Bioefficacy Evaluation of Non-edible Parts of Aronia melanocarpa for the Use of Functional Biomaterials (기능성 바이오소재 활용을 위한 아로니아 비가식 부위 추출물의 생리활성 평가)

  • Total polyphenol content and biological activities of water and methanol extract from Aronia melanocarpa berry, leaf, stem and twig were analyzed to investigate the potential for a bio-functional material. The polyphenol content of leaf and twig extracts were higher than those of berry and stem extracts. DPPH radical and ABTS radical scavenging activities of both extracts, were measured in order of twig, leaf, berry and stem. Tyrosinase inhibitory activities of water extract were measured in order of berry, leaf, twig and stem. Those of methanol extract were measured in order of twig, leaf, berry and stem. The nitrite-scavenging ability of water and methanol extract were measured in order of berry, leaf, twig and stem. Consequently it was shown that the non-edible parts of aronia, leaf and twig, had antioxidant activities and nitric oxide scavenging activities. It is expected that these materials could be used as functional bio-materials in bio-health care products including cosmetic products.

Effects of Astragali Radix Herbal Acupuncture Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity (황기 약침액의 사람 섬유아 세포에서 Collagen 생성과 Tyrosinage 활성에 미치는 영향)

  • 목적 : 본 연구는 황기약침액의 collagen 대사과정과 tyrosinage 활성에 대한 효과를 관찰하여 항노화 약침소재로의 개발 가능성을 알아보고자 계획되었다. 방법 : 황기 약침액이 사람 정상 섬유아세포 HS68에 UVB 조사 후 type I procollagen의 생산량 회복과 collagenage 활성에 미치는 효과를 ELISA법을 이용하여 측정하였다. 황기약침액의 tyrosinage 활성도에 미치는 영향을 측정하였다. 결과 : 황기 약침액은 사람 섬유아세포 HS68 실험에서 UVB 조사로 감소된 type I procollagen생성을 회복시키고, 통계적으로 유의하게 collagenage 활성을 억제하는 것을 관찰하였고 tyrosinage 활성을 통계적으로 유의하게 억제하지만, L-DOPA 산화는 억제하는 경향이 있는 것을 관찰하였다. 결론 : 본 연구 결과에 의하면, 황기약침액은 collagenage 활성을 억제하고, tyrosinage 활성을 억제하므로 주름개선과 미백효과가 있어 미용약침 소재로 개발가능성이 있을 것으로 사료된다.

Effect of Fruit Extract of Prunus mume on the Scavenging Activity of Reactive Oxygen Species and Melanin Production in B16F1 Cells (매실추출물이 활성산소종 소거효과와 B16F1 세포에서 멜라닌 생성에 미치는 영향)

  • Prunus mume has been traditionally used as a medicinal food in Korea, Japan, and China. In particular, this fruit has been reported to have beneficial biological effects on gastritis and gastric ulcers. However, its action in relation to skin whitening has remained unclear. Accordingly, the effects of fruit extract of P. mume related to antioxidation and skin whitening were examined in this study. First, using the MTT assay, it was observed that fruit extract of P. mume below 0.1% has no cytotoxicity in B16-F1 cells as a result of cell viability. Second, the direct scavenging effects and the reducing power of the fruit extract of P. mume were evaluated in vitro on DPPH radicals, hydrogen peroxide, and superoxide. It exhibited high reducing power and scavenging activity on the aforementioned reactive oxygen species. Furthermore, we found that its protective effect against genomic DNA damage related to oxidative stress was increased in a dose-dependent manner. In addition, the fruit extract of P. mume had an inhibitory effect on melanin production induced by L-dopa. In addition, it reduced the expression level of NRF-2, SOD-1, and SOD-2 related to antioxidation in western blot analysis. These results suggest that fruit extract of P. mume could exert a whitening effect through inhibition of melanin production by its antioxidant effect.

Effect of the Ethanol Extract from Lavandula vera on ${\alpha}$-MSH Induced Melanogenesis (라벤더 에탄올 추출물이 ${\alpha}$-MSH 유도 멜라닌 생성에 미치는 효과)

  • Down-regulation of melanin synthesis is required for recovery of pigmentary disorders and it is well known that ${\alpha}$-MSH induced melanin synthesis and dendrite outgrowth on melanocytes. This study was conducted to evaluate the depigmenting properties of ethanol extract from a Lavandula vera. The ethanol extract from Lavandula vera inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the ethanol extract of Lavandula vera effectively suppressed the ${\alpha}$-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the ${\alpha}$-MSH-induced mRNA expression of tyrosinase was significantly attenuated by Lavandula vera treatment. These results suggest that Lavandula vera exerts its depigmenting effects through the suppression of tyrosinase and cytoplasmic dendricity. And it may be a potent depigmetation agent in hyperpigmentation condition.

Skin Whitening and Anti-Wrinkle Effects of Extract from Jubak of Oriental Herbal Liquor (한방 발효주 주박 추출물의 미백 및 피부 주름 개선 효과)

  • Oriental herbal liquor (Yakju) is a type of Korean traditional alcoholic beverage that uses Nuruk and oriental herbs for fermentation. The purpose of this study was to develop cosmetic ingredients using Jubak, which is a by-product of alcoholic fermentation of oriental herbal liquor. To investigate antioxidant, whitening, and anti-aging effects of Jubak, we prepared extract of Jubak and its solvent fractions. Ethyl acetate fraction (KSD E4-3) showed the most prominent free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH)] scavenging activity ($SC_{50}$: 0.75 mg/mL). KSD E4-3 significantly inhibited in vitro mushroom tyrosinase activity ($IC_{50}$: 0.82 mg/mL) and reduced the melanin contents in mouse melanoma melanocyte, B16F10 cells. KSD E4-3 down-regulated protein expression of tyrosinase related proteins (TRP)-1, -2, which play key roles in melanogenesis. For anti-aging effects, inhibition of matrix metalloproteinase (MMPs) expression was evaluated using human keratinocyte, HaCaT cells. Treatment of HaCaT cells with KSD E4-3 reduced expression of MMP-1, -2, -9 and inhibited proteolytic activities of MMP-2, -9. These results suggest that KSD E4-3 induces down-regulation of cellular melanogenesis and protects against photoaging induced by UVB-induced damage. Thus KSD E4-3 could potentially be a valuable cosmetic ingredient.

Whitening Effect and Skin Regeneration Effect of Red Sea Cucumber Extract (홍해삼 추출물의 멜라닌 형성 억제를 통한 미백효과 및 피부 재생효과에 관한 연구)

  • Recently, several researchers have been developing cosmetics from natural ingredients for skin whitening and anti-aging products. The red sea cucumber (RSC), Apostichopus japonicas, is a species of sea cucumber in the family stichopodiae, which is widely distributed in China, Japan, and Korea. To use Red Sea Cucumber as a cosmetic ingredient, its inhibitory effects on melanogenesis and the anti-aging effects of RSC extracts were investigated. First, a tyrosinase activity assay was performed, which showed that RSC inhibited tyrosinase activity at a concentration of $200{\mu}g/ml$. An MTT assay was carried out to evaluate cell toxicity, and the results showed that RSC extract has no cytotoxicity in HaCaT cells. Furthermore, the mRNA expression levels of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), microphthalmia-associated transcription factor (MITF), and matrix metalloproteinase (MMPs) genes treated with RSC extract in B16F10 and HaCaT cells decreased. Moreover, a wound-healing assay was performed to identify the cell regeneration effect of RSC extracts. Also, a skin turnover effect was confirmed by creating a three-dimensional cell culture with HaCaT and human fibroblasts. Altogether, the results suggested that Red Sea Cucumber may possess a high ability to induce whitening and anti-wrinkle effects as a cosmeceutical ingredient.

Synthesis of 2',4'-Dimethoxyflavone and Its Anti-pigmentation Effect (2',4'-Dimethoxyflavone의 합성과 멜라닌 생성 저해 효과)

  • We synthesized 2',4'-dimethoxyflavone and investigated the effects on melanogenesis. To determine the effects as a whitening agent, various in uitro tests were performed such as free radical scavenging activity, melanin assay, tyrosinase activity and expression of tyrosinase, TRP-1 and TRP-2 (western blot and RT-PCR) in Bl6 melanoma cells. 2',4'-Dimethoxyflavone showed neither free radical scavenging activities against 1,1- diphenyl -2-picrylhydrazvl (DPPH) radical and inhibition of mushroom tyrosinase activity, 2',4'-dimethoxyflavone significantly inhibited melanin production in B16 melanoma cells. 2',4'-Dimethoxyflavone treatment (48h) suppressed the biosynthesis of melanin up to 27% at $5{\mu}g/mL$ and reduced tyrosinase activity up to 20% at $5{\mu}g/mL$ in B16 melanoma cells. 2',4'-Dimethoxyflavone was also able to significantly inhibit tyrosinase and TRP-1 expression in protein and mRNA level. These results suggest that 2',4'-dimethoxyflavone inhibits melanin biosynthesis at the level of enzyme activity and protein mRNA expression B16 melanoma cells. Therefore, 2',4'-dimethoxyflavone may be useful as a new whitening agent in cosmetics.

Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening (왕겨초액의 대량생산과 항노화 및 미백 효과)

  • Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.

Inhibitory Effects of Polyopes affinis Ethanol Extract on Melanogenesis in B16F10 Melanoma Cells (참까막살 에탄올 추출물이 B16F10 흑색종 세포에서의 멜라닌합성에 미치는 영향연구)

  • Polyopes affinis is a kind of red algae found in the South coast and near Jeju Island of Korea. The purpose of this study was to investigate the effects of Polyopes affinis ethanol extract (PAEE) on melanogenesis in ${\alpha}-MSH$ stimulated B16F10 melanoma cells. Melanoma cells were cultured for 72 hr treated with PAEE. Total melanin content and the activity of tyrosinase, a key enzyme in melanogenesis, were measured. When the melanin content in B16F10 melanoma cells was tested, PAEE was decreased in a dose-dependent manner: treatment with 100, 300, and $500{\mu}g/ml$ caused 25%, 30%, and 35% reduction, respectively. Treatment of 100, 300, and $500{\mu}g/ml$ of PAEE caused 6%, 12%, and 21% reduction of tyrosinase activities in B16F10 melanoma cells. Also, PAEE suppressed the expression of tyrosinase, tyrosinase-related protein-1, tyrosinase-related protein-2, and melanocyte-inducing transcription factor in B16F10 melanoma cells. A concentration of $500{\mu}g/ml$ of PAEE showed a greater decrease in tyrosinase activity, melanin content, and melanogenic enzyme protein expression. These results indicate that PAEE inhibits melanin synthesis and tyrosinase activity, and Polyopes affinis ethanol extract could be used as a functional whitening agent.

Synthesis and Biological Evaluation of Water-Soluble Oleanolic Acid Derivatives for use as Melanogenesis Inhibitors (멜라닌 생합성 억제제로서 수용성 Oleanolic Acid 유도체의 합성 및 활성 평가)

  • This study was focused on the synthesis of methoxy polyethylene glycol-oleanolic acid ester (mPEG-OA derivative) and investigation of its water solubility and anti-melanogenic effects. mPEG-OA derivative was identified by 1H and 13C NMR and FT-IR spectroscopic measurements. The water solubilities of mPEG-OA derivative and OA were found to be 13 and 0.013 mg/mL and that of mPEG-OA was found to be 1000-fold higher than that of OA. The effects of mPEG-OA derivative and OA on cell viability were measured using B16F10 melanoma cells. The viability of cells treated with mPEG-OA derivative (250 μM) increased 4-fold compared to that of cells treated with OA (62.5 μM). At mPEG-OA derivative and OA concentrations where the cell viability was unaffected, the inhibitory effect of mPEG-OA derivative and OA on the melanogenesis in B16F10 melanoma cells were 36 and 35% at 50 and 10 μM, respectively. The expression level of microphthalmia-associated transcription (MITF) was also reduced in B16F10 melanoma cells treated with mPEG-OA and OA. Overall, mPEG-OA derivative showed excellent water solubility and inhibitory effects of the melanogenesis, which could be used as a potential formulation for use in whitening functional cosmetic material.
