Production of chicken egg yolk antibody to Canine parvovirus (개의 파보바이러스에 대한 난황 항체 생산)

  • 산란계에 불활화 개 파보바이러스 백신을 근육내로 1주 간격으로 4회 접종하여 면역화시키고 최종 접종 2주후에 채란하여 $4^{\circ}C$에 보관하며 사용하였다. 난황항체는 5% HPMCP를 이용하여 분리하였고 0.5% HPMCP 용액은 lipid 침전에 매우 효과적이었으며 희석배수 10배에서 투명한 상층액을 나타내었다. 1차분리한 상층액의 단백질 농도는 2.5mg/ml이었고 최종 단백질 용액의 경우는 26.53mg/ml이었다. SDS-PAGE 전기영동상에서 분자량 60~70 KD 및 30~40 KD의 2 band가 나타났으며 non-reducing 전기영동에서는 닭 혈청 IgG와 같은 120~160 KD의 분자량을 보인 band가 각각의 분리용액에서 나타났다. 난황 항체의 개 파보바이러스에 대한 혈구응집억제반응 항체역가는 혈청의 역가에 비해 1주의 차이를 주며 증가했으며 난황 항체는 1:640에서 1:2560, 혈청은 1:640에서 1:5120을 나타내었다.

  • 구제역 방역정책은 국가적으로 운영되어야하고 백신의 사용도 정부 통제하에 사용되어야 한다. 만약 백신생산시 강한 병원성을 지닌 구제역바이러스가 항원으로 사용된다면 적절한 생물안전절차를 수행할 수 있는 생산시설에서 실시되어야 한다. 일반적으로 백신접종은 구제역이 유행되는 지역이나 나라에서 사용된다. 구제역 청정국은 백신접종은 하지 않으나 엄격한 이동제한과 살처분 조치를 취한다. 그럼에도 많은 청정국가가 불활화 항원을 비축하여 필요시 사용하는 전략으로 긴급백신 접종을 선택할 수 있다. 우리나라의 경우도 전국을 대상으로 백신접종을 실시하는 정책을 2011년 1월 이후 시행되고 있고, 이에 따라 구제역 백신의 국내에서 생산을 준비하고 있다. 구제역 백신의 생산과 관리는 주변지역에서 유행하거나 유행되었던 바이러스형에 대한 항원 개발과 원활한 백신의 공급을 통해 우리나라에서 구제역을 근절하고 재유입을 예방하는 데 매우 중요한 요소가 될 것이다. 본 글에서는 백신에 사용되는 종독주, 항원의 생산 및 관리에 대한 세계적인 기준으로 사용되고 있는 세계동물보건기구 (OIE)에서 권고되는 백신에 대한 관리 사항을 알아보고자 하였다.

Studies on the combined inactivated oil emulsion vaccine of Newcastle disease and avian infectious bronchitis in chickens (닭의 뉴캣슬·전염성 기관지염 바이러스 혼합 불활화 오일 에멀션 백신의 생산시험)

  • A single inoculation of combined vaccines of Newcastle disease and avian infectious bronchitis of chicken, in a form of gel-oil emulsion type (gel-OEV) was tested their immunogenecity in chickens. The results were summerized as follows: 1. Average minimum and maximum ELISA antibody titers of ND were recorded 2407 and 13144 respectively. In the case of IB, 1824 and 4496 were recorded as minimum and maximum titers. 2. The distribution of average proportional groups, in the lowest and the highest, were 1.6 and 7.0 in ND ELISA and 1.4 and 2.8 in IB ELISA antibody titers. 3. ND ELISA antibody titers were significantly increased upto 7th week after the vaccination. On the other hand, IB ELISA antibody titers were raised upto 4th week after the vaccination.

Efficacy of Genetic Adjuvant (Plasmid-Expressed Chicken Interleukin-6) and Chemical Adjuvant (Levamisole) on the Protective Immunity of Genetic Vaccine against Infectious Bursal Disease Virus (닭의 전염성 F낭병 바이러스 유전자백신에 의한 방어 면역에 Genetic Adjuvant (Chicken Interleukin-6)와 Chemical Adjuvant (Levamisole)의 효과)

  • Infectious bursal disease (IBD) caused by the infectious bursal disease virus (IBDV) has an important economic impact on the poultry industry worldwide. This study examined the adjuvant effects of a plasmid encoding chicken interleukin-6 (pcDNA-ChIL-6) and levamisole (LMS) on in ovo prime-boost vaccination using a genetic vaccine (pcDNA-VP243) to prime in chicken followed by a killed-vaccine boost. A pcDNA-VP243 was injected into the amniotic sac alone or in combination with a pcDNA-ChIL-6 or LMS at embryonation day 18, followed by an intramuscular injection of killed IBD vaccine at 1 week of age. The chicken were orally challenged with very virulent IBDV (vvIBDV) strain at 3 weeks of age and observed for 10 days. No mortality was observed in the groups that received the pcDNA-VP243 alone and pcDNA-VP243 plus pcDNA-ChIL-6 or LMS compared to 100% mortality in unvaccinated challenge control group. However, as determined by bursal damage (the presence of IBDV RNA, B/B ratio, and lesion score), a pcDNA-VP243 alone group was superior to pcDNA-VP243 plus pcDNA-ChIL-6 or LMS groups in the protection against post-challenge. These findings suggest that in ovo priming with genetic vaccine and boosting with killed vaccine is an effective strategy for protecting chicken against vvIBDV and the addition of pcDNA-ChIL-6 or LMS did not enhance protective immunity.

Industrial-Scale Production of High-Purity Antihemophilic Factor IX from Human Plasma (사람 혈장으로부터 고순도 혈액응고 제9인자의 산업적 생산)

  • The use of antihemophilic factor IX complex has been associated with a variety of thrombotic complications, the major cause of which was the contamination of thrombogenic proteins such as vitamin K-dependent clotting factors II, VII, and X. In order to produce a commercial factor IX (GreenNine VF) free from thrombogenic potential, industrial-scale production process for high-purity factor IX from human plasma has been developed. The purification process contains cryo-precipitation, DEAE-sephadex A-50 anion-exchange chromatography, DEAE-toyopearl 650M anion-exchange column chromatography, heparin-sepharose 6FF affinity column chromatography, and CM-sepharose FF cation-exchange column chromatography. Also the process includes two viral inactivation and removal procedures, solvent/detergent treatment and nanofiltration using Viresolve NFP filter. The purification yield was 35.4%. The specific activity in the purified concentrate was 190.8 IU/mg which exceeded that in the factor IX complex (FacNine) by a factor of 48. The activities of factor II, VII, and X were not detected in GreenNine VF. SDS-PAGE analysis showed that GreenNine VF had the highest purity in comparison with commercially available high purity factor IX concentrates, Mononine, Octanyne, Berinin HS, and Immunine STIM plus 600. One batch size of the production was 2,400 vials of 250 IU product or 1,200 vials of 500 IU product from 1,600 L cryo-poor plasma.

Preparation of an Inactivated Influenza Vaccine Using the Ethanol Extracts of Medical Herbs (한약재 식물 에탄올추출물을 이용한 인플루엔자 불활화백신 제작)

  • As seen in the COVID-19 pandemic, unexpected emergence of new viruses presents serious concern on public health. Especially, the absence of effective vaccines or antiviral drugs against emerging viruses significantly increases the severity of disease and duration of viral circulation among population. Natural products have served as a major source for safe and effective antiviral drugs. In this study, we examined the virucidal activity of medical herb extracts with a view to discover novel antiviral agents with desired levels of safety and antiviral efficacy. Ethanol extracts of ten selected medical herbs were tested for antioxidant activity and in-vitro cytotoxicity in various animal cell lines. Of note, the herbal extracts showed broad and potent virucidal activities against rotavirus, hepatitis A virus, and influenza A virus. The extracts of Sorbus commixta and Glycyrrhiza uralensis showed strong virucidal activities against influenza A virus. We also examined whether the extracts of Sorbus commixta and Glycyrrhiza uralensis can be used as inactivating agents to prepare an inactivated viral vaccine. In a mouse model, influenza A virus inactivated by the extracts elicited high levels of neutralizing antibodies, and the vaccination provided complete protection against lethal challenge. These results suggest that herb-derived natural products can be developed to antiviral drugs as well as inactivating agents for preparation of inactivated viral vaccines.

Immunosuppressive effect of Cryptosporidium baileyi infection on vaccination against Newcastle disease in chicks (닭와포자충 감염이 닭의 뉴캣슬병 예방접종에 대한 면역억제 효과)

  • Hemagglutination-inhibition titers (log2) to Newcastle disease (ND) vims were chronologically observed in chicks, which were orally inoculated with 5 × 105 oocysts of Cwptospori,drum bcileyi at 2 days of age and subsequently vaccinated with inactivated ND virus at 4 and 21 days postinoculation. In general, the titers were considerably lower in the infected chicks than those in the uninfected control throughout the experimental period (p < 0.01), and rapid negative seroronversions were observed in the infected chicks. The titers reached a peak on weeks 2 and 4 post-booster-vaccination in the control and infected chicks, respectively. Thus, C. bciLeWi infection was shown to have an immunosuppressive effect on ND vaccination when the agent was given to 2-day-old chicks. It is suggested that C. bniLeWi infection in chicks may increase the host susceptibility to ND virus.

Evaluation of safety and immunogenicity of a new octavalent inactivated vaccine containing porcine parvovirus, erysipelas, and leptospira (돼지 파보바이러스, 단독 및 렙토스피라 8가 불활화 백신의 안전성 및 면역원성 평가)

  • Porcine parvovirus, Erysipelothrix (E.) rhusiopathiae, and Leptospira (L.) interrogans are considered major etiologic agents of reproductive failure in pigs, causing economic loss in the swine industry. In this study, the safety and immunogenicity of a new octavalent inactivated vaccine were evaluated. The vaccine contained inactivated porcine parvovirus, E. rhusiopathiae, and six L. interrogans serovars (Bratislava, Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, and Pomona). Safety test results showed no notable side effects or clinical signs after vaccination in mice, guinea pigs, and sows. In addition, we assessed immunogenicity of the vaccine in 25 sows under field conditions. The vaccinated group (n = 20) had a significantly higher antibody level than the non-vaccinated group (n = 5). Moreover, the stillbirth rate decreased in piglets born from vaccinated sows, resulting in an increased fertility rate. The results of this study demonstrate that the new octavalent inactivated vaccine can be applied safely and effectively to improve reproductive performance in sows.

Depression of Immune Response by Newcastle Disease Virus Infection (Newcastle병(病) 바이러스감염(感染)에 의(依)한 면역반응억제(免疫反應抑制))

  • The immunosuppressive activity of newcastle disease virus(NDV) and some possible role of interferon(C-IF) in viral suppression of immune response were evaluated by SRBC-induced delayed-type hypersensitivity(DTH), rosette formation in spleen cells, number of lymphocytes in peripheral blood, hemagglutinin and hemolysin response to SRBC in ICR mice sensitized with SRBC. When NDV was inoculated before or after sensitization of mouse with SRBC, virus caused a marked inhibition of DTH, and its depressive effect was dependent on the time of virus inoculation in relation to SRBC sensitization or challenge. Rosette formation of spleen cells was significantly reduced by NDV infection. The degree of the depression of rosette formation was more prominent in mice inoculated before sensitization than after sensitization and could be related to the amount of serum interferon induced by the virus. Humoral response to SRBC of virus infected mouse was significantly depressed when NDV was inoculated 24 or 48 hours before sensitization. However, there was no difference in the response when the virus was inoculated 9 hour before and at the same time of sensitization or even after that. Lymphocytes in peripheral blood of mice were markedly diminished in numbers when NDV was inoculated 48 and 24 hour before sensitization with SRBC, but they were slightly augmented when the virus was inoculated 9 hour before and at the same time of sensitization. When UV-inactivated or heat-inactivated NDV was injected to the mouse at the same time of sensitization with SRBC, DTH and rosette formation of spleen cells were slightly depressed. DTH and rosette formation in mice treated with crude-IF were generally depressed as com pared with those of control mice. These studies suggest that the NDV causes a significant depression of cell-mediated immunity, whereas the humoral immune response is not inhibited markedly, and that the depression of immune response by NDV infection may be caused by interferon produced by NDV and direct viral activity.

A Field Efficacy Trial of Inactivated Hantaan Virus Vaccine ($Hantavax^{TM}$) Against Hemorrhagic Fever with Renal Syndrome (HFRS) in the Endemic Areas of Yugoslavia from 1996 to 1998 ($1996{\sim}1998$년 유고슬라비아에서 실시한 한탄바이러스 불활화 백신($Hantavax^{TM}$)의 면역효과에 대한 야외시험)

  • In Yugoslavia, hemorrhagic fever with renal syndrome (HFRS) is one of the important national health problem, but no vaccine has been used to prevent HFRS. Since first HFRS case in 1952, sporadic cases of HFRS occurred every year and over 4,000 registered cases with $1{\sim}16%$ mortality so far. We performed a prospective, randomized double-blind placebo-controlled trial to evaluate the effectiveness of $Hantavax^{TM}$ against HFRS in 3,900 healthy adults living in the endemic areas of Yugoslavia. 1,900 people were given 0.5 ml of Hantavax subcutaneously twice at one month interval and a booster shot at one year after. For controls other 2,000 healthy people were given 0.5 ml of physiolosical saline as a placebo. We investigated HFRS cases in both the vaccinated and nonvaccinated groups by monitoring the program for patient registration in the areas from 1996 to 1998, and the effect of vaccine was analyzed epidemiologically. No confirmed case of HFRS was observed among 1,900 Hantavax vaccinees, while 20 confirmed cases were observed among 2,000 nonvaccinated control group. There were no remarkable side effects among the vaccinees either locally or in general after inoculation of the vaccine. The Hantavax vaccine showed statistically significant protective efficacy against HFRS among Yugoslavian people.
