Aerial Distribution of Bacterial Groups of Xanthomonas oryzae (Uyeda et Ishiyama) Dowson in Korea (벼 흰빛잎마름병균의 균군의 분포에 관한 연구)

  • Studies on bacterial groups of X. oryzae in Korea has been continued annually since 1975. The present report maintained the result of recent Studies covering on 224 isolates collected front 191 portions of all over the country during the period from 1977 to 1978. The four bacterial groups, I, II, III and V, were found with the ratio of 79, 16, 3 and 3 per-cent, respectively. Variable bacterial groups were found in Chung- nam province while in Jeon- nam province the group II was more pre4valent than group I. Variety-group 'Kimmaze' maintained much more of heterogeneous bacterial groups, and it was significant, especially, on cultivar Milyang $\#23$. Variety-group 'Kogyoku' maintained group II, only while it was possible to isolate group I, II, and III from variety group 'Rantai-emas.' Isolates from Kresek symptom, mostly on Milyang $\#23$, belonged to group I and group II, and their occurring incidence were 20 and 8 out of 28 isolates, respectively. Variability of bacterial group was differed with the location of fields. There were homogeneous and heterogeneous groups, respectively, regardless of host varieties in a given locations.

Development of Drainage Water Disinfection System by Electric Shock in Recirculating Soilless Culture (순환식 수경재배에서 배액의 전기충격살균법 연구)

  • This study was conducted to develop the disinfection system using electric shock for recycled nutrient solution in recycling soilless culture. Stainless steel (SUS 316) was found as the most appropriate electrode material for electrical disinfection system from the view of high electrical conductivity, low electric resistance, and low price. There were no changes in nutritional elements when the electric shock passed through the nutrient solution by stainless steel electrode. The amount of electric current increased with width than thickness of the electrode. The farther the distance between the electrodes was increased the time to reach out the aimed amount of current. The electric shock was applied to Ralstonia solanacearum and Fusarium oxysporum as representative bacteria and also fungi. Any of those pathogens were killed with the percentage of higher than 97% in the condition of 15VDC or 24VDC.

Lichen Diseases on Prunus mume Caused by Colonization of Consortium of Several Crustose Lichens Including Parmotrema autrosinense (지의류 복합체에 의한 매실나무 지의류병)

  • Lichen is a symbiotic association which lichen-forming fungi associate with green algae or cyanobacteria capable of producing food by photosynthesis. Lichen is one of the most widely distributed eukaryotic organisms on every kind of surface of soils, rocks and trees. Lichen-forming fungi commonly colonize on the surface of barks of dead or dying trees, which they use merely as a means of support without parasitizing. Lichen consortium of Parmotrema autrosinense and several crustose lichens heavily colonized on the surface of P. mume in Suncheon, Jeonnam province in Korea. The lichen consortium on the surface of P. mume gave the affected trees a dull gray appearance and suppressed their normal growths. Severely affected twigs or branches resulted in the death when the lichen thalli were not rubbed off the barks of P. mume. Twigs or branches of P. mume were recovered from blight syndrome when the lichen thalli were rubbed off, The recovered twigs or branches became healthy and clean when $Tween^{(R)}$ 20 or thiophanate-methyl paste were treated on the barks of P. mume. This is the first report of lichen diseases on P. mume caused by colonization of consortium of several crustose lichens including P. autrosinense.

Detection of Xanthomonas axonopodis pv. glycines and Survey on Seed Contamination in Soybean Seeds Using PCR Assay (PCR Assay 이용 콩 종자에서 Xanthomonas axonopodis pv. glycines 검출 및 종자오염 조사)

  • Xanthomonas axonopodis pv. glycines is the causal agent of bacterial pustule of soybean(Glycine max. (L.) Merr), which is one of the most prevalent bacterial diseases in Korea. In this study, Polymerase Chain Reaction (PCR) assay was applied to detect Xanthomonas axonopodis pv. glycines and to survey on seed contamination in 36 soybean cultivars of Korea. And we have to compare PCR assay with dilution-plating assay of detection and identification. We confirmed detection of pathogen from artificial infected seeds and natural Infected seeds using PCR assay. This assay gave results similar to a seed-wash dilution plating assay and proved more effective than classical methods. Results of survey on seed contamination by X. axonopodis pv. glycines from 36 cultivar seeds showed that the pathogen was detected from Pungsan-namulkong, Mallikong, Taekwangkong, Daemangkong, Ajukkarikong using PCR assay. Therefore, The PCR assay provides a sensitive, rapid tool for the specific detection of X. axonopodis pv. glycines in soybean seeds.

Screening of Seed Treatment Agents against Leaf Blight and Black Root Rot for Carrot Organic Cultivation (당근 유기재배를 위한 검은잎마름병과 검은무늬병 종자소독제 선발)

  • This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.

Detection of the Causal Agent of Bacterial Wilt, Ralstonia solanacearum in the Seeds of Solanaceae by PCR (가지과 종자에서 Ralstonia solanacearum의 검출을 위한 PCR 방법)

  • Ralstonia solanacearum, a causal agent of bacterium wilt is very difficult to control once the disease becomes endemic. Thus, Ralstonia solanacearum is a plant quarantine bacterium in many countries including Korea. In this study, we developed PCR assays, which can detect Ralstonia solanacearum from the Solanaceae seeds. Primers RS-JH-F and RS-JH-R amplified specifically a 401 bp fragment only from Ralstonia solanacearum race 1 and race 3. The nested PCR primers, RS-JH-F-ne and RS-JH-R-ne that were designed inside of 1st PCR amplicon amplified specifically a 131 bp fragment only from Ralstonia solanacearum race 1 and race 3. The primers did not amplify any non-specific DNA from the seed extracts of the Solanaceae including tomato and pepper. When detection sensitivity were compared using the Solanaceae seeds inoculated with target bacteria artificially, the nested PCR method developed in this study 100 times more sensitive than ELISA and selective medium. Therefore, we believe that the PCR assays developed in this work is very useful to detect Ralstonia solanacearum in the Solanaceae seeds.

Effects of Fusarium circinatum on Disease Development and Gas Exchange in the Seedlings of Pinus spp. (푸사리움가지마름병균 Fusarium circinatum이 소나무류 묘목의 병 진전과 침엽의 가스교환에 미치는 영향)

  • Four-year-old seedlings of Pinus thunbergii, Pinus densiflora and Pinus rigida were inoculated with Fusarium circinatum isolate (FT-7), the pitch canker fungus, from P. thunbergii, to evaluate the effects of the pathogen on disease development and gas exchange rate. Needle dehydration was evident on 2 of 10 seedlings of P. thunbergii and P. rigida at 18 and 21 days after inoculation, respectively, while no symptoms were observed in P. densiflora seedlings throughout the experiment. Gas exchange stopped completely in 4 of 5 measured seedlings of P. thunbergii and 2 of 5 measured seedlings of P. rigida at 25 days after inoculation, and in the remaining 3 seedlings of P. rigida at 39 days after inoculation. Disease development in P. thunbergii seedlings was faster than that in P. rigida seedlings. By the time, the experiment was ended at 78 days after inoculation, 9 of 10 seedlings of P. rigida and 8 of 10 seedlings of P. thunbergii seedlings treated with FT-7 was almost dead, but all seedlings of P. densiflora were still healthy. We suggest that P. densiflora is resistant to F. circinatum in the current study, and gas exchange rate of the species after inoculation does not differ significantly compared to that of untreated control.

Vegetative Compatibility Grouping of Sclerotinia homoeocarpa Isolates Infecting Turfgrass in South Korea (한국의 잔디에서 분리한 Sclerotinia homoeocarpa 균의 체세포화합성군 분류)

  • Sclerotinia homoeocarpa, the causal agent of dollar spot, is one of the most common pathogens of cool season turfgrasses in South Korea. The vegetative compatibility group (VCG) assay was carried out using nitrate-nonutilizing (nit) mutants recovered from 13 South Korean isolates with various geographical origins. The mutants were divided into four phenotypic classes based on mutation loci associated with nitrogen assimilation: nit1, nit2, nit3, and NitM. The recovered number of nit mutants greatly varied among the isolates, ranging from 0 to 15 mutants. Of the mutants isolated, nit1 and nit2 mutants were most common (80%) while NitM and nit3 were relatively rare. One dominant and four minor VCGs were determined from 18 mutant isolates tested. To study population structures of Korean S. homoeocarpa isolates and increase our understanding of its ecological and epidemiological aspects for dollar spot management on turfgrass, more generated mutants should be tested with more diverse isolate collections.

Contamination Level of Ralstonia solanacearum in Soil of Greenhouses Cultivating tomato Plants in Chungbuk Province and Characteristics of the Isolates (충북지역 토마토 시설재배지의 풋마름병균(Ralstonia solanacearum) 오염도 및 분리균주의 특성)

  • Contamination level and characteristics of Ralstonia solanacearum in soil of greenhouses cultivating tomato plants in Chungbuk province was determined. R. solanacearum was isolated with the semiselective media in 27 greenhouse soil samples out of 133 greenhouses soil investigated, which indicates 20.3 % of tomato cultivating greenhouses in Chungbuk contaminated with the bacterial wilt pathogen. Density of R. solanacearum was estimated to 10$^{2.4}$ cfu/g in the contaminated soil. All 71 isolates of R. solanacearum which containing 12 isolates from the diseased tomato plants were race 1, ann 35 isolates of them were biovar 3 and 36 isolates were biovar 4. More than 88% of 71 isolates were inhibited growth on nutrient agar containing oxolinic acid 0.5 $\mu\textrm{g}$/ml, streptomycin 25 $\mu\textrm{g}$/ml, tetracycline 5 $\mu\textrm{g}$/ml and cupric sulfate 375 $\mu\textrm{g}$/ml (1.5 mM). The 11.3%, 4.2% and 5.6 % of the isolates can grow on nutrient agar containing 10 times more oxolinic acid, streptomycin, tetracycline than minimal inhibitory concentration of the sensitive strains. Five isolates were resistant to 2 bactericides and one isolates was resistant to all 3 bactericides.

  • In 2005, average temperature was lower, and average rainfall was less than those of previous year. The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Bacterial blight, bacterial grain rot, and panicle disease of rice, black rot of pear, and white rot and bitter rot of apple were severe. Especially, brown rot of rice occurred four times higher than those of previous year. Panicle blight of rice increased about 3 times, compared with the previous year, presumed that the higher rainy days, rainfall and RH promoted spread of the fungal pathogens to panicles of rice. The diseases of rice leaf blast, sudden wilt syndrome, downy mildew and powdery mildew of chinese melon in plastic greenhouse, and virus diseases of hot pepper occurred distinctly less than those of the previous year. Another diseases surveyed occurred similar or less.
