Material and Heat Balances of Bioethanol Production Process by Concentrated Acid Saccharification Process from Lignocellulosic Biomass (목질계 Biomass로부터 강산 당화 공정에 의한 Bioethanol 생산 공정의 물질 및 열수지)

  • The process for bioethanol production from lignocellulosic biomass was studied through process simulation using PRO/II. Process integration was conducted with concentrated acid pretreatment, hydrolysis process, SMB (simulated moving bed chromatography) process and pervaporation process. Energy consumption could be minimized by the heat recovery process. In addition, material and energy balance were calculated based on the results from the simulation and literature data. A net production yield of 4.07 kg-biomass and energy consumption value of 3,572 kcal per 1 kg ethanol were calculated, which is indicating that 26% yield increase and 30% energy saving compared to the bioethanol production process with dilute-acid hydrolysis (SRI report). In order to make it possible, sugar conversion yield of cellulose and hemi-cellulose is to be reached up to 90% and fermentation of xylose needs to be developed. In order to reduce the energy consumption up to 30%, the concentration of acid solution after being separated by 5MB should exceed 20%. If acid/sugar separation by SMB process is to be practical, the bioethanol process designed in this study can be commercially feasible.

Functional Properties of Sunmul (Soybean Curd Whey) Concentrate by Ultrafiltration (한외여과에 의한 순물 농축액의 기능적 특성)

  • This study was carried out in order to investigate the feasibility of utilizing concentrated sunmul (soybean curd whey), which is a waste by-product of soybean curd processing, as a functional food ingredient. Sunmul Powder was concentrated by ultrafiltration and spray dried with or without dextrin. Oil adsorption capacity of UF retentate powder was similar to that of ISP (Isolated Soy Protein) and higher than that of sunmul powder, whereas water holding capacity of UF retentate powder was lower than that of ISP. Protein solubility of all types of UF retentate powder was significantly higher than that of ISP at pH 2.0-10.0 with the lowest protein solubility seen at pH 4.0 and solubility increasing as the conditions became more acidic or alkaline. Emulsifying activity indexes of UF retentate powder at pH 2.0-10.0 were not influenced by pH. Emulsion stability of 4% sunmul solution was lowest at pH 4.0, but that of UF retentate powder was higher at acidic pH values and decreased with increasing pH. Foaming capacities of sunmul and UF retentate powder were high at pH 4.0-6.0, but the foam of UF retentate powder disappeared within 20 minutes in all conditions of pH.

Fabrication and characterization of 3-D porous scaffold by polycaprolactone (폴리카프로락톤을 이용한 3차원 다공성 지지체 제조 및 특성 분석)

  • This study was a preparatory experiment aimed the development of membrane scaffolds for tissue engineering. A PCL composite solution contained sodium chloride(NaCl). PCL porous membrane scaffolds were formed on a glass casting plate using a film applicator and immersed in distilled water to remove the NaCl reaching after drying. NaCl was used as a pore former for a 3 dimensional pore net-work. The dry condition parameters were $4^{\circ}C$, room temperature (RT) and $40^{\circ}C$ for each different temperatures in the drying experiment. SEM revealed the morphology of the pores in the membrane after drying and evaluated the in vitro cytotoxicity for basic bio-compatibility. The macro and micro pores existed together in the scaffold and showed a 3-dimensional pore net-working morphology at RT. The in vitro cytotoxicity test result was "grade 2" in accordance with the criterion for cytotoxicity by ISO 10993-5. The dry condition affected the formation of a 3 dimensional pore network and micro and macro pores. Therefore, these results are expected provide the basic process for the development of porous membrane scaffolds to control degradation and allow drug delivery.

Quality Characteristics of Mul-kimchi Prepared Using Eastern Deep Seawater Added with Chitosan-Ascorbate (Chitosan-Ascorbate 함유 동해심해수로 담근 물김치의 품질특성)

  • Quality characteristics of mul-kimchi prepared using eastern deep seawater added with $0.1\%$ chitosan-ascorbate (CA) were investigated during fermentation for 12 days at $10^{\circ}C$, in an effort to effectively utilize the eastern deep seawater in Korea. Four water sources were considered for preparation of mul-kimchi; (1)distilled water added with $1\%$ mineral salt (DW), (2) eastern deep seawater (containing $6\%$ salt) diluted to $1\%$ salt concentration with distilled water (ED), (3) DW+CA (DW-CA), and (4) ED+CA (ED-CA). Juice of mul-kimchi prepared with ED-CA as a water source showed higher PH and antioxidant activity, and lower acidity, total microbe numbers, turbidity and alcohol insoluble substance compared with those Prepared with other three water sources. The former also showed higher overall acceptability compared with the latter. This study demonstrated the possibility of use of eastern deep seawater added with chitosan-ascorbate as a water source for extension of shelf-life of mul-kimchi.

Effect of Saccharides and Incubation Temperature on pH and Total Acidity of Fermented Black Tea with Tea Fungus (배양액 제조에 사용된 당의 종류와 농도 및 배양 온도가 Tea Fungus발효 홍차의 pH 변화와 Total Acids생성에 미치는 영향)

  • The fermented black tea with tea fungus (FBTF) known to cure various diseases was prepared by culturing tea fungus biomass in black tae with different kinds (sucrose, glucose, fructose and corn syrup) and concentrations (10-60% m/v) of saccharides. pH changes and total acid production of FBTF were investigated during 14 dats incubation at $5-45^{\circ}C$ The patterns of acid production in sucrose and fructose black tea solution were similar each other, and those in glucose and corn syrup black tea solution were similar each other showing that tea fungus biomass utilizes fructose than glucose more efficiently. The optimum incubation temperature for the formation and growth of tea fungus Biomass, and the acid production was $30^{\circ}C$ Low contents of total aids (0.1%-0.2%) were produced in 20% of higher concentrations of sucrose and fructose black tea solution, while 1.8% and 0.68% were produced in the 10% of sucrose and fructose black tea solutions, respectively, pH of FBTF dropped to 2.74-3.56 after 2 days of incubation of all the culture solution and Kept this level to 14 days incubation.

A Study on the Change of Streptococcus Mutans in Dental Plaque after Use of $0.05\%$ NaF in Orthodontic Patients (NaF $0.05\%$ 양치액 사용시 고정성 교정장치 장착 환자에서의 Streptococcus mutans 변화에 관한 연구)

  • The purpose of this study was to evaluate the effectiveness of gargling solution with 0.05% NaF and 10% Xylitol in orthodontic patients with fixed appliance. The sample consisted of 20 patients who were classified into an experimental group and a control group, 10 patients each. Experimental group was used experimental gargling solution and the control group was used with placebo solution. The change of S. mutans was analysed by culture on MSB and BHI agar plate pre and post 3, 6, 9 weeks. The results were as follows. 1 There were significant reduction in the number of S. mutans C.F.U. between pre and post 3 weeks(p<0.01), 9 weeks(p<0.05) in experimental group 2. There were significant reduction in the ratio of S. mutans C.F.U. to total C.F.U. between pre and post 3, 6, 9 weeks(p<0.01) in experimental group. 3. S. mutans, which were reduced until 3 weeks, did not show significant change after 3, 6, 9 weeks. 4. S. mutans were strongly suppressed until 3 weeks after gargling solution with 0.05% NaF and 10% Xylitol.

  • 1. 목적 생체재료의 생체친화성을 증진시키고 치유를 촉진하기 위한 목적으로 생체재료의 생화학적 표면개질에 관한 연구가 널리 진행되고 있다. 이와 같은 목적으로 이용되어 온 부착분자에는 아미노산, 펩타이드, 단백질, 효소 및 성장인자들을 들 수 있으며, 이들 분자들을 금속, 골대체물질 및 폴리머와 같은 생체재료의 표면개질에 이용하여 왔다. 이 연구의 목적은 생체적합성이 우수하고 생분해성을 지닌 키토산으로 얇은 막을 제작한 후, 세포외 기질의 구성성분 중 세포부착에 관여하는 RGD 펩타이드를 부착시킨, 표면개질 키토산막의 생물학적 영향을 MG-63 조골양세포를 이용하여 관찰하는 것이다. 2. 방법 2% acetic acid에 키토산 가루를 녹여 만든 2% 키토산 용액으로 24-well 배양접시의 표면을 도포 후 24시간 동안 건조시켜 키토산막을 제작하였다. GRGDS 펩타이드를 cross-linker(EDC, NHS) (Sigma, MO, USA) 용액과 반응시켜서 펩타이드의 카르복실기를 활성화시켰다. 이들을 PBS 완충용액으로 수화시킨 키토산막과 결합시켜 펩타이드의 활성화된 카르복실기와 키토산의 아민기 간에 안정적인 아미드 결합(amide bond)이 형성되도록 하였다. 하루 동안 반응을 일으킨 후 PBS 완충용액과 증류수로 씻어내고 냉동 건조시킴으로써 GRGDS가 결합된 키토산막을 제작하였다. 재료 표면의 화학 성분을 알아보는데 사용되는 방법의 일종인 X-ray photoelectron spectroscopy(XPS) 분석을 통하여 부착분자가 키토산막에 결합된 여부를 확인하였다. GRGDS 펩타이드에 요오드를 결합시킨 후, 이것을 키토산막에 공유 결합시키고 XPS를 통해 요오드가 재료 표면에서 검출되는지를 검사하였다. 요오드가 검출된다면 이것은 키토산막 표면에 실제로 GRGDS 펩타이드가 존재하는 것을 의미하게 된다. 표면개질된 키토산막에 사람조골양세포인 MG-63을 접종하여 이를 실험군으로 하였고, 표면이 개질 되지 않은 키토산막을 대조군으로 하였다. 세포부착의 최적화 농도를 확인하기 위하여 GRGDS를 0.01, 0.05, 0.1, 0.25, 0.5, 1.0mg/ml의 농도로 준비하였다. 배양 후 1일, 7일째에 각 well에서 trypsin EDTA를 이용하여 세포를 분리한 후, 이를 원심 분리하여 세포수측정기를 이용하여 부착 세포의 수를 측정하여 세포의 부착 정도를 비교하였다. 배양 2시간, 24시간 후 주사전자현미경을 이용하여 키토산막에 부착된 세포의 양상을 관찰하였다. 3. 결과 XPS를 통한 표면의 화학 성분 분석 결과 GRGDS 펩타이드를 결합시킨 키토산막에서 요오드가 검출되었으며 펩타이드를 부착하지 않은 대조군에서는 검출되지 않았다. 따라서 cross-linker를 이용한 펩타이드와 키토산막의 공유결합을 확인할 수 있었다. 세포 배양 후 1일째 부착된 세포 수를 측정한 결과 0.1mg/ml 이상의 GRGDS 펩타이드 농도로 공유 결합시킨 키토산막에서 부착 세포 수가 다른 농도에 비해 유의성 있게 많이 관찰되었다. 이 농도 이하에서는 대조군과 실험군간에 세포부착의 유의한 차이가 없었다. 따라서 주사전자현미경을 이용한 부착 세포의 양상에 관한 관찰은 0.1mg/ml 농도의 펩타이드를 이용하였다. 세포 배양 7일째, 부착된 세포 수 측정 결과 GRGDS의 농도에 따른 유의성 있는 차이가 없었으며, 실험군과 대조군간에도 유의성 있는 차이가 없었다. 주사전자현미경 관찰결과 2시간 및 24시간 배양된 실험군 모두에서 별모양의 세포들이 키토산막 표면에 편평하게 잘 부착되어 있으며 많은 위족이 발달된 소견을 보인 반면, 대조군에서는 원형 또는 다각형 모양의 세포들이 실험군에 비해 부착이 덜 되어있는 양상을 보였다. 이 연구를 통하여 기능성 펩타이드를 생체재료의 표면에 공유결합 시키는 방법을 확립할 수 있었으며, RGD 펩타이드의 공유결합으로 표면개질된 키토산막이 조골세포의 부착능을 증진시킬 수 있음을 확인하였다. 표면개질된 생체재료를 소, 중동물에 적용시켜 생체 내에서의 생물학적 영향을 평가할 필요가 있으며, 이 실험의 결과는 향후 다양한 기능성 부착분자를 선발, 고안하여 임플란트용 생체재료의 표면개질에 이용하는 이른바 모방생체재료분야에 널리 활용될 수 있을 것으로 생각된다.

Morphological Characteristics of Seed Coat and Enzyme Activity of Aleurone Layers in Soybeans (콩 종피의 형태적 특성과 호분층 세포의 효소활성 연구)

  • Soybean seed coat consisted of three layers, and the aleurone layer was attached to the innermost part of seed coat. It showed the different morphological characteristics with single cell layer compared with many cell layers in barley aleurone layer. The structural difference in aleurone cell among varieties was not detected in this experiment. The hole of middle part of hilum consisted of net formed material in order to pass water and gas. In the experiment, it was not studied whether the varieties with hard seed consist of the same structure or not, but the detailed study on this would be necessary. The activity of acid phosphatase showed a tendency to increase in process of imbibition in distilled water. There was no significant difference in the enzyme activity among the varieties tested, but the enzyme activity of Danyoupkong was slightly higher than that of Hwanggeumkong. In germinability, Danyoupkong is higher than Hwangkeumkong, so it might be attributed to the higher activity. There was no difference in acid phosphatase activity between released from the aleurone cell and accumulated in the aleurone cell from 6 to 12 hours incubation of the medium in the absence of GA$_3$, but the difference was detected after 12 hours incubation. And enzyme activity was the highest in the 18 hours incubation. The increase in the release of acid phosphatase from the aleurone cell would be considered as passive diffusive effect due to the increase in turgo pressure of aleurone cell. The acid phosphatase released from aleurone layer increased greatly after 24 hours incubation of the medium in the presence of GA$_3$ and the accumulation within the aleurone cell decreased linearly after 18 hours incubation. The result indicates that GA$_3$ enhance the rate of enzyme release from aleurone layer, suggests that the aleurone cell wall be digested by the introduction of GA and the digested wall act as the channels for enzyme release.

Ameliorating effect of the ethyl acetate fraction of Pteridium aquilinum on glucose-induced neuronal apoptosis (포도당으로 유도된 신경세포 손상에 대한 고사리 아세트산에틸 분획물의 개선 효과)

  • The protective effect of Pteridium aquilinum on high glucose-induced cytotoxicity was examined in vitro to investigate the relationship between diabetic condition and neuronal dysfunction. The ethyl acetate fraction of P. aquilinum (EFPA), with total phenolic content of 265.08 mg gallic acid equivalent/g, showed higher 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)/2,2-diphenyl-1-picrylhydrazyl radical scavenging activities and lipid peroxidation inhibitory effect than any other fraction. In addition, EFPA showed a significant reduction in the inhibitory effect on ${\alpha}$-glucosidase activity ($IC_{50}$ value=$205.26{\mu}g/mL$) compared to the acarbose positive control. The anti-oxidative effect in PC12 cells, protective effects on high glucose-induced oxidative stress in neuronal cells, and neurotoxicity were measured using 2',7'-dichlorofluorescin diacetate, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazoliumbromide, and lactate dehydrogenase assays, respectively. EFPA showed conspicuous inhibitory effect on cellular reactive oxygen species production and neuronal cell apoptosis. Finally, kaempferol-3-glucoside was identified as the main phenolic compound of EFPA using high performance liquid chromatography.
