Antioxidant Effects of Ethanol Extracts of Aplidium pliciferum and Herdmania momus (만두멍게와 분홍멍게 에탄올 추출물의 항산화 효과)

  • This study aims to verify the potential of functional cosmetic materials by examining the antioxidant effects of Aplidium pliciferum and Herdmania momus, used to determine the antioxidant efficacy of Aplidium pliciferum and Herdmania momus ethanol extracts, Mean and standard deviation were calculated using Minitab® 18 (IBM, USA). After various radicals were removed from the extracts of Aplidium pliciferum and Herdmania momus, DPPH was detected in 30.87±1.98% in Aplidium pliciferum, 88.27±0.84% in Herdmania momus, and ABTS in 56.23±2.50% in Aplidium pliciferum and 94.80±0.91% in Herdmania momus in the concentration of 20 mg/ml. Also, NO was detected in 88.57±0.81% in Aplidium pliciferum and 97.53±0.34% in Herdmania momus, respectively, thus all showing statistically significant differences. So we can confirm the efficiency of the solvent acts as an important factor through the total poly phenol content

  • Ciona intestinalis and Ciona savignyi have been designated as hazardous species because they have caused great damages as they appeared in ports, ships, and farms in Korea in large amount. Therefore, in terms of using them, the possibility of use as cosmetic ingredients was examined. C. intestinalis and C. savignyi were treated with 70% ethanol to make an extract and then their antioxidant activity was measured. C. intestinalis and C. savignyi showed 71.80% and 21.40% of DPPH at 20mg/mL of extract, respectively. ABTS was 95.47% and 27.53%, while nitric oxide production inhibitory ability was 97.47% and 88.90%, respectively. Total phenol content was 0.081 and 0.041 mg gallic acid/mg extract, respectively. As a result, C. intestinalis showed somewhat higher antioxidant activity than C. savignyi. It is confirmed that these extracts are animal-derived antioxidants that can be used as cosmetic materials.

Occurrence of bi-flagellated protists in the tunics of ascidians Halocynthia roretzi with tunic-softness syndrome collected from Tongyeong, south coast of Korea (통영산 물렁증 멍게 피막의 편모충 감염)

  • The edible ascidian Halocynthia roretzi is a commercially important fisheries resource in Korea. However, for the last several years, outbreaks of mass mortalities of the species have been occurring along the south and east coasts of Korea, where most ascidians are produced. Although it is known that tunic-softness syndrome is associated with these mortality events, the agent causing the syndrome has not yet been confirmed. To determine the agent causing tunic-softness syndrome, healthy and diseased ascidians were collected in March 2011 from Tongyeong, on the south coast of Korea, and were used for biological and pathological investigations. The results showed that diseased ascidians exhibited remarkably reduced body fluid, fatness index, and tunic index compared with healthy specimens. Interestingly, bi-flagellated protozoans were observed specifically in the tissue imprints and tunic cultures of diseased ascidians at an occurrence rate of 97.5%. Histological observation showed that the thickness of the tunics of diseased ascidians was reduced by half, and irregular structure and breakdown of the tunic fiber bundles were observed. In particular, flagellate-like cells were observed in the diseased ascidians. Our study clearly shows that bi-flagellated protists are present only in the softened ascidians, suggesting that the flagellates are partly or entirely associated with soft-tunic syndrome. Accordingly, further investigations to verify the effects of the flagellates found in the present study on soft-tunic syndrome should be conducted.

Gene Promoter Variation of Phosphoglycerate Kinase, a Glucose Metabolism Enzyme, is a Biomarker for Selection of Disease-resistant Sea Squirt, Halocynthia Roretzi (당 생합성 효소 PGK 유전자 프로모터 변이와 물렁증 저항성 멍게의 선별)

  • The sea squirt, Halocynthia roretzi, has experienced mass mortality due to softness syndrome. The identification of disease-induced genes can provide insights into the development of this syndrome. To identify the genes, we performed differentially expressed gene (DEG) analysis. The expression of the phosphoglycerate kinase (HrPGK) gene was significantly decreased in diseased sea squirts compared to normal ones. We confirmed the result of the DEG analysis through RT-PCR and real-time PCR. In addition, we detected single nucleotide polymorphisms at position -106 (A/T) and -254 (G/T) in the HrPGK gene promoter by genotyping analysis. At the -106 site of the HrPGK gene, the frequency of the AA allele in disease-resistant sea squirts was about two-fold higher than that of sensitive ones, and the frequency of the TT allele in the disease-resistant sea squirts was about six-fold lower. At the -254 site of the HrPGK gene, the frequency of the GT and the GG allele was approximately two-fold higher and two-fold lower, respectively, in the disease-resistant sea squirts compared to the disease-sensitive ones. Analysis of the relationship between the genotypic variation at the -106/-254 promoter and the expression of HrPGK mRNA showed that HrPGK mRNA expression was higher in the -106/-254 AA/GT genotype samples than in the -106/254 TT/GG genotype ones. These results show that sea squirts harboring the AA/GT genotype may have more resistance to mortality than the sea squirts with other genotypes.

릿테르개멍게, Halocynthia hilgendorfi ritterif의 생식주기

  • 릿테르개멍게는 척색동물문 해초강에 속하는 멍게류로서 고착생활을 하며, 보통 해조류나 히드라 등으로 덮여 있어 눈으로 식별하기가 어렵고, 생태 및 분류에 관한 연구만이 있을 뿐이다. 이 연구는 제주도 연안에 서식하는 릿테르개멍게의 번식생태에 대한 기초자료를 얻고자 배우자형성과정 및 생식소 발달 단계 등을 조직학적으로 조사하였다. 실험 재료는 제주도 용담 연안에서 2001년 11월부터 2003년 1월까지 매월 20개체씩 총 300개체를 채집하여 생식소의 조직학적 변화를 관찰하였다. 릿테르개멍게는 자웅동체로서 생식소는 체벽부에 붙어 있고, 성숙시기에 난소는 적색을, 정소는 유백색을 띠고 있다. 생식소와 체벽부를 포함해 절단된 조직표본에서 보면 외측체벽은 근섬유를 포함한 섬유성결합조직이 발달해 있고 여기에 이어진 방형의 난소낭과 그 주변부에 자리 잡고 있는 여러 개의 정소낭을 식별할 수 있었다. 생식소 발달 및 배우자형성과정, 월별 생식소변화를 토대로 생식주기를 구분하면 성장기 (3-5월),난황기 (5-7월), 성숙기 (7-11월), 방출기 (11-2월), 회복기 (1-3월)로 구분되었으며, 주 산란시기는 11-1월로, 연중 1회의 산란시기를 갖는다.

Ascidians of Tangsa and its Adjacent Waters in Korea Strait (대한해협의 당사 및 인근 수역 해초류의 분류)

  • The ascidians specimens were collected from Tangsa and its adjacent waters in Korea by scuba divers during the period from 1994 to 1999. They were identified into 30 species which are reported for the first time in Tangsa. Out of them one species, Molgula hozawai, was turned out to be new to the Korean fauna, and six species, such as Eudistoma illotum, Symplegma connectans, Boltenia trasversaria, Microcosmus nultitentaculatus, Pyura squamata and Molgula hozawai, were newly recorded in Chundo Island and Geojedo Island, which are influenced by warm waters of the Kuroshio Current.

Quality Characteristics and Antioxidant Activity Research of Halocynthia roretzi and Halocynthia aurantium (우렁쉥이와 붉은멍게의 품질특성 및 항산화활성 연구)

  • In this study we investigated the antioxidant activities and quality characteristics of Halocynthia aurantium and Halocynthia roretzi. The pH of H. aurantium was higher than that of H. roretzi. The volatile basic nitrogens of H. roretzi and H. aurantium were 22.41 mg% and 16.80 mg%, respectively. Lightness and yellowness of H. roretzi were higher than those of H. aurantium, but redness of H. aurantium was higher. The results of sensory evaluation showed that the H. aurantium was better for color, odor, taste and acceptability. Total combined amino acid contents of H. roretzi and H. aurantium were $36368.23\;{\mu}mol/g$ and $36500.12\;{\mu}mol/g$, respectively. Our results showed that H. roretzi had relatively higher contents of Asp, Glu, Gly, DPPH radical scavenging activity, ABTS radical scavenging activity and reducing power. Also total phenol content of H. roretzi was higher than that of H. aurantium. The organoleptic properties of the H. aurantium were superior but the antioxidant activities were relatively lower than those of H. aurantium. For commercial usage, additional study would be helpful in the two ascidians to recommend.

Embryonic Development and Metamorphosis of the Ascidian Halocynthia aurantium (붉은멍게(Halocynthia aurantium)의 배발생과 변태)

  • The ascidian Halocynthia aurantium (sea peach), which belongs to the phylum Chordata, is thought to be a valuable organism of aquaculture like H. roretzi (sea pineapple), but its biological characteristics such as development and ecology are not well known. In this study, in order to obtain basic data for H. aurantium farming, the development processes of H. aurantium inhabiting the east coast of Gangwon-do were investigated and compared with those of H. roretzi, a related species. As a result, the morphology and developmental stages of the fertilized eggs, embryos and larvae of H. aurantium were very similar to those of H. roretzi. Fertilized eggs of H. aurantium took about 42.1 hours to hatch at 11℃, almost similar to 40.9 hours of H. roretzi. The time required for larvae to metamorphose into juveniles after hatching was very similar between the two species. The hatched larvae of the two species became juveniles with oral and atrial siphons after 23 days at 11℃. Both types of embryos developed slowly in seawater at low temperatures and rapidly developed at high temperatures. Fertilized eggs of H. aurantium hatched in an average of 62.3 hours at 9℃, 42.1 hours at 11℃, and 36.3 hours at 13℃, whereas those of H. roretzi hatched in an average of 60.4 hours, 40.9 hours, and 35.2 hours. Most of H. aurantium embryos did not develop normally above 15℃, so it is thought that attention is needed in the seed production processes.

In Vivo Evaluation of Chondroitin Sulfates from Midduk (Styela clava) and Munggae Tunics (Halocynthia roretzi) as a Cosmetic Material (In vivo에 의한 미색류 콘드로이틴황산의 기능성 화장품 소재로서의 가능성)

  • Crude chondroitin sulfates extracted from midduck tunics (Styela clava) and munggae tunics (Halocynthia roretzi) were examined in vivo in order to be utilized as a cosmetic material which was followed by an in vitro assay. Examinations, such as acute oral toxicity, skin sensitization, acute eye irritation, and primary skin irritation, were peformed with a variety of laboratory animals. Phototoxic and photosensitization tests were not conducted since all chondroitin sulfates failed to absorb U.V. light at the range of 280 to 420 nm. In acute dermal and eye irritation, both specific clinical signs and dead cases were not demonstrated during the test period, but crude chondroitin sulfates from midduck and munggae tunics, and standard chondroitin sulfate from bovine trachea were showed 2.5, 1 and 1.25 of acute ocular irritation index (A.O.I.), respectively. In the case of skin sensitization, crude chondroitin sulfate from midduck tunics exhibited neither specific clinical signs nor dead cases in the entire course of the examination. While in acute oral toxicity, crude chondroitin sulfates from both midduck and munggae tunics found neither specific clinical signs nor dead cases during the test, and LD50 was suspected to be over 2 g/kg. Based on this study, it was proven that crude chondroitin sulfates from either midduck or munggae tunics can be used safely as a cosmetic material.

Effects of Temperature and Salinity on Egg Development of Ascidiella aspersa (Ascidiacea, Phlebobranchia, Ascidiidae) (거친대추멍게(Ascidiella aspersa: Ascidiacea, Phlebobranchia, Ascidiidae)의 알 발육에 미치는 수온과 염도의 영향)

  • This study was performed to investigate the effects of water temperature and salinity on the egg development and larval attachment of Ascidiella aspersa. The egg development and larval attachment were examined in 12 different water temperatures (6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26 and $28^{\circ}C$) and two salinity conditions(30 and 34 psu). The hatching and developmental rates of A. aspersa showed a tendency to increase with increasing water temperature regardless of salinity and to decrease after the optimal water temperature range. The optimal water temperatures for the hatching and development of egg of A. aspersa were in the range of $20-22^{\circ}C$. The low threshold water temperature was not different between 1.5 and $1.8^{\circ}C$ at 30 and 34 psu, respectively. The attachment rate showed the optimal water temperature range of $16-22^{\circ}C$ irrespective of the salinity and the attachment time increased continuously with increasing water temperature. Experimental results showed that optimum development and survival temperature of the egg and larvae of A. aspersa were in the range of $20-22^{\circ}C$ regardless of the salinity conditions. The results can be used to predict the distribution and occurrence of A. aspersa, and to prevent economic damages caused by its spread.
