Free phenolic acid, soluble phenolic acid ester and insoluble bound phenolic acid were extracted from defatted perilla flour. Their antioxidative effects were compared with those of BHA, AE and TBHQ for soybean oils by measuring acid and peroxide values at 60$^{\circ}C$ for 25 days. The patterns of these extracts were compared by using gas chromatography. Free phenolic acid and soluble phenolic acid ester extracts showed a higher antioxidative effects than BHA and AP. Among phenolic extracts, free phenolic acid showed the most effective antioxidant activity in soybean oil. Six types of free phenolic acid, 3 types of soluble phenolic acid ester, and 2 types of insoluble phenolic acid were found in the extract.
In this study, enzymes were screened for hydrolysis of defatted perilla seed residue (DPSR) and optimal conditions for enzymatic treatment were determined to produce the hydrolysate of DPSR. Also its antioxidant activity and utilization as a culture medium were examined. The combined treatment of Alcalase and Ceremix is most effective for solubilization of protein and carbohydrate in DPSR. The optimal dosage, pH, and reaction time for enzymatic treatment were found to be 2.0% (w/w), 7.0, and 2 h, respectively. Treatment with optimal conditions of enzymes dramatically increased reducing sugar, soluble protein, and total phenolic content. The hydrolysate of DPSR possessed better scavenging activity against cation and free radicals than enzyme-untreated extract. When Leuconostoc mesenteroides 310-12 was cultured in the hydrolysate of DPSR, cell population rapidly increased compared to enzyme-untreated extract, and titratable acidity increased in proportion to the bacterial growth. In conclusion, these results imply that the hydrolysate of DPSR could be utilized as a bacteria culture medium as well as a physiologically active material with antioxidant activity.
This study was investigated physiological activities of enzyme hydrolysates in 80% ethanol extracts from sesame, black sesame and perilla cake. Total phenol contents in 80% ethanol extracts of sesame, black sesame and perilla cake were 116.90, 102.20 and 141.90 mg/g, respectively. Whereas total flavonoid contents were 64.10, 32.00 and 131.90 mg/g, respectively. Total phenol contents of enzyme hydrolysates in 80% ethanol extracts from sesame, black sesame and perilla cake were 413.30, 221.20 and 409.10 mg/g, respectively. Whereas total flavonoid contents were 361.80, 103.30 and 345.80 mg/g, respectively. Electron donating effects, nitrite-scavenging abilities, ferrous ion chelating effect and SOD-like activities increased by emzymatic hydrolysis. The order of Electron donating effects, nitrite-scavenging abilities and SOD-like activities by emzymatic hydrolysis was perilla > sesame > black sesame cake. And the order of nitrite-scavenging abilities(pH 1.2) was sesame > perilla > black sesame cake.
Purpose: This study was conducted to investigate the antioxidative, collagenase and elastase activities of defatted perilla ethanol extracts. In addition, the possibility of its use as a functional cosmetic material has been studied. Methods: In order to investigate the antioxidant function of the perilla extract, electron spin resonance (ESR) was measured with a spectrometer by analyzing the scavenging ability of diphenyl-β-picrylhydrazyl (DPPH) free radicals. Results: The antioxidant activity of DPPH free radical scavenging activity of the perilla extract was about 68% at 85 ㎍/ml, 85% at 20 ㎍/ml, 90% at 40-100 ㎍/ml, and showed antioxidant ability. The inhibitory activity of collagenase was increased to 4.1% at 20 mg/ml, 12% at 40 mg/ml and 26.7% at 100 mg/ml. The inhibitory activity increased in proportion to the concentration. The inhibitory activity of elastase was increased to 3.8% at 20 mg/ml, 15% at 40 mg/ml, 28% at 80 mg/ml and 35.7% at 100 mg/ml. Conclusions: These results suggest that the ethanol extract of perilla may inhibit the antioxidant activity and the activity of collagenase and elastase to improve the skin aging and wrinkles.
The free, ester and insoluble bound phenolic acids in the extracts from defatted perilla seed flour were isolated and their antioxidative activities were evaluated in comparison with commercial synthetic antioxidants. Total phenolic content of the perilla seed was 0.75% as chlorogenic acid. Each percent ratio of the content of free, ester, and insoluble bound phenolic acid to total phenolic content was 87.5, 7.5 and 5.0% respectively. Chlorogenic acid was identified as a major phenolic acid and a small amount of caffeic acid was also identified in the free phenolic acid extract, but they were not found in soluble ester and insoluble bound phenolic extracts by two dimensional paper chromatography. Each type phenolic extract from 30g of deffated perilla flour showed antioxidant activity similar to that of BHT (0.02%, w/w) in 200g of soybean oil substrate inspite of the difference of each phenolic content.
The antioxidative effect of the 75% ethanol extracts of preliminary selected Fritillaria ussuriensis Max(Fs), Houttuynia cordata Thunb(Hc), Portulaca oleracea L.(Po) and Perilla frutescene Var javanica Hara cake(Pf) were tested on palm oil and lard by rancimat test. Each solvent fraction of chloroform, ethyl acetate(EtOAc), butanol and water, was also evaluated its antioxidative effect with some synergists, like as ascorbic acid, citric acid and ${\delta}-tocopherol$. Po extract showed higher antioxidative effect on lard and the fraction of all test plants were the most effective on palm oil and lard with ascorbic acid and ${\delta}-tocopherol$. When 200 ppm of EtOAc fraction of Fs, He and Po extract each with 200 ppm of ascorbic acid were added to palm oil, the antioxidative index(AI, induction time of oil containing each extract/induction time of test oil) were 1.60, 1.53 and 1.47 respectively and 200 ppm of EtOAc fraction of Po and Pf extract each with 200 ppm of tocopherol to lard, the AI were 3.19 and 3.63 respectively.
The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.

Elevation of the activities of phase 2 enzymes such as quinone reductase(QR) provides protection against several types of neoplasia. In this study, we performed partial purification of QR inducer(s) from roasted and defatted perilla meal by solvent fractionation and thin layer chromatography. Cellular QR induction was most notable in chloroform fraction of roasted perilla extract, compared with other solvent fractions. QR inducer(s) was partially purified by TLC, with 0.8 of $R_f$ value in n-butanol : n-propanol : 2N-ammonium hydroxide(10 : 60 : 30). AHH-inducing activity in TLC fractions isolated from methanol extracts of roasted perilla comigrated with QR-inducing fraction, suggesting that QR and AHH are induced by the same compound. TLC fractions shown strong QR-inducing activity also had a potent antioxidative activity, suggesting that cellular QR enzyme is induced by antioxidant(s) present in roasted perilla.
The induction of phase II enzymes including quinone reductase [NAD(P)H dehydrogenase(quinone): NAD(P)H : (quinone acceptor) oxidoreductase, EC 1.6.99.2] is a major mechanism of whereby a large group of heterogeneous compounds prevent the toxic, mutagenic, and neoplastic effects of carcinogen. Using murine hepatoma cells(Hepalclc7 cells), quinone reductase(QR) inducers as the possible chemopreventive agents were screened from rice bran, wheat bran, soymilk residue, defatted soybean cake, defatted sesame and perilla residues. The 80% methanol extracts of defatted sesame and perilla residues induced quinone reductase significantly while the others did have little effect on the enzyme induction. Thin layer chromatography of the extracts showed that the fastest moving band(Rf=0.70) in the developing solvent of n-butanol : n-propanol : 2N ammonia(10 : 60 : 30) was responsible for the enzyme induction by the 80% methanol extracts of defatted sesame and perilla residues. Further identification of active component(s) is in progress.
The antioxidant activity of ethanol-extracts of defatted soybean, sesame, and perilla flours was compared with that of 0.02% BHT in a soybean oil-water emulsion system. The emulsion substrates and control were stored at $46.0{\pm}0.5^{\circ}C$ for 25 days. The peroxide and TBA values of the substrates and control were determined regularly. The activity of the oilseed flour extracts and BHT was estimated by comparing the POV development of the substrates with that of the control. The POVs of the substrates containing the soybean, sesame, and perilla flour extracts and BHT and that of the control after 25 day storage were respectively $43.3{\pm}0.1,\;22.6{\pm}0.7,\;21.5{\pm}0.2,\;38.6{\pm}0.4,\;and\;80.1{\pm}0.8$. The TBA values after 20 day storage were $0.91{\pm}0.05,\;0.67{\pm}0.02,\;0.68{\pm}0.01,\;0.38{\pm}0.01,\;and\;0.62{\pm}0.01$ The soybean, sesame, and perilla flour extracts exhibited considerable antioxidant activity in the oil-water emulsion system. The activity of the sesame and perilla flour extracts was far stronger than that of 0.02% BHT in the emulsion system. The abnormally high TBA values of the oilseed flour extracts in the present study might be attributed to the interference of some carbonyl compounds in the extracts in the TBA value determination.
