Effect of Eleutherococcus senticosus supplementation on laccase and cellulase activity of Ganoderma lucidum (가시오가피 첨가가 영지버섯의 라카아제 및 셀룰라아제 효소 활성에 미치는 영향)

  • The aim of this study is to investigate the effect of Eleutherococcus senticosus (ES) on the laccase and cellulase activity of Ganoderma lucidum mycelia. Following the addition of ES, the laccase activity of Ganoderma mushroom mycelia was found to be 0.84-2.18 times, 0.61-2.37 times, and 0.78-2.17 times the activity of mycelia treated with sawdust in Yeongji-1 (Y1; ASI-7004), Yeongji-2 (Y2; ASI-7071), and nokgak (GN; ASI-7013), respectively, with the laccase activity of Y2 being the highest at 0.947 U/min. Using the Congo-red assay, a wider clear zone was formed because of the CMCase activity of mycelia treated with ES than that of mycelia treated with sawdust. Cellulase activity was found to be 1.84-2.24 times, 1.77-1.87 times, and 2.74-2.81 times that of mycelia treated with sawdust in Y1, Y2, and GN, respectively, with the cellulase activity of GN being the highest at 0.172 U/min. However, the addition of ES did not affect the growth of G. lucidum mycelia.

The Development of Enzymatic Mordanting Using Laccase for Phenolic Natural Dye (라카아제 촉매 활성에 의한 홍차 염색물의 매염효과)

  • This study aim is to provide new coloration method by laccase-catalyzation on natural phenolic dyeing process. In this study, silk was dyed with black tea, which is one of polyphenolic dye, extracted in distilled water. The dyed samples were catalyzed by laccase as the eco-friendly mordanting process. To optimize the conditions of laccase-catalyzed coloration, conditions were varied by different mordanting methods (one-bath, two-bath), temperature and treatment time. The dye affinity in terms of the value of K/S, $L^*$, $a^*$, $b^*$, and H, V, C was measured by Computer Color Matching System (CCM, CM-2600d, Spectra Magic NX, Korea). The effect of laccase-catalyzed coloration on washing fastness was evaluated and compared with the synthetic mordant (Al, Cu, and Fe). As the result of color analysis of dyed silk, the optimum conditions of laccase-catalyzed coloration were determined to post-mordanting by one-bath at $50^{\circ}C$ for 3 hours. Under the optimum laccase-catalyzed conditions, the dyed silk was shown the color of yellowish-red. After laccase-catalyzed coloration on the dyed silk, the improvement of washing fastness was obtained compared with mordanted silk by synthetic mordant (Al, Cu, and Fe). Therefore, the present study was demonstrated that the effective enzymatic mordanting method by laccase for phenolic natural dyeing with vivid color and good fastness.

Mycelial Culture and Fruiting Analysis of Panellus edulis Strains Collected in Korea (Panellus edulis 수집 균주의 균사배양 및 자실체 특성 분석)

  • Molecular analysis using the internal transcribed spacer region sequences revealed that the strains used in this study, which were formerly identified as Panellus serotinus, are Panellus edullis. After Universal Fungal PCR Fingerprinting (UFPF) analysis, eight strains of P. edulis were divided into two groups. We conducted fundamental research on mycelial growth and sawdust cultivation to understand the cultural characteristics of eight wild P. edulis strains collected from Korean forests. All strains showed faster and denser mycelial growth on potato dextrose agar (PDA) than on other media (malt extract agar, Sabouraud dextrose agar). Optimal conditions for mycelial growth were: $20^{\circ}C$ on PDA, $25^{\circ}C$ on potato dextrose broth (PDB), and pH 5~8 on PDB at $25^{\circ}C$. Two strains (NIFoS 2407, 3993) were selected as excellent strains based on mycelial growth and density on PDA. NIFoS 2792 showed high cellulase activities on carboxymethyl cellulose (CMC) agar, and NIFoS 2387 and 2804 exhibited high laccase activities on ABTS-containing agar media. The mycelial growth of P. edulis was the fastest on Quercus acutissima and Q. mongolica sawdust media, and mycelial density was the highest on Quercus spp. sawdust-containing media. Sawdust cultivation of P. edulis was successful. The conditions were 80~85 days of cultivation period after spawn inoculation, 10~11 days for primordial formation at $17{\sim}18^{\circ}C$, and 15~20 days for fruiting growth. NIFoS 2804 and 3993 were selected as good strains in terms of cultivation period and mushroom production. These results could be useful for the artificial cultivation of P. edulis.
