Effect of commercial sanitizers on microbial quality of fresh-cut iceberg lettuce during storage (세척용 시판 살균제 종류에 따른 신선편의 양상추의 저장 중 미생물 변화)

  • This study was investigated the effects of various commercial sanitizers on microbial characteristics in fresh-cut iceberg lettuce during storage. For screening sanitizer, lettuce was cut and dipped in chlorine water ($0.2ml{\cdot}L^{-1}$), solution of organic acids such as ascorbic acid, citric acid, acetic acid, mixture of ascorbic acid and acetic acid (1-6%), and solutions of commercial sanitizers such as Formula 4$^{TM}$ (1,3,4%), Fresh produce wash$^{TM}$ (1,3,4%), Cleancol$^{TM}$ (1%), Chitochol$^{TM}$ (1%) and Natural Ca$^{TM}$ (0.1%) for 3 min, respectively. Washing lettuce with selected sanitizers resulted in reduction of aerobic bacteria of more than 2 log CFU/g. Initial pH of lettuce was related with the pH of sanitizers. pH ranged from 4.7 to 6.1 in Formula 4 (4%, pH 1.7) and Natural Ca (0.1%, pH 12.0), respectively. Chlorine water showed consistent and significant inhibition effect in all of microorganisms except total coliform. Over 3% of Formula 4 and Fresh produce wash were found to have high bactericidal activity among sanitizers. The sanitizers of chlorine water, Fresh produce wash, Chitochol and Natural Ca were effective in reducing yeast and mould populations. As coliform and E. coli, Formula 4 (4%) showed the highest bactericidal activity. The bactericidal effect of commercial sanitizers during storage varied with the kinds and concentrations of tested sanitizers. Although inhibition effect was not showed during storage, these results suggest that commercial sanitizers could be an alternative to chlorine for washing fresh-cut produce.

Change of Microbial Communities in Kimchi Fermentation at Low Temperature (김치의 저온 발효 중 미생물 변화 양상)

  • The diversity and change of microbial communities during kimchi fermentation at $4^{\circ}C$ were analyzed by denaturing gradient gel electrophoresis (DGGE). Kimchi samples were taken every 5 days over the fermentation periods (for 60 days) to extract total DNA for DGGE analysis. Touchdown polymerase chain reaction was performed to amplify the V3 region of 16S rRNA gene. Sequencing results of partial 16S rDNA amplicons from DGGE profiles revealed that lactic acid bacteria (LAB), especially Weissella koreensis, Lactobacillus sakei and Leuconostoc gelidum were dominants in kimchi fermentation at $4^{\circ}C$. And we knew that W. koreensis steadily existed throughout the whole fermentation period, also Lb. sakei and Leuc. gelidum appeared from 10th day and 30th day of fermentation time, respectively and then these species were to be dominant microorganisms.

Changes in Physico-chemical and Microbiological Parameters during Active Composting of Cattle Manure (우분 퇴비화의 주발효과정 중 이화학적 및 미생물학적 파라미터의 변화)

  • Various physico-chemical and microbiological parameters of a composting system were compared with respect to their potential use for the monitoring and evaluation of composting processes for cattle manure. The temperature changed within a range of $30-65^{\circ}C$ during the whole composting process, and the period of active composting (>$40^{\circ}C$) persisted for 16 days. The concentrations of total carbon, total nitrogen, and organic matter decreased by 15% during active composting, but significant changes in C/N ratio were not observed. The decrease of temperature in the latter period of active composting caused a decrease of $NH_4^+-N$ and an increase of $NO_3^--N$ in the composting pile. When temperature exceeded $50^{\circ}C$, the population of thermophiles was higher than that of mesophiles by more than 1 or 2 orders of magnitude. Correlation analyses showed that amylase activity correlated positively with the population of mesophiles and reducing sugar content, but negatively with the population of thermophiles. Amylase activity was higher at the beginning of active composting, whereas cellulase, xylanase and ligninase activities which showed close relationship with each other, increased continually during active cornposting, suggesting the distinction of temporal niches between amylose-degrading and lignocellulose-degrading bacteria in the same habitat.

복합미생물제제를 이용한 RCM공법의 선박오수 처리장치 적용에 관한 연구

  • 본 연구에서는 유입수의 변동이 심하고 전문가가 부재한 환경인 선박에서 발생하는 오수의 효과적인 처리를 위하여 RCM공법을 선박오수처리장치에 적용하는 실험실 규모의 실험을 수행하였다. 질소 인의 고도처리 효율과 선박이라는 특수환경과의 접목성을 검토한 결과 RCM공정에 유효미생물을 주입하는 방법은 선박환경에 적합한 것으로 평가되었다. 또한 RCM공정은 활성슬러지 공정에서 배출되는 슬러지는 배출시키지 않고 슬러지액화분해조(SDC)에서 재분해하여 순환함으로써, 최근 해양투기가 금지됨으로 인해 문제가 되고 있는 슬러지의 발생량을 최소한으로 하여 친환경적인 수처리가 가능하다. 복합미생물제제 주입 후 미생물 관찰결과 고도처리에 유리한 미생물종의 출현을 확인하였으며 이들의 상호기작으로 질소 인의 처리에 도움을 주어 처리효율이 높은것이라 판단된다. 유기물 제거효율 실험결과 $BOD_5$, CODcr T-N, T-P의 처리효율이 각각 96, 97, 78, 81.68 %로 나타나 Membrane이나 Filter없이도 강화되어가는 해양오염기준을 충족시킬 수 있는 공정으로 판단된다.

옻나무 추출액의 급여가 육계의 생산성 및 장내미생물에 미치는 영향

  • 참옻나무 추출액이 육계의 생산성과 장내미생물 변화 및 영양소 소화율에 미치는 영향을 구명하고자 1일령 육계(Ross) 960수를 공시하여 평사에서 5주간 사양시험을 실시하였다. 증체량은 추출액 급여구가 첨가수준이 증가할수록 증가하는 양상을 보였으나 통계적 유의성은 인정되지 않았다. 사료섭취량 및 사료요구율 역시 차이가 없었다. 장내미생물 변화에서 맹장내 유산균, E. coli 및 Salmonella는 처리간 차이가 없었다. 영양소 소화율은 추출액 첨가구가 향상되는 결과를 보였으나 통계적 유의성은 인정되지 않았다.

Characteristics of temperature variation in the collection area of Amanita virgineoides (흰가시광대버섯(Amanita virgineoides) 발생지역의 기온변화 특성)

  • Amanita virgineoides Bas is an ectomycorrhizal mushroom belonging to the family Amanitaceae in the order Agaricales. This fungus usually grows solely on damp soil in coniferous or deciduous forests from summer to autumn. Although Korean people consume A. virgineoides, some studies indicate that it can be poisonous. During our biodiversity survey of four specific regions i.e., Yongin, Eumseong, and Chungju from 2016 to 2018, A. virgineoides specimens were collected on the sixth rotation. We analyzed the temperature, precipitation, relative humidity, and vegetation surrounding the A. virgineoides fruiting bodies to reveal the environmental factors that influenced mushroom formation. It was found to usually grow in August, the hottest season in Korea. For 60 days before forming the fruiting body, temperatures were found to be between 20 and $30^{\circ}C$. More evidence is necessary to ensure that change in temperature is the major environmental factor that determines A. virgineoides fruiting.

  • Effects of nitrogen souses and C/N ratio were investigated on the production of extracellular microbial surfaclant, sophorolipid, from C. bombiocola. Organic nitrogen sources, such as urea, peptone and yeast extract was found to be more effective for sophorolipid production, than inorganic nitrogen sources. Depending on the nitrogen sources, sophorolipid production pattern varied by increasing C/N ratio. Increased production of sophrolipid could be obtained up to 90g/L by feeding carbon source again 2 days after cultivation.

Changes of Microbial Community Depending on Different Dissolved Oxygen in Biological Nitrogen Removal Process (생물학적 질소제거 공정에서 용존산소변화에 따른 미생물의 군집변화)

  • PCR-DGGE method was applied to analyze changes of microbial community in simultaneous nitrification and denitrification (SND) bioreactor with various DO concentrations. In the analysis of eubacterial community, band profiles of DGGE were similar with 2 or 1 mg/L DO concentrations in the reactor. Experimental results led to 16 different bacteria being identified, including 5 dominant strains(3 strains of Uncultured Bacterium, 1 strains of Bacillus, 1 strains of Uncultured Bacteroidetes). DGGE results at 0.5 mg/L DO concentration led to 12 strains being identified, including 7 dominant strains(5 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). DGGE results at 0.1 mg/L DO concentration led to 11 strains being identified, including 3 dominant strains(1 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). In DGGE band profiles of $\beta$-AOB($\beta$-Ammonia Oxidizing Bacteria), only one band was observed. This band had 97% similarity with Nitrosomonas sp. done DNB Y20. This band was clearly observed at the 2, 1 and 0.5 mg/L DO concentrations, while the brightness of the band at 0.1 mg/L DO concentration was mostly dimmed. In DGGE band profiles of denitrification process, 5 bands(3 strains of Uncultured organism containing nirS, 2 strains of Uncultured organism containing nirK) were observed. Among those bands, the brightness of one band was gradually increased at the lower DO concentrations. This band has 86% identity with Uncultured organism clone eS1 cd1 nirS gene, partial cds. Based on this result, it could be concluded that Uncultured organism clone eS1 cd1 nirS gene, partial cds is a predominant microorganism in the denitrification process.

Characteristics and Distribution of Microorganisms in a Rice Straw Compost for Cultivation of Button Mushrooms (Agaricus bisporus) (양송이 재배에서 볏짚 배지의 발효 단계별 관여 미생물의 분포양상 및 특성)

  • In this study, we analyzed the densities and taxonomic characteristics of various microorganisms that play important roles in Agaricus bisporus culture medium composting, and examined changes in the levels of decomposition-related enzymes secreted by these microorganisms. Various microorganisms such as thermophilic bacteria, actinomycetes, fluorescent Pseudomonas spp., and filamentous bacteria are closely associated with culture medium composts of Agaricus bisporus. The population densities of microorganisms change, and harmful bacteria disappear during thermophilic composting. Psychrobacter sp., Pseudomonas sp., Bacillus sp., and Pseudoxanthomonas sp. accounted for the highest proportion of bacteria in the culture media during outdoor composting, whereas Bacillus sp. and Psychrobacillus sp. were dominant after pasteurization. Cellulose and hemicellulose enzymes of the microorganisms were important at an early stage of rice straw composting and after decomposition of carbon sources, respectively. Microorganisms that secreted these enzymes were present in the second and third turning stage of composting.

Changes in the oral environment during four stages of orthodontic treatment (교정치료 4단계 동안의 구강 내 환경의 변화)

  • Objective: To identify clinical, salivary, and bacterial changes during orthodontic treatment with follow-up to 24 months. Methods: In 30 patients, clinical (decayed, missing, and filled surfaces [DMFS], O'Leary's plaque index, and plaque pH), salivary (unstimulated and stimulated saliva, buffer capacity, pH, and occult blood), and bacterial (Streptococcus mutans and Lactobacillus) markers were evaluated. A questionnaire was employed to evaluate their hygienic-dietary habits. Data were analyzed by ANOVA, logistic regression and Spearman's correlation. Results: DMFS increased significantly, whereas the plaque index diminished, plaque pH was more acidic (p = 0.23), and unstimulated salivary flow showed significant differences during the treatment (p = 0.013). Stimulated saliva flow increased in females after the placement of appliances; buffer capacity was diminished in males during the therapy; salivary pH remained at basal values. Bacterial levels and occult blood increased to high-risk levels and were not statistically significant different between genders (p > 0.05). Two major relationships were confirmed: initial plaque with use of dental aids (r = 0.429; p = 0.018) and final DMFS with unstimulated salivary flow (r = -0.372; p = 0.043). Conclusions: The increase in retentive surfaces increased the bacterial levels, plaque pH became acidified, and gingival damage was greater. Buffer capacity was altered but maintained a healthy salivary pH during the treatment.
