Genetic Comparison Between Crucian Carp (Carassius auratus Linnaeus) and Crucian Carp (C. cuvieri Temminck and Schlegel) (붕어(Carassius auratus Linnaeus)와 떡붕어(C. cuvieri Temminck and Schlegel)의 유전적 비교)

  • Genomic DNA isolated from two crucian carp species obtained from Yesan (Carassius auratus) and Dangjin (Carassius cuvieri) in Korea were amplified at several times by polymerase chain reaction (PCR). The amplified products were separated by agarose gel electrophoresis (AGE) with oligonucleotides decamer primer and stained with ethidium bromide. The seven arbitrarily selected primers OPC-11, OPC-14, OPC-18, OPD- 02, OPD-11, OPD-15 and OPD-20 generated the shared loci by each species, the polymorphic and specific loci. The seven primers generated the total 458 loci that can be scored from the crucian carp obtained in C. auratus species. 358 fragments were generated from the species obtained in C. cuvieri species. The size of DNA fragments varies from 150 to 1,600bp. The complexity of the banding patterns varies dramatically between the primers and two locations. In this study, 458 loci were identified in the crucian carp species from Yesan and 358 in the crucian carp species from Dangjin: 84 polymorphic loci (18.3%) in the C. auratus species and 48 (13.4%) in the C. cuvieri species. 154 shared loci by each species, the average 22 per primer, were observed in the C. auratus species and 187 loci, the average 26.7 per primer, in the Dangjin species. Based on the average bandsharing (BS) values of all samples, the similarity matrix ranged from 0.434 to 0.868 in the C. auratus species and from 0.449 to 0.924 in the C. cuvieri species. The average BS value was 0.641±0.013 within the C. auratus species and 0.684±0.013 within the C. cuvieri species. The average BS value between two crucian carp species 0.484 ± 0.007, ranged from 0.307 to 0.682. The BS value between the individual No. 09 and No. 16 was 0.682, which was the highest between two crucian carp species. Compared separately, the BS value of individuals within the C. cuvieri species was higher than the C. auratus species. The dendrogram obtained by the seven primers, indicates three genetic clusters: cluster 1 (AURATUS No. 01, 02, 03, 04, 05, 06, 07, 08, 09, 10 and 11), cluster 2 (CUVIERI No. 12, 13, 14, 15, 16, 17, 18, 19, 20 and 21) and cluster 3 (CUVIERI no. 22). The shortest genetic distance displaying significant molecular difference was between the individual AURATUS No. 09 and AURATUS No. 08 from Yesan (genetic distance=0.064). The longest genetic distance displaying significant molecular differences was between the individual CUVIERI No. 17 and AURATUS No. 11 between two crucian carp species (0.477). Phylogenetic Analysis of Carassius auratus and C. cuvieri in Lake Yedang Based on Variations of Mitochondrial CYTB Gene Sequences (예당호 붕어와 떡붕어의 CYTB 유전자를 이용한 유연관계 분석)

  • Two crucian carp species (Carassius auratus and C. cuvieri) inhabit Lake Yedang in South Korea, and C. auratus is known to be native to Korea. Classification of these two freshwater fish species is often confused because of their morphological similarity. To distinguish the two species, we conducted phylogenetic and population genetic analyses of C. auratus and C. cuvieri based on their mitochondrial DNA sequences of the cytochrome b gene (CYTB). We also compared our partial CYTB sequence (<1,056 bp) with 10 Chinese, nine Japanese, and two Russian crucian carp fishes. The results of our phylogenetic analysis showed that C. auratus and C. cuvieri were clearly divided into two phylogroups. The nucleotide diversity (π) of C. auratus from Korea, China, and Japan showed a range of 0.146%~0.421%, while the range of π of C. cuvieri from Korea and Japan was lower than those of C. auratus (0.0%~0.054%). Moreover, the comparison of CYTB divergence among crucian carp fishes in China, Japan, and Korea indicated that Korean Carassius fishes were distantly related to those from China and Japan, with two exceptions: the pairwise Fst value between Korean C. auratus and northern Chinese C. auratus was not significantly different. In addition, no significant genetic divergence between Korean and Japanese C. cuvieri was detected. We conclude that, despite the morphological similarities, C. auratus and C. cuvieri should be considered as separate freshwater fish resources in conservation efforts for genetic diversity.

Comparative Morphology and Morphometry of the Olfactory Organ of Carassius auratus and Carassius cuvieri (Cypriniformes, Cyprinidae) (붕어 Carassius auratus와 떡붕어 Carassius cuvieri 후각기관의 형태 및 형태계측학적 비교 연구)

  • The olfactory organ of Carassius auratus and C. cuvieri was compared morphologically and morphometrically using stereomicroscopy and statistical program (SPSS version 18.0). The external morphology of the olfactory organ consists of the open semicircular anterior and posterior nostril, and nasal flap. The internal structure showed the oval rosette consisting of several lamellae arranged radiately. In statistical analysis of standard length (SL), olfactory lamellar number (LN), and SL/LN ratio between two species using independent two sample t-test and Pearson's correlation and coefficient, C. cuvieri is the longer SL than that of C. auratus and C. auratus LN (14~20) is lager than that of C. cuvieri (14~16) (P<0.001) and C. auratus LN/SL ratio (12.7±0.7%) is larger than that of C. cuvieri (8.2±0.6%). These results suggest that i) open semicircular nostrils is functional morphology to offset the boundary layer and ii) the lager LN of smaller C. auratus may be a morphological adaptation to reflect its higher olfactory dependence than C. cuvieri and iii) such interspecific difference in LN and LN/SL ratio could be applied as a new taxonomic trait for identification.

Evaluation of the Potential Human Health Risk Associated with the Microcystin Bioaccumulation in the Freshwater Fish from Lake Yeongcheon and Lake Daecheong (영천호와 대청호에서 담수어류의 microcystin 농축에 따른 인체 건강위해성 평가)

  • This study evaluated the potential human health risk on the basis of the level of bioaccumulation and EDI (Estimated Daily Intake) of microcystin-LR, one of hepatotoxic, in organs, including liver, muscle, viscera and gill, of fish from Lake Yeongeheon and Lake Daecheong when the cyanobacterial water-blooms broke out. The result has confirmed that Carassius cuvieri from Lake Yeongcheon contains higher level of microcystin-LR in its organs including liver. In Lake Daecheong, omnivorous Hemibarbus labeo and phytoplanktivorous Carassius cuvieri have shown high microcystin-LR level on average, especially higher for viscera, and Carassius cuvieri has appeared to contain higher level of microcystin-LR in the liver and the gill compared with other species. As a result of comparison between EDI of microcystin-LR from each organs and TDI (Tolerable Daily Intake) of WHO (Chorus and Bartram, 1999) to evaluate human health risk, the EDI levels from Carassius cuvieri's organs except museles have exceeded TDI level at the both lakes. Consequently, the study has proved that microcystin was bioaccumulated in the various parts of fish, and it can be ingested by human resulting in risking human health. Continuous monitoring and reducing consumption of fish, especially Carassius cuvieri, during the cyanobacterial water-blooming period will be needed to protect human health.

Fish Fauna and Community in Cheongpyeong Reservoir (청평호의 어류상 및 어류군집)

  • The fish fauna and community structure were investigated in the Lake Cheongpyeong, Korea, from August 2003 to April 2004. During the period, 43 species belonging 10 families were collected. There were 18 Korean endemic species 41.9%. The five dominant species numerically were A. gracilis (17.4%), Zacco platypus (14.6%), A. yamatsutae (13.2%), Hemibarbus labeo (11.9%), Rhinogobius brunneus (11.7%), and Zacco temmicki (3.0%). The five dominant species in biomass were Erythroculter erythropterus (15.07 kg), H. labeo (13.21 kg), Carassius cuvieri (6.47 kg), A. gracilis (6.05 kg), and Z. platypus (4.71 kg). Of 6 exotic fishes in the lake C. cuvieri, Lepomis macrochirus and Micropterus salmoides were originated from foreign countries but Anguilla japonica, Tridentiger brevispinis, and Chaenogobius urotaenius were introduced from other watershed in Korea.

  • The introduced exotic species has often caused severe problems to the native ecosystem. One of such species is the freshwater fish gengorobuna (Carassius cuvieri) introduced from Japan. The first step to assess harmful effects of this species on the Korean freshwater ecosystem is to discriminate it from the most similar native crucian carp (Carassius auratus). Because traditional morphological identification often gives unreliable results due to their highly similar phenotype, a new more efficient method is needed. For this purpose, molecular markers produced by the efficient one-step PCR method using three primers (DDF, DDR and DDR1) were developed and tested in the present study. This molecular marker will play an important role in monitoring fish community of Korean freshwater ecosystem.

Fish fauna and characteristics of Carassius auratus population in the Boryeong Reservoir (보령호의 어류상 및 붕어 개체군 특성)

  • This study was conducted to investigate the fish fauna and characteristics of the Carassius auratus population in the Boryeong Reservoir in Chungcheongnam-do from October 2017 to June 2018. The collected fish were identified as 3,506 individuals of 15 species from a total of nine families. The dominant and subdominant species were H. nipponensis with 1,706 (48.6%) individuals and C. auratus with 1,021 (29.1%) individuals, respectively. The biomass of C. auratus (246,130g), P. fulvidraco(50,610g), C. cuvieri (14,730 g), S. asotus (11,560 g), and C. carpio (10,930 g) was analyzed. The results of the community analysis showed a dominant index value of 0.87 (±0.2), a diversity index value of 0.78 (±0.5), an evenness index value of 0.47 (±0.2), and a richness index value of 0.99 (±0.5). The length-weight analysis of C. auratus showed a regression coefficient b of 3.06, and a condition factor (K) of 0.0004 with a positive slope. The frequency distribution of the total length analysis of the C. auratus population inhabiting the Boryeong Reservoir showed a high distribution of lengths between 140-160 mm and a low distribution between 230-280 mm. The normalized difference water index (NDWI) was analyzed over the Boryeong Reservoir water surface from 2013 to 2014 using Landsat 8 channel data. The areas where the NDWI was decreased were located at the inflow site of Ungcheon Stream.

Study of the Fish Community of Lake Uiam (의암호의 어류군집)

  • The fish fauna and the dynamics of fish community structure in Lake Uiam, Korea, were investigated from July 2003 to May 2004. During the surveyed period, 41 species belonging to 13 families were collected. There were 11 Korean endemic species (26.8%), including Rhodeusuyekii, Acheilognathus yamatsutae, Sarcocheilichthys nigripinis morii, Squalidus gracilis majimae, Squalidus japonicus coreanus, Microphysogobio yaluensis, Iksookimia koreensis, Koreocobitis rotundicaudata, Silurus microdorsalis, Liobagrus andersoni, and Odontobutis platycephala. Dominant species were Hemibarbus labeo (16.2%), Zacco platypus (13.8%), A. yamatsutae (11.8%), S. gracilis majimae (10.4%), Pungtungia herzi (5.8%), and Hypomesus olidus (4.3%). The biomass of collected fishes were H. labeo (35.9 kg), Cyprinus carpio (22.5 kg), Micropterus salmoides (5.5 kg), Lepomis macrochirus (5.2 kg), Carassius cuvieri (3.3 kg) and Z. platypus (2.4 kg). Of the 9 introduced fishes in Lake Uiam, Cyprinus carpio (Israeli type), Carassius cuvieri, Oncorhynchus mykiss irideus, Lepomis macrochirus, and Micropterus salmoides originated in foreign countries, but Anguilla japonica, Hypomesus olidus, Chaenogobius urotaenius, and Rhinogobius giurinus were introduced from other native river systems.

Field Survey of River Environment at River Basin of Cheongmi-Cheon (청미천 유역의 하천환경 조사 - 2008년 식생 및 어류모니터링 결과를 바탕으로 -)

  • 본 연구에서는 하천 식생 및 어류 모니터링 결과를 바탕으로 청미천 유역의 본류에 대한 하천 수변환경을 평가하였다. 하천 식생은 홍수 전과 후로 구분하여 2회 조사하였다. 식생 조사는 Braun-Blanque의 조사 방법에 따라 표본구 내의 출현식물에 대해 조사하였고 표본구 면적은 자생지 주변의 특징과 형성된 군락의 최소면적에 근거하여 $2{\ast}2m^2$ 결정하였다. 1, 2차 조사 결과 공히 국화과 식물이 많이 출현하였고 다음으로 벼과 식물이 많이 출현하였다. 청미천 조사구간에서 볼 수 있는 식물은 21개과 38종이 출현하고 있는 것으로 조사되었다. 이중 국화과 7종과 벼과 5종, 콩과 4종으로 출현종이 많이 조사되었다. 화봉천 합류점에서는 총 123마리를 포획하여 동종 분류한 결과 1개과 3개아과 7개종이 출현하였다. 1지점의 경우 출현어종은 모두 잉어과로 피라미가 48%로 우점종을 차지하고 있었다. 죽산천 합류지점에서는 총 83마리를 포획하였으며, 출현어종은 1개과 2개아과 7개종의 어류이며 그중 붕어와 피라미가 각각 31%와 23%로 우점하고 있었다. 청미천 조사구간의 출현어종의 개체수를 보면 피라미 68.5%, 붕어 44.3%, 떡붕어 18.1%, 모래무지 14.3%, 왜매치 14.1% 순으로 나타나고 있으며 기타 출현어종은 대부분 4% 내외로 피라미가 우점종으로 조사되었다.

Morphological Development of Eggs, Larvae and Juveniles of the Carassius cuvieri (Cypriniformes: Cyprinidae) (떡붕어, Carassius cuvieri (Cypriniformes: Cyprinidae)의 난발생 및 자치어 형태발달)

  • This study was carried out to clarify the egg, larvae and juveniles development of Carassius cuvieri in Nakdong River, Gyeongbuk. The amount of spawning of female C. cuvieri was about 30,400~44,900 (average 37,650±7,250). The egg shape was circular and the size was 1.24~1.37 mm (average 1.31±0.04 mm, n=30). At 100 hours after fertilization, more than of the total embryos were hatched. The newly hatched larvae had an 4.69~5.65 mm in total length (average 5.15±0.31 mm, n=10) and had egg yolk in the abdomen. At 3 days after hatching, the preflexion larvae absorbed all egg yolk was 6.27~6.70 mm in total length (average 6.59±0.08 mm, n=10). On the 10 days after hatching, the postflexion larvae were 8.71~8.92 mm in total length (average 8.81±0.07 mm, n=10), and the tip of the caudal fin was bent at 45°. On the 42 days after hatching, the total length of 15.1~16.8 mm (average 15.8±0.57 mm, n=10) was transferred to juvenile as the number of fins was (iii17 dorsal fins, iii4 anal fins) reached a constant number of each part.
