless of the environment conditions, the shortest variety was L-93 and the longest one T-1. Considering germinating vigor, early germination characteristics, germination speed and germination peak time, Penn A-1, Penn A-4 and Penncross were regarded as excellent varieties under ISTA conditions in terms of early establishment characteristics, while L-93, Penn A-1 and Penncross under natural conditions. These results suggest that an intensive germination test be needed prior to planting, for the early germinating vigor, germination speed and germination peak time. Also, a proper variety selection and comprehensive site analysis for the growing environmental conditions should be done before golf course construction.

Effects of Temperature, Seed Water Content and Osmoconditioning on Germination and Seedling Elongation of Soybeans (온도.종자수분함량 및 삼투처리가 콩의 발아 및 묘신장에 미치는 영향)

  • Germination and seedling elongation of soybeans [cult. ‘Williams’] were measured at two temperatures (15 and 3$0^{\circ}C$), three seed water contents (30, 50, and 70%), and with and without polyethylene glycol-8000 (PEG) treatments. A split-plot design was used with four replications. Observations were made from two hours to eight days for each treatment combination. Seeds were soaked with 30% PEG solution with 0.2% thiram at 15 C for six days, rinsed with deionized water and dried at 25.5$^{\circ}C$ for three days. Ten treated or untreated seeds were placed on Whatman No. I (9 cm) filter paper in plastic petridishes to which sufficient deionized water was added to adjust seed moisture content to the desired level. The dishes were then sealed with parafilm and placed in a continuous programmed temperature chamber under dark condition. Seedling growth did not occur at either temperature when moisture content was 50 percent or less. Osmo-conditioning with PEG showed positive effects on seedling moisture uptake and seedling growth at 15$^{\circ}C$ but little effect at 3$0^{\circ}C$. Seedling moisture content increased rapidly early in the germination period and reached 60% at 15 C for 56 hours and at 3$0^{\circ}C$ for 28 hours. Seedling growth started when seed moisture reached a critical point of 60% at 15$^{\circ}C$, however, growth started after 20 hours of germination at 3$0^{\circ}C$. Seedlings of soybeans elongated more than four times faster at 3$0^{\circ}C$ than at 15$^{\circ}C$. Water uptake during germination was characterized by two phases in this experiment.
