Echinacea purpurea, an indian traditional plant medicine has been widely used as herbal remedy for the treatment of disease such as colds or other infections. However, Echinacea purpurea extracts recently have been applied as a cosmetic ingredient for skin care. We artificially cultured Echinacea purpurea by using the bioreactor culture system for this study. We induced callus from Echinacea purpurea and separated adventitious roots, harvested and extracted after cultured in bioreactors. Previously, several studies have been reported on anti-oxidant and immuno-enhancing effects of Echinacea purpurea extract but other efficacies were not well known. In this study, we investigated the whitening, anti-wrinkle and anti-oxidant effects to know applicable value of tissue-cultured Echinacea purpurea adventitious roots extract(TCEPARE) as a cosmetic ingredient. TCEPARE did not show cytotoxicity until a concentration of 2% and showed the anti-oxidative effect in DPPH and NBT tests. Also, the extract decreased tyrosinase expression in a dose-dependent manner and inhibited melanin synthesis in B16 melanoma cells. TCEPARE reduced protein levels of MMP-1, 2 secreted in culture medium or in cell lysates. From these results we suggest that TCEPARE has potential benefits applicable as to cosmetic ingredient for skin care products.

Yuza (Citrus junos Sieb ex TANAKA) is a citrus fruit that is cultivated in northeast Asia. Citron is known for containing abundant antioxidants such as vitamin C, flavonoids, for example hesperidin and hesperetin, and terpenoids such as limononin. When mature citron is processed for tea or other beverage food products in Korea, massive amounts of seeds and pericarp are remained as waste. This study aimed to exploit the processed remnant of Citron for developing functional cosmetic applications. Ethanol extracts of Yuza seed and pericarp did not show significant radical scavenging activities measured by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. But they contained significantly high phenolic compounds. Cultured human dermal fibroblasts and HaCaT keratinocytes were irradiated with 25 mJ UVB and the citron extracts were added to the medium of each culture. Cellular damages caused by UVB irradiation were prevented by the addition of the Yuza extract. In addition, the reduction of the enhanced MMP-1 expression after irradiation of UVB in human dermal fibroblasts was observed. Also the increased level of pro-inflammtory TNF-$\alpha$ in the UVB irradiated HaCaT cells was decreased. The collagen expression was enhanced by the extract. Yuza extract markedly inhibited melanin production from $\alpha$-MSH treated B16F1 melanoma cells. Melanin assay, tyrosinase zymography results indicated that Yuza extract had strong depigmenting activity. In conclusion, Yuza ethanol extracts have good anti-photoaging and strong anti-melanogenic efficacies.

We investigated the in vitro antioxidant and antimelanogenesis effects of ethanol extracts from eggplant (Solanum melongena L.) peels. The total phenolics and chlorogenic acid in ethanol extracts were 2,465 mg/100 g and 2.08 mg/100 g, respectively. ABTS radical scavenging activity, ferric reducing/antioxidant power assay, and malondialdehyde (MDA) inhibitory effect of the extracts increased in a dose-dependent manner. In addition, the extracts generally showed strong UV absorption in the range of UV-B (290-320 nm). The $IC_{50}$ of mushroom tyrosinase inhibitory activity of ethanol extracts from eggplant peels was 870 ${\mu}g/mL$. Importantly, the melanin syntheses of B16/F10 melanoma cells were decreased by extracts in a concentration-dependent manner. Overall, these results suggest that eggplant peels can be potentially applied as a anti-melanogenic agent as well as an antioxidant resource.

Plants and their extracts can be utilized as inexpensive and rich resources of active constituents in the cosmetic field, as well as the food, pharmaceutical and medicinal fields. Until now, Aster glehni Fr. Schm. had no known active effect, except on anti-oxidation, that was found during investigations for application in the cosmetic field. In this study, we examined the inhibition of enzymatic reactions to protein levels in inclusive B16F10 melanoma cell lines. Significant inhibition of enzymatic reactions was observed in the EtOAc extract, which advanced to tyrosinase protein and TRP-1 in the B16F10 melanoma cell line. These results indicated that the effect of EtOAc extract inhibited expressions of tyrosinase protein and TRP-1 in the B16F10 melanoma cell line by 30.5% and 41.5% at 100ug/ml respectively. On the other hand, antimicrobial activity was evaluated to the four fractions in normal flora of the skin. Hexane extract was only exhibited in the higher clear zone in all strains. In conclusion, any cosmeceutical effects of Aster glehni Fr. Schm. may have a potential meaning, as well as possible value for further studies regarding the effects of chemical constituents of Aster glehni Fr. Schm.
Human skin is constantly exposed to ultraviolet (UV) radiation, polluted air, and chemical products. UV rays, in particular, will affect the skin in a variety of ways, including causing wrinkles, fine lines, rough skin, and xeroderma, thereby resulting in skin aging. This study aimed to investigate the whitening effects of Juniperus rigida Sieb., which is a cedar tree that is found throughout the world. The whitening efficacy that was measured by tyrosinase inhibition revealed 49.4% efficacy in water extract and 80.0% efficacy in ethanol extract. Among the B16F10 black cells, the effect of the ethanol extract was higher than the effect of the water extract in the restrain creation of melanin pigment, tyrosinase, microphthalmia-associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2). Thus, the results of these studies demonstrated that the ethanol extract had greater efficacy than the water extract and Juniperus rigida Sieb. Ethanol extracts could be utilized as materials for functional cosmetics, such as whitening products.
UV-protection skin whitening and immune activities several parts of Acer mono and Acer okamotoanum were investigated. The bark of both Acer mono and Acer okamotoanum had higher yields than other parts as 2.67% and 2.45%. The cytotoxicity of the extracts were lower than 21.64% against human skin cell(CCD-986sk) line in adding 1.0 mg/mL of the highest concentration. The bark extracts of Acer mono greatly reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 30%. At 1.0 mg/mL of bark extration of Acer mono, $PGE_2$ expression was also significantly decreased. Generally, the bark extracts of Acer mono and Acer okamotoanum had higher activity than other parts, but, interestingly, wood extract of Acer okamotoanum showed strong inhibition effect on melanin production by Clone-M3 cells as 79.25%. From these results, we could conclude that the bark extract from Acer mono and Acer okamotoanum had skin-whitening activity as well immune enhancement activity.

The purpose of this experiment was to know the healing effects of saualene on the burned mouses. The experimental groups were divided into seven groups: the control group, the burned groups, and 50 treated groups after burned. All groups except the control group burned with second degree on a dorsoanterior part. All groups that treated with 50 adapted three drops of pure squalene at 10 seconds interval; one time a day. The histological and ultrastructural changes during 10 days after burned were observed by light and electron microscope. Under the light microscope, all parts of epidermal layer of the burned groups were wounded deeper than the 50 treated groups. At 6 and 10 days groups with 50 treated, especially 10 days, the basal layer was greatly differentiated, and the prickle cells in the spinous layer were greatly increased in number. Under the electron microscope, the cell divisions of basal layer in all groups that treated with 50 were more activated and rapidly regenerated than the all burned groups. Especially, some fantastic results obtained from the 10 days group with SQ treated; much thicker spinous layer than the control group, many prickle cells, fine intercellular bridges, and healthy basal layer contained melanocytes. These results suggest that squalene may active the epidermis growth factor (EGF), acts as scavenger, and provide sources of energy in the membrane system. The results of this experiment consider that squalene has specific effects for burn healing and regeneration.
In this study, functionality of products of silkworm hemolymph fermented by Bacillus species was studied such as cell viability, antioxidant effect, and inhibitory effect on tyrosinase activities. A matured silkworm hemolymph was degraded by fermentation with Bacillus subtilis 10854 and Bacillus amyloliquefaciens M27. Especially, proteins of matured silkworm hemolymph were degraded to 3,000 Da by fermentation with B. amyloliquefaciens M27. Cell viability for MTT assay was higher than PBS in hemolymph and fermented hemolymph. A DPPH free radical scavenging activity of fermented hemolymph was as higher as Vitamin C and dependent on sample concentrations. Thus, these results suggest that degraded hemolymph fermented by B. amyloliquefaciens M27 may have antioxidant properties as a material for cosmetics.
This study was performed to determine the antimelanogenic effect and tyrosinase inhibitory activities of anthocyanin rich fraction (AN-SLP) from Liriope platyphylla Wang et Tang seeds. Anthocyanins isolated from L. platyphylla seeds revealed the presence of four major anthocyanin components, which were tentatively identified as delphinidin-3-Oglucoside, delphinidin-3-O-rutinoside, petunidin-3-O-rutinoside, and malvidin-3-O-rutinoside using semipreparative HPLC, $^1H$-NMR, $^{13}C$ NMR, FAB-MS and LC/ES-MS. The inhibitory effect of AN-SLP on tyrosinase activity was studied using in vitro (against mushroom tyrosinase) and ex vivo (against B16 melanoma cell tyrosinase) models. Cellular tyrosinase activity was decreased by AN-SLP treatment in B 16 melanoma cells through dose dependent manner, but AN-SLP did not inhibit mushroom tyrosinase and L-DOPA oxidation directly. AN-SLP showed melanin inhibition by 53.2% at 50 ${\mu}g/m{\ell}$ which was 0.7 times more efficient than the antimelanogenic effect of commercial arbutin and kojic acid (36.5%) also did not show cell toxicity. Additionally, AN-SLP inhibited the activity of ${\alpha}$-glucosidase and the glycosylation of tyrosinase in melanoma cell. The resulting unsaturated glycosylation of tyrosinase makes it unstable and disturb correct transportation. From theses results, we conclude that AN-SLP could be used as anti-melanogenic agent for skin whitening.
In order to develop a new starter for fermented milk, Lactobacillus plantarum M23 was isolated from raw milk and investigated for physiological characteristics. It showed good tyrosinase inhibitory activity compared with commercial lactic acid bacteria. The optimum growth temperature of Lactobacillus plantarum M23 was $40^{\circ}C$ and cultures took 17 hr to reach pH 4.3. Lactobacillus plantarum M23 showed more sensitivity to Penicillin-G, Oxacillin, Novobiocin, Chloramphenicol in a comparison of 12 different antibiotics, and showed most resistance to Vancomycin. It showed higher leucine arylamidase and ${\beta}-galactosidase$ activities compared to 16 other enzymes. It was comparatively tolerant to bile juice and able to survive at pH 2 for 3 hours. It showed high resistance to Escherichia coli, Salmonella typhimurium and Staphylococcus aureus with rates of 77.8%, 86.5% and 83.8%, respectively. Based on these and previous results, Lactobacillus plantarum M23 could be an excellent starter culture for fermented milk with high resistance to melanin.
