A nonlinear Co-rotational Quasi-Conforming 4-node Shell Element Using Ivanov-Ilyushin Yield Criteria (이바노브-율리신 항복조건을 이용한 4절점 비선형 준적합 쉘요소)

  • A co-rotational quasi-conforming formulation of four- node stress resultant shell elements using Ivanov-Ilyushin yield criteria are presented for the nonlinear analysis of plate and shell structure. The formulation of the geometrical stiffness is defined by the full definition of the Green strain tensor and it is efficient for analyzing stability problems of moderately thick plates and shells as it incorporates the bending moment and transverse shear resultant force. As a result of the explicit integration of the tangent stiffness matrix, this formulation is computationally very efficient in incremental nonlinear analysis. This formulation also integrates the elasto-plastic material behaviour using Ivanov Ilyushin yield condition with isotropic strain hardening and its asocia ted flow rules. The Ivanov Ilyushin plasticity, which avoids multi-layer integration, is computationally efficient in large-scale modeling of elasto-plastic shell structures. The numerical examples herein illustrate a satisfactory concordance with test ed and published references.

Detect of Hypericin (HyH) gene in Hypericum erectum in Korea and Comparison of H. perforatum in Europe (한국내 고추나물의 하이퍼리신 유전자(HyH)의 탐색과 유럽의 서양고추나물과 비교)

  • Hypericin (HyH) is a substance which is isolated a medicinal herb, Hypericum perforatum L., commonly known as St. John's Wort. Hypericum erectum is a long-lived herb that is distributed in Korea. Cloned HyH genes H. erectum of were conformed by sequencing. The cDNA Hyp-1 sequence has 732 bp with an open reading frame of 567. Thus coding for a protein of 152 amino acid residues. A BLAST re-search using the deduced nucleotide sequences in HyH gene produced significant alignments with the H. perforatum. Sequences in HyH gene showed significant homology with Rubus idaeus putative allergen Rub-i-1 mRNA, Protein sequence comparisons revealed significant homology between Hyp-1 and the phenolic oxidative coupling protein hyp-1 of H. perforatum (98%). Additionally, Hyp-1 showed sig-nificant homology with various other classes of allergens, including Pru-av-1 (62%) from Prunus avium and allergen Bet-vl-Sc3 from Betula pendula (60%). Thus, the result of this study may offer an important information to establish an assay system for chemicals of the herbal medicines for H. erectum as well as H. perforatum.

Development of Non-toxic Recombinant Ricin Vaccine and Evaluation of Vaccine Efficacy (독성을 제거한 재조합 리신 백신 개발 및 효능평가)

  • Ricin is a highly toxic protein which is produced in the seeds of the castor oil plant. Ricin toxin A chain has ribosomal RNA N-glycosylase activity that irreversibly hydrolyses the N-glycosidic bond of the adenine residue at position 4324 within the 28S rRNA. In this study, we developed non-toxic recombinant ricin vaccine(R51) in E. coli expression system, and evaluated efficacy of the R51 according to adjuvants. When the R51 was administered using aluminum hydroxide as an adjuvant, the vaccine efficacy was higher than that of TLR agonists or aluminum phosphate. Because it is time-consuming to administer the vaccine three times at three-week intervals, we investigated the survival rate and antibody titer of mice according to the change of time interval of vaccination. Interestingly, there was no difference in survival rate and antibody titer when R51 was administered at 0, 1, and 3 weeks or 0, 2, and 4 weeks compared to when administered at 0, 3, and 6 weeks. Therefore, the developed R51 vaccine is promising to protect soldiers from Ricin attack.

Growth Inhibitory Activity of Sulfur Compounds of Garlic against Pathogenic Microorganisms (마늘 황화합물의 병원성미생물 번식억제작용)

  • Efforts have been made to explore the possibility of using garlic as an antimicrobial therapeutic agent since garlic extract and its individual sulfur compounds show antimicrobial activities against all kinds of microorganisms including bacteria, molds, yeasts and protozoa. Staphylococcus aureus has been the most studied bacteria along with many other Gram positive and negative pathogenic bacteria, including species of the genera Clostridium, Mycobacterium, Escherichia, Klebsiella, Bacillus, Salmonella and Shigella. Candida albicans has been the most studied among the eukaryotic microorganisms. A pathogenic protozoa, Giardia intestinalis, was also tested. All the microorganisms tested was inhibited by garlic extract or its sulfur components. Garlic has been known to be growth inhibitory only when fresh garlic is crushed, since allicin-generating reaction is enzyme-catalyzed. Allicin is known to be growth inhibitory through a non-specific reaction with sulfhydryl groups of enzyme proteins that are crucial to the metabolism of microorganisms. Another plausible hypothesis is that allicin inhibits specific enzymes in certain biological processes, e.g. acetyl CoA synthetase in fatty acid synthesis in microorganisms. Allicin transforms into other compounds like ajoene and various sulfides which are also inhibitory to microorganisms, but not as potent as their mother compound. It is reported recently that garlic heated at cooking temperatures is growth inhibitory especially against yeasts, and that the growth inhibitory compound is allyl alcohol thermally generated from alliin in garlic.

Effect of Substituted Residue 24 on Folding of Tryptophan Synthase $\alpha$ Subunit (트립토판 중합효소 $\alpha$ 소단위체의 폴딩에 미치는 24번 잔기 치환효과)

  • In order to elucidate a role of residue 24 in the folding of tryptophan synthase $\alpha$ subunit, mutant proteins in which Thr 24 was replaced by Met, Ala, Ser, Leu or Lys were overexpressed in E. coli, and the extents of accumulated proteins as soluble or aggregated forms were examined. The mutant proteins with Met or Leu at residue 24 were predominantly accumulated as soluble forms as the native protein. On the other hand, mutant proteins with Ser, Ala or Lys at residue 24 were expressed as aggregated forms as well. This result suggests that residue 24 of tryptophan synthase $\alpha$ subunit may be implicated in the folding of this protein.

Analysis of Evaluation Methods for the Efficacy of Protein Removal Agents for Soft Contact Lens (소프트콘택트렌즈 단백질제거제의 효능 평가법 분석)

  • Purpose: The present study was conducted to establish the experimental condition for the proper evaluation of protein removal efficacy when developing protein removal agents. Its protein removal efficacy was further analyzed and compared with the result from protein removal efficacy against protein deposition on contact lens to suggest the evaluation method for efficacy of protein removal agents. Methods: Protein digestibility assay presented in the Korean pharmacopoeia was selected to establish the evaluation method for efficacy of papain, pancreatin, subtilisin A and protease itself as a ingredient and protein removal tablets or solution containing those enzymes and find a suitable test conditions. Furthermore, the cleaning efficacy of commercially available protein removal tablets and solution on balafilcon A lens deposited with protein artificially was measured and the correlation between two evaluation methods was further analyzed. Results: When pancreatin itself and the product containing pancreatin was evaluated by protein digestibility assay, both reached 28 IU/mg, the standard value of protein digestibility suggested by the Korean pharmacopoeia. In case of protease and subtilisin A tested with trichloroacetic acid B solution, both of them met the enzyme activity level proposed by the manufacturers when they were evaluated by protein digestibility assay however, papain and subtilisin A tested with trichloroacetic acid A solution were not reached the enzyme activity level. Among protein removal agents, three products except a product containing pancreatin did not meet the enzyme activity value specified by the manufacturer when they were evaluated by protein digestibility assay. However, actual protein removal efficacy of three products except a papain-containing product on the lens was greater than 90% protein removal. In the case of papain-containing protein removal product, its effect was not measured by protein digestibility assay however, its actual protein removal efficacy on the lens reached 73.72%. Conclusions: From the results, it was confirmed that the efficacy of protein removal agents for contact lens should be evaluated by different method according to the type of proteolytic enzyme contained. That is, the protein removal agents containing pancreatin, protease and subtilisin A can be evaluated by protein digestibility assay and protein removal efficiency evaluation and the products containing papain can be effectively evaluated by only the evaluation method for protein removal efficiency employing the lens.
