A Study on Whitening and Anti-inflammatory Effects of Eriobotrya Japonica Leaf Extracts with Different Extraction Methods (추출 방법에 따른 비파엽 추출물의 미백 및 항염활성에 관한 연구)

  • In this study, we prepared Eriobotrya japonica leaf extracts by several extraction processes and then evaluated their biological activities for their potential application as a new raw material of functional cosmetic. Their whitening effects were measured by tyrosinase inhibitory activities, and anti-inflammatory effects were determined by inhibitory activities of nitric oxide (NO) production. Among the several extracts obtained from E. japonica leaf, supercritical fluid extract showed tyrosinase inhibitory activities at the concentration of 10%. Inhibitory activity on NO production effect related to anti-inflammatory efficacy was in the order: supercritical fluid extract > ethanol extract > hot water extract. According to the results of MTT assay, cell cytotoxicity was not observed at all concentrations except for a 5% concentration of the 70% ethanol extract. For whitening effects, 30% ethanol and 70% ethanol extract showed mushroom tyrosinase inhibitory activity at the concentration of 5%. These results indicated that E. japonica leaf extracts could have the functional effects when they are added as ingredients in cosmetics.

  • 목적 : 상지, 상백피, 상엽은 모두 뽕나무에서 유래한 것으로 한의학에서 수천 년 동안 사용되어왔다. 최근 이들의 미백효능이 발견되면서 더욱 주목을 받고 있다. 피부의 멜라닌 색소 침착은 자선에 대한 정상적인 방어기전이다. 그러나 비정상적인 색소침착은 심각한 미용적 문제를 가져온다. 최근 약침이 피부 미백에 이용되고 있다. 또한 발효와 열처리는 미백화장료 가공의 최신 경향이다. 이 연구에서는 상지, 상백피, 상엽 약침액이 발효와 열처리 후에 피부 미백에 어떠한 영향을 미치는지 알아보고자 한다. 방법 : 상지, 상백피, 상엽을 각각 4개의 그룹으로 나누었다. (1) 비발효, 비열처리, (2) 비발효, 열처리, (3) 발효, 비열처리, (4) 발효, 열처리한 그룹들에 타이로시나제 활성 및 도파 산화 활성, 그리고 멜라닌 함량을 측정하여 비교하였다. 결과 : 타이로시나제 활성 및 도파 산화 활성, 그리고 멜라닌 함량에 있어서, 발효, 열처리가 대체로 효과가 있는 것으로 나타났다. 특히 멜라닌 함량에 있어서 발효, 열처리한 군이 대조군에 비해 유의성 있는 감소를 보였다. 결론 : 주근깨, 흑색종, 간반(肝斑) 등 비정상적인 색소 침착 질환을 치료하기 위한 상지, 상백피, 상엽 약침액 제조에 있어, 발효 및 열처리 방식을 적용하면 보다 나은 미백효능을 발휘할 수 있을 것으로 생각된다.

Whitening Effect of Androsace umbellata Extract (봄맞이 추출물의 미백 효능 연구)

  • The purpose of this study is to identify the whitening effect of Androsace umbellata extract. To discover the anti-pigmentation effective, we investigated Androsace umbellata extract on tyrosinase and melanogenesis inhibition. As results, it reduced tyrosinase activity and melanin contents in B16F1 melanoma cells in a dose-dependent manner with decreased to about 32% at a concentration of $25{\mu}g/mL$. To reveal how it works in inner-cellular level, we performed Western blot method. We found out that it also inhibited the protein expression in tyrosinase, tyrosinase related protein 1 (TRP-1), and microphthalmia associated transcription factor (MITF) in melanocytes. Therefore, we successfully identified the whitening effect of Androsace umbellata extract, and this finding suggested that Androsace umbellata extract is a considerable potent cosmetics ingredient for whitening. Based on this, we anticipated further researches about Androsace umbellata extract for gene levels and additional mechanisms to develop not only for functional cosmetics but for medicines or healthcare food.

Anti-inflammatory and Tyrosinase Inhibition Effects of Amaranth (Amaranthus spp L.) Seed Extract (아마란스 씨앗 추출물의 항염 및 Tyrosinase 억제 효과)

  • This study examined the anti-inflammatory and whitening effects of Amaranth (Amaranthus spp L.) seed extract. Amaranthus spp L. seeds were extracted using 70% ethanol and then fractionated sequentially with n-hexane, dichloromethan, ethyl acetate and butanol. For the study of anti-inflammatory activity in RAW 264.7 cells, EtOAc fraction of Amaranthus spp L. seeds significantly inhibited nitrogen oxide production as well as the protein level of iNOS. Furthermore, EtOAc fraction of Amaranthus spp L. seeds inhibited expression of $TNF-{\alpha}$, PGE2 and the protein level of COX-2 in a dose-dependent manner. Inaddition, the tyrosinase inhibitory activities of the Amaranthus spp L. seed 70% ethanol extract and subfractions were also measured to see if these extracts can be used as an ingredient for whitening cosmetics. Tyrosinase is an oxidase that is a rate-limiting enzyme for controlling the production of melanin. Therefore, tyrosinase inhibitors have become increasingly important in cosmetics and medical products with regards to hyperpigmentation. EtOAc fraction of Amaranthus spp L. seeds showed mushroom tyrosinase inhibitory activity in a dose-dependent manner. This activity was more potent than that of a positive control cynandione A. These results suggest that Amaranthus spp L. seeds may be a valuable natural ingredient for the food and cosmetics industries.

Evaluation of Alcea rosea L. Callus Extract as a Natural Cosmetic Ingredient (접시꽃 캘러스 추출물의 천연화장품 원료로서의 효능 평가)

  • In this study, we investigated the biological effects of Alcea rosea L. callus extract for the development of natural cosmetics ingredients. The antioxidant activities of A. rosea L. callus extract was measured through DPPH, ABTS and FRAP assay. As a result, A. rosea L. callus extract were found to have a strong antioxidant ability in a dose dependent manner. In addition, A. rosea L. callus effectively reduced the intracellular oxidative stress induced by AAPH at a concentration of 10 mg/mL. In a tyrosinase activity assay, we found that A. rosea L. callus extract reduced tyrosinase activity by 51% at 10 mg/mL. Based on these results, A. rosea L. callus extract is considered as a promising natural ingredients for cosmetics with antioxidant and whitening functions.

  • Novel whitening agents were prepared using peptide-Natural origin compound derivatives. The peptide could be an antagonist of MC1R and Natural origin compound were well-known material as a Tyrosinase inhibitor. We also suggest the new assay method which could evaluate the Antagonistic effectiveness to MC1R using cAMP signaling pathway. 24 candidates were synthesized and 11 peptide derivatives were selected by cAMP assay method. To evaluate cAMP assay, the selected peptide derivatives were assayed to evaluate their melanogensis inhibitory activity. At this work, we could know that the sequences which include -RW- have a melanogensis inhibitory activity, and cAMP assy could use as a evaluating method of MC1R antagonist. But, to evaluate the whitening activity of some material, cross-checking with melanin inhibitory assay method was recommended.

Studies on Whitening and UV Damage Protection Effect of the Pterin Compounds (테린계 화합물의 미백 및 자외선 손상방어 효능에 관한 연구)

  • (6R)-5,6,7,8-tetrahydrobiopterin ($6-BH_4$) cofactor is essential for various process, and is present in probably every cell or tissue of higher organism. $6-BH_4$ is required lot various enzyme activities, and for less defined functions at the cellular level. And it is well known about the antioxidant effects as a non-protein compound. Recently, scientists proposed another roles for $6-BH_4$ in melanogenesis. $6-BH_4$ is a well known tyrosinase inhibitor. In this study, we found that methyl-$BH_4$ and $6-BH_4$ have antioxidant activities and inhibitory activity for melanin synthesis. These pterin compounds were not toxic in HaCaT and B16F10 cells and showed scavenging activity against DPPH radicals. We also showed that pterin compounds decreased protein levels of tyrosinase and TRP-1. In a clinical test, pterin compounds showed the significant skin whiteining effect after treatment for 3 weeks. Furthermore pterin compounds significantly suppressed the UVB-induced expression of $PGE_2$ and IL-6 genes induced UVB In HaCaT and inhibited UVB-induced melanogenesis in B16F10 cells. These results showed the effect of pterin compounds as a cosmeceutical ingredient.

  • Melanin plays an important role in protecting human skin from UV radiation and determines the race and skin color. Melanin is also major target for developing skin-whitening cosmeceuticals. Recently, as the market size of skin-whitening cosmeceuticals has rapidly expanded, related researches and developments are also focused on maximizing the safety and efficacy. Also, patents of skin-whitening materials have been increasing steadily for ensuring the competitive power. Patent also shows the research trend of industry and institutes. In this review, we analyze the trend of research and development based on the patent application of skin-whitening cosmeceuticals.

  • 목적 : 본 연구는 합환피약침액(合歡皮藥鍼液)이 사람 피부 섬유아세포의 콜라게나제 활성 및 프로콜라겐 합성에 미치는 영항과 티로시나제 활성에 미치는 효과를 측정하고자 실시하였다. 방법 : HS68 사람 정상 섬유아세포에 UVB 조사 후 합환피(合歡皮) 약침액(藥鍼液)가 type I procollagen 생성과 콜라게나제 효소활성에 미치는 효능과 티로시나제 효소활성에 미치는 효능을 평가하였다. 결과 : 합환피약침액(合歡皮藥鍼液)은 UVB 조사된 세포의 콜라게나제 효소활성을 통계적으로 유의하게 억제하였고, 티로시나제 활성을 통계적으로 유의하게 억제하였다. 그러나 티로시나제 억제활성의 정도는 미백효능으로 활용하기에 약간 약한 경향이 있었다. 결론 : 합환피약침액(合歡皮藥鍼液)의 콜라게나제 억제효능은 주름개선 약침치료에 활용이 가능할 것으로 생각된다.
