  • All immature stages, eggs, larvae, and pupae of Korean Thiaspida, T. cribrosa and T. lewisii, are described and Illustrated for the first time. Two species belonging to genus Thiaspida In the subfamily Cassidlnae; T. cribrosa and T. lewisii.Observatiors on bionomics of immature stages and on their taxonomic charaders are also given.

  • The present study was conducted to gain insights into oocyte maturation and early embryo development, SELDI-TOF-MS was used to find the protein candidates that are specifically or prominently expressed in mouse oocytes at the in vitro matured metaphase II (MIl) and germinal vesicle (GV) stages. By using selected CM10 chip, found 16 candidates which were up-regulated in GV stage oocytes compared with in MIl stage oocytes, molecular weight are 8180 (2 candidates), 10226 (5 candidates), 15767 (5 candidates) and 16770 (4 candidates) Da respectively. And 29 candidates were higher in MIl than in GV stage oocytes, molecular weight are 10832 (3 candidates), 17744(8 candidates), 20122 (3 candidates), 22131 (3 candidates), 24857 (7 candidates) and 33507 (5 candidates) Da, respectively. All (45) candidate (0.2 and 1.0 % error tolerances) were performed real time RT-PCR analysis and further selected 13 more potential candidates.

  • 본 연구는 발정주기에 따른 개의 미성숙난자의 체외배양 0시간째 핵의 발달단계를 비교ㆍ검토하였다. 발정주기가 다른 개의 난소를 적출 하여 항생제가 첨가된 38℃의 0.9% 생리식염수가 들어있는 보온병에 넣어 2시간 이내에 연구실로 운반하고 난소를 면도날로 세절하여 난포란을 회수하였고, 난구세포 제거 후 Chohan과 Hunter (2003) 방법에 준하여 10 ug/ml Hoechst 33342를 이용한 핵염색을 실시하여 핵의 모양을 판명하였다. 조사된 결과는 SAS 8.0 Package를 이용하여 통계분석을 실시하였다.   • The present paper is a part of the study on the immature stages of genus Tipula in Korea. It described and illustrated egg, each larval stage and pupa of Tipula nova. It is the first to deal the all immature stages of the species in Korea. Their taxonomic remarks and habitats are also given.

  • 많은 연구에서 돼지의 난포란 혹은 배아가 낮은 온도에 대해 민감한 것은 세포질 내에 함유된 지방구와 관련이 있다고 하였으며, 이런 세포질내의 지방구를 제거함으로 동결능력이 향상된다고 하였다. 이렇게 지방구가 제거된 돼지의 난포란과 배아를 효과적으로 동결보존하기 위해서는 새로운 개념의 동결보존 기술이 필요하다고 하겠다. 따라서, 본 연구는 미성숙 단계에서 지방이 제거된 돼지 난포란의 유리화 동결에 적합한 방법을 찾기 위하여 straw를 이용한 유리화 동결법과 electron microscopic grid(EM grid)를 사용한 유리화 동결법을 실시하여 효율적인 동결방법을 검토하였다.

Influence of Stage of Maturation of Bovine Oocytes at Time of Vitrification on In Vitro Development and Viability (한우 미성숙 난자의 체외성숙 단계가 Vitrification 동결시 체외발생 및 생존성에 미치는 영향에 관한 연구)

  • This study was carried out to study the viability of oocytes when vitrified at various maturation stages. Bovine cumulus-oocyte complexes were recovered from ovaries at a slaughter and then divided into five groups: control group(unvitrified oocytes), 0 hr. group(composed of oocytes vitrified before the onset of maturation) and 10, 14, and 20 hrs groups(vitrified at 10, 14 and 20 hrs after the onset of maturation, respectively). The oocytes remained vitrified for 24 hrs, and then were thawed in 3$0^{\circ}C$. Survival and cleavage rates were investigated by results of in vitro culture and aceto-orcein staining or FDA test. No difference in the incidence of diploid oocytes was observed among the control, non-vitrified group(3.6%) and oocytes vitrified at 14 hrs(6.7%) or 20 hrs(1.7%). However, more diploid oocytes were detected after vitrification at 0 hr.(26.7%) and 10 hrs(21.7%) post maturation. The survival rate of all vitrified immature oocytes(12.0~38.0%) was low, 48.0% of unvitrified oocytes and oocytes vitrified before maturation or 0~ 10 hrs after the onset of maturation were higher than that of other groups. The overall fertilization and cleavage rates of vitrified immature oocytes (32.3 ~ 64.6% and 4.6 ~ 32.3%) were low, and 55.0% of unvitrified oocytes and the rate of immature oocytes were very higher than that of mature oocytes.
