Isolation and Tyrosinase Inhibitory Activity of Wild Yeasts Obtained from Soil in the Fields of Medicinal Plants, Ginseng and Korean Angelica (인삼과 당귀 재배 토양으로부터 야생효모들의 분리 및 미백성 Tyrosinase 저해활성)

  • The goal of this study was to isolate wild yeasts from the fields of medicinal plants and investigate its tyrosinase inhibitory activities. Wild yeasts isolated from soil in the ginseng and Korean angelica fields of Geumsan, Chungcheongnam-do, Korea were identified by comparing the nucleotide sequences of the D1/D2 domain of 26S rDNA. In total, 43 yeast strains belonging to 21 species were isolated from 50 soil samples obtained from two medicinal plant fields. From the ginseng field, six strains of Rhodotorula glutinis and four strains of Sampaiozyma ingeniosa were isolated, out of which Rhodotorula glutinis strains were dominant. In the Korean angelica field, six strains of Cyberlindnera saturnus, three strains of Piskurozyma taiwanensis, and three strains of Saitozyma podzolica were isolated, out of which Cyberlindnera saturnus strains were dominant. We prepared cell-free extracts of the isolated wild yeasts and their tyrosinase inhibitory activities were investigated. Among 43 yeast strains, cell-free extracts of Naganishia globosa G1-7 showed the highest tyrosinase inhibitory activity (28.0%).

Potent whitening effects of rutin metabolites (루틴 대사체의 미백 효능)

  • The aim of this research was to investigate the whitening effects of rutin and rutin metabolites including 3,4-dihydroxyphenyl acetic acid (DHPAA), 3-hydroxyphenyl acetic acid (HPAA), 3,4-dihydroxytolene (DHT) and homovanillic acid (HVA). The potent whitening effect of rutin and rutin metabolites were determined by mushroom tyrosinase inhibition assay and expressed as the half maximal inhibitory concentration ($IC_{50}$) against tyrosinase activity in vitro. The HVA showed the highest inhibitory effect ($IC_{50}=37.10{\mu}M$) of tyrosinase activity, followed by DHPAA ($IC_{50}=45.87{\mu}M$), HPAA ($IC_{50}=50.96{\mu}M$), rutin ($IC_{50}=57.98{\mu}M$), and DHT ($IC_{50}=66.09{\mu}M$), respectively. To evaluate cell cytotoxicity, MTT assay was performed with JB6 P+ mouse epidermal cells and expressed as a relative percentage of untreated control. The results showed that rutin and rutin metabolites had no cytotoxic effects on JB6 P+ cells up to $100{\mu}M$ except for DHT (up to $50{\mu}M$). These results suggests that rutin metabolites may be utilized as a potential tyrosinase inhibitors and the whitening agents for the future.

Whitening Effects of Adamantyl Benzamide Derivatives (Adamantyl Benzamide 유도체의 미백효과)

  • The structure activity relationship of polyhydroxylated benzamide derivatives for whitening effects was examined. The adamantyl benzamide derivatives with catechol (3,4-dihydroxyphenyl) of B-ring part showed good anti-melanogenesis activity, but the inhibitory activity of mono-hydroxyphenyl (3-OH or 4-OH) or 3,4-dimethoxyphenyl substituted derivatives was decreased or lost. Therefore the catechol unit was appeared to be the crucial factor for the inhibition of melanogenesis. And the existence of 2-OH of A-ring part had minor influence on the activity, the length of carbon chain between A-ring and B-ring was also not the major factor for the anti-melanogenesis activity.

Antioxidant and Anti-Melanogenic Activities of Hyssopus officinalis Extracts (히솝 추출물의 항산화 효과 및 멜라닌 생성 저해효과)

  • Hyssopus officinalis is a herbaceous plant of the genus Hyssopus. Due to its properties as an antiseptic, cough reliever and expectorant, it is commonly used as an aromatic herb and medicinal plant. This study was performed to investigate the anti-oxidative and anti-melanogenic properties of Hyssopus officinalis extracts (HE) using in vitro assays and cell culture systems. As a result, HE showed higher DPPH and ABTS radicals scavenging activity in a dose-dependent manner. Also, HE inhibited the prodution of intracellular ROS and melanin contents in B16F10 melanoma cell as well as tyrosinase activity. We also found that HE inhibit mRNA expression of MITF, tyrosinase and TRP-2 gene. These findings suggest that HE may be beneficial for preventing oxidative damage and melanogenesis of skin.

A Study on the Physiological Activity of Agastachis herba Extract and Fermented Products as Cosmetic Materials (화장품소재로서 곽향 추출물 및 발효물의 생리활성 연구)

  • Agastachis herba is so similar in use to herbs that it is called Korea Mint in the West. Western herbs are commonly used as ingredients in cosmetics. However, in Korea, Agastachis herba is primarily used as a medicinal herb. This study aims to compare the effectiveness of one cosmetic ingredient with another cosmetic ingredient. Antioxidant, whitening, polyphenol, flavonoid, and antibacterial experiments were conducted on lactobacillus, yeast ferments, essential oils, and floral water using extracts from Agastachis herba. Antioxidants exhibited significant effects in all samples, while essential oils displayed the highest efficacy in terms of whitening, polyphenol content, and antibacterial properties. Flavonoids exhibited the strongest effect in the 70% ethanol extract. The essential oil had the most significant functional impact. However, it was confirmed that the fermented products that underwent yeast fermentation with extracts were as effective as essential oils.

Inhibition of Melanogenesis by Ramalin from the Antarctic Lichen Ramalina terebrata (남극 지의류 Ramalina terebrata로부터 분리된 라말린의 미백효과)

  • Ramalin (${\gamma}$-glutamyl-N'-(2-hydroxyphenyl)hydrazide) isolated from the Antarctic lichen Ramalina terebrata has been shown to have strong antioxidant activities in the previous study. To investigate additional activities of ramalin, we studied the effects of ramalin on melanogenesis in melan-a cells, a non-tumorigenic melanocyte cell line. At a non-cytotoxic concentration, ramalin dramatically decreased melanin synthesis in melan-a cells in a dose-dependent manner, which was more potent than arbutin, a well-known tyrosinase inhibitor. Ramalin inhibited cell-free tyrosinase activity directly and intracellular tyrosinase activity as well. Its inhibitory mechanisms on melanin production were further assessed, and we found that ramalin significantly decreased the protein levels of melanogenic enzymes such as tyrosinase, tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2). However, the mRNA levels of these enzymes were not altered. In a clinical study, application of 0.2 % ramalin on human skin significantly improved the degree of skin brightness after 3 weeks. In conclusion, ramalin has strong anti-melanogenic activity that is exerted both by the direct inhibition of tyrosinase activity and by down-regulation of melanogenic proteins. Furthermore, ramalin showed skin brightening effect in a clinical study. Collectively, these results suggest that ramalin may be a useful inhibitor for melanogenesis in skin.

Inhibitory Effect of Active Compounds Isolated from Impatiens balsamina(Garden Balsam) for Melanogenesis (봉선화 내에 함유 된 레톡시나프타퀴논의 미백활성에 미치는 영향)

  • 본 논문은 피부미백에 관한 실험적 연구로서, 봉선화(Impatiens balsamina Linne)의 성분 중에서 하이드록시-나프타-퀴논(hydroxynaphthoquinone)이 멜라닌 생성억제 효능, 색소침착 저해 효과 등의 미백 활성이 있는 것으로 밝혀졌다. 피부흑화(melanogenesis)는 멜라닌 색소생성세포(melanocyte)에서 자외선 등의 자극에 대한 방어기작으로 멜라닌 생성 활동이 증가되고 이로 말미암아 만들어진 다량의 멜라닌이 각질형성세포(keratinocyte)로 전이되어 피부 표피층에 축척된 결과이다. 비록 멜라닌이 피부에 보호작용을 하나 피부의 과색소 침착은 기미, 주근깨, 피부염증 후의 피부흑화, 노인성 색소반점 등을 일으키며 이로 인해 피부미용 상의 불편뿐만 아니라 정신적으로 부정적인 영향을 미쳐 사회활동에 불편을 초래하기도 한다. 멜라닌 생성 과정은 아미노산의 일종인 티로신(tyrosine)에 티로시나제(tyrosinase)라는 효소가 작용하여 도파(DOPA), 도파퀴논(dopaquinone)으로 바뀐 후 비효소적인 산화 반응을 거쳐 만들어 지며, 이것이 피부 내에 이상 침착하여 기미, 검버섯 등이 생기는 것이라고 알려져 있다. 이와 같은 색소침착, 기미, 반점 등의 완화, 예방 및 치료에는 멜라닌 생성을 억제하는 물질, 예를 들면 하이드로퀴논(hydroquinone), 알부틴, 비타민 C 및 그 유도체 등이 개발되어 사용되고 있으나 이 중, 하이드로퀴논은 일단 효과가 인정되고 있지만 감작성이 있기 때문에 일반적으로 사용이 제한되고 있다. 아스콜빈산은 쉽게 산화되어, 이를 배합한 제품에는 변색, 변취가 되는 문제를 야기하고, 식물추출물 유래의 물질들은 식물의 산지에 따라 효능의 차이가 심하여 제품의 균질성이 유지되기 어렵다. 이러한 이유로 많은 멜라닌 생성을 억제하는 물질을 개발하기 위한 연구들이 계속 진행하고 있다. 2-히드록시-[1,4]나프토퀴논 유도체는 다음과 같은 구조를 나타낸다. 상기 구조에서 R은 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬로서, 바람직하게는 C1 내지 C10의 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬이고, 보다 바람직하게는 C1 내지 C5의 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬이다. 실험에 활용된 화합물은 다음과 같은 반응식으로 얻어졌다. 2-히드록시-[1,4]나프토퀴논과 포타슘카보네이트(K2CO3)를 아세토니트릴에 용해한 후, 저온수조로 냉각하고 알킬할라이드를 적당한 당량비로 천천히 적하하여 히드록시기와 에테르(ether) 반응을 통하여 얻어질 수 있다. 상기 반응식에서 R은 C1~10의 포화 또는 불포화된 알킬기로서 직쇄형 또는 분지쇄형 모두 가능하다. 상기 화합물인 2-히드록시-[1,4]나프토퀴논(2-hydroxynaphthoquinone)은 배양된 쥐의 멜라노마 세포에 대하여 하이드로퀴논과 대등한 멜라닌 생성 억제효과를 보였다. 하이드로퀴논은 저농도에서 강력한 멜라닌 생성 억제효과를 보이지만 상기 화합물들은 보다 낮은 농도에서도 세포독성을 나타내지 않으며 하이드로퀴논보다 높은 멜라닌 생성 억제 효과를 보였다.

Anti-Inflammatory and Whitening Effect of the Lyophilized Powder of Oriental Plant Extracts Fermented with Streptococcus thermophilus (Streptococcus thermophilus로 발효한 한약재 발효분말의 항염증 및 미백 효과)

  • To identify new anti-inflammatory and whitening material, this study investigated the whitening and anti-inflammatory effects of the lyophilized powder from 6 oriental plant extracts (OPE; Citrus junos Tanaka, Mori cortex Radicis, Schisandra chinensis Baillon, Coix lachrymajobi var. mayuen, Angelica gigas NAKAI, and Sophora japonica L.) fermented with Streptococcus thermophilus by assessment of cytotoxicity on human dermal fibroblast, inhibitory effect of nitric oxide (NO) prodction, tyrosinase activity and melanin formation. The OPE was fermented with Streptococcus thermophilus at $37^{\circ}C$ for 2 days and the lyophilized powder was manufactured by freezing-dryer. OPE didn't show cytotoxicity at concentration of $500{\mu}g/mL$ using a cytopathic effect reduction method. OPE also exhibited inhibitory effect on nitric oxide (NO) prodction by Griess reagent system. Furthermore, OPE showed inhibitory effect on tyrosinase activity with dose dependent manner, and exhibited significant inhibition of melanin formation by measurement of melanin from culture media (p < 0.05). From these results, 6 OPE extracts showed anti-inflammatory and whitening effect and may be used as an active ingredient for cosmetics.

Development of Cosmeceutical Cosmetics Using Enzyme Bio-Conversion System (효소 생전환 시스템을 이용한 기능성 화장품 개발)

  • This study is about the cosmeceutical products using enzyme induced bio-conversion system. In general, ascorbic acid (AA) has the higher reducing activity and can be used for various purpose in the cosmetics. But it is very unstable in the aqueous system and difficult to maintain its stability in the cosmetics product. 2-O-$\alpha$-D-Glucopyranosyl-L-ascorbic acid (AA2G) is the stabilized form of AA and showed the less whitening activity than AA. In this study, we developed bio-conversion system improving the stability and efficacy of AA2G and AA, respectively. In this system, AA2G (over $80\%$) can be converted to AA and glucose within 30 min. The converted product showed higher anti-tyrosinase activity like AA (AA2G showed no anti tyrosinase activity) and depigmenting activity in the artificial tanning test. From these results, we could conclude this system is a brand new method to increase the activity of AA and maintain its stability.

Antimelanogenic Effect of Ligularia fischeri, Solidago virga-aurea, Aruncus dioicus Extracts from Ullung Island in Murine Melanoma Cells (마우스 악성흑색종세포에서 울릉도 곰취, 미역취, 삼나물 추출물의 멜라닌 생성 억제 효과)

  • The objective of the present study was to evaluate the skin whitening effect of the extracts of 3 herbs, Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus, which were collected from Ullung island. Tyrosinase inhibition activities were 33% in pre-fermented extracts and 45% in post-fermented ones. When tyrosinase activities in B16F10 murine melanoma cells were tested, activities in pre- and post-fermented extracts were 41 and 56.5%, respectively. Thus, the post-fermented extracts might have greater skin whitening effects. The protein expression of MITF, TRP-1, TRP-2, and tyrosinase, which are all skin-whitening related transcription factors, showed that both pre- and post-fermented herbs inhibited protein biosynthesis in B16F10 melanoma cells. Post-fermented herb extracts especially showed a greater decrease of protein expressions. The expression of MITF, a regulatory transcription factor, was also decreased by both extracts but was greater in the post-fermented ones. From the results, it can be concluded that the 3 herb extracts from Ullung island may inhibit melanin biosynthesis by the suppression of MITF activity in a signaling pathway. Results indicate that the post-fermented herbs tested in the present study had skin whitening activities and can be used as functional ingredients for food and cosmetic compositions.
