Identification and Chromosomal Reshuffling Patterns of Soybean Cultivars Bred in Gangwon-do using 202 InDel Markers Specific to Variation Blocks (변이영역 특이 202개 InDel 마커를 이용한 강원도 육성 콩 품종의 판별 및 염색체 재조합 양상 구명)

  • The areas of soybean (Glycine max (L.) Merrill) cultivation in Gangwon-do have increased due to the growing demand for well-being foods. The soybean barcode system is a useful tool for cultivar identification and diversity analysis, which could be used in the seed production system for soybean cultivars. We genotyped cultivars using 202 insertion and deletion (InDel) markers specific to dense variation blocks (dVBs), and examined their ability to identify cultivars and analyze diversity by comparison to the database in the soybean barcode system. The genetic homology of "Cheonga," "Gichan," "Daewang," "Haesal," and "Gangil" to the 147 accessions was lower than 81.2%, demonstrating that these barcodes have potentiality in cultivar identification. Diversity analysis of one hundred and fifty-three soybean cultivars revealed four subgroups and one admixture (major allele frequency <0.6). Among the accessions, "Heugcheong," "Hoban," and "Cheonga" were included in subgroup 1 and "Gichan," "Daewang," "Haesal," and "Gangil" in the admixture. The genetic regions of subgroups 3 and 4 in the admixture were reshuffled for early maturity and environmental tolerance, respectively, suggesting that soybean accessions with new dVB types should be developed to improve the value of soybean products to the end user. These results indicated that the two-dimensional barcodes of soybean cultivars enable not only genetic identification, but also management of genetic resources through diversity analysis.

  • 본 연구는 가상 캐릭터인 3D Pooh 와의 감성 교감이 이루어지는 유아용 감성 교육 게임 개발이 목적이다. Web 카메라를 이용하여 사용자의 위치 정보 및 영역을 검출하고 사용자는 화면에 보이는 가상 공간에 손과 마커를 이용하여 행동을 가하게 된다. 사용자의 행동 입력 방식에 따라 Pooh 의 상태를 나타내는 게이지와 행동의 다양한 변화를 제공하며, 이로써 사용자들이 가상 캐릭터인 3D Pooh 와 인터렉션을 할 수 있도록 제공한다. 기존의 감성 교감 연구 방법 분석 결과 뇌파와 체온 등의 생리신호를 이용한 방법과 사용자의 행동패턴을 분석하는 방법 등이 있으며, 현재는 생리신호를 이용하는 방법이 더 많이 활용되고 있다. 생리신호 측정 시 사용자는 신체에 센서를 착용해야 하므로 착용 부담감과 움직임이 자유롭지 못하다는 단점이 있다. 본 프로그램은 기존 연구의 단점인 센서 착용의 부담감을 줄이고 자유롭고 편리한 인터렉션이 가능하도록 개발하였으며, 직관적인 인터페이스를 기반으로 유아들도 쉽고 편리하게 즐길 수 있도록 구현된 감성 교육 게임이 될 것으로 기대된다. 또한 카메라 이외의 다른 하드웨어 장치 사용을 최소화하여 경제성과 사용 편의성을 동시에 고려한 효율적인 유아용 감성 교육 게임으로 활용될 수 있다.

Improved Motion-Recognizing Remote Controller for Realistic Contents (실감형 컨텐츠를 위한 향상된 동작 인식 리모트 컨트롤러)

  • This paper describes the improvements made on hardware and software of the remote controller for realistic contents. The controller can provide vibrotactile feedback which uses both of a voice-coil actuator and a vibration motor. A vision tracking system for the 3D position of the controller is optimized with respect to the marker size and the camera parameters. We also present the improvements of motion recognition due to the effective motion segmentation and the fusion of vision and acceleration data. We apply the developed controller to realistic contents and validate its usability.

  • This paper presents the pose estimation of a small UAV utilizing visual information from low cost cameras installed indoor. To overcome the limitation of the outside flight experiment, the indoor flight test environment based on multi-camera systems is proposed. Computer vision algorithms for the proposed system include camera calibration, color marker detection, and pose estimation. The well-known extended Kalman filter is used to obtain an accurate position and pose estimation for the small UAV. This paper finishes with several experiment results illustrating the performance and properties of the proposed vision-based indoor flight test environment.

  • 환경오염의 조기 경보 시스템으로 생체지표를 이용한 생태독성 모니터링이 최근 널리 연구되고 있다. 환경내의 생물종에서 측정한 생체지표를 이용한 환경 모니터링은 생태계 수준의 영향에 대한 예측 정보를 제공해 줄 수도 있다. 이를 위해서는 생체지표와 개체군 수준에서의 반응과의 인과관계가 밝혀져야 한다. 오염물질에 대한 생체의 반응은 분자, 세포, 생화학, 생리적, 개체, 개체군, 군집 수준에서 나타나게 되며, 이러한 각 단계별 반응은 반응 시간의 규모와 독성학적, 생태적 관련성에 따라 구분 지어 볼 수 있다. 각 개별 수준에서의 반응을 종합하면 오염물질에 노출된 생체의 전제적인 영향을 이해할 수 있으며, 이러한 이해를 바탕으로 개체군에서 나타나는 영향에 대한 인과관계를 추론할 수 있다. 생체지표와 개체군 수준의 반응과의 인과관계 정립은 효율적인 환경오염 예방기능 수행에 필수적인 과정이며, 다단계 생체 지표는 각 단계별 반응의 인과관계를 밝히기 위해 가장 적절한 접근 방법이다. 수서 무척추 생물인 Chironomus의 유충은 이러한 다단계 바이오마커 연구에 매우 적절한 생물학적 모델이다. 이 논문의 첫 번째 부분은 생체지표를 이용한 환경 모니터링을, 두 번째 부분은 Chironomus의 유충에서 생체지표의 적용에 대해서 다룬다.

Efficient Cell Tracking Method for Automatic Analysis of Cellular Sequences (세포동영상의 자동분석을 위한 효율적인 세포추적방법)

  • The tracking and analysis of cell activities in time-lapse sequences plays an important role in understanding complex biological processes such as the spread of the tumor, an invasion of the virus, the wound recovery and the cell division. For automatic tracking of cells, the tasks such as the cell detection at each frame, the investigation of the correspondence between cells in previous and current frames, the identification of the cell division and the recognition of new cells must be performed. This paper proposes an automatic cell tracking algorithm. In the first frame, the marker of each cell is extracted using the feature vector obtained by the analysis of cellular regions, and then the watershed algorithm is applied using the extracted markers to produce the cell segmentation. In subsequent frames, the segmentation results of the previous frame are incorporated in the segmentation process for the current frame. A combined criterion of geometric and intensity property of each cell region is used for the proper association between previous and current cells to obtain correct cell tracking. Simulation results show that the proposed method improves the tracking performance compared to the tracking method in Cellprofiler (the software package for automatic analysis of bioimages).

Identification of Diagnostic PCR Markers for Honeybee Foulbrood Disease from Specific Genes of Paenibacillus larvae (부저병 원인균 Paenibacillus larvae 특이 유전자 분석을 통한 진단마커 발굴)

  • Foulbrood disease is infected by Paenibacillus larvae on larval stage of honeybee, and is lethal disease to result in population death. This disease was manifested in 2008 in Korea, is still suffered by the secondary damages. In this study, we are to examine diagnostic PCR approaches to manage the Foulbrood disease. PCR amplification of 16S rRNA is generally using for microbial infection, but the specificity is little poor for the correct diagnosis. Therefore, we are to identify specific genes expressed in Paenibacillus larvae, and perform PCR analysis. We selected five distinct genes from literature references. Those genes are commonly known as toxic genes for host infection, and include Toxin1, Toxin2A & 2B, SplA, CBP49, and SevA&SevB. PCR amplification for these genes is difficult to detect at the first time. So, we performed the second PCR using the first PCR product as a template. This approach using the nested PCR was very useful for detecting large marker genes. When Paenibacillus larvae was cultured in the medium containing plant extracts, PCR amplification of the identified genes is correlated with the microbial growth inhibition. Therefore, these results suggest that the identified genes might be useful to study diagnostic PCR markers for honeybee Foulbrood disease.

Current status and prospects of citrus genomics (감귤 유전체 연구 동향 및 전망)

  • Citrus is an economically important fruit tree with the largest amount of fruit production in the world. It provides important nutrition such as vitamin C and other health-promoting compounds including its unique flavonoids for human health. However, it is classified into the most difficult crops to develop new cultivars through conventional breeding approaches due to its long juvenility and some unique reproductive biological features such as gamete sterility, nucellar embryony, and high level of heterozygosity. Due to global warming and changes in consumer trends, establishing a systematic and efficient breeding programs is highly required for sustainable production of high quality fruits and diversification of cultivars. Recently, reference genome sequences of sweet orange and clementine mandarin have been released. Based on the reference whole-genome sequences, comparative genomics, reference-guided resequencing, and genotyping-by-sequencing for various citrus cultivars and crosses could be performed for the advance of functional genomics and development of traits-related molecular markers. In addition, a full understanding of gene function and gene co-expression networks can be provided through combined analysis of various transcriptome data. Analytic information on whole-genome and transcriptome will provide massive data on polymorphic molecular markers such as SNP, INDEL, and SSR, suggesting that it is possible to construct integrated maps and high-density genetic maps as well as physical maps. In the near future, integrated maps will be useful for map-based precise cloning of genes that are specific to citrus with major agronomic traits to facilitate rapid and efficient marker-assisted selection.

Varietal characteristics of new Cordyceps militaris 'Dowonhongcho 2ho' improved by mating type molecular markers (교배형 분자마커를 이용한 신품종 밀리타리스 동충하초 '도원홍초 2호'의 품종 특성)

  • The mushroom species Cordyceps militaris has been studied and cultivated as a medicinal mushroom due to its multiple valuable biological and pharmaceutical activities. For breeding new strains of C. militaris, multiplex PCR assays were performed using primers specific for its mating type genes, MAT1-1 and MAT1-2. Mating types and mating status were confirmed, as evidenced by DNA bands at 233-bp and 191-bp for MAT1-1 and MAT1-2 respectively. The novel 'Dowonhongcho 2ho' was developed through mating; they were found to possess high-quality fruiting bodies when grown in artificial media. The stromata of the new strain were club-shaped, with a bright orange-red color, and measured 7.1 cm in length. They had an average cordycepin content of 0.33%. Compared to 'Dowonhongcho,' the new strain had a 7% higher yield, as well as firm fruiting bodies. The optimum temperature for mycelial growth was $20{\sim}25^{\circ}C$, and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. The fruiting bodies developed after 49.1 days from inoculation. The use of mating type molecular markers improved the breeding efficiency of the new strain 'Dowonhongcho 2ho.' Thus, they may be valuable for artificial cultivation and industrial-scale production of C. militaris with excellent characteristics.

Copyright Protection for Fire Video Images using an Effective Watermarking Method (효과적인 워터마킹 기법을 사용한 화재 비디오 영상의 저작권 보호)

  • This paper proposes an effective watermarking approach for copyright protection of fire video images. The proposed watermarking approach efficiently utilizes the inherent characteristics of fire data with respect to color and texture by using a gray level co-occurrence matrix (GLCM) and fuzzy c-means (FCM) clustering. GLCM is used to generate a texture feature dataset by computing energy and homogeneity properties for each candidate fire image block. FCM is used to segment color of the fire image and to select fire texture blocks for embedding watermarks. Each selected block is then decomposed into a one-level wavelet structure with four subbands [LL, LH, HL, HH] using a discrete wavelet transform (DWT), and LH subband coefficients with a gain factor are selected for embedding watermark, where the visibility of the image does not affect. Experimental results show that the proposed watermarking approach achieves about 48 dB of high peak-signal-to-noise ratio (PSNR) and 1.6 to 2.0 of low M-singular value decomposition (M-SVD) values. In addition, the proposed approach outperforms conventional image watermarking approach in terms of normalized correlation (NC) values against several image processing attacks including noise addition, filtering, cropping, and JPEG compression.
