Production of Pyruvate Dehydrogenase Complex-E2 Specific Human Monoclonal Antibody in Fed-batch Culture Systems with High Cell Density Recombinant Escherichia coli (고농도 재조합 대장균의 Fed-batch 배양 시스템을 이용한 Pyruvate Dehydrogenase Complex-E2 특이성 인간 모노클론 항체의 생산)

  • Several culture systems including batch, two-stage CSTR, semi-fed batch, and two-stage cyclic fed-batch were investigated for the efficient production of the Fab fraction of PDC-E2 specific human monoclonal antibody using high cell density recombinant E. coli. A two-phase batch system and a two-stage continuous system were examined to overcome plasmid instability problems, by separating the growth and the production stages. The cell density and productivity of the two-stage continuous culture was better than that of the two-phase batch fermentation. In the two-stage continuous culture system with DO-stat, the cell growth and the productivity were superior to those of the system without the DO control. Also, almost total plasmid stability was maintained in the two-stage continuous culture system. Modified M9 medium was selected as an optimum feeding medium for the fed-batch process, and the optimum C/N ratio determined to be 2:3. The optimum feeding rate was $0.6g/\ell/hr$ for a constant feeding strategy in semi-fed batch system. When the feeding medium was fed by pulsing, it was observed that more frequent pulsing resulted in improved cell growth. The linear feeding method was the most efficient of the various feeding methods tested. Finally, high cell density culture using a two-stage cyclic fed batch system with pH-stat was tried because the linear feeding method showed limitations in terms of obtaining high cell densities, and a cell density of $54 g/\ell$ was achieved. It was concluded that the two-stage cyclic fed batch system was the most efficient system for high cell density culture of the systems tested. However, productivity improvements were lower than expected due to the extremely high accumulations of acetate, although the low levels of residual glucose were maintained.

A Study of 'Yokagura of Takachiho': A sacred Music and Dancing Performed in Takachiho, the Land of Japanese Myths (일본신화의 고장 다카치호(高千穗)의 요가구라(夜神樂))

  • A ritual ceremony accompanied by a song and dance is called as Kagura in Japan. The word Kagura is originally derived from Kamukura, which means the place where spirits reside. However, later it came to refer to the whole process of the ritual ceremony itself. Through the field studies, this paper examines Takachiho-kagura, helded in Takachiho, known as the village of myths, located in northern Miyazaki-ken in Japan. Kagura takes place all night through in each and every village in Takachiho and it normally runs from the end of November when the harvest season ends until early February the next year. One ordinary house is especially chosen for this ceremony, in which Kagura performs 33 repertoires. Takachiho-kagura is sometimes called as Yokagura, since the performance takes place over-night. A song and dance is performed by ordinary village people called hoshyadong, who inherited Takachiho-kagura. Currently, the ceremony is held in more than 20 villages and designated by the government as "Important Intangible Folk Cultural Property" in Japan. Takachiho-kagura follows the example of rituals held in Sada shrine in Izumo, which is now eastern Hiroshima-ken. It is the dance using a bell, a sword, a fan, which signify Norimono and is usually combined with mask-play called as Shinno. From the shrine of village, itwelcomes the gods who defense their village, called Ujikami and other 8 million gods, called Yaoyorozunokami, in Kagurayado, where Kagura of 33 repertoires is played in order. Kagura starts from dances for attendance of the gods, Hikomai, Daidono, Kamioroshi, performs dances of Amanoiwato, the gate of heaven's cave in Japanese myths, Dazikara, Uzume, Dotori, and continues dances for the old ghosts Shibahiki, Yatsubachi mixed with acrobatics. Finally, this performance ends with dances to send off the gods, Hinomae, Gurioroshi, Gumooroshi, until the dawn the next morning. This paper explores Takachiho-kagura from the perspective of folk performance with ethnography. These days, ecstasy and oracle do not happen in Takachiho-kagura. However, it kept the old form of folk performance as Kagura held in ordinary house. Especially, in Takachiho, remarkable venue of the Japanese myths, Takachiho-kagura is developed artistically. The first field study was held in Gokamura, Iwato-zone and Ashakabe, Mitai-zone between December 6th and December 12th 1997. Afterwards, the second field study was conducted in the area of Shiba and Ashakabe from December 17th until December 19th 1997 and from December 1st until December 10th 2000.

Thermotropic Liquid Crystalline Behavior of α,ω-Bis(4-nitroazobenzene-4'-carbonyloxy)alkanes (α,ω-비스(4-니트로아조벤젠-4'-카보닐옥시)알칸들의 열방성 액정 거동)

  • A homologous series of linear liquid crystal dimers, ${\alpha},{\omega}$-bis(4-nitroazobenzene-4'-carbonyloxy)alkanes (NATWESn, n = 2~8, 10, the number of methylene units in the spacer) have been synthesized, and the thermal behavior of the series has been investigated. All the dimers formed enantiotropic nematic phases. The nematic-isotropic transition temperatures of the dimers and their entropy variation at the phase transition showed a large odd-even effect as a function of n. This behavior was rationalized in terms of the change in the average shape of the spacer on varing the parity of the spacer. The thermal stability and degree of order in the nematic phase and the magnitude of the odd-even effect of NATWESn were very similar to those of the corresponding ether compounds, while they were significantly different from those of the monomesogenic compounds, 4-{4'-(nitrophenylazo)phenoxy}alkanoyl chlorides and the side-chain liquid-crystalline polymers, the poly[1-{4-(4'-nitrophenylazo) phenoxycarbonylalkanoyloxy}ethylene]s. The results were discussed in terms of the 'irtual trimer model'by Imrie.

Characteristics of Wheat Germ Oil during Enzymatic Ethanolysis in Supercritical Carbon Dioxide (초임계 이산화탄소에서 밀배아유의 효소적 에탄올화 반응 특성)

  • Enzymatic ethanolysis of wheat germ oil with immobilized lipase was investigated for enhancing the function of wheat germ oil. Ethanolysis reactions were carried out in two different systems; non-pressurized and pressurized system. In non-pressurized system, the enzymatic ethanolysis was carried out in an erlenmeyer flask(25 ml) containing a mixture of wheat germ oil and 99.90% ethanol using 1~5 wt% immobilized lipase as Lipozyme TL-IM and Lipozyme RM-IM and the reaction mixtures were incubated at $40{\sim}70^{\circ}C$ with 120 rpm shaking. In pressurized system, the enzymatic ethanolysis was carried out at various condition; immobilized lipase concentration(2 wt%), reaction time(24 h), reaction temperature($40{\sim}60^{\circ}C$) and reaction pressure(75, 100, 150, 200 bars). The samples obtained from each fraction were analyzed by HPLC for analysing contents of monoglyceride, diglyceride, and triglyceride. The conversion of wheat germ oil relied on the reaction temperature and the concentration of immobilized lipase. The optimum condition of enzymatic ethanolysis in non-pressurized and pressurized systems was at $50^{\circ}C$ and 100 bar.

STUDY OF ${\beta}$-GLUCURONIDASE FROM SULL SEMINAL PLASMA:PURIFICATION AND PROPERTIES (황소의 정액에서 베타-굴룩 유로니다아제의 정제 및 그 성질에 관한 연구)

  • ${\beta}$-Glucuronidase from bull seminal plasma was partially purified by $(NH_4)_2SO_4$fractionation, two successive DEAE-cellulose columns, isoelectric focusing (pH 4 to 6) and Gel filtration on Sephadex G-200. Only one form of ${\beta}$-glucuronidase was obtained by isoelectric focusing at pH 5.13. Highly purified ${\beta}$-glucuronidase had specific activity of 34 units/mg protein and showed one major and some minor contaminants by disc gelk electrophoresis. The enzyme showed maximum activity at pH 5.2 and at $48^{\circ}C$. The enzyme was completely inhibited by 1,4 saccharo-${\alpha}$-lactone (5 mM). Albumin and 0.15 M NaCl increased the ${\beta}$-glucuronidase activity. Km of ${\beta}$-glucuronidase using phenolphthalein mono-${\beta}$-glucuronic acid as substrate was 2.9 mM and Vmax was $0.8{\mu}$mole/min. The enzyme appeared to be a glycoprotein by its binding to concanvalin·A. Rabbit and human sperm-acrosomal extracts and seminal plasma showed high ${\beta}$-glucuronidase activity.

광학 분석 시스템용 수.발광 소자 집적 모듈 개발

  • 본 발표에서는 광학적 분석 시스템에 적용 가능한 발광소자(광원)과 수광소자(광센서)를 집적화시키는 모듈(수 발광 집적모듈) 기술을 제시하고자 한다. 이러한 수-발광 집적모듈은 다양한 응용 분야에 적용 될 수 있다. 예를 들어, 광신호 감지를 위한 광통신용 송-수신 모듈(optical communication), 의료/진단 분야에서 단백질/DNA/박테리아 등의 검출 및 분석에 관한 바이오 센서(bio-sensor), 그리고 대기(가스)/수질 모니터링에 관한 환경센서 등 매우 광범위한 분야에 해당되는 요소 기술이라 할 수 있다. 특히, 이들 분야들 중 바이오 물질을 분석하고 검출하는 광학적 바이오 센서 기술은 높은 경제적 가치와 산업적 성장 잠재력으로 인해 오랫동안 활발한 연구가 진행되어 오고 있다. 이러한 광학적 바이오 센서에서 가장 범용적인 방법 중 하나가 항온-항체 면역반응을 기반으로 하는 형광 검출(fluorescence detection) 기법이다. 이러한 시스템은 전체적으로 광원, 광학계, 그리고 센서로 구성되는데 기존에 일반적으로 사용되고 있는 형광 현미경의 경우는 민감도가 우수하다는 장점은 있으나 상당히 고가이고 부피가 크며 복잡한 광학구성으로 이루어져 있다는 한계점을 가지고 있다. 이러한 맥락에서 고민감도를 확보하면서 휴대성, 고속처리, 저가 등의 특성을 가진 시스템에 대한 요구가 갈수록 증가하고 있다. 이를 해결하기 위한 핵심기술 중의 하나가 수-발광 부분을 집적화 시키는 기술이라 할 수 있다. 본 연구에서는 바이오 센서 기술의 하나로서 형광을 측정하여 혈액내의 진단 지표인자를 검출할 수 있는 휴대용 혈액진단기기에 적용되는 소형 수 발광 집적 모듈을 개발하였다. 혈액내의 검출 성분의 양에 따라 형광의 세기가 변화하게 됨으로써 정량적인 검출이 가능한 원리이다. 모듈의 구조는 크게 광원(발광소자), 광학계, 그리고 광센서(수광소자) 세 영역으로 나누어 진다. 광원은 635 nm 적색 레이저다이오드로서 형광체(Alexa Fluor 647/발광파장: 668 nm)를 여기 시키는 기능을 하며 장착된 볼렌즈 의해 샘플의 형광체 영역으로 집광된다. 광학계는 크게 시준렌즈(collimating lens)와 광학필터로 구성됨으로써 샘플로부터 발생되는 광을 적절하게 수광소자로 전달하는 기능을 하게 된다. 여기서 광학필터의 경우는 기본적으로 Distributed Bragg's Reflector(DBR) 구조로써 실리콘(Si) 포토다이오드 상부에 모노리식(monolithic)하게 형성되며 검출 샘플로부터 진행되는 레이저 광(잡음의 주원인)은 차단하고 형광(광신호)만 통과 시키는 기능을 하게 된다. 따라서 신호 대 잡음비(S/N ratio)를 향상시키기 위해서는 정밀한 광 필터링 기능이 요구됨으로써 박막의 세밀한 공정 조건과 구조적-광학적 특성 분석이 수행되었다. 마지막으로 포토다이오드 소자는 일반적인 구조 이외에 중앙에 원형 구멍이 형성된 특별한 구조가 적용된다. 이것은 포토다이오드 구조에 변화를 줌으로써 모듈 구조를 효율적으로 응용할 수 있다는 의미를 갖는다. 또한 포토다이오드의 전기적-광학적 측정 분석을 통해 잡음 및 감도 특성이 세부적으로 조사되며 형광신호를 효과적으로 측정할 수 있음을 확인하였다. 최종적으로 제작된 모듈은 약 $1{\times}1{\times}1cm^3$ 내외 정도의 크기를 갖는다. 요약하자면 본 발표에서는 광학적 바이오센서에 적용할 수 있는 소형 수-발광 소자 집적모듈을 소개한다. 전체 모듈 설계는 최소한의 부피를 가짐과 동시에 측정의 정밀성을 향상시키는데 초점을 맞추어 진행하였다. 세부요소인 광학필터와 포트다이오드의 경우 잡음 및 민감도에 미치는 중요성 때문에 세밀한 공정 및 특성분석이 수행되었다. 결론적으로 독자적인 설계 및 공정을 통해 휴대성 및 정밀성 등의 목적에 부합한 경쟁력 있는 수-발광 소자 집적모듈 제작 기술을 확보하였다.

Effect of Surfactants on ZnO Synthesis by Hydrothermal Method and Photocatalytic Properties (계면활성제 첨가에 의한 산화아연의 수열합성과 광촉매 특성)

  • Zinc oxide is, one of metal oxide semiconductor, harmless to human and environment-friendly. It has excellent chemical and thermal stability properties. Wurtzite-zinc oxide is a large band gap energy of 3.37 eV and high exciton binding energy of 60 meV. It can be applied to various fields, such as solar cells, degradation of the dye waste, the gas sensor. The photocatalytic activity of zinc oxide is varied according to the particle shape and change of crystallinity. Therefore, It is very important to specify the additives and the experimental variables. In this study, the zinc oxide were synthesized by using a microwave assisted hydrothermal synthesis. The precursor was used as the zinc nitrate, the pH value was controlled as 11 by NaOH. Surfactants are the ethanolamine, cetyltrimethylammonium bromide, sodium dodecyl sulfate, sorbitan monooleate was added by changing the concentration. The composite particles had the shape of a star-like, curcular cone, seed shape, flake-sphere. Physical and chemical properties of the obtained zinc oxide was characterized using x-ray diffractometer, field emission scanning electron microscopy, thermogravimetric analysis and optical properties was characterized using UV-visible spectroscopy, photoluminescence and raman spectroscopy.

A Study on the Figurative Universality of Religious Costume -Centering on the Religious Costume of High Religions- (종교복식의 조형적 보편성에 관한 연구 -고등종교의 종교복식을 중심으로-)

  • Just as various religions of the world have multiple systems based on their own belief system respectively, religious costumes, which are the expression of religion, are varied in forms according to different religions. Nonetheless, this research attempts to examine the universal features of the variety of religious costumes. Since the range of this research is broad and the limit of study is clear, this research confines the study objects into world's four high religions. The purposes of this research are as followings; first, the investigation of the world's high religions, second, the study of the figurative attributes of religious costume to study and discuss the universality of figurative beauty and aesthetic value. Figurative attributes are distinct in religious costume. First, the non structural feature of composition, and the manner in which the costume is worn. Second, the rich silhouette covering the body. Third, the restraint and inhibition of decoration, and fourth, the preference of achromatic color and monotones. 'The beauty of concealing', derived from the religious absolute and chastity, 'The beauty of chastity' influenced by the restraint of decoration and design, and 'The beauty of nature' as the drapery and non structural feature are the universal aesthetic values. Human beings tend to contact the divine beings by pursuing the essential thing and concealing the body through religion. The forms of concealment and chastity, mentioned above are reflected in the usual costumes, affected by religion as well as religious costume.

Preliminary Design and Implementation of 3D Sound Play Interface for Graphic Contents Developer (그래픽 콘텐츠 개발자를 위한 입체음 재생 인터페이스 기본 설계 및 구현)

  • Due to the advance of H/W and S/W techniques to play 3D sound, the virtual space contented by 3D graphics and sounds can provide users more improved realities and vividness. However for the small 3D contents developers and companies, it is hard to implement 3D sound techniques because the implementation requires expensive sound engines, 3D sound technical understanding and 3D sound programming skills. Therefore 3D-sound-playing-interface is necessary to easy and cost-effective 3D sound implementation. Using this interface, graphics experts can easily add 3D sound techniques to their applications. In this paper, the followings are designed and implemented as a preliminary stage in the way of developing the 3D sound playing interface. First, we develop 3D sound S/W modules converting mono to 3D sound in PC based systems. Second, we develop the interconnection modules to map 3D graphic objects and sound sources. The developed modules in this paper can allow the user to percept sound source position and surround effect at the moving positions in the virtual world. In the coming works, we are going to develop the more completed 3D sound playing interface consisted of the synchronization technique for sound and moving objects, and HRTF.

Changes' of glycolipids and phospholipids during maturation of perilla seed(Perilla frutescens) (들깨종자의 성숙과정중 당지방질과 인지방질의 변화)

  • To investigate changes in fatty acid and lipid composition of maturing perilla (Perilla frutescens var. japonica Hara) seeds, Suweon 10 and Jeju varieties were subjected to lipid analysis. The results were summarized as follows; As perilla seeds matured, content of glycolipid and phospholipid decreased. Glycolipid and phospholipid of mature seed were 25.4% and 4.5% of total lipids in Suweon 10 and those from Jeju were 44.5%, 4.0%, respectively. Cerebroside, galatosyl diglyceride and monogalatosyl diglyceride were major constitutents of ether-extractable glycolipids in developing perilla seeds. Monogalatosyl diglyceride, the richest constitutent in the early stage of seed development, decreased rapidly as seeds matured. In ether-extractable glycolipid of mature Suweon 10 seeds, content of linolenic acid was 51.1% which was higher than 19.4% of oleic acid of total acids. However, in Jeju variety, content of oleic acid was 31% which was higher than 18.6% of linolenic acid. Phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol and lysophosphatidyl inositol were major constituents of phospholipids. Content of phosphatidyl glycerol in developing seeds showed irregular changes in Jeju but in Suweon 10, it decreased after rapid increase in the early stage of seed development. Olieic acid ranges $28.7{\sim}35.2%$, linolenic acid $8.2{\sim}11.2%$, linoleic acid $16.4{\sim}32.5%$ and Palmitic acid $22.7{\sim}29.9%$ of total fatty acids in methanol-ertractible phospholipid of mature perilla seeds.
