Antioxidant and Nitrite Scavenging Activities of Acanthopanax senticosus Extract Fermented with Different Mushroom Mycelia (버섯균사체를 달리한 발효가시오가피 추출물의 항산화 활성 및 아질산염 소거능)

  • This study was designed to investigate the antioxidant activities (ORAC value, DPPH and ABTS radical scavenging activity, FRAP activity, and reducing power) and nitrite scavenging activities of Acanthopanax senticosus extracts fermented with the mycelia of three different mushroom species: Ganoderma lucidum, Phellinus linteus, and Hericium erinaceus. The highest total phenol content (42.09 GAE mg/g) and ORAC value ($74,912{\mu}M$ TE/g) were observed in a hot water extract of A. senticosus fermented with G. lucidum. The highest DPPH radical and nitrite scavenging activities were observed in a 70% ethanol extract of A. senticosus fermented with G. lucidum. In addition, ABTS radical scavenging activity (8-88%), FRAP activity (0.1-0.2), and reducing power (0.3-0.7) were increased by ethanol addition in all samples in a dose-dependent manner. These results provide a basic understanding of the antioxidant and nitrite-scavenging activities of A. senticosus extracts fermented with different mushroom mycelia.

Antibacterial and Antioxidative Activities of Various Extracts from Basidiomycetes (담자균 추출물의 항균작용 및 항산화작용)

  • The water and methanol extract were obtained from fruit body of 8 kind of edible mushrooms. The antibacterial activity of extracts on the growth of pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O 157:H7 and Salmonella typhimurium) was determined. Methanol fraction of Gyrophora esculenta showed excellent antibacterial activity alai t 5 strains of pathogenic bacteria. The 80% methanol extract of Gyrophora esculenta and Phelinus were fractionated with diethylether, chloroform, ethyl acetate, butanol and water. The diethylether, ethyl acetate and butanol fractiorl of Gyrophora esculenta had excellent antibacterial activity and ethyl acetate and butanol fraction of Phellinus linteus had weak antibacterial activity against 5 strains of pathogenic bacteria. Electron donating ability of each fraction of Gyrophora esculenta was increased in order of ethylacetate, chloroform, butanol, diethylether and water. Nitrite scavenging ability was observed in ethyl acetate fraction of Gyrophora esculenta and other fractions showed no activities.

Antioxidant and DNA Damage Protective Activities of Freeze-Dried Blue Mussel (Mytilus edulis) (동결건조 진주담치 추출물의 항산화 및 DNA 손상 보호 활성)

  • Blue mussels (Mytilus edulis) are widely distributed among the world's oceans in various habitats. The purpose of this study was to investigate the effects of freeze-drying on the antioxidant and antigenotoxic activities of blue mussels collected in the Gyeongnam coast area of Korea. Raw (RM) and freeze-dried blue mussel flesh (FRM) were extracted with ethanol, methanol, and water. Antioxidant activities were evaluated on the basis of DPPH radical scavenging activity, oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC), and antigenotoxic activity (comet assay). Except for the water extract, RM and FRM showed DPPH radical scavenging activities, which increased upon freeze-drying in MeOH extract. The highest ORAC value was observed in water extract of RM and MeOH extract of FRM. CAC was protected against AAPH-induced oxidative stress in HepG2 cells by both RM and FRM extracts. Freeze-drying lowered ORAC value of water extract, whereas it increased CAC activity, suggesting that antioxidant activities varied according to the generated radicals. All extracts from RM and FRM showed antigenotoxic activities by reducing $H_2O_2$-induced DNA damage in human leukocytes. Freeze-drying had no effect on antigenotoxicity of blue mussels. Taken together, these results indicate that blue mussels possess antioxidant and antigenotoxic properties, and freeze-drying might be a useful processing method for blue mussels to retain their maximum physiological potential as a functional food.

Production of Bioactive Components and Anti-Oxidative Activity of Soybean Grit Fermented with Bacillus subtilis HA according to Fermentation Time (Bacillus subtilis HA를 이용한 soybean grit의 고체발효 기간에 따른 생리활성물질 생산 및 항산화 효과)

  • Soybean grits, fortified with various bioactive components, were produced by solid-state fermentation using Bacillus subtilis HA. ${\alpha}$-Amylase activity gradually increased during fermentation over 5 days. Fibrinolytic and protease activities were highest in the soybean grits fermented for 7 days. The grits fermented for 5 days also showed the highest tyrosine content, indicating a higher peptide content. Peptides of low molecular weight (below 3,000 daltons) and browning pigments increased with increasing fermentation time. The fermented soybean grits showed higher contents of total phenolic compounds, to approximately 18 mg/g. DPPH free radical scavenging effects were higher in the soybean grits fermented for 3 days. Also, ABTS radical scavenging effects were greater in the fermented grits compared to the unfermented grits. Overall, the soybean grits fermented by solid-state fermentation for 5 days showed enhanced production of bioactive compounds and greater antioxidant properties.

식품 발효중 기능성 물질의 생전환 및 기능성 변화

  • 본 연구에서는 프로바이오틱스로 사용하는 유용 미생물과 식용으로 안전하게 사용 가능한 미생물 자원을 이용하여 배당체의 전환 연구를 실시하여 전환체의 새로운 생리활성 작용에 대하여 연구하였다. 연구에 사용한 주요 소재로는 인삼의 사포닌 배당체, 길경의 platycodin 배당체, 호로파의 사포닌 배당체, 칡과 두유의 이소플라본 배당체 등이었다. 사용 균주로는 bifidobacteria, lactobacilli, leuconostocs, yeasts, aspergilli 등이었다. 유산균 중에는 Bifidobacterium sp. Int-57의 전환능이 우수하였고 식용 곰팡이 중에는A. niger의 전환능이 우수하였다. 다양한 미생물 또는 효소를 이용한 전환체에 대하여 아질산 소거능, 항산화능, a-glucosidase와 a-amylase 저해능 등을 조사하였을 때 일반적으로 전환 후의 활성이 중가하였다. 길경 사포닌은 전환 후에 세포 독성이 줄어들었으며 관능성이 유의적으로 개선되었다. 결론적으로, 프로바이오틱스와 식용 미생물을 이용하여 천연물 중에 존재하는 배당체의 생리활성 및 관능성을 증가시킬 수 있어 새로운 기능성 식품의 개발에 활용할 수 있을 것이다.

Physicochemical Properties and Biological Activities of Protaetia brevitarsis seulensis Larvae Fermented by Several Kinds of Micro-organisms (유용 미생물을 이용한 발효굼벵이 추출물의 이화학적 특성 및 생리활성효과)

  • In this study, the biological activities of aqueous, ethanol, and methanol extracts of larvae of the edible insect Protaetia brevitarsis seulensis, fermented using several kinds of microorganisms, were tested in in vitro experimental models. Six effective microorganisms were used for fermentation, namely Lactobacillus plantarum JBMI F3, Lactobacillus plantarum JBMI F5, Lactobacillus gasseri Ba9, Aspergillus kawachii KCCM 32819, Saccharomyces cerevisiae KACC 93023, and Bacillus subtilis KACC 91157. Biological activities (${\alpha},{\alpha}^{\prime}-diphenyl-{\beta}-picrylhydrazyl$ [DPPH] free radical scavenging activity, reducing power, and fibrinolytic activity), and biochemical properties (phenolic compounds and flavonoids) were examined in aqueous, ethanol, and methanol extracts from P. brevitarsis seulensis powder and fermented P. brevitarsis seulensis powder. The total phenolic compounds and flavonoid contents were highest in the aqueous extract of B. subtilis-fermented P. brevitarsis seulensis powder. DPPH radical scavenging activity and reducing power were stronger in the fermented group than the nonfermented group. Fibrinolytic activity were highest in the extract from B. subtilis-fermented P. brevitarsis seulensis powder. The ${\alpha}-amylase$ activity in starch was higher in the fermented group than the nonfermented group, but there was no significant difference. These results provide basic data to understand the biological activities of bioactive materials derived from fermented P. brevitarsis seulensis larvae for the development of functional foods.

Physicochemical Characteristics and Antioxidant Activities of Fermented Garlic Husk (발효 마늘껍질의 이화학적 특성 및 항산화 활성)

  • This study was carried out to evaluate the possibility for functional characteristics of garlic husk and its products fermented by Lactobacillus plantarum. Each water and 80% ethanol extracts made from garlic husk and its fermented products were tested for antioxidant activities. Total mineral content of fermented garlic husk was 2,056.23 mg/100 g and its content was 1.5 times higher than the unfermented sample. Total phenol contents were the highest in the ethanol extracts made from fermented garlic husk. Activities of electron donating, reducing power and NO radical scavenging in the ethanol extracts of fermented product were significantly higher than the unfermented sample. ABTS radical scavenging activity of fermented garlic husk was higher than the unfermented sample. Also, its activity was higher in the ethanol extracts than the water extracts. Nitrite scavenging activity of garlic husk was significantly different between fermented and not fermented sample. Because antioxidative effects of fermented garlic husk were elevated, these results recommended its utilization as a functional materials for animal feed.

Evaluation of radical scavenging and diasestive enzyme inhibitory capacities of peach twigs fraction extract (Prunus persica L. Bastch) (복숭아 나뭇가지 분획추출물의 라디칼 소거 및 소화효소 저해활성 평가)

  • We investigated the free radical scavenging and digestive enzyme inhibitory activities of the hot water extract of peach twig (Prunus persica L. Bastch). This extract of the peach twigs was further split up into n-hexane, ethyl acetate (EtOAc), and n-butyl alcohol(n-BuOH), which resulted in three solvent-soluble fractions. Free radical scavenging activity was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+) assay systems, while hypoglycemic effect of the peach twig extract and the solvent-soluble fractions were tested using α-glucosidase and α-amylase inhibition assays. Accordingly, the EtOAc layer showed a greater free radical scavenging activity compared to other solvent-soluble fractions. Furthermore, based on the α-glucosidase and α-amylase assays, the IC50 values were determined to be 38.2±1.6 and 69.6±6.1 ㎍/mL for the EtOAc-soluble fractions, respectively. Taken together, these results suggest that the fractions obtained from the peach twig extract can be considered as a potential source of natural antioxidant and hypoglycaemic constituents.

Antioxidative and Anti-Diabetes Activity, and Free Amino Acid and Mineral Contents of Beverage with Gugija (Lycii fructus) Extracts (구기자 추출물 함유 음료의 산화방지활성, 항당뇨효과, 유리아미노산과 무기질 함량)

  • Free amino acid and mineral analysis of beverages with Gugija (Lycii fructus) extracts performed to determine the antioxidative and anti-diabetes activities of the beverages. The four major free amino acids in the water- or ethanol-extracts of the Gugija beverages were asparagine (115.23, 51.95 mg%), methionine (20.02, 22.07 mg%), aspartic acid (19.65, 13.72 mg%) and taurine (18.64, 22.44 mg%). The mineral contents of the Gugija beverages with water- and ethanol-extracts were in the order K>Mg>Na>Ca>Zn>Fe. Antioxidant activity determined by DPPH and ABTS radical scavenging activity. There was no significant difference in DPPH and ABTS radical scavenging activities between the water-extract beverages and the ethanol-extract beverages, although more phenolics and flavonoids were found in the ethanol-extract beverage. Higher anti-${\alpha}$-glucosidase activity was observed in the ethanol-extract beverage compared to acabose, which was used as the control and is an inhibitor of ${\alpha}$-glucosidase, suggesting that the Gugija beverage with ethanol extract could be a potential hypoglycemic agent.

  • The increase in the elderly population and growing interest in skin beauty, alongside specialized scientific examination of natural substances, have led to a rise in the utilization of natural materials. This paper investigates the types of natural substances, the functionality of extracts, and extraction technologies based on the literature that utilizes natural ingredients to mitigate skin aging. To directly assess the impact of functional materials derived from natural products on skin aging mitigation, the functionality of the extracts was determined based on their ability to synthesize Procollagen and reduce MMP-1. Each natural product is categorized into plants, herbal medicines, and microalgae in the literature utilizing the above evaluation methods. This paper describes the extraction technologies for securing functional materials from each ingredient and the main outcomes, providing trends in research on extraction technologies for functional materials aimed at alleviating skin aging
