Quality Characteristics of Makgeolli Added with Kiwifruit (Actinidia deliciosa) (키위를 첨가한 막걸리의 품질특성)

  • This study was conducted to investigate the physicochemical and microbial properties of Makgeolli supplemented with kiwifruit (Actinidia deliciosa). Four hundred grams of kiwifruit were added to 3.1 L of distilled water, followed by 2.0 kg of rice, 40.0 g of Nuruk, and 14.0 g of yeast. The mixed rice solution was then fermented at $28^{\circ}C$ for 6 days to prepare the kiwifruit Makgeolli. The pH values of the kiwifruit Makgeolli decreased from 5.31 to 4.37, but the total acidity values increased from 0.05 to 0.34% during fermentation. The total viable cells ($3.18{\times}10^7$ and $2.88{\times}10^7$, respectively), lactic acid bacteria ($1.51{\times}10^6$ and $1.50{\times}10^6$, respectively), and yeast counts ($1.96{\times}10^7$ and $1.90{\times}10^7$, respectively) of the kiwifruit Makgeolli and control were similar throughout the fermentation process. Glucose was the major free sugar in the control and kiwifruit Makgeolli and significantly decreased during fermentation. Succinic acid was the highest organic acid in both the control (24.6 mg/mL) and kiwifruit Makgeolli (26.3 mg/mL). In a volatile compound analysis, 3-methyl-1-butanol, 2-methyl-1-propanol and ethyl acetate were the major volatile compounds in the kiwifruit Makgeolli.

  • Makgeolli is Korean traditional alcoholic beverage that has historically been brewed. In this study, we analyzed the profile of organic acids in makgeolli and also evaluated its physiological characteristics. Makgeolli contained excess lactic acid, which is produced by lactic acid bacteria (LAB). Anti-obesity effects of makgeolli were investigated in 3T3-L1 preadipocytes. Compared to the negative control, makgeolli inhibited the differentiation of preadipocyte as quantified by Oil red O dye. In particular, $100{\mu}g/mL$ makgeolli reduced 40 to 70% of differentiation. To evaluate the anti-angiogenic and anti-inflammatory effects of makgeolli, we performed chorioallantoic membrane assay and measured nitric oxide production from lipopolysaccharide-induced RAW264.7 cells. Most makgeolli interrupted the formation of neo-vasculature and significantly inhibited NO production in a dose-dependent manner. Taken together, these findings suggest that commercial makgeolli has inhibitory activities against adipogenesis, neo-vascularization, and inflammation, and also they are influenced by second metabolites from nuruk microflora containing fungi and LAB.

Inhibition Effect against the Rat Blood Plasma Oxidation of the $Makgeolli$ ($Takju$) Korean Rice Wine (막걸리(탁주)의 쥐 혈장 과산화물 생성 억제효과)

  • The antioxidant activities of $makgeolli$ and other alcoholic beverages were compared. Based on the same volume (70 ${\mu}L$ eq.) of the alcoholic beverages, the 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid ammonium salt) (ABTS+) radical-scavenging activities were as follows: whisky > $makgeolli$ crude extract (MCE) > rice wine (RW) > clarified $makgeolli$ (CM) > soju. Based on the same alcohol concentration (6%) of the alcoholic beverages, however, $makgeolli$ showed the highest activity. In addition, based on the same volume (70 ${\mu}L$ eq.), the inhibition effects against the formation of cholesteryl ester hydroperoxide (CE-OOH) were as follows: soju > whisky > RW > MCE > CM. Based on the same alcohol concentration (6%), however, the inhibition effects against the formation of CE-OOH were as follows: RW > MCE > soju > whisky > CM. Therefore, it was suggested that $makgeolli$ may contain radical-scavenging- and metal-ion-chelate-type antioxidants and may increase the antioxidant activity in the blood.

  • In recent years Mag-Geol-Li is becoming popular again and the consumption layer of Mag-Geol-Li is definitely younger. Nowadays, Mag-Geol-Li is the main traditional alcohol and without consideration of ages and genders whoever can enjoy Mag-Geol-Li. Producers should ensure fair price of goods and improve production facilities to maximize products to meet the needs of consumers who will buy. In the studies I discuss about the ways to improve the distribution of Mag-Geol-Li, the theories about Mag-Geol-Li, Social backgrounds and effects of Mag-Geol-Li why it is becoming popular and improvement of the distribution structure of Mag-Geol-Li and impact of big Mag-Geol-Li company in Mag-Geol-Li industry. Lately, people can enjoy Mag-Geol-Li without burden because of low alcohol. It is gaining a good response by consumers because Mag-Geol-Li contains lactobacillus, yeasts and dietary fiber, also it is well balanced with nutritional aspects. There are not only huge factory to produce Mag-Geol-Li but also most are still cottage industry. Therefore, quality management does not work well because the majority of Mag-Geol-Li is small businesses. In spite of there are a lot of ways to improve the distribution structure and problems of Mag-Geol-Li, but none provide any improvement that make consumer awareness about the main cause of the problem is inaccurate. The ultimate goal of improving the distribution structure of Mag-Geol-Li is activating the market, the improvement of production facilities and safe for consumers with affordable price and good quality product. As mentioned above, to improve and change the logistics systems and hygiene, government support is required and Mag-Geol-Li companies should be recognized the distribution system as a major problem.

Changes of Physicochemical Properties and Microbial during Storage of Commercial Makgeolli (시판막걸리의 저장기간에 따른 품질 특성 및 미생물의 변화)

  • The aim of this study was to determine changes of physicochemical properties and microorganisms of commercial Makgeolli during storage at $10^{\circ}C$ and $25^{\circ}C$ During storage at $10^{\circ}C$ for 11 days, ethanol contents did not increased up to 7.0%, and pH and total acidity were ranged with 3.12-3.99 and 0.22-0.28%, respectively. Makgeolli which was storaged at $25^{\circ}C$ for 11 days showed more than 7.0% of alcohol contents, pH of 3.96-4.17 and total acidity of 0.27-0.30%. Yeast cell counts showed maximal $2.90-16.00{\times}10^7$ CFU/mL at $10^{\circ}C$ for 3 days and $1.12-29.40{\times}10^7$ CFU/mL at $25^{\circ}C$ for 1 day. Lactic acid bacteria and total bacteria cell counts of Makgeolli which was storaged at $25^{\circ}C$ for 11 days were higher than those $10^{\circ}C$ storage. In preference test, Makgeolli from storage at $10^{\circ}C$ for 11 days was showed the level of 4.9-5.2 points however, Makgeolli from storage at $25^{\circ}C$ for one day showed below 5.0 point.

Processing and Antioxidant Activity of Makgeolli Using Mistletoe Cultivated on Prunus mume (매실나무(Prunus mume) 겨우살이 막걸리의 제조 및 항산화활성)

  • 인공재배한 매실나무겨우살이를 이용한 기능성 막걸리를 개발하기 위해, 겨우살이, 기장과 쌀을 활용하여 원료의 배합비율 및 발효방법에 따른 총 폴리페놀, 플라보노이드 함량 및 DPPH free radical 소거능을 비교하였다. 최적의 배합비율을 찾기 위해 백미 100%, 백미와 기장 7:3, 5:5, 3:7 및 기장 100%의 비율로 막걸리를 제조하였다. 발효방법은 Shaking incubator($30{\sim}35^{\circ}C$, 100 rpm, 19 days), Hand Shaking($21^{\circ}C$, per 12hr, 19days)하였고, 발효가 끝난 막걸리는 80 mesh 망으로 여과하였다. 총 폴리페놀 함량은 Machine shaking과 Hand shaking 모두 쌀과 기장의 비율 7:3에 겨우살이를 10% 첨가한 막걸리가 각각 $795.83{\mu}g/ml$, $757.87{\mu}g/ml$로 가장 높았고, 기장 100% 막걸리가 각각 $503.73{\mu}g/ml$, $435.3{\mu}g/ml$로 가장 낮았다. 총 플라보노이드 함량은 Machine shaking, Hand shaking 모두 쌀과 기장의 비율이 7:3에 겨우살이를 10% 첨가한 막걸리가 $93.48{\mu}g/ml$, $84.56{\mu}g/ml$로 가장 높았고, 기장 100% 막걸리가 각각 $37.75{\mu}g/ml$, $21.86{\mu}g/ml$로 가장 낮았다. DPPH free radical 소거능은 겨우살이를 10% 첨가한 막걸 리가 가장 높았는데 Machine shaking은 평균 92.08%, Hand shaking은 평균 91.63%로 가장 높았으며 통계적으로 유의한 차이가 없었다. Machine shaking은 쌀과 기장의 비율이 7:3일 때, 73.37%, Hand shaking은 기장 100% 막걸리가 64.03%로 가장 낮았다. 결론적으로, 총 폴리페놀 및 플라보노이드 함량은 기장의 비율이 증가함에 따라 감소하는 경향을 보였고, DPPH free radical 소거능은 겨우살이를 첨가한 모든 막걸리에서 비교적 높았다. 발효방법은 Machine shaking이 Hand shaking 보다 총 폴리페놀 및 플라보노이드 함량, DPPH free radical 소거능이 비교적 높아 발효방법으로 더 적합하였다.

Fermentation and Sensory Characteristics of Korean Traditional Fermented Liquor (Makgeolli) Added with Citron (Citrus junos SIEB ex TANAKA) Juice (유자즙을 첨가하여 제조한 막걸리의 발효기간 중 이화학적 특성 및 제조된 막걸리의 관능적 특성)

  • This study was performed to examine the physicochemical characteristics of Korean traditional fermented liquor added with citron juice and the levels (3, 6 and 9%) of citron (Citrus junos) juice during two-step-brewing. The pH of traditional Korean fermented liquor added with citron juice increased after 3 days of fermentation and then the pH values decreased from the sixth day. pH increased again at the end of fermentation. Total acidity increased after 5 days of fermentation until the sixth day, then decreased rapidly but increased slightly up to 12 days. Alcohol content increased to 12.76-14.32% after 12 days of fermentation. As the amount of added citron juice increased, alcohol content decreased. Amino acids increased after 12 days of fermentation. A sensory evaluation appearance, flavor, sourness, sweetness, bitterness and overall acceptance of Korean traditional fermented liquor added with 6% citron showed higher values than those of the control.

  • Today's liquor culture in Korea is basically divided into a general liquor culture and a functional liquor culture. In particular, the market of the latter has been going through a rapid change. In the previous days, it was a crop for a special purpose or a fruit which was mostly used to make liquor but now, many other regular crops are being used in the liquor market. At present, while the demand for Makgeolli has risen sharply, the designs of Makgeolli bottles have never satisfied needs from consumers nor the trends in the consumer market. As a result, Makgeolli industry should come up with new designs for the bottles immediately if it dose not want to lose in a competition with other liquor products. The study has looked into existing designs of Makgeolli bottles, investigating how the designs have changed over the past years and how competitive they are in the market. Based on results from the investigation, the study focuses on finding out designs which would fit the trends in the consumer market to develop Makgolli bottles which would be equipped with both functionality and practicality. In other words, to make Makgeolli survive in these rapid changes in the liquor market, the study works on offering a Makgeolli bottle design which is appropriate for certain characteristics of Makeolli and also competitive over other liquor bottles.

A Survey of Drinking Habits and Perception of Makgeolli Targeting the Chinese Students Studying in Korea (한국거주 중국인 유학생의 막걸리 음용 실태 및 인식 조사)

  • This study was intended to investigate the alcohol drinking characteristics, Makgeolli drinking behavior, and perception of Makgeolli focusing on the Chinese students studying in Korea. 550 copies of self-administrated questionnaire for a survey were distributed from May 2nd to June 30th, 2012, and then 461 copies were adopted for the analyses. The drinking frequency and drinking amount at a time increased as the length of stay in Korea and monthly pocket money increased, and Makgeolli was second preferred, following beer. 97.5% of the students experienced drinking Makgeolli, the drinking rate and drinking amount of Makgeolli increased with the length of stay in Korea, and they mostly drank at restaurants and bars. Common motivations for drinking Makgeolli were taste and atmosphere while drinking it, and bad taste and headache were the most common reasons for disliking Makgeolli. The awareness of commercial Makgeolli types was low, and for the price and health promoting effects of Makgeolli, 92.4% and 85.8% of the students replied positively, respectively. These results suggest that the development of high quality Makgeolli adapted to the tastes of Chinese and relieving hangover, and advertising the health benefits would promote Makgeolli consumption in Chinese market.

Isolation of Pediococcus Strain from Nuruk and Anti-Lipid Accumulation Effect of Ornithine-Containing Makgeolli on 3T3-L1 Cells (누룩으로부터 오르니틴 생성능을 갖는 Pediococcus 속 균주의 분리 및 오르니틴 함유 막걸리의 3T3-L1 세포의 중성지질 축적 억제 효과)

  • To evaluate the functional effect of ornithine produced by isolated lactic acid bacteria, we examined the anti-lipid accumulation effect of ornithine produced by isolate lactic acid bacteria on 3T3-L1 cells. Lactic acid bacteria (Pediococcus strain) were isolated from nuruk, which is made from wheat, rice, and barley (whole grain, grits, or flour) by fermenting microorganisms (Aspergillus, Rhizopus, and yeasts). Pediococcus strain was identified by 16S rDNA sequencing analyses, and cells were collected by centrifugation and developed as an ornithine starter. makgeolli, an ornithine-containing Korean traditional alcoholic beverage, was made with isolated lactic acid bacteria and arginine. makgeolli was made with the help of ornithine starter using a makgeolli making kit. We evaluated the anti-proliferation effect of ornithine makgeolli on 3T3-L1 cells. To determine the anti-proliferation effect of ornithine makgeolli on preadipocytes, lipid droplets were quantified and stained with Oil Red O. makgeolli made with ornithine starter and arginine showed a 3-fold higher concentration of ornithine compared to makgeolli without starter and arginine. In the results of 3T3-L1 cell line experiment, lipid accumulation was significantly reduced by adding 0.05 mg/mL of ornithine makgeolli compare to the control (adipocyte without sample). In conclusion, ornithine makgeolli containing ornithine starter isolated from nuruk showed an anti-lipid accumulation effect with increased ornithine content without toxicity.
