Changes of Microbial Community Structure According to a Changes of Season and Influent Characteristics in Biological Wastewater Treatment (생물학적 폐수처리 공정에서의 계절 및 유입수 성상 변화에 따른 미생물 군집 특성 변화)

  • The bacterial community structure in biological reactor in wastewater treatment system was investigated by denaturing gradient gel electrophoresis (DGGE) and fluorescent in situ hybridization (FISH). Samples were collected at different three points in wastewater treatment system. Through treatment processes, BOD (biochemical oxygen demand) and COD (chemical oxygen demand) of was removal efficiency was 83.1~98.6%, 67.2~85.2% respectively. Microbial community of aerobic tank and oxic tank were similar but anoxic tank was different (RRP group was increased about tripple) by DGGE and FISH in sludge (2007 October and 2008 January). Samples in 2007 October and 2008 January were dominant ${\alpha}$-Proteobacteria and CF group respectively. Sludge in 2008 April were different comparing former results dominant others as 65~80%. Others group was dominant. Eubacteria by FISH with the probe EUB338 was about $1.7{\sim}7.6{\times}10^9\;cells/mL$. It could be successfully observed bacterial community in biological wastewater system.

A Comparison of Bacterial Diversity Associated with the Sponge Spirastrella abata Depending on RFLP and DGGE (RFLP와 DGGE에 따른 해면 Spirastrella abata 공생세균의 다양성 비교)

  • Culture-dependent RFLP and culture-independent DGGE were employed to investigate the bacterial community associated with the marine sponge Spirastrella abata. A total of 164 bacterial strains associated with the sponge were cultivated using Zobell and Natural sea salt media. PCR amplicons of the 16S rDNA from the bacterial strains were digested with the restriction enzymes HaeIII and MspI, and then assigned into different groups according to their restriction patterns. The 16S rDNA sequences derived from RFLP patterns showed more than 95% similarities compared with known bacterial species, and the isolates belonged to four phyla, Proteobacteria (Alphaproteobacteria, Gammaproteobacteria), Actinobacteria, Firmicutes, and Bacteriodetes, of which Alphaproteobacteria was dominant. DGGE fingerprinting of 16S rDNAs amplified from the sponge- derived total gDNA showed five major DGGE bands, and their sequences showed more than 96% similarities compared with available sequences. The sequences derived from DGGE bands revealed high similarity with the uncultured bacterial clones. DGGE revealed that bacterial community consisted of four phyla, including Proteobacteria (Alphaproteobacteria, Gammaproteobacteria), Actinobacteria, Spirochetes, and Chloroflexi. Alphaproteobacteria, Gammaproteobacteria, and Actinobacteria were commonly found in bacteria associated with S. abata by both RFLP and DGGE methods; however, overall bacterial community in the sponge differed depending on the analysis methods.

  • 유기농업에서 유기물은 양분의 공급, 토양의 이화학성 개선, 토양의 생물학적 건전성 유지 등 중요한 역할을 한다. 토양의 생물학적 건전성은 토양의 생태계적 기능을 지속적으로 유지시키는 토양미생물이 관여하고 있다. 따라서 본 연구는 유기물의 장기연용에 따른 밭토양 미생물의 다양성을 비교 분석하였다. 여러 가지 유기자원을 동일한 기준으로 매년 동일 장소에 처리하였다. 사용된 유기자원은 가축분퇴비, 채종유박인 유기질비료, 볏짚으로만 퇴비화한 볏짚퇴비와 겨울철 휴한기에 헤어리베치를 재배하여 이듬해 봄에 예취한 후 토양에 환원한 녹비처리구, NPK구, 가축분퇴비를 혼용처리한 NPK퇴비군, 양분을 전혀 시용하지 않은 무비구 등 총 7처리구였다. 각각의 처리구에서 토양(0-20 cm)을 채취하여 배양성 토양미생물은 희석평판법으로 해당 선백배지에 시료를 도말 하여 조사하였고 비배양성 미생물은 토양으로부터 genomic DNA를 추출하여 세균의 16S rDNA를 증폭시킨 후 denaturing gradient gel electrophoresis (DGGE)를 수행하여 분석하였다. 주요결과를 요약하면 밭토양에 서식하는 토양미생물의 균수는 처리별간의 차이를 보였으며 유기물처리구가 화학비료처리구보다 높았다. DGGE 분석을 통해 유기물 처리에 따른 군집의 다양성을 살펴본 결과 Fig. 1에서 보는바와 같이 Gel 상에서 다양한 위치의 밴드를 확인할 수 있었고 처리별로 특이 밴드가 있음을 확인할 수 있었다. Fig. 1에서 얻은 DGGE profile상의 밴드 강도와 수를 비교하여 Fig 2와 같은 dendrogram을 나타낼 수 있었다.

Analysis of a Sulfur-oxidizing Perchlorate-degrading Microbial Community (황 산화를 통해 퍼클로레이트를 분해하는 미생물 군집 분석)

  • Perchlorate (ClO4) is an emerging pollutant detected in surface water, soil, and groundwater. Previous studies provided experimental evidence of autotrophic ClO4 removal with elemental sulfur (S0) particles and activated sludge, which are inexpensive and easily available, respectively. In addition, ClO4 removal efficiency was shown to increase when an enrichment culture was used as an inoculum instead of activated sludge. PCR-DGGE was employed in the present study to investigate the microbial community in the enrichment culture that removed ClO4 autotrophically. Microorganisms in the enrichment culture showed 99.71% or more ClO4 removal efficiency after a 7-day incubation when the initial concentration was approximately 120 mg ClO4/l. Genomic DNA was isolated from the enriched culture and its inoculum (activated sludge), and used for PCR-DGGE analysis of 16S rRNA genes. Microbial compositions of the enrichment culture and the activated sludge were different, as determined by their different DGGE profiles. The difference in DGGE banding patterns suggests that environmental conditions of the enrichment culture caused a change in the microbial community composition of the inoculated activated sludge. Dominant DGGE bands in the enrichment culture sample were affiliated with the classes β-Proteobacteria, Bacteroidetes, and Spirochaetes. Further investigation is warranted to reveal the metabolic roles of the dominant populations in the ClO4 degradation process, along with their isolation.

Findings of Microbial Community Structure and Dominant Species in Soils Near Army Bases and Gas Stations (군부대와 주유소 주변에서 채취한 토양에서의 미생물 군집구조와 우점종의 파악)

  • This study examined microbial community structures (MCSs) according to environmental factors through DGGE analysis and comparison in various soils collected from near army bases and gas stations. As a result, the similarities based on DGGE band profiles showed the closer relationship in regional properties than in pollution characteristics, probably due to the degree of weak contamination. The highly contaminated samples with oil revealed low MCS similarities with others in the same region and very low with all the other samples in the other regions. Thus the microbial community structure would more be affected by region-based natural factors than by contamination factors in case of minor pollution. All the dominant culturable bacterial species were involved in firmicutes or high GC Gram+ in a major portion of soil samples and the highly oil-contaminated samples contained Arthrobacter, Bacillus, Methylobacterium, Clavibacter, Streptomyces and Nocardia as reported genera, and Leifsonia as a unreported genus.

  • An anaerobic PCE(tetrachloroethylene) dechlorinating bacterial culture from a landfill soil was enriched and characterized. The enrichment culture could dechlorinate 60$\mu$mol/$m\ell$ of PCE during a month of incubation and cis-DCE(cis-dichloroethylene) was observed as a main product of PCE dechlorination. Microbial analysis of the dechlorinating enrichment culture by rising PCR-DGGE (Polymerase chain reaction-Denaturing gradient gel electrophoresis) method showed that at least three microorganisms were related to the anaerobic PCE dechlorination.

  • The bacterial community of water stream, soil and guano in Gossi cave was examined by using PCR amplified the 16S rDNA-denaturing gradient gel electrophoyesis (DGGE). In this study, the genetic diversity and the similarity of bacterial community between open area and non - open area toy cave tour were investigated, and the seasonable variation pattern was compared each other. DGGE is attractive technique, as it sepayate same length dsDNA according to sequence variation typical 16S rDNA genes. The diversity and similarity of bacterial community in cave was analyzed by GC341f and PRUN518r primer sets foy amplification of V3 region of eubacteria 16S rDNA. The specific DGGE band profile of the cave water gives the possibility that the specific bacterial cell can be adapting to the specific cave environment and living in the cave. The DGGE band profiles of all samples with guano were compared and analyzed by image analyzer, in which mutual band profile was compared to be and the band intensity of guano was the highest. From these result, it is thought that the guano was main nutrient source and influenced on the community structure of the cave environment where is nutritionally limited. Pseudomonas sp. NZ060, Pseudomonas pseudoalcaligenes, uncultured Variovorax sp. and soli bacterium NS7 were identified to be on some sample from analysing DNA sequence of some DGGE band.

PCR-DGGE and PCR-RFLP Analyses of the Internal Trascribed Spacer(ITS) of Ribosomal DNA in the Genus Rhizopus

  • To estimate genetic relationships within the genus Rhizopus, genetic variations in 20 strains were investigated by DGGE and PCR-RFLP of rDNA ITS region (ITSI, ITS2,5.8S). The size of the amplified products showed the interspecific polymorphisms, 650 bp,700 bp, and 900 bp. The DGGE approach allowed the separation of PCR amplicons of the same length according to their sequence variations. When the rDNA ITS region was digested with six restriction enzymes, 20 strains were classified into five RFLP haplotypes. The range of similarity between the 20 strains by PCR-RFLP was 42.3-100%. Based on the results of DGGE aud PCR-RFLP, the 20 strains were divided into four groups, R. oryzae, R. stolonifer, R. microsporus and R. homothallicus. Further study of R. homothallicus is required.

Analysis of Microbial Diversity in Makgeolli Fermentation Using PCR-DGGE (PCR-DGGE를 이용한 막걸리발효에서 미생물 다양성 분석)

  • Kumjungsansung-Makgeolli$^{(R)}$ is a traditional Korean rice wine that is fermented from traditional nuruk and rice. In this study, we performed the PCR-denaturing gradient gel electrophoresis (DGGE) analysis targeting the 16S and 28S rRNA genes to characterize bacterial and fungal diversity during Makgeolli fermentation. The predominant bacteria in the PCR-DGGE profile during Makgeolli fermentation were Lactobacillus spp. (Lactobacillus curvatus, L. kisonensis, L. plantarum, L. sakei, and L. gasseri), Pediococcus spp. (P. acidilactici, P. parvulus, P. agglomerans, and P. pentosaceus), Pantoea spp. (P. agglomerans and P. ananatis), and Citrobacter freundii; these were identified on the base of analysis of 16S rRNA gene sequences. The dominant bacterium during Makgeolli fermentation was L. curvatus. The predominant fungi in PCR-DGGE profile during Makgeolli fermentation were Pichia kudriavzevii, Saccharomyces cerevisiae, Asidia idahoensis, Kluyveromyces marxianus, Saccharomycopsis fibuligera, and Torulaspora delbrueckii, and these were identified on the basis of analysis of 28S rRNA gene sequences. The dominant fungal species during Makgeolli fermentation changed from P. kudriavzevii at 0-2 days incubation to S. cerevisiae at 3-6 days incubation. This study suggests that PCR-DGGE analysis could be a suitable tool for the understanding of microbial diversity and structure during Makgeolli fermentation.

유류오염 지역내 지층 특성이 지하수 수질에 미치는 영향 연구

  • 유류누출지역의 수리지질, 수리지화학 및 미생물 분석을 통하여 지층특성이 지하수 흐름, 수질 및 미생물 특성에 미치는 영향을 고찰한 결과 지표에서 심도 $1.8{\sim}3.5m$ 구간에 수십 cm에서 2m 정도의 두께를 갖는 투수성이 상대적으로 양호한 지층이 존재함을 확인할 수 있었다. 이러한 지층의 존재에 의하여 상하부의 지하수대의 수두차이가 현저함을 관측하였으며 유류 수송관에서 누출된 유류가 이 투수성 지층을 따라 이동하여 이 구간의 토양 및 지하수가 오염되었다고 판단할 수 있다. 지하수 수질은 이러한 지층의 특성에 따라 다른 이온성분 및 동위원소 특징을 보여주었으며 이는 다변량통계분석에 의해서도 확인할 수 있었다. 미생물 DGGE 분석결과 역시 유사한 특징을 보여주어 이러한 수리지화학, 수리지질 및 미생물 특징이 서로 밀접한 상관성을 보여줌을 확인할 수 있었다.
