상부 Au 전극 면적 Size에 따른 PbI2 필름의 전기적 특성 평가

  • 의료용 X-ray는 과거 analog 방식과, 연구가 진행 중이며 현재 많이 사용되고 있는 digital 방식으로 나누어진다. 최근, 광도전체와 형광체 기반의 flat panel X-ray detector의 발전에 따른 상용화가 이루어지고 있으며, 많은 발전 가능성이 제기되고 있다. flat panel X-ray detector 검출방식은 direct method (직접 방식)와 indirect method (간접 방식)로 나누어진다. 본 연구는 일반적으로 상용화 되어있는 amorphous seleinum (비정질 셀레늄)의 큰 일함수에 의한 저 해상력이라는 단점을 보완하기 위해, 작은 일함수를 가지는 물질을 사용하여, 영상을 얻을 시에 높은 해상력으로 표현할 수 있도록 하고, 원자번호가 높은 물질을 사용하여 X-ray 흡수율을 높일 수 있도록 기존 direct method에 많이 사용되고 있는 amorphous seleinum 기반 digital X-ray detector가 아닌, 이러한 장점을 충족시킬 수 있는 PbI2 물질 층을 사용하여 시편을 제작 하였다. PbI2를 같은 두께로 올린 후, 물질 층 상부에 Au 전극 면적을 다른 size로 제작한 시편으로 X-ray에 노출 시켰다. 이는 상부 전극 size 차이에 따른 신호 차이를 측정하여 전기적 특성을 평가하기 위한 것이다. 전도성을 띠고 있는 ITO (Indium - Tin - Oxide) glass를 이용하여 screen printing 방법으로 제작하였다. PbI2층을 약 160~180 um두께, $3cm{\times}3cm$ size로 5개 제작하였으며, 상부 전극으로는 Au를 진공 증착 시켰다. 상부 전극 size는 각각 시편 5개에 $0.5cm{\times}0.5cm$, $1cm{\times}1cm$, $1.5cm{\times}1.5cm$, $2cm{\times}2cm$, $2.5cm{\times}2.5cm$로 PbI2 물질 층 중앙에 증착 시켰다. 이러한 설정으로 X-ray 노출 시 관찰할 수 있는 PbI2의 전기적인 특성을 평가할 수 있었다. 관전압을 40 kVp, 60 kVp, 80 kVp, 100 kVp, 120 kVp, 140 kVp로 설정하고, 관전류는 100 mA로 설정하였으며, Dark current, Sensitivity를 측정하였다. Dark current와 Sensitivity를 측정한 뒤, 그 값을 이용하여 SNR (신호 대 잡음 비)값을 구해보았다.실험 결과 단위면적당 signal과 SNR을 분석할 수 있었다. 80 kVp로 기준을 잡고 결과 값을 보면 $0.5cm{\times}0.5cm$ 시편에서 2.92 nC/cm2, $2.5cm{\times}2.5cm$ 시편에서 0.84 nC/cm2로 상부 전극 크기가 작을수록 더 좋은 신호를 측정할 수 있었다. 똑같은 기준에서 SNR을 계산 해 보았을 때, $0.5cm{\times}0.5cm$ 시편에서 6.46, $2.5cm{\times}2.5cm$ 시편에서 1.91로 SNR역시 상부 전극 크기가 작을수록 더 큰 값을 확인할 수 있었다. 이러한 결과는 edge-effect의 영향으로 인해 나온 결과라고 할 수 있다. 이러한 실험 결과, detector 제작 시, 같은 물질을 사용하여 더 높은 효율을 내기 위해서는 큰 size의 상부 전극 보다는 작은 size의 상부 전극을 증착 시키는 것이 전기적 특성을 더욱 효율적으로 평가할 수 있을 것이라고 사료된다.

반도체 검출기의 절연 최적화를 위한 다층 절연막 평가

  • 반도체 검출기는 입사되는 X선 에너지에 의하여 이온화되어 발생하는 전자 전공쌍을 수집함으로 방사선 정보를 확인하는 선량계로써 많은 연구와 활용이 이루어지고 있다. 하지만, X선 에너지에 의하여 반도체 검출기에서 발생하는 전기적 신호량이 높지 않기 때문에 누설 전류의 저감이 필수적이다. 누설 전류를 저감시키기 위한 방안으로 반도체 층과 전극 층의 Schottky Contact 구조의 설계, Insulating Layer의 사용, 높은 비저항의 반도체 물질 연구 등이 이루어지고 있다. 하지만, 기존에 누설 전류 저감을 위하여 Insulating Layer를 전극층과 반도체 층 사이에 형성하는 연구에 있어서 Insulating Layer와 반도체 층의 계면 사이에서 발생하는 Charge Trapping으로 인하여 생성되는 신호의 Reproducibility 저하, 동영상 적용의 제한 등의 문제점을 겪어왔다. 이에 본 논문에서는 누설 전류를 저감시킴과 동시에 Charge Trapping의 최소화를 이루기 위하여 Insulating Layer의 두께 최적화 연구를 수행하였다. 본 연구에서 사용한 Insulating Layer는 검출기 표면에 입사하는 X선 정보 손실을 최소화 시키는 동시에 누설 전류와 Charge Trapping을 최소화 시키는 방법으로써 CVD방법으로 검출기 표면에 균일하게 Insulating Layer를 코팅하였다. Insulating 물질은 Parylene을 사용하였으며, 그 중 온도, 습도 등 외부환경에 영향을 적게 받는 type C를 사용하였다. 증착에 사용한 장비의 진공도는 Torr로 설정하여 증착되는 Parylene의 두께가 약 $0.3{\mu}m$가 되게 하였으며, 실험에는 반도체 물질 PbO를 사용하였다. Parylene의 절연 특성은 Dark Current와 Sensitivity를 측정한 SNR을 이용하여 Parylene코팅이 되지 않은 동일 반도체 검출기와의 신호를 비교하였으며 또한 Parylene를 다층 제작한 검출기의 수집 신호량을 비교하였다. 제작한 검출기의 X선 조사 시의 수집 전하량 측정 결과, 100 kVp, 100mA, 0.03s의 X선 조건에서 $1V/{\mu}m$의 기준 시, Parylene를 코팅하지 않은 PbO 검출기의 Dark current는 0.0501 nA/cm2, Sensitivity는 0.6422 nC/mR-cm2, SNR은 12.184이었으며, Parylene단층의 두께인 $0.3{\mu}m$로 증착된 시편의 Dark current는 0.04097 nA/cm2, Sensitivity는 0.53732 nC/mR-cm2으로 Dark current가 감소되고 sensitivity도 감소하였지만 SNR은 13.1150으로 높아진 것을 확인할 수 있었다. Perylene이 $0.6{\mu}m$로 증착된 시편의 경우, Dark Current는 0.04064 nA/cm2, Sensitivity는 0.31473 nC/mR-cm2, SNR은 7.7443으로써 Insulating Layer가 없는 시편보다 SNR이 약 40% 낮아진 것을 확인할 수 있었다. Parylene이 $0.9{\mu}m$로 증착된 시편의 경우 Dark current는 0.0378 nA/cm2, Sensitivity 0.0461 nC/mR-cm2로 Insulating Layer가 없는 시편에 비해 SNR은 약 1/12배 감소한 1.2196이었고, Parylene이 $1.2{\mu}m$로 증착된 시편의 SNR은 1.1252로서 더 감소하였다. 따라서 Parylene을 다층 코팅한 검출기일수록 절연 효과의 영향이 커짐으로써 SNR 비교 시 수집되는 신호량이 줄어드는 것을 확인하였다. 반도체 검출기의 누설 전류를 저감시킴과 동시에 신호 수집율에 영향을 최소화시키기 위하여 Insulating Layer의 두께를 적절하게 설정하여 적용하면 Insulating Layer가 없는 검출기에 비해 누설전류를 최소한으로 줄일 수 있고 신호 검출효율이 감소하는 것을 방지할 수 있을 것이라 사료된다.

An analysis of the genetic diversity of a riparian marginal species, Aristolochia contorta (수변 경계종인 쥐방울덩굴의 유전적 다양성 분석)

  • Northern pipevine (Aristolochia contorta) commonly inhabits marginal areas between waterside and terrestrial vegetation. In particular, A. contorta is ecologically important in the marginal areas as a food plant of dragon swallowtail butterfly (Sericinus montela), which is designated as vulnerable species in the Republic of Korea. For long-term sustainability of the plant population, assessment of the genetic diversity of exist populations should be conducted. Genomic DNA of A. contorta leaf samples were extracted from four populations where the vigorous growth were observed in the South Korea. Intra-population genetic diversity and inter-population genetic distance were assessed using randomly amplified polymorphic DNA (RAPD) with five polymorphic random primers. Overall genetic diversity was lower, compared to other wetland species (h: 0.0607 ~ 0.1401; I: 0.0819 ~ 0.1759), while GP showed the highest intra-population genetic diversity. Despite of the geographical distance, GP showed the larger genetic distance from other populations. This result seemed to be caused by the fragmented habitat and lower sexual reproduction of A. controta. Mixture of the different source populations and construction of the proper environmental condition such as shade and physical support for sexual reproduction should be considered for conservation of A. contorta population.

Biotic and Abiotic Effects on the Growth and Reproduction of Aristolochia contorta (생물 및 비생물적 요인이 쥐방울덩굴의 생육과 생식에 미치는 영향)

  • For the conservation of both Sericinus montela (vulnerable species by red book of Korea) and A. contorta, we investigated environmental factors affecting the growth and reproduction of Aristolochia contorta. We selected four A. contorta habitats at Gapyeong, Pyeongtaek, Cheongju, and Yeoju in Korea and surveyed community and habitat characteristics of companion species, herbivore appearance, support types, and soil physicochemical properties in July and October, 2018. Habitat environments and the growth and reproduction characteristics of A. contorta were different according to four habitats of different regions. In particular, the fastest growth speed and earliest flowering and fruiting were observed in Pyeongtaek. Growth of A. contorta in Cheongju and Yeoju were interrupted by aboveground damage from human disturbance. In this study, support types seemed to be important for growth speed of A. contorta. Flowering and fruiting timing were likely to be related to combined effects of soil cation contents and competitive and herbivore stresses. Therefore, providing effective support and reducing biological stress should be necessary for stable growth and proper flowering and fruiting timing of A. contorta. In addition, experimental evidence would be needed to figure out the effect of soil cation and biological stress on flowering and fruiting of A. contorta in detail.

Construction of a Pure Cryparin-null Mutant for the Promoter Analysis of Cryparin Gene (Cryparin 유전자의 promoter 분석을 위한 cryparin 유전자 치환체의 순수 제조)

  • The cryparin of Cryphonectria parasitica belongs to a cell wall associated fungal hydrophobin. The cryparin, though it is encoded by a single copy gene, is known for the high expression during the liquid culture of C. parasitica, and it turns out that 22% of total mRNA was transcribed for cryparin at 48hr after the liquid culture. In addition, it is also known as one of down-regulated fungal proteins by the presence of double stranded RNA virus, Cryphonectria hypovirus 1. In previous studies (Kim et al., 1999), we have constructed a cryparin-null mutant by replacing the cryparin gene with hygromycin B resistance gene due to site directed homologous recombination. In order for the promoter analysis of cryparin which seems to be very strong as well as mycoviral specific, it is preferable to have a strain with only a target promoter replaced and a discernable target site for incoming vectors. However, the cryparin-null mutant revealed the presence of an additional copy of transforming vector except the one which replaced the cryparin gene. In addition, the cryparin-null mutant did not contain any markers for targeted integration of incoming vectors. This prompts us to design an experiment to obtain a strain for promoter analysis of cryparin gene. A different mating type strain EP6(Mata, $met^-$) was mated with the cryparin-null mutant ${\triangle}$Crp194-7(MatA, Crp${\triangle}$::hph) to make the progenies with only a single replacement vector and $met^-$ characteristic remained. Nutritional assay as well as Southern blot analysis revealed that the progeny, ${\triangle}$Crp194-a6, was the methionine auxotroph with a single replacing vector in genome. Northern blot analysis and PAGE showed that there was no cryparin produced in this bred strain either.

Chlorogenic Acid Enhances Glucose Metabolism and Antioxidant System in High-fat Diet and Streptozotocin-induced Diabetic Mice (고지방식이와 스트렙토조토신으로 유도한 당뇨마우스에서 Chlorogenic Acid의 혈당강하 및 항산화 효과)

  • This study investigated dose-response effects of chlorogenic acid (CA) on glucose metabolism and the antioxidant system in streptozotocin (STZ)-induced diabetic mice with a high-fat diet (HFD). Male ICR mice were fed with a HFD (37% calories from fat) for 4 weeks prior to intraperitoneal injection with STZ (100 mg/kg body weight). Diabetic mice were supplemented with two doses of CA (0.02% and 0.05%, wt/wt) for 6 weeks. Both doses of CA significantly improved fasting blood glucose level, glucose tolerance and insulin tolerance without any changes in plasma insulin and C-peptide levels. Plasma leptin concentration was significantly higher in the CA-supplemented groups than in the diabetic control group. Both doses of CA significantly increased hepatic glucokinase activity and decreased glucose-6-phosphatase activity compared to the diabetic control group. The ratio of glucokinase/glucose-6-phosphatase was dose-independently higher in CA-supplemented mice than in diabetic control mice. CA supplementation dose-independently elevated superoxide dismutase and catalase activities, whereas it lowered lipid peroxide levels compared to the diabetic control mice in the liver and erythrocyte. These results suggest that low-dose CA may be used as a hypoglycemic agent in a high-fat diet and STZ-induced diabetic mice.

Effect of Dandelion (Taraxacum officinale) Leaf Extracts on Hepatic Antioxidative System in Rats fed High Cholesterol Diet (고콜레스테롤 흰쥐의 간조직 항산화효소계에 미치는 민들레잎 추출물의 영향)

  • This study was conducted to investigate the effects of dandelion leaf (Taraxacum officinale) extracts on hepatic antioxidative system in high cholesterol-fed rats. Four groups of rats were given high cholesterol diets containing 10 g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received a diet without dandelion leaf extract and the other three groups received dandelion leaf extracts, ie, water, ethyl acetate and ether extracts, respectively. There were no significant difference in cytochrome P-450 contents among four groups. Hepatic xanthine oxidase activity was significantly lower in water extract group than the other three groups. Superoxide dismutase activity was significantly lower in three dandelion leaf extract groups, but catalase activity was significantly higher in three dandelion leaf extract groups than control group. Glutathione peroxidase and glutathione S-transferase activities were significantly increased in water extract group than control group. Lipid peroxide content was decreased in water extract group than control group.

The Effect of Puerariae thubergiana Bentham Extract on Brain Tissue in Alcohol-Treated Rats (칡추출물이 알코올을 급여한 흰쥐의 뇌조직에 미치는 영향)

  • This study investigated the effect of Puerariae Flos (PF; flower of Puerariae plant) and Puerariae Radix (PR; root of Puerariae plant) water extracts on the activities on the activities of ethanol-metabolizing enzymes and free radical generating/scavenging enzymes of brain in ethanol-treated rats. Five groups of male Sprague-Dawley rats were orally administered ethanol (25%, v/v) 5 g/kg body weight/day, and sacrificed 5 weeks post treatment. PF and PR water extracts were supplemented in a diet based on 1.2g (I) or 2.4 g (II) raw PF or PR/kg body weight/day. Alcohol dehydrogenase activity of brain was significantly lowered in PF of PR groups, whereas aldehyde dehydrogenase activity was significantly higher in PR groups than those of control and PF groups. Cytochrome P-450 content, aminopyrine D-methylase and aniline hydroxylase activities were decreased in both PF and PR groups compared to control group. Aldehyde oxidase and xanthine oxidase activities tended to decrease by Puerariae plant extract supplemented goups and degree of decrease predominated in PRI. Superoxide dismutase and glutathione S-transferase activities were increased in PF or PR groups, whereas glutathione peroxidase and catalase activities were significantly decrased by Puerariae plant extracts supplement. These results indicated that supplementation of PF or PR lowers free radical generating enzymes activities. It was suggested that the activities of ethanol metabolizing emzymes and antioxidant enzymes in brain can be enhanced by PF or PR supplement in ethanol-treated rats.

Effect of Dandelion Leaf Extracts on Lipid Metabolism in Rats Fed High Cholesterol Diet (민들레잎추출물의 흰쥐 체내 지질대사 개선 효과)

  • The effect of dandelion leaf extracts on lipid metabolism was investigated in high cholesterol-fed rats. Four groups of rats were given a high cholesterol diet containing 10g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received without dandelion leaf extracts and the other three groups received with one of dandelion leaf extracts (water, ethyl acetate, ether), respectively. Weight gain and feed intake were significantly higher in water group than control group. Serum triglyceride, total cholesterol, cholesteryl ester, chylomicron and VLDL-cholesterol concentratins were lower, whereas HDL-cholesterol and phospholipid concentrations were significantly higher in dandelion leaf extract supplemented-groups than control group. Dandelion extract increased HDL-C/total-C ratio and lowered atherogenic index. Hepatic triglyceride, total cholesterol, free cholesterol and cholesteryl ester concentrations lowered in dandelion leaf extract groups compared to control group. Especially, hepatic triglyceride and free cholesterol levels were lower in ethyl acetate extract group than in water and ether extract groups. Dandelion leaf extract supplements did not affect hepatic phospholipid cencentration. Hepatic HMG-CoA reductase activity was significantly lower in water extract group than the others. ACAT activity was significantly decreased by dandelion extract supplements.

  • Liparis ingens (Gilbert et Burke) is a demersal fish belonging to Liparidae, widely distributed in the coastal waters of the east coast of Korea. On January 16, 1986, mature adults of Liparis ingens were captured by a demersal gill net near Chongdongchin-ri, Kangwon-do, Korea. The authors stripped eggs and milt, and fertilized on the ship. The eggs were incubated and the larvae were reared in laboratory. The eggs of this species are demersal and adhesive, and are pale yellow, containing many oil globules. Egg diameters were varied from 1.55 to 1.65 mm. The hatching took place in ca. 747 hours after fertilization at the water temperature of $7.09{\sim}10.99^{\circ}C$ and salinity $30.62{\sim}33.98\%0$. The newly hatched larvae were elongate in shape and $4.40{\sim}4.85mm$ in total length with $12{\sim}13$ (abdominal)+40(caudal)=$52{\sim}53$ myomeres. Many melanophores were distributed on the lower jaw, pectoral fins, yolk sac, and ventral line of the tail part. Three days after hatching, the larvae reached 5.15 to 5.25 mm in total length, and the yolk sac was almost absorbed. Some melanophores appeared on the back of auditory vesicles. Seven days after hatching, the larvae attained $5.75{\sim}5.85mm$ in total length.
