Effects of Grain Size Distribution on the Shear Strength and Rheological Properties of Debris Flow Using Direct Shear Apparatus (직접전단장비를 이용한 토석류의 전단강도 및 유변학적 특성에 대한 입도분포의 영향 연구)

  • In this study, effects of grain size distribution on the shear strength and rheological properties are investigated for coarse- and fine-grained soils by using direct shear apparatus. Shear strengths are estimated for fine-grained soils with the maximum particle size of 0.075 mm and coarse-grained soils with the maximum particle size of 0.425 mm and fine contents of 17% prepared at dry and liquid limit states. The direct shear tests are conducted under the relatively slow shear velocity, which corresponds to the reactivated landslide or debris flow after collapse according to the landslide classification. In addition, for the evaluation of rheological properties, residual shear strengths for both fine- and coarsegrained soils prepared under liquid limit states are obtained by multiple reversal shear tests under three shear velocities. From the relationship between residual shear strengths and shear rates, Bingham plastic viscosity and yield stress are estimated. The direct shear tests show that cohesions of fine-grained soil are greater than those of coarse-grained soil at both dry and liquid limit states. However, internal friction angles of fine-grained soil are smaller than those of coarse-grained soil. In case of rheological parameters, the plastic viscosity and yield stress of fine-grained soils are greater than those of coarse-grained soils. This study may be effectively used for the prediction of the reactivated landslide or debris flow after collapse.

  • To investigate the variations of physico-chemical factors and microbial population, in ten stations at water region of coastal area of Chagwi-Do, Nutritive salts, water temperature, transparency, suspended solid, salinity, COD, DO, pH, heterotrophic bacteria, coliform group and Vibrio spp. were analysed three times in September, November in 2004 and February in 2005. Heterotrophic bacteria in surface water was $3.5X10^1{\sim}1.16X10^3cfu/ml,\;1.0X10^2{\sim}5.2X10^1cfu/ml\;2.0X10^1{\sim}7.6X10^1cfu/ml$ and bottom water counted $7.0X10^2{\sim}1.0X10^3cfu/ml,\;1.4X10^1{\sim}2.5X10^2cfu/ml\;2.0X10^2{\sim}4.2X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively. The cell number of total coliform bacteria in the surface water amounted to $0{\sim}4.3X10^2cfu/ml,\;0{\sim}6.0X10^1cfu/ml,\;0{\sim}1.0X10^1cfu/ml$ and bottom water amounted $0{\sim}2.2X10^2cfu/ml,\;0{\sim}5.4X0^2cfu/ml,\;0{\sim}2.0X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively. As for Vibrio spp., the cell number in the surface water was $1.0X10^1{\sim}2.5X10^2cfu/ml,\;1.0X10^1{\sim}2.0X10^1cfu/ml,\;0cfu/ml$ and bottom water counted $1.0X10^1{\sim}5.2X10^2cfu/ml,\;0cfu/ml,\;2.0X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively.

Analysis of Bacterial Community Structure of Biological Activated Carbon Process in Drinking Water Treatment Plant Using FISH (FISH법을 이용한 정수처리장 내 생물활성탄 공정의 세균군집 구조 분석)

  • The bacterial community structure in biological activated carbon (BAC) process in drinking water treatment plant was investigated by Fluorescent in situ Hybridization (FISH) with rRNA-targeted oligonucleotide probe. Samples were collected at different three points in BAC process every month for one year. They were hybridized with a probe specific for the alpha, beta, gamma subclass of the class Proteobacteria, Cytophaga-Flavobacteria group and Gram-positive high G+C content (HGC) group. Total numbers of bacteria in BAC process counted by 4',6-diamidino-2-phenylindole (DAPI) staining were $5.4{\times}10^{10}$ (top), $4.0{\times}10^{10}$ (middle) and $2.8{\times}10^{10}$ cells/ml (bottom). The number of the culturable bacteria was from $1.0{\times}10^7$ to $3.6{\times}10^7$ cells/ml and the culturability was about 0.05%. The faction of bacteria detectable by FISH with the probe EUB338 was about 83% of DAPI counts. Gamma and alpha subclass of the class Proteobacteria were predominant in BAC process and their ratios were over 20% respectively. In top and middle, alpha, beta and gamma subclass of the class Proteobacteria competed with each other and their percentages was changed according to the season. In bottom, gamma subclass of the class Proteobacteria was predominant all through the year. It could be successfully observed the seasonal distribution of bacterial community in biological activated carbon process using FISH.

  • The purpose of this study was to evaluate critically the effects of forest experience on young children using meta-analysis. To test this, a total 30 studies were identified, and mean effect size of variables was calculated by applying Fixed-effect model. As a result, the overall effect size of forest experience on young children was 0.797. The effect sizes of daily type as a program type had an effect than experience type at the level of .939. And also the effect sizes of period program (more 4month), number of participants (10-20people), program time (180minute), dependant variables (multiple intelligence) yield effects 0.972, 0.980, 1.138 and 0.957 respectively. As the result, this study implies a way which is to derive effectively further studies on the forest experience on young children.

Rapid Detection of the pathogenic agent of Bacterial white enteritis of Larval and Juvenile Stages in Olive flounder (Paralichthys olivaceus) (넙치(Paralichthys olivaceus)자치어 장관백탁증(Bacterial white enteritis) 원인균의 신속 검출)

  • Bacterial wihte enteritis ocurred by infection of V. ichthyoenteri is a devastating disease in olive flounder (Paralichthys olivaceus) hatcheries in Korea. Since white enteritis has been a problem in aquqtic industries, necessity of a rapid detection method is increased. In an attempt to develop rapid PCR method the detection of V. ichthyoenteri, we examined the 16S-23S rRNA intergenic spacer region(ISR) of V. ichthyoenteri and developed species-specific primer for V. ichthyoenteri. The intergenic spacers were amplified by primers complementary to conserved region of 16S and 23S rRNA genes. The intergenic spacer region between the 16S and 23S rRNA genes of V. ichthoenteri were investigated by PCR fragment length typing and DNA sequencing. Analysis of the ISR sequences showed that V. ichthyoenteri contains one types of polymorphic ISRs. The size of ISRs ranged 348bp length and not contains tRNA genes. Mutiple alignment of representative sequences from different Vibrio species revealed several domains of high sequence variability, and allowed to design species-specific primer for detection of Vibrio ichthyoenteri. PCR. The specific of the primer was examined using genomic DNA prepared from 19 different Vibrio species, isolated 18group Vibrio species. The results showed that the PCR reaction using species-specific primer designed in this study can be used to detect V. ichthyoenteri.

Static Culture Condition for Production of Bacterial Cellulose, Environment-Friendly Functional Material, by Acetic Acid Bacteria (초산균에 의한 환경친화적 기능성소재인 세균 셀룰로오스 생산을 위한 정치배양조건 최적화)

  • Bacterial cellulose (BC) has played important role as new functional material for food industry and industrial products based on its unique properties. The interest in BC from static cultures has increased steadily in recent years because of its potential for use in medicine and cosmetics. In this study, we investigated culture condition for BC production by Acetobacter sp. F15 in static culture. The strain F15, which was isolated from decayed fruit, was selected on the basis of BC thickness. The optimal medium compositions for BC production were glucose 7%, soytone 12%, $K_2HPO_4$ 0.2%, $NaH_2PO_4{\cdot}_2H_2O$ 0.2%, lactic acid 0.05% and ethanol 0.3%, respectively. The strain F15 was able to produce BC at $26^{\circ}C-36^{\circ}C$ with a maximum at $32^{\circ}C$. BC production occurred at pH 4.5-8 with a maximum at pH 6.5. Under these conditions, a maximum BC thickness of 12.15 mm was achieved after 9 days of cultivation; this value was about 2.3-fold higher than the thickness in basic medium. Scanning electron micrographs showed that BC from the optimal medium was more compact than plant cellulose and was reticulated structure consisting of ultrafine cellulose fibrils. BC from the optimal medium was found to be of cellulose type I, the same as typical native cellulose.

Evidence-based Field Research on Health Benefits of Urban Green Area (과학적 근거를 바탕으로 한 도시녹지의 건강편익에 관한 연구)

  • With increasing interest in health promotion and quality of life, growing attention has been focused on the beneficial effects of urban green area. However, very few evidence-based approaches have been conducted on the health-related benefits of urban greenery. Therefore, this study examined the health-related benefits of green areas using physiological and psychological indices to obtain evidence-based data on these benefits. Twenty male university students were selected as subjects. Data were collected when participants viewed landscapes in a green area or an urban area for fifteen minutes. This research was reviewed and approved by the Ethics Committee of School of Medicine, Chungnam National University. Physiological data in the green area revealed significantly decreased heart rates, significantly increased high-frequency value of heart rate variability, an index of parasympathetic activity, and reduced salivary cortisol concentration, a stress hormone, compared to the urban area. Psychological tests showed the green area significantly reduced the negative mood state and psychological symptoms, and significantly increased the positive mood state. Our data provided evidence for the health-related benefits of green areas, and the findings of this study support that green areas can play a critical role in health promotion for urban residents, by positively affecting autonomic nervous and endocrinal activities.

Efficacy of Supplemental Lecithin-free Egg Yolk at Different Dietary Calcium Levels on Growth Performance and Ca Absorption of Laying Hens (계란 난황 레시틴 추출 부산물이 사료 내 Ca 함량을 조절한 산란계의 생산성과 Ca 흡수에 미치는 영향)

  • A 3-weeks feeding trial with 288 laying hens were conducted to determine the efficacy of lecithin-free egg yolk at different levels of dietary Ca on performance and Ca absorption. Laying hens were divided into 6 groups according to calcium level and testing agent; 0% calcium feed (A), 0.2% calcium feed (B), 0.4% calcium feed (C, normal feed), 0.6% calcium feed (D), 0.4% calcium feed + 0.2% egg byproduct (C+0.2), 0.4% calcium feed + 0.4% egg byproduct (C+0.4). The final body weight gain of C+0.2 and C+0.4 groups were higher by 1.5% and 7.4% respectively than group C. Tibia ash contents did not show significantly difference, but calcium contents increase (p<0.05) in C+0.2 and C+0.4 groups. Parallel undecalcified tibia joint sections were stained for calcium absorption by the von Kossa's stain. This result show that lecithin free egg byproduct supplementation to normal calcium feed improved growth performance and calcium utilization in laying hens.

Pretreatment of Cane Molasses for Production of Bacterial Cellulose and Its Physico-Chemical Properties (미생물 셀룰로오스 생산을 위한 당밀의 전처리 및 생산된 셀룰로오스의 물리화학적 특성)

  • The aim of this study is to investigate cane molasses pretreatments for the production of cellulose by Acetobacter sp. V6, which has excellent bacterial cellulose (BC) producing capacity in the shaking culture. Among pretreatments of cane molasses, 1% (w/v) tricalcium phosphate (TP) treatment was more efficient in BC production. The physico-chemical properties of BCs that were produced in static and shaking cultures were also investigated. Although BC had an emulsifying ability, its emulsion stability was low. Water holding capacity (WHC) of BC was high; the WHC of BC produced in static culture was 14 times higher than that of $\alpha$-cellulose. In addition, the viscosity of BC was higher than that of $\alpha$-cellulose. Composition analysis by FT-IR showed no difference in composition between BC and plant cellulose. In the crystallinity analysis by XRD, all BC samples showed crystallinity. All BC samples showed reticulated structures consisting of ultrafine cellulose fibriles. Microfibriles of cellulose from static culture were especially more compact than those of cellulose from shaking culture.

Evaluation on Skin Irritation and Sensitization of Oxyresveratrol and Oxyresveratrol-3-O-glucoside Produced by Biotransformation of Morus alba Extract (상백피 추출물을 효소전환 하여 생성한 oxyresveratrol 및 oxyresveratrol-3-O-glucoside의 피부 자극성 및 감작성 평가)

  • Stilbenes are known as antioxidants and some of them demonstrate anti-pigmentation activity. The purpose of the present study was to investigate whether two stilbene compounds produced by biotransformation of the extract of $Morus$ $alba$ root show skin irritation and sensitization. In skin irritation test, 1% oxyresveratrol (OXY), and 5% OXY, and 1% oxyresveratrol-3-$O$-glucoside (OXY-3) showed a P.I.I score of 0, 0.04, and 0, respectively. Accordingly, the two stilbenes were evaluated to be virtually 'non-irritant' materials. In a skin sensitization study by GPMT, 1% OXY, 5% OXY, and 1% OXY-3 did not cause edema and erythema at 24 h and 48 h after topical application and exhibited a sensitization score of 0 and a rate of 0%. Consequently, it was confirmed that OXY and OXY-3 had no contact allergic sensitization in GPMT. Therefore, OXY and OXY-3 might be potential candidates as skin-whitening agents without posing any serious side effects.
