Rediscovery of haploid breeding in the genomics era (유전체 시대에 반수체 육종의 재발견)

  • Advances in DNA sequencing technologies have contributed to revolutionary understanding of many fundamental biological processes. With unprecedented cost-effective and high-throughput sequencing, a single laboratory can afford to de novo sequence the whole genome for species of interest. In addition, population genetic studies have been remarkably accelerated by numerous molecular markers identified from unbiased genome-wide sequences of population samples. As sequencing technologies have evolved very rapidly, acquiring appropriate individual plants or populations is a major bottleneck in plant research considering the complex nature of plant genome, such as heterozygosity, repetitiveness, and polyploidy. This challenge could be overcome by the old but effective method known as haploid induction. Haploid plants containing half of their sporophytic chromosomes can be rapidly generated mainly by culturing gametophytic cells such as ovules or pollens. Subsequent chromosome doubling in haploid plants can generate stable doubled haploid (DH) with perfect homozygosity. Here, classical methodology to generate and identify haploid plants or DH are summarized. In addition, haploid induction by epigenetic regulation of centromeric histone is explained. Furthermore, the utilization of haploid plant in the genomics era is discussed in the aspect of genome sequencing project and population genetic studies.

Evaluation of pH and Compressive Strength Development of Alpha-Calcium Sulfate Hemihydrate-based Binder (알파형 반수석고 기반 결합재의 pH 및 강도발현 평가)

  • This study examined the compressive strength development and pH values of alpha-calcium sulfate hemihydrate(${\alpha}-CH$)-based binders developed for vegetation concrete with neutral pH between 6~7. Considering cost down and strength enhancement of the prepared binders, the ${\alpha}-CH$ was partially replaced by ground granulated blast furnace slag(GGBS), fly ash(FA), or ordinary Portland cement(OPC) by 25% and 50%. The compressive strength of mortars using 100% ${\alpha}-CH$ was 50% lower than that of 100% OPC mortars. With the increase of the replacement level of GGBS or FA, the compressive strength of ${\alpha}-CH$-based mortars tended to decrease, whereas the pH values were maintained to be 6.5~7.5. The main hydration products of ${\alpha}-CH$-based binders with GGBS or FA were a gypsum($CaSO_4$), whereas portlandite($Ca(OH)_2$) was not observed in such binders. Meanwhile, the pH values of ${\alpha}-CH$-based binders with OPC exceeded 11.5 due to the formation of $Ca(OH)_2$ phase as a hydration product. From the thermogravimetric analysis, the amount of $Ca(OH)_2$ in ${\alpha}-CH$-based binders with OPC was evaluated to be approximately 10% of the cement content.

  • This research was carried out to investigate the history and development process of the Bansu(泮水) which appeared uniquely at the Chinese 'temple-school(廟學)', Confucian schools as the common and necessary facility through the literature reviews and field surveys. The earliest record about the Bansu was shown in "The Book of Odes(詩經)". Originally Bansu was the water system which flowed near the 'Banpalace(泮宮)', but, in the following time, Bansu has become the symbol of the school which was built by the feudal lords and a very important element in the temple-school landscape. Temple-schools were started at Song(宋) Dynasty and at that time there was no certain form of Bansu, just the natural water course near the structure. Until Ming Dynasty, the "Picture of the Feudal lords' Ban-su" in the book "Samjedohoe(三才圖會)" that compiled by Wang-Xi(王圻), the form of Bansu was appeared to be transformed as the half-round pond. And the half-round pond as called Banji(泮池) of today's form was all rebuilt after Ming Dynasty. The half-moon pond appeared at the private houses, shrines, temples and villages were influenced by the book. From this research we can get the conclusions that Banji seemed one of the sacred one and used in the space arrangement formally for the various functions. This research has found the cultures of half-moon ponds which were used uniquely in China.

Effect of the Fatty Acid Synthase Gene for Beef Quantity Traits in Hanwoo Breeding Stock (한우 Fatty Acid Synthase (FASN) 유전자 반수체형의 후대검정우 육량 및 육질에 미치는 영향)

  • A previous study has shown that the g.17924G>A polymorphism of fatty acid synthase (FASN) is associated with unsaturated fatty acid composition in the Hanwoo beef, hence this study was conducted to evaluate the effect of single nucleotide polymorphisms (SNPs) within FASN gene on the selection phenotypes of Hanwoo breeding stock. A total of 925 progeny test steers were used to genotype g.11280G>A, g.13125T>C, and g.17924G>A polymorphisms and significant associations were found among g.11280G>A, g.17924G>A, and carcass traits, such as carcass weight, backfat thickness, and beef quantity index. No significant association was found between g.13125T>C and carcass traits. Although the degree of linkage disequilibrium (LD) was not strong among g.11280G>A, g.13125T>C, and g.17924G>A in the LD analysis, four major haplotype classes were formed with the genotypic information within the FASN gene; the frequencies of the halpotypeswere -GCG-[0.378], -ATG-[0.301], -GTA-[0.191], and -ACG-[0.063], respectively. Phenotypic association was performed among these haploptypes, and the haplotype 2 (-ATG-)was significantly associated with higher carcass weight when compared to the other haplotypes, i.e. haplotype 1 (-GCG-) and haplotype 3 (-GTA-). A copy number of the FASN haplotype 3 (-GTA-) had also a significant association with carcass weight of subjects. In conclusion, it was observed that two polymorphisms (g.11280G>A and g.17924G>A) and their haplotypes within the FASN gene are consistently associated with carcass traits. Therefore, it is desirable to use the FASN polymorphisms for pre-selection program as genetic marker with improved carcass yield and beef quality of the Hanwoo sire at the Hanwoo Improvement Center as well as for commercial Hanwoo producers, the FASN genotypic information can be used for a part of selecting Hanwoo dam for superior calf production.

Characteristics of Haploid Plants Derived from Interspecific Cross between Cytoplasmic Male-sterile Tobacco $F_1$(Nicotiana tabacum) and Nicotiana africana (연초(Nicotiana tabacum L.) 새포질 웅성불임 $F_1$과 Nicotiana africana의 종간 교배에 의한 반수체 식물의 특성)

  • This study was conducted to investigate the utility and agronomic characteristics and use of cytoplasmic. male-sterile (CMS) haploid plants derived from interspecific cross between (male-sterile NC82$\times$burley21) F$_1$ plant (Nicotiana tabacum L.) and Nicotiana africana. Abundant seeds of high germinability were obtained when Nicotiana tabacum (cytoplasmic male-sterile F$_1$ plants) is pollinated by Nicotiana africana. Most of seedlings died at the cotyledonary stage. The remaining seedlings are viable F$_1$ hybrids or maternal haploids that can be easily distinguished. Number of interspecific Fl hybrids and matermal haploids per capsule obtained from the interspecific cross between cytoplasmic male-sterile tobacco F$_1$ plants and N.africana yielded 2.2 and 0.5 plants, respectively. Out of 149 CMS haploid plants obtained from interspecific cross, 102 plants showed green type while the others were yellow type for leaf and stem. This results agreed with the genetic ratios expected among haploid plants from the F$_1$ hybrids heterozygous for two recessive genes of yellow color of burley tobacco plant. Out of 83 CMS haploid plants inoculated with TMV, 48 plants showed resistant, while the others was susceptible. It agreed with the expected genetic ratios for a single dominant gene of TMV resistance. CMS haploid plant will be useful as a source material for breeding of CMS doubled haploid lines

  • Induced androgenesis, a form of artificial parthenogenesis is an important tool for the generation and use of genetically isogenic or clonal fish strain. An optimized protocol for the genetic inactivation of mud loach (Misgurnus mizolepis) oocytes (i.e. production of androgenetic haploid) was developed using UV-irradiation. Various dose levels of UV significantly affected the fertilizing capacity of the eggs, hatchability of embryos and incidence of haploidy. Based on the extensive examinations of treatment conditions on embryo viability and haploid incidence, the optimum dose level of UV irradiation was turned out to be $10,800ergs/mm^2$ with 56.9% of hatching success and 94.6% of haploidy. The overall yield of putative androgen under optimized treatment condition was more than 50% out of total eggs inseminated. The success of androgenetic reproduction of haploid genome was verified by flow cytometry and PCR amplification of transgene that is exclusive to either one of parental sexes. However, a small portion $(8\sim11%)$ of presumed androgenetic haploid larvae was proven to contain residual DNA fragment(s) from maternal parent.

  • Polyclads (Stylochus ijimai and Pseudostylochus obscurus) which damage oyster were treated with calcium hypochlorite to determine the TLm concentration. In this experiment the TLm and the extermination density as well as the growth of the oyster after the treatment were checked with following results: The higher the temperature and the longer the period of the treatment is, the lower the TLm is. The greater the concentration of sea water is, the higher TLm is because of the weakened action of calcium hypochlorite. When the oyster is treated by calcium hypochlorite at the oyster farm, its extermination concentration is 2.4-7.1 times higher than that of TLm in fresh water. The growth rate was not affected after the treatment.

  • When $Al_2(SO_4)_3$ as an accelerator was added to $\beta$-hemihydrate gypsum, the setting time, mobility and compressive strength properties of $\beta$-hemihydrate gypsum were examined with the adding of two types grounded gypsum crushed by ball mill. By 15wt% adding of 7% $Al_2(SO_4)_3$ dilute solution, the setting time of $\beta$-hemihydrate gypsum was sharply accelerated than that of non-added $\beta$-hemihydrate gypsum. When ground phospho gypsum(PG) and chemical gypsum(CG) were added to $\beta$-hemihydrate gypsum, the initial and final setting time of $\beta$-hemihydrate gypsum were accelerated markedly with the increasing of grinding time and added amount of ground phospho gypsum. Especially, this trend largely presented when ground phospho gypsum was added to $\beta$-hemihydrate gypsum. The compressive strength of $\beta$-hemihydrate gypsum added by ground phospho and chemical gypsum was largely increased at initial curing time such as 1, 3 days. Particularly, the compressive strength of $\beta$-hemihydrate gypsum added by ground phospho gypsum was increased by 15~20% than that of ground chemical gypsum.

Changes of Chloroplast Number per Guard Cell pairs of Leaves by Ploidy Level in Nicotiana tabacum L. cv. BY-4 (담배 식물체[Nicotiana tabacum L. cv. BY-4]의 배수성에 따른 공변세포의 엽록체 수 변화)

  • We evaluated a possibility of the use of chloroplast number per guard cell pairs as a measure for ploidy level in the different ploidy levels of tobacco plant (Nicotiana tabacum L. cv. BY-4) . The guard-cell chloroplast numbers of leaves of haploid plant were a half of wild-type plant. Furthermore, the number of chloroplast per guard cell pairs of the leaves of doubled-haploid plant increased in two times compared with that of haploid plant. In addition, the chloroplast number was not changed in the F$_1$ progenies. The change of the chloroplast number by leaf stage was not observed. The results indicate that there is a strong relationship between ploidy level (2x and 4x) and chloroplast number per guard cell pairs. This relationship was also, observed in both in vitro and pot cultured plants. It was determined that the measurement of chloroplast number in guard cells of leaf epidermis is simple to use and less labour intensive, and hence can be considered a practical alternative to the chromosome counting methods or flow cytometry in the tobacco plant.

Rheological Properties and Hydration of Belite-rich Cement Paste with Various Calcium Sulfate Types (석고형태에 따른 Belite-rich Cement의 수화 및 유동특성)

  • 본 논문에서는 석고형태에 따른 belite-rich cement(BRC)의 수화성상 및 혼화제의 흡착거동으로부터 시멘트의 유동성에 미치는 영향성을 조사하였다. 또한, 보통 포틀랜드 시멘트(OPC)의 경우와도 그 영향성을 비교 검토하였다. 유동특성을 측정한 결과, 폴리카르복실산계 혼화제 사용시 반수와 이수석고를 첨가한 BRC가 미니슬럼프 값이 약 $\pm$20cm 근방의 수치를 보였으며, 무수석고를 첨가한 것과 OPC에서는 낮은 슬럼프치를 보였다. 초기 수화의 경우, XRD와 DSC에서 Ca(OH)$_2$의 생성량의 정도가 BRC 2-C(반수)>BRC 1-C(이수)>BRC 3-C(무수)의 순서로 줄어들기 때문에 장기강도측면에서도 반수석고와 이수석고를 동시에 사용하는 것이 우수할 것으로 추정된다. 또한 이 결과는 비표면적의 데이터와도 일치함을 보였다.
