Microscopic Anatomy of Male Reproductive Organ in the Long Arm Octopus Octopus minor (Cephalopoda: Octopodidae) (낙지 Octopus minor 수컷 생식기관의 미세해부학적 구조)

  • This study was described the microscopic anatomy of male reproductive organs and spermatophore necessary for understanding the reproductive ecology of the long arm octopus Octopus minor. The long arm octopus was a species that has sexual dimorphism that can distinguish between sex based on the presence of hectocotylus. Male reproductive organs consisted of testis, primary spermatic duct, spermatic gland, secondary spermatic duct, spermatophoric gland and spermatophoric sac. Histologically, the testis was testicular tubule type and male germ cells showed a layered arrangement. The primary spermatic duct was a tube connecting the testis and spermatic gland, and consisted with epithelial layer and connective tissue. The spermatic gland was located between the primary and secondary spermatic duct, and the epithelial layer was composed of epithelial cells and mucous cells. Mucous cells reacted blue in the AB-PAS (pH 2.5) reaction and purple in the AF-AB (pH 2.5) reaction. The secondary spermatic duct was a short tube connecting spermatic gland and spermatophoric gland, and folds were developed in lumen. The spermatophoric gland consisted of numerous tubular glands and secretory cells had eosinophilic granules. The spermatophoric sac was shape of pouch, folds were developed in lumen, and vacuolar secretory cells were present in the epithelial layer. The spermatophore was 83.5 mm long and consisted of cap thread in anterior portion, ejaculatory apparatus and cement body in medial portion, sperm mass in posterior portion.

Anti-corrugation activity of micosporine-like amino acid mixtures from Chlamydomonas sp. (Chlamydomonas sp. 유래 mycosporine-like amino acid 혼합물의 항주름 활성)

  • To examine the effects of a mycosporine-like amino acids (MAAs) mixture from microalgae, Chlamydomonas sp, on the anti-wrinkle activities, the expression levels of genes that are associated with skin aging, including type I procollagen, elastin and involucrin, were analyzed. Asterina 330+palythine (A+P) and shinorine+palythine (S+P) mixtures were purified from Chlamydomonas sp using the following steps: 80% methanolic extraction, column purification, and HPLC analysis. As a result of the MTT assay, A+P and S+P did not induce cellular cytotoxicity with up to 0.1 mg/mL of both MAAs. In addition, the treatment of UV-exposed fibroblasts with A+P (0.05 mg/mL) and S+P (0.01 mg/mL) increased the levels of the PCOLCE mRNAs by 2.7 and 3.6 fold compared to the control group, respectively, The levels of elastin gene expression were elevated 5.59 and 3.1 fold in the A+P and S+P treated (0.01 mg/mL) cells, respectively. In particular, at a concentration of 0.01 mg /mL, the A+P and S+P expression levels of Involucrin mRNAs were increased 3.5 and 2.5 fold in the UV-exposed cells compared to the control, respectively. In conclusion, the MAAs derived from Chlamydomonas sp can be utilized as functional cosmetic materials with anti-wrinkle effects on the skin.

Beauty activity of extracts from Saurus chinensis by ultra-fine ground (초미세 분쇄한 삼백초(Saurus chinensis) 추출물의 미용 활성)

  • In this study, beauty food activities were determined for extracts of Saurus chinensis as functional beauty food. Tyrosinase inhibitory effect was increased as more 4 folds by ultra-fine grind technology than normal grind technology in ethanol extracts from Saurus chinensis. A ultra-fine ground technology was used to increase the extraction yield for bioactive compounds from medicinal plants. Tyrosinase inhibitory activity, which is related to skin-whitening, was confirmed to be 100% for ethanol extracts at 1,000 ppm of phenolics. Anti-wrinkle activity was determined showing 30% of inhibition effect at above 1,000 ppm phenolic concentration. The elastase inhibitory activity of water and ethanol extracts from ultra-fine ground Saurus chinensis showed higher inhibition activity than compared to extracts from normal ground Saurus chinensis. Astringent activity in ethanol extracts was determined showing over 70% at 250 ppm of phenolics. Based on these results, Saurus chinensis extracts can be used as a functional beauty food and cosmetic with whitening, anti-wrinkle and pore contraction effects.

  • Boophilus microplus (Canestrini, 1887) female specimens were subjected to scanning electron microscopy for the observation of surface fine structures. Morphological characteristics observed were summarized as follows: 1. The palpi were wider than long, and compressed, ridged dorsally and laterally. There were usually four pairs of ventro-internal setae on the articles. 2. The hypostome dentition was usually 4/4 but occasionally 5/5. 3. The basis capituli were hexagonal dorsally and the porose area was distinct with the holes developed well. 4. There were numerous folds resembling finger prints around the female genital pore. 5. A pair of anal lobes with specified setae on them was also rocognized as one of the morphological characteristics.

  • The ancient paper document we chose as a case study for our research is "One Hundred Poets of the Tang Dynasty", which is housed in the Ryukoku University Library. The purpose of this research is to introduce a method to analyze the surface roughness and microstructure at a high resolution. In addition, we attempt to quantitatively measure the surface unevenness of the types and curve structure. We used a tridimensional digital microscopy as a non-contact and a non-destructive method to study ancient cultural paper. The information contained in the paper may be lost in the process of applying strong pressure to clean and lining or press. However, this microscopic measurement method can non-destructively analyze a large amount of data in old printed books. Moreover, it enables observing them directly with reflected light. Therefore, this method may be useful for collecting printed information remaining on the surface of the paper.

Ultrastructural Changes on the Cuticular Surface, Excretory and Digestive Organs of Anisakis simplex Larvae Chronologically Recovered from Experimental Cats (실험감염 고양이로부터 경시적으로 회수한 고래회충 유충의 표피, 배설기 및 소화기의 미세구조 변화)

  • The ultrastructural changes of cuticular surface, excretory and digestive organs of Anisakis simplex larvae chronologically recovered from experimental cats were observed with a SEM and TEM. The larva recovered from an experimental cat at 3 days post-infection (PI) retained the cuticular surface with regular transverse striations and a longitudinal groove on the lateral side of body. This finding suggests that the molting of the 3rd stage larva of A. simplex to 4th one occurred from the 3rd day after infection in cats. The excretory organ (renette cell) consisted of a large cell with numerous ductules ramified from the main duct, mitochondria and secretory granules in cytoplasm. Secretory granules in the renette cell of larvae recovered at 24 hours PI were round whereas those of control and larvae recovered at 6 hours PI were amorphous. Muscular esophagus and ventriculus also retained many secretory granules in the cytoplasm. The secretory granules in these organs of larvae recovered at $6\sim24$ hours PI were electron-dense and widely distributed whereas those of control worm were packed in a pocket and retained various electron densities. In the cytoplasm of intestinal epithelial cells, numerous fine glycogen particles and mitochondria were distributed. The chronological changes of secretory granules in renette cell, muscular esophagus and ventriculus seem to be related with the worm penetration into host tissue.

Comparative Studies on the Ultrastructure of Salivary Ducts between the Two Species of Snails, Achatina fulica and Incilaria fruhstorferi (두 종의 달팽이류 (Achatina fulica and Incilaria fruhstorferi) 사이의 타액관의 미세구조에 관한 비교연구)

  • We observed the salivary ducts of two species of snails, Achatina fulica and Incilaria fruhstoferi with an electron microscope, and obtained the following results. The intralobular and interlobular ducts of Achatina fulica assume the forms of round or ellipsoidal doughnuts. The boundaries between the endothelial cells are not clear. It is also found that the cytoplasm of the endothelial cells consists of the membrane infolded in interdigital form, and there are well -developed microvilli at the apical portion of the cytoplasm. On the other hand, the intralobular and interlobular ducts of Incilaria fruhtoferi consist of the irregular simple columnar epithelia. The high electron dense cytoplasm is filled with the irregular round granules. The microvilli at the apical portion of the cytoplasm are not so well-developed as those in Achatina fulica. In the salivary duct of Achatina fulica, the lumen has narrow and long tubular structure. The boundaries between the endothelial cells are not clear. The cytoplasm is full of many vacuoles and electron lucent granules. At the apical portion of the cytoplasm, lots of short and thin microvilli are found. The salivary duct of Incilaria fruhstorferi is wider ($65\times250{\mu}m$ in diameter) than that of Achatina fulica, and consists of endothelial cells of the same structures. At the apical portion of those endothelial cells, a lot of junction apparatus such as desmosomes are observed. The vessels in the salivary ducts of Achatina fulica and Incilaria fruhstoferi are observed mainly in the connective tissues between the salivary glands. The endothelial cell of the vessel has the irregular structure and looks dark due to the high electron density. These cells protrude their filopodia and phagocytosize foreign bodies.

Ultrastructural Changes in the Mantle of the Equilateral Venus, Gomphina veneriformis (Bivalvia: Veneridae) Exposed to TBTCl (TBTCl 노출에 의한 대복, Gomphina veneriformis (Bivalvia: Veneridae) 외투막의 미세구조적 변화)

  • Equilateral venus, Gomphina veneriformis exposed to tribultyltin chloride (TBTCl) for 36 weeks was showed ultrastructural changes of the mantle. The fine mantle had 4-folds and its epidermal layer consisted of simple columnar epithelial cells and ciliated cells and secretory cells. Inner and outer epidermal layer covered connective tissue. The mantle exposed to TBTCl at 12 weeks was decreased cilia in the inner epidermal layer, and observed extension of the hemolymph sinus and destruction of the septum. At 20 weeks, it revealed vacuole formation and pycnosis in the cytoplasm, and scattered muscular fiber. After 28 weeks of exposure, the mantle revealed partially degenerative changes in the epidermal layer. In the ciliated cells, basal body was isolated from the cilia and rootlet complex and basal foot were scattered. The sarcolemma had debris fiber. At 36 weeks, it observed degenerative cells that it revealed disappearance of the cilia, atrophic nucleus, poorly membrane and destruction of the cresternae in the mitochondria, and increasing heterophagosome. The outer epithelial cell had necrotic nuclus, numeous lysosome and disappearance of the microvilli. Therefore, results of this study suggested that chronical TBTCl exposure in the Gomphina veneriformis induced the disorders of shell growth and physiological function with histopathological changes of the mantle.

  • Seed morphological studies of 12 species of sect. Zygophyllidium and related Poinsettia using light and scanning electron microscopy were conducted to test the hypotheses of the sectional boundaries and species relationships. Characteristics of seeds, and the keys of identification of 12 species were presented. Seeds of 12 species were divided into 6 types based on the UPGMA tree from numerical analyses using nine seed characters. Type I, including E. uniglandulosa, is characterized by the foveolate seed with granulate testa cells. Type II, including species E. exstipulata, E. lagunensis, E. bilobata, E. hexagonoides and E. chersonesa, was recognized by the presence of inter-cellular granules among testa cells. Type III, including E. cyathophora, has a seed with acute apex, and concentric circles on the surface of testa cells. Type IV, including species of E. dentata and E. pentadactyla, has a seed with caruncle and lacking inter-cellular granules among testa cells. Type V, including E. hexagona, was characterized by the ovate seeds and lacking inter-cellular granules. Type VI, including E. eriantha and E. lacera, was recognized by the seeds with rectangular shape with caruncle. Based on the seed morphology, sect. Zygophyllidium should be defined only species with E. exstipulata, E. lagunensis, E. bilobata, E. hexagonoides and E. chersonesa. E. hexagona, traditionally included in this section, was closely related to E. dentata from Poinsettia.

Scanning Electron Microscopical Findings of Echinochasmus japonicus Tegument (Echinochusmus juponicus 표피 미세구조의 주사전자현미경적 관찰)

  • Tegumental ultrastructures of Echinochasmus japonicus were observed by scanning electron microscopy. The worms were recovered from albino rats which were experimentally infected with the metacercariae obtained from Pseudorasbora parva. Followings are summarized findings. 1. The worms were minute and plumpy gourdshaped with attenuated anterior and round posterior end. The tegument of whole body was wrinkled transversely and covered with cobblestone-like cytoplasmic processes. 2. Head crown was armed with 24 collar spines which were embedded in cytoplasmic pockets. The spines were arranged in a row with an interruption at dorsomedian line, however, the 2nd and the 4th spines were outstretched more than others. Oral and ventral suckers were muscular with numerous type II sensory papillae, and genital pore opened between the two suckers. 3. Tegumental spines were spade-shaped with broad base and pointed tip. They were compact in ventra-lateral tegument or dorsal surface of anterior body. They were not found between the two suckers and dorsal surface of posterior body. 4. Two types of sensory papillae, uni-ciliated (Type I) and roundly swollen sensory papillae (Type II), were observed. The type I papillae were chiefly distributed on ventral surface of tegument and type II were on the lips of suckers. Arrangement of collar spines, shape and distribution of tegumental spines or sensory papillae are regarded as characteristic features of E. japonicus.
