본 연구에서는 현장규모의 biopile을 제작하여 유류로 오염된 토양을 정화하였다. 오염토양의 TPH농도는 평균 2,800mg/kg(최고 3,590mg/kg)이었으며, 오염토양의 양은 $746m^3$이었다. Biopile $(17{\times}20{\times}2.5m)$을 약 3개월간 운전하며 일정 시간 간격으로 토양가스($O_2,\;CO_2)$ 및 VOCs) 및 토양의 TPH농도를 분석하였다. 지속적인 공기의 주입/추출에 의해 biopile내부는 호기조건을 유지하였으며, 미생물의 활성도 증가에 따라 이산화탄소의 농도가 증가하는 경향을 나타내었다. 또한 토양가스 분석결과 휘발성 유기오염물질(VOCs)은 약 40일 경과 후 90% 제거되었으며, 90일 경과 후 토양의 TPH 제거율은 98%로 나타났다.

유류로 오염된 토양에 대해 토양경작법(Landfarming)을 적용하여 복원효율을 평가하였다. 복원 초기에는 주로 휘발에 의한 오염물질의 저감이 이루어졌으며, 이 후 미생물성장에 필요한 토양뒤집기와 질소원 영양물질의 공급으로 인해 미생물수가 복원초기에 비해 약 200배 증가하면서 약 30여일 경과 후 초기 20,000ppm에서 복원목표인 TPH의 토양오염우려기준 2,000ppm의 이하인 1,910ppm으로 감소하였다. 시간이 경과할수록 탄소원 섭취기질 부족으로 완만한 오염물질 감소속도를 나타내면서 최종적으로 TPH를 측정한 결과 1,288ppm으로서 제거효율 94%이상을 나타내었다.

Since oil leakage is one of the most common nonpoint pollution sources that contaminate soil in Korea, the capacity of soil microbial community for degrading petroleum hydrocarbons should be considered to assess the functional value of soil resource. However, conventional methods (e.g., microcosm experiments) to assess the remediation capacity of soil microbial community are costly and time-consuming to cover large area. The present study suggests a new approach to assess the toluene remediation capacity of soil microbial community using a microbial diversity index, which is a simpler detection method than measuring degradation rate. The results showed that Shannon index of microbial community were correlated with specific degradation rate ($V_{max}$), a degradation factor. Subsequently, a correlation equation was generated and applied to Michaelis-Menten kinetics. These results will be useful to conveniently assess the remediation capacity of soil microbial community and can be widely applied to diverse engineering fields including environment-friendly construction engineering fields.
The objective of this study was to model the kinetics of S. aureus survival on high risk foods in school foodservice operations. After inoculating S. aureus ATCC25923 onto the various high risk foods, the effects of competitive microorganism, storage temperatures($25^{\circ}C$, $35^{\circ}C$), and initial contamination levels ($1.0{\times}10^2\;CFU/g$, $1.0{\times}10^5\;CFU/g$) on the growth of S. aureus were investigated. Lag time decreased and specific growth rate increased with a storage temperature ($25^{\circ}C$<$35^{\circ}C$) and with a higher initial inoculation level ($1.0{\times}10^2\;CFU/g$<$1.0{\times}10^5\;CFU/g$). Previously it was shown that S. aureus is a weaker competitor than other organisms, but it proliferates aggressively in a noncompetitive environment. However, in our study, when S. aureus was used to inoculate japchae (glass noodles with sauteed vegetables) and meat ball, the growth of S. aureus was similar and more active with competitive organisms than that without competitive organisms. Regardless of other factors, the initial level of S. aureus was a more significant factor of the growth. High inoculation levels of S. aureus were reached at 6 log CFU/g within 3 hours. An incubation temperature of $35^{\circ}C$ and the animal protein component of menu items also were identified as significant factors influencing the growth of S. aureus. Therefore, the duration of time meals are stored before serving should be considered a critical control point. Food service providers must control time and temperature to insure the safety of cooked foods.
There is growing interest in groundwater resources to overcome the loss of surface water resources due to climate change. An understanding of the microbial community of aquifers is essential for monitoring and evaluating groundwater contamination, as well as groundwater management. Most microorganisms that inhabit aquifer ecosystems are attached to sediment particles rather than planktonic, as is the case in groundwater. Since sampling aquifer sediment is not easy, groundwater, which contains planktonic microorganisms, is generally sampled in microbial community research. Although many studies have investigated microbial communities in contaminated aquifers, there are only a few reports of microbial communities in uncontaminated or pristine aquifers, resulting in limited information on aquifer microbial diversity. Such information is needed for groundwater quality improvement. This paper describes the ecology and community structure of groundwater bacteria in uncontaminated aquifers. The diversity and structures of microbial communities in these aquifers were affected by the concentration or distribution of substrates (e.g., minerals, organic matter, etc), in addition to groundwater characteristics and human activities. Most of the microbial communities in these uncontaminated aquifers were dominated by Proteobacteria. Studies of microbial communities in uncontaminated aquifers are important to better understand the biogeochemical processes associated with groundwater quality improvement. In addition, information on the microbial communities of aquifers can be used as a basis to monitor changes in community structure due to contamination.

PCBs를 분해하는 bphABC유전자를 plasmid vactor pMFB2에 유전자조작한 Pseudomonas putida AC30(pMFB2)를 PCBs에 오염된 연안해역의 해수와 저니로 만든 microcosm에 도입한 결과, 각각 도입 4일과 7일만에 사멸하였다. 그러나, 도입한 P. putida AC30(pMFB2)는 사멸하였지만, 연안해수와 저니 microcosm에서 plasmid pMFB2가 전이한 토착미생물이 검출되었다. 도입한 P. putida AC30(pMFB2)의 생잔실패의 원인을 분석한 결과 공경 0.2$\mu\textrm{m}$의 filter를 통과하는 물질과 생물이 가장 크게 영향을 미치는 것으로 나타났다. 유전자조작 P. putida AC30(pMFB2)의 도입과 bphABC유전자의 토착미생물로의 전이에 따른 토착미생물군집에 미치는 영향을 개체수 변동으로 조사한 결과, 토착미생물 군집에 미치는 영향은 보이지 않았다. P. putida AC30(pMFB2)의 도입에 의한 PCBs의 생분해성을 분석하였다. 그러나, 도입한 유전자조작 균주가 생잔에 실패함으로써 잔류하고 있는 PCBs의 농도변화는 보이지 않았다.
The effects on arsenic geochemistry of indigenous microorganisms isolated from an area contaminated with high concentration of arsenic were investigated. Arsenite exerted higher inhibitory effects on the microbes' growth than arsenate. During incubation of the microbes in an arsenate-spiked medium over 24 hours, decrease in microbial growth was observed as arsenate content increased. Arsenate of 150 mM or over apparently inhibited cell growth. However, further incubation for up to 4 days in the high arsenate concentration medium resulted in cell growth, implying that the microorganisms adjusted their biochemical functions to detoxify arsenic and maintain growth. Two types of microbes were observed during 20 hours to reduce arsenate to arsenite in solution through a detoxification mechanism. As well, decrease in the total arsenic content occurred over a 4-day incubation with the same microbes in an arsenate-spiked medium. Therefore it is suggested that microorganisms can influence arsenic speciation in natural settings and this may be applied to efficient bioremediation of arsenic-contaminated sites.
In this study, membranes were coupled to a sequencing batch reactor for simultaneous removal of organic matter and nitrogen, and the influences of MLSS (mixed liquor suspended solid) concentration and the sludge loading rate on membrane fouling and bioactivity were investigated. The amount of membrane fouling slightly increased with MLSS concentration at both non-aeration and aeration conditions, but effect of MLSS concentration was more significant at aeration condition. Although the effect of MLSS concentration on membrane fouling was found to be insignificant at low concentration level, extremely low sludge loading, which were generated by the maintenance of large amount of biomass in the reactor, caused severe membrane fouling, and air scouring effect decreased significantly in this condition. Specific bioactivity was constantly reduced as sludge loading rate decreased. In spite of high MLSS concentration over 17,000 mg/L, the activity of the reactor decreased at extremely low sludge loading rate presumably due to the lower oxygen transfer and the competition of biomass to deficient substrate.
We evaluated the microbiological quality of a facility in which freshly cut onions were prepared. The total plate counts on walls, equipment, and raw materials were ND (not detected) to $10^1$ CFU/100 $cm^2$, $10^0{\sim}10^3$ CFU/100 $cm^2$, and $10^3{\sim}10^4$ CFU/g, respectively. No coliforms were detected on walls however, coliforms were detected at concentrations of ND to $10^3$ CFU/100 $cm^2$ and $10^3{\sim}10^4$ CFU/g on equipment and raw materials, respectively. The total plate counts for falling and floating bacteria in the processing plant were ND to $10^0$CFU/plate and $10^1{\sim}10^2$$CFU/m^3$, respectively. Pathogenic microorganisms such as Escherichia coli, Salmonella spp, Staphylococcus aureus, and Listeria monocytogenes were not detected on walls, equipment, or raw materials. Overall, the results of the study indicate that hygiene control at the fresh-cut processing plant should be improved.
In this study, it was investigated that the isolation and identification of spoilage bacteria from contaminated aseptic packaged cooked rice, the potential for application of acidic electrolyzed saline solution (AESS) as water-for-cooked rice and the microbiological safety of AESS-based cooked rice. Five strains of Bacillus subtilis and a B. cereus strain among the total six isolates were partially identified by biochemical method and by Microbial Identification System (MIS). The bactericidal effect of AESS was similar as 0.1% NaOCl and 70% ethanol solution, or better than that. All of the test microorganisms except Bacillus spp. that were exposed to AESS for five seconds were destructed. The effect of AESS against Bacillus spp. was much better than that of the two solutions and all of them were destructed or inhibited on exposure for five minutes. The pH value of cooked rice prepared using AESS was in the range of 3.6 to 4.3 and was not almost changed through the storage period. Various concentrations of cell suspension of Bacillus isolates were inoculated to cooked rice, which were prepared with tap water and AESS, and stored at $37^{\circ}C$ for two weeks. The result was shown that the bacteria in tap water-based cooked rice appeared normal cell growth, while they were completely repressed in AESS-based cooked rice.
