  • The objective of this study was to investigate the effect of hot water extract from Cornus walteri Wanger (CWE) on tert-butyl hydroperoxide (TBHP)-induced oxidative stress in HepG2 cells. Generation of reactive oxygen species (ROS), concentrations of cellular lipid peroxidation products and reduced glutathione, and antioxidant enzyme activity were used as biomakers of cellular oxidative status. Cells pretreated with CWE (25~200 ${\mu}g/mL$) showed an increased resistance to oxidative stress in a dose-dependent manner, as revealed by a higher percentage of surviving cells compared to control cells. ROS generation induced by TBHP was significantly reduced when cells were pretreated with 200 ${\mu}g/mL$ CWE for 4 h. Pretreatment with CWE (5~50 ${\mu}g/mL$) prevented the decrease in reduced glutathione and the increase in malondialdehyde and ROS evoked by TBHP in HepG2 cells. Finally, CWE pretreatments prevented the significant increase of glutathione peroxidase, catalase, glutathione reductase, and superoxide dismutase activities induced by TBHP. These results show that CWE has significant protective ability against a TBHP-induced oxidative insult and that the modulation of antioxidant enzymes by CWE may have an important antioxidant effect on TBHP-induced oxidative stress in HepG2 cells.

  • This studies were carried out to investigate the optimum shading for seedling cultivation of Cornus controversa and C. walteri. The experiment was performed under five different shading scheme such as 100%, 50%, 30%, 10%, and 2% of the natural full sun light intensity for 7 months in the field condition. Two species showed the highest height growth under 50% of relative light intensity, and the highest diameter growth at root collar was observed both under 100% and 50% of relative light intensity. C. controversa seedlings grown under 50% of relative light intensity produced more total biomass than control seedlings, but root biomass was less in the seedlings under 50% of relative light intensity. In case of C. walteri, total biomass production of the seedlings under 50% of relative light intensity was lower than that of the seedlings under full sun light intensity, but leaves and shoot biomass was slightly high under 50% of relative light intensity. Great reductions of height growth and diameter growth at root collar, and biomass production were observed below 30% of relative light intensity in the both species. T/R ratio of the two species was highest under 30% of relative light intensity, and the chlorophyll content of the seedlings tended to be increased by reduction of relative light intensity.

  • This study was conducted to verify the types of village forest in Gyeongsan City, North Gyeongsang Province, and a total of 32 village forests were investigated. The result indicated that village forests in Gyeongsan City can be classified as 8 village enterance forests(25.0%), 6 hillside forests(18.8%), 7 streamside forests(21.9%), and 11 village peripheral forests(34.4%). The major tree species in the village enterance forests are as follows: Zelkova serrata(34.3%), Ginkgo biloba(11.2%) and Robinia pseudoacacia(10.5%). The major tree species in hillside forests are: Pinus densiflora(56.8%), Quercus acutissima(26.4%) and Zelkova serrata(4.3%). The major tree species in the streamside forests are: Zelkova serrata(40.4%), Salix chaenomeloides var. chaenomeloides(27.1%) and Cornus wateri(5.5%). The major tree species in the village peripheral forests are: Zelkova serrata(21.7%), Quercus acutissima(10.8%) and Cornus walteri(8.9%). The habitat fragmentation due to transportation infrastructure, the function of habitats for village forests has deteriorated. In order to improve it, the management plan should be implemented and the seedlings of village forest trees should be carefully selected and propagated. Furthermore, the public should be educated about the importance of preserving and developing those village forests.

  • This study was conducted to determine the characteristics and utilization of pruned kiwifruit branches as wreath materials. Amount of pruned branches produced from one 11~15 year-old kiwi plant cv. 'Hayward' can be used for one wreath with 76cm in diameter. Making a wreath with 76cm in diameter with the pruned kiwifruit branches required 75 branches with 31~40cm length. And the cost to make a wreath with 76cm in diameter for the Keaki and the paper bush were cheeper than those of winged spindle tree and Korean dogwood. Floral designers evaluated that the glossy, the delicacy and the unique form of cut branches, and their various hue were lower than that of the commonly-used cut branches, however, the beauty of curved line, the characteristics after drying, and the utilization and prospect of pruned kiwi branches were higher by 'over good' than that of the commonly-used branches. Consequently, it was assumed that the utilization of pruned kiwifruit branches can be contributed to the improvement of side income for a farm household, and the diversity wreath materials.

  • Cornus walteri Wanger has been used in folk medicine in Korea. Ultraviolet (UV) irradiation has been known as a major cause of photo damage in skin. In the present study, research on how to cure damaged cells by UVB was conducted using an extract of Cornus walteri Wanger leaves (CWE), which was treated with an enzyme. CWE was applied to human dermal fibroblasts (HDFs) affected by UVB. UVB-irradiated HS68 cells showed increased caspase-3 activity, phosphorylation of p53, ${\gamma}H2AX$, cyclobutane pyrimidine dimers (CPDs) formation, and DNA fragmentation compared with non-irradiated cells. However, all these effects were inhibited by treatment with CWE for 12 h after UVB irradiation. Furthermore, CWE has proved not to cause primary skin irritation through the human patch test. Collectively, these results suggest that CWE could be a new potential candidate as photoprotective agent against UVB-induced cellular damage in HDFs.

  • ${\alpha}-Amylase$ inhibitor is used to control blood glucose level by inhibiting starch digestion in the small intestine and delaying the absorption of glucose. In this study, we investigated the effect of the ethanol extracts from more than 1400 species of plants against ${\alpha}-amylase$ with the aim of developing a new ${\alpha}-amylase$ inhibitor. In the results, Cornus walteri extracts showed the highest inhibition activity. The inhibitory effect of Cornus walteri extract on the carbohydrate hydrolysis enzymes has different sensitivities against ${\alpha}-amylase$ from salivary and pancreatin and against ${\alpha}-glucosidase$ from yeast and porcine small intestine. In the study of inhibition kinetics of ${\alpha}-amylase$ and ${\alpha}-glucosidase$, Cornus walteri extract showed competitive inhibition against salivary and pancreatin while showing the combination of uncompetitive and noncompetitive inhibition against ${\alpha}-glucosidase$. The Cornus walteri extract was stable at acidic and thermal conditions. As for the blood glucose and body weight levels of Cornus walteri extract, we confirmed anti-hyperglycemic and anti-obesity effects. Also, in the investigation of the mRNA lever, Cornus walteri extract upregulated the level of GLUT4 mRNA in the quadriceps muscle.
