Changes of microflora in processing and preservation of dried persimmon (곶감의 제조 및 저장 중 미생물상의 변화)

  • We investigated the change of microfloral states during drying processing and storage of dried persimmon. Initial stave of drying, a total bacteria counts of dried persimmon which was made the rural region of Sangju province were higher than that of downtown but after 7 weeks, there was no differents. A total cell count of bacteria decreased during drying processing but mold and yeast increased. In a storage test of dried persimmon at a different temprature, 3$0^{\circ}C$, room temp. 4$^{\circ}C$, respectively, low temprature storage was effectvie not only to retard the growth of microorganism but also maintain the suitable appearence.

A Study on the Types and Growth Patterns of Microorganisms and Quality Characteristics in Cherry Tomatoes and Head Lettuces According to Storage Period and Temperature (저장기간과 온도에 따른 신선편이 방울토마토와 양상추의 미생물 종류와 성장패턴 및 품질특성에 관한 연구)

  • The purpose of this study is to investigate the quality changes and contamination of microorganisms such as Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) in cherry tomatoes and head lettuces during the storage of different temperatures and periods. This study determines the pH levels, color changes and the growth patterns of microorganisms in cherry tomatoes and head lettuces during storage of 14 days at $5^{\circ}C$, $10^{\circ}C$ and $15^{\circ}C$. According to the results, the pH level is being reduced with storage periods in cherry tomatoes and head lettuces. The $L^*$, $a^*$ and $b^*$ values of cherry tomatoes are decreased with storage period, whereas the $a^*$ and $b^*$ values have increased with storage of the head lettuces. With regards to the types of microorganisms, the aerobic count plate (ACP), coliform count (CC), mold and yeast are being detected when cherry tomatoes and head lettuces are stored at $5^{\circ}C$, $10^{\circ}C$ and $15^{\circ}C$, whereas the S. aureus and E. coli are not being detected at 14 days of storage. The ACP, CC and yeast of cherry tomatoes increase with storage period, whereas the mold of cherry tomatoes was decreased after 14 days of storage. For the head lettuces, APC and CC have significantly increased with storage, whereas the mold stored at $5^{\circ}C$, $10^{\circ}C$ and $15^{\circ}C$ decreased after 21 days of storage. From these studies, we can confirm that changes in quality characteristics and the types of microorganisms existed in cherry tomatoes and head lettuces during storage were the ACP, CC, mold and yeast, whereas the E. coli and S. aureus are not detected.

Succession of bacterial community structure during the early stage of biofilm development in the Antarctic marine environment (남극 해양에서 생물막 생성 초기 단계의 세균 군집 구조 변화)

  • Compared to planktonic bacterial populations, biofilms have distinct bacterial community structures and play important ecological roles in various aquatic environments. Despite their ecological importance in nature, bacterial community structure and its succession during biofilm development in the Antarctic marine environment have not been elucidated. In this study, the succession of bacterial community, particularly during the early stage of biofilm development, in the Antarctic marine environment was investigated by pyrosequencing of the 16S rRNA gene. Overall bacterial distribution in biofilms differed considerably from surrounding seawater. Relative abundance of Gammaproteobacteria and Bacteroidetes which accounted for 78.9-88.3% of bacterial community changed drastically during biofilm succession. Gammaproteobacteria became more abundant with proceeding succession (75.7% on day 4) and decreased to 46.1% on day 7. The relative abundance of Bacteroidetes showed opposite trend to Gammaproteobacteria, decreasing from the early days to the intermediate days and becoming more abundant in the later days. There were striking differences in the composition of major OTUs (${\geq}1%$) among samples during the early stages of biofilm formation. Gammaproteobacterial species increased until day 4, while members of Bacteroidetes, the most dominant group on day 1, decreased until day 4 and then increased again. Interestingly, Pseudoalteromonas prydzensis was predominant, accounting for up to 67.4% of the biofilm bacterial community and indicating its important roles in the biofilm development.

Serological Analysis of the Antigenicity during Cultivation of Streptomyces Strains (Streptomyces 속 균주들의 생장에 따른 외피 항원성 변화의 혈청학적 분석)

  • The changes in antigenicity during cultivation of streptomycetes were determined by immunodiffusion assay and indirect ELISA. New precipitin lines in immunodiffusion assay began to appear in the growth period of the soluble pigment production and became thickened thereafter. The increase in the antigenicity was also confirmed by ELISA. The antigenic development was relatively weak for S. lavendulae and S. viridochromogenes while that was strong for S. lavendulae and S. viridochromogenes. The results indicated that Streptomyces strains, even though not proved for some strains, changed the compositions of cell envelope during submerged growth and this could be estimated quantitatively by serological method.

Total Microbial Biomass Measured by ATP in Three Marine Sedimentary Environments (아데노신 3인산(ATP; Adenosine-5′ triphosphate)을 이용한 심해저 및 연안퇴적토의 총 미생물 생체량 측정)

  • ATP concentrations far estimating total microbial biomass in the sediment were measured in three different marine sedimentary environments. ATP concentrations were highest in the surface sediment and decreased with increasing sediment depth and distance from the land. The results indicated that the benthic microbial biomass is primarily controlled by nutrient inputs from the overlying water column. Because of the longer residence time and adsorption to the sediment, the variations in organic carbon (OC) contents with sites and depths were not as distinct as that of ATP, and the correlation between OC and ATP was not significant in the coastal sediments. No significant correlation between OC and ATP in the coastal sediments also suggested that microbial biomass in the labile organic-enriched coastal sediment is suppressed by the grazing of higher trophic level such as meiofauna. Overall regional and vertical distribution of ATP indicated that h\`w can be a relevant tool for measuring total microbial biomass in various marine sedimentary environments.

Advanced Treatment of Shipboard Sewage by RCM Process with BM (복합미생물제제를 이용한 RCM공법의 선박오수 처리장치 적용에 관한 연구)

  • Lab scale experiment was carried out to study biological wastewater treatment technology developed for shipboard. RCM process using BM(Beneficial Microorganisms) was investigated for practical application on shipboard sewage treatment. RCM process is an environmental friendly treatment system, with minimum production of sludge. In the test, BOD5, CODcr, T-N and T-P were reduced a 96%, 97%, 78% and 81.68% respectively. From the result it was suggested that RCM process with BM might be a suitable process for shipboard sewage treatment, maintenance of useful microorganisms and creating special environment as the SDC tank is circulating in the non-biodegradable organics sludge generated during the wastewater treatment, thus reducing the amount of sludge generated. Therefore, the RCM process does not require additional equipment to strengthen it to meet the marine pollution standards.

Effect of Dandelion (Taraxacum Officinale) Extracts on the Intestinal Microorganisms of Streptozotocin-Induced Diabetic Rats (민들레추출물이 당뇨유발 흰쥐의 장내 미생물 균총에 미치는 영향)

  • This study was designed to test the effect of dandelion (Taraxacum officinale) extracts on the intestinal microorganisms of streptozotocin-induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively. Diabetes was induced by injecting streptozotocin (55 mg/kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45g of raw dandelion / kg diet for 50 days. The growth of Lactobacillus was more enhanced in dandelion leaf-water and leaf-ethylacetate extract group than that of the diabetic control group, whereas the growth of E. coli decreased. Results indicate that the dandelion extracts would be effective to improve the intestinal microorganisms.

Bacterial Abundance and Heterotrophic Activity in Sudong Stream (수동천에서의 세균의 분포와 생리적 활성도)

  • The density of heterotrophic bacterial population and heterotrophic activity were measured at monthly interval from March, 1986 to March, 1987 at four sites in the Sudong Stream, a tributary of North Han River. Total bacterial numbers and maximum uptake velocity ($V_{max}$) of glucose ranged as 0.8-$25.2\times 10^{5}$ cells/ml, 0.0006-24.39.$\mu$gC/1/hr, respectively. $V_{max}$ of glucose showed marked seasonal periodicity, with highest values in summer. But density of viable bacteria varied considerably, with no definite seasonal pattern. At the nucontaminated site which located in upstream, heterotrophic bacterial population and activities were relatively low, and small variations were observed downstream flowing except the site where animal originated sewage inputs occurred. And this large input of allochthonous materials and bacteria was an important factor for the stream condition.

  • Diacylglycerol kinase (DGK) phosphorylates the second messenger diacylglycerol (DAG) to phosphatidic acid and it may play a role in signal transduction in Escherichia coli as well as in eukaryotic cells. In addition, DGK is important for microorganisms to adapt to several physiological stimuli. In Bacillus subtilis, the effect of stress on dgk transcription was examined by northern hybridization. The high level of dgk transcription was induced against high osmolarity, low pH value and low temperature. Transcriptional analysis revealed that the dgk gene and dgk upstream locus (ORF2, ORF3 and ORF4) were transcribed as a polycistronic mRNA to form an approximately 2.5 kb transcript.

Nitrogen Removals according to Aeration/Non-aeration Periods in the Intermittent Aeration Reactor and Analysis of Microbial Community (간헐포기공정에서 포기/비포기 구간에 따른 질소제거 및 미생물 군집분석)

  • In this study, variations of the organic and nitrogenous compounds in wastewater were investigated in a single reactor with intermittent aeration. Over 90% of organic and nitrogen removals are accomplished with C/N ratio of 3 : 1 and 20/20 min of aeration/non-aeration period. Longer non-aeration period on the aeration/non-aeration cycle showed more stable nitrogen removal, showing various microbial community in the reactor. From PCR-DGGE analysis, it is conclusive that Dysgonomonas mossii strain Melo40, Eubacterium sp. oral clone JN088, Uncultured bacterium clone SPESB2_718, and Bacterium enrichment culture clone LE are related with the organics and nitrogen oxidation. Uncultured Acidobacteria bacterium clone AKYG487, Lactobacillus harbinensis strain FQ003, Erythrobacter litoralis strain Gi-3, Phytobacter diazotrophicus strain Ls8, and Mycobacterium sp. enrichment culture clone GE10037biofNNA are distinctly appeared under denitrification condition.
