Gemifloxacin is a synthetic fluoroquinolone antimicrobial agent that exhibits potent activity against most Gram-negative and Gram-positive organisms, and has a comparatively low chondrotoxic potential in immature animals. This study examined the effects of gemifloxacin on the Achilles tendons in immature Sprague-Dawley rats treated by oral intubation once daily for 5 consecutive days from postnatal week 4 onward at doses of 0 (vehicle), and 600mg/kg body weight Ofloxacin was used for comparison. The Achilles tendon sperimens were examined by electron microscopy. In comparison with the vehicle-treated controls, there were ultrastructural changes in all samples from the gemifloxacin- and ofloxacin-treated rats. Degenerative changes were observed in the tenocytes, and the cells that detached from the extracellular matrix were recognizable. The degree of degenerative changes and the number of degenerated cells in the Achilles tendon were significantly higher in the treated group than in the control group. Moreover, among the quinolone treated groups, these findings were more significant in the ofloxacin treated group, and less significant in the gemifloxacin treated group. It is unclear what these findings mean with respect to the possible risk ill juvenile patients treated with gemifloxacin or other quinolones. However, these results show that gemifloxacin causes fewer changes in the connective tissue structures.
Although the vegetable Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Korean population, its functionalities are not very well defined. In this study, we examined the effects of methanol extract of AK in rats on the biochemical changes induced by two hepatotoxins, D-galactosamine (GalN) and carbon tetrachloride ($CCl_4$). AK was orally administered once daily for 7 days to male rats at 200 and 500 mg/kg, before hepatotoxins. Effects of AK were assessed 24 hr later. AK pretreatments at 200 and 500 mg/kg significantly blunted GalN-induced elevation in liver lipid peroxidation, plasma aspartate-transaminase (AST) and alanine-transaminase (ALT) activities. AK also prevented, after 500 mg/kg but not after 200 mg/kg, the GalN-induced elevation in triglyceride, total cholesterol and LDL-cholesterol levels. Differently from against GalN-induced toxicity, AK did further elevate the $CCl_4$-induced rise in AST, ALT and lipid peroxidation. These results suggest that AK, when pre-administered prior to GalN, exerted protective effects against GalN-induced hepatotoxicity, in contrast however, AK exacerbated that induced by $CCl_4$. To explore possible mechanism for the toxicity-potentiating effects of AK on $CCl_4$, the activity of hepatic drug metabolism after AK treatment was assessed. It was observed that AK increased the activity of aniline hydroxaylase, a cytochrome P450 isoenzyme responsible for metabolic activation of $CCl_4$. This finding suggests that hepatoprotective effects of AK are not equally expected depending on hepatotoxins employed.
The objective of this study was to compare the biocompatibility for local irritation and bioavailability of $Gentacol^{TM}$ and $Collatamp^{TM}$, after single intramuscular loading in rats. Sixty-six male Sprague-Dawley rats were divided into 4 groups; (1) any test materials were not applied into the quadriceps muscles (control group, N = 6), (2) Gentamicin was injected into the quadriceps muscles (Gentamicin group, N = 6), (3) Collatamp was applied into the quadriceps muscles (Collatamp group, N = 27), and (4) Gentacol was applied into the quadriceps muscles (Gentacol group, N = 27). The concentration of gentamicin in muscles was gradually decreased with time after loaded in the both $Gentacol^{TM}$ and $Collatamp^{TM}$ loaded regions. No detectable gentamicin was observed in the plasma of $Gentacol^{TM}$ and $Collatamp^{TM}$ loaded rats. Histologically, moderate to severe inflammatory cell infiltrations including PMN, lymphoid cells and macrophages were observed with slight to moderate edematous changes of muscle fibers after intramuscular injection of gentamicin. However, these histopathological changes of gentamicin injection induced local irritations were dramatically decreases after intramuscular loading of $Collatamp^{TM}$ and $Gentacol^{TM}$. These results suggest $Gentacol^{TM}$ may show favorable local bioavailability and induce only slight local irritations as comparable as $Collatamp^{TM}$ without systemic exposures in the condition of this experiment.
The pharmacokinetic properties of KR-V compounds, recently developed as new anti-HIV agents, were studied after i.v. and p.o. administration in rats. The concentrations of the KR-V series were determined in rat plasma using an high-performance liquid chromatography (HPLC)-UV detection system. Of the 19 KR-V compounds investigated in the present study, only KR-V 3, 10, 14, 16 and 18-1 showed oral bioavailability. The plasma concentration-time data could be adequately described by an one-compartment open model. In the i.v. kinetic study (10mg/kg), the CLt of KR-V 3, 10, 14 and 16 (>4L/hr/kg) were significantly higher than that of KR-V 18-1 (1.1 L/hr/kg). The AUC of KR-V 18-1 was greater ($8.97{\mu}g{\cdot}hr/ml$) than that of the other compounds, but the Vd (0.58 L/kg) was lower. In the p.o. kinetic study (50mg/kg), although the t-1/2 of KR-V 18-1 was shorter than that of the other compounds, the AUC ($3.659{\mu}g{\cdot}hr/ml)$ and $C_{max}(1.891{\mu}g/ml$) were markedly higher. In a seperated in vitro experiment, only KR-V 18-1, of the 5 compounds with bioavailibility, exhibits potent activity against HIV-1 mutant strains. Therefore, KR-V 18-1 is expected to become a new potent anti-AIDS drug candidate/lead compound.
The purpose of the present study was to determine the preventive effects of the two antioxidant vitamin E and catechin on DEHP-induced disturbance of spermatogenesis in male rats. Rats at 4 weeks of age were randomly allocated into five groups with 20 animals per group. The first group was not any administrated as control. The second group was administrated DEHP (2 g/kg) daily for 14 days. The third group was administrated vitamin E (500 IU/kg) following DEHP treatment by the same method (daily for 14 days). The fourth group was administrated catechin (200 mg/kg) following DEHP treatment by the same method. The fifth group was co-administrated vitamin E (500 IU/kg) and catechin (200 mg/kg) following DEHP treatment by the same method. In order to determine the preventive effects, we examined pathological changes of testis with apoptotic index, and characteristics of sperm with computer assisted sperm analysis (CASA). Vitamin E and catechin supplementation were significantly prevented the testicular atrophy, apoptosis of germ cells in the seminiferous tubules and abnormal rate of sperm. Moreover, sperm concentration, viability and motility was significantly recovered in groups of alone and along with vitamin E and catechin. The results suggest that preventive effects of alone and along administration of vitamin E and catechin on DEHP-induced testicular atrophy damages have been demonstrated.

In this study, we evaluated whether Lunasin Enriched Soybean (LES) suppressed 1-chloro-2,4-dinitrochlorobenzene (DNCB) induced allergic contact dermatitis in vivo. In the group treated with DNCB alone, erythema and hemorrhage in the skin were observed, while the application of LES after DNCB treatment modulated erythema and hemorrhage similar to the untreated group. In histopathological analysis of the skin, DNCB-treated groups showed increased skin thickness compared to the untreated groups, while the treatment of LES dose-dependently modulated skin thickness. Also, hypertrophy and hyperkeratosis of the epidermis, intracellular edema and infiltration of inflammatory cells were observed in the group treated with DNCB alone, while treatment of LES inhibited these inflammatory changes. DNCB also induced a significant up-regulation of iNOS and COX-2 expression, as compared to the untreated groups. However, the treatment of LES significantly decreased the down-regulation of iNOS and COX-2 expression, as compared to DNCB-treated groups. Also, LES reduced the IgE level in DNCB-stimulated groups. And lunasin from LES translocated the nucleus of the cells in the skin of Sprague-Dawley rats. Taken together, it is thought that LES has the potential to be used as an alternative treatment for allergic contact dermatitis.
For evaluating the anti-allergy activity of mushrooms, forty two mushroom extracts were studied their inhibitory activities on the IgE-mediated degranulation in rat basophilic leukemia cell line (RBL-2H3 cell). Effects of mushroom extracts on the release of interleukin-4 (IL-4) and beta-hexosaminidase, and the cell viability of the IgE-sensitized were measured. From the analysis, five mushroom extracts such as the water extract from Flammulina velutipes (Curt.:Fr.) Sing showed the suppressive activities on IL-4 release as 20%. Eight extracts including the water extract of Ganoserma lucidum showed the suppressive activities on ${\beta}$-hexosaminidase release as 20%. Almost all of the extracts stimulated the proliferation of RBL-2H3 cell. The water extracts of Flammulina velutipes and Phellinus linteus were examined against the inhibitory activity in the production of IL-4 and ${\beta}$-hexosaminidase. Additionally, the extracts from Ganoserma lucidum, Isaria japonica, Phellinus linteus and Pleurotus ostreatus inhibited dose-dependently on ${\beta}$-hexosaminidase production. In conclusion, the result suggests that the mushrooms with the potent inhibitory efficacies on the degranulation of the mast cell would be candidate resources for the anti-allergy resources, and thus need to study for their utilization.
Thread lifting has become popular as a minimally invasive technique for facial rejuvenation. Commercially available threads are composed of poly-L-lactic acid (PLLA), polycaprolactone (PCL), or polydioxanone (PDO). However, the histological changes that occur in response to implanted threads are unclear. The aim of this study was to evaluate histological changes that occur in response to implantation with three types of bioresorbable threads (PLLA, PCL, PDO) in rat skin. PLLA, PCL and PDO threads were implanted in the dorsal skin of Sprague Dawley rats and tissue samples were harvested 2, 4, 8 and 12 weeks post-implantation. To evaluate histologic changes induced by bioresorbable face-lifting threads, tissue samples were stained with hematoxylin & eosin, Masson's trichrome stain and Herovici's collagen stain. All three threads induced neocollagenesis of type 3 collagen in the rat skin. The amount of collagen induced by the threads was dependent on the thread surface area. The PDO cavern-type thread was most effective in inducing neocollagenesis due to its extensive surface area. Our results suggest that type 3 collagen induced by bioresorbable threads depends on the thread surface area to uphold the dermis and contributes to facial rejuvenation.

This study investigated action mechanism and biological effect of Jijang-kimch, including its anti-obesity effect and blood lipid-decreasing effect in a high-fat diet-induced obese model animals. There were four treatment groups: CD (chow diet as normal control), HFD (high fat diet as obesity control), HFDCK (HFD plus commercial kimchi extracts), and HFDJK (HFD plus Jijang-kimchi extract). Kimchi extracts were orally administered for 28 days. Body weight, liver, and adipose tissue weight declined in HFDJK compared to those in HFDCK(p<0.05). Blood triglyceride, total cholesterol, low density lipoprotein-cholesterol (LDL-C), and glucose level decreased in CD, HFDJK, and HFDCK compared to those in HFD(p<0.05). Those in HFDJK were lower than those in HFDCK(p<0.05). Sizes of liver and adipose cells increased in HFD, HFDCK, and HFDJ that those in CD(p<0.05). Those in HFDJK were greatly decreased than those in HFDCK(p<0.05). These results indicate that ingestion of Jijang-kimchi in obese model animals has anti-obesity effect by lowering blood lipid and glucose levels and decreasing adipocyte size compared to that of commercial-kimchi.
The present study, to evaluate the effect of vitamin E on the oxidative stress in STZ-treated rat and BB rat, was investigated the biochemical enzyme activity in the serum, and malondialdehyde and carbonyl group in the RBC membrane, liver and microsomal fraction after vitamin E and/ or insulin treatment. Results obtained through the experiments were summarized as follows; 1. Effect of vitamin E and/or insulin treatment in STZ-treated rat 1) Lipid peroxidation level in RBC membrane, liver and microsomal fraction was significantly decreased in vi. tamin E and/or insulin treatment group, and especially more significantly decreased in vitamin E with insulin treated group. 2) Protein oxidation level in RBC membrane, liver and microsomal fraction was significantly decreased in vitamin E and/or insulin treatment group. And it was especially more significantly decreased in RBC membrane and liver of vitamin E with insulin treated group. 3) In the enzyme activity in the serum, the activity of AST and ALT was not altered in all experimental group. The increased ALP activity in STZ-treated group was significantly decreased in insulin treated group and vitamin E with insulin treated group. 4) Decreased level of albumin and creatinine after STZ treatment was significantly increased in vitamin E and/or insulin treated group. 5) Level of glucose, cholesterol and triacylglycerol in serum: Glucose level was not significantly different in vitamin E treated group compared to STZ control group. But it was significantly different in the insulin treated group and vitamin E with insulin treated group compared to STZ control group. The cholesterol content in the serum was significantly increased in STZ control group compared to normal control group. And except low dose vitamin E treatment group, it was significantly decreased in vitamin E and/or insulin treated group compared to STZ control group. The triacylglycerol content in the serum was significantly decreased in STZ control group and increased in high dose vitamin E treated group and vitamin E with insulin treated group. But it was not significantly different in low dose vitamin E treated group and insulin treated group compared to STZ control group. 2. Effect of vitamin E and/or insulin treatment in BB rat 1) Lipid peroxidation level in liver was decreased by vitamin E with insulin treatment compared to insulin treatment. But it was not different in microsomal fractions. 2) Protein oxidation level in liver and microsomal fraction was decreased by vitamin E with insulin treatment compared to insulin treatment only in microsomal fractions. These results suggest that the combination treatment of vitamin E and insulin could prevent the oxidative change of lipid and protein of the RBC membrane, liver and microsomal fraction in STZ-treated rats and BB rats.
