Higher Fungi of Cheju-do(1) - Unrecorded mushrooms - (제주도의 고등균류(1) - 미기록종 버섯 -)

  • The mushrooms of this study were collected mainly on Mt. Halla on Cheju-do from April of 1995 to October of 1997. A total of 292 species from 127 genera were identified. Among them, 103 species from 41 genera of the Basidiomycota and 88 species from 13 genera of the Ascomycota were recorded for the first time on Cheju-do. Five species were recorded for the first time in Korea. They are: Typhula phacorriza, Cordyceps crinalis, Cordyceps cochlidiicola, Cordyceps longissima and Cordyceps prolifica.

Effects of LED on the growth of P. tenuipes (LED광을 이용한 눈꽃동충하초 생산)

  • The effects of LED irradiation on growth and functional components of crops are actively being studied. We investigated the effects of red LED on the cordyceps cultivation because Cordyceps is very difficult to find in the wild and to cultivation. The synnemata formation rate of P. tenuipes was 100% under red LED. Also weight, length, and number of the synnemata were significantly increased. We expect that the red LED treatment will be of help to improve farm income.

Biological activities of an entomogenous fungus, Paecilomyces tenuipes grown on silk worm (눈꽃동충하초의 생리활성에 관한 연구)

  • This study was carried out to investigate the biological activities of P. tenuipes formed on silkworm (Bombyx mori) variety. The mean content of total amino acid in the fruiting bodies of P. tenuipes was $1.03{\mu}mole/g$. The distribution rate of amino acid components decreased in the order of Arginine(12.2%) > Glycine(10.5%) > Proline(9.6) > Tyrosine(8.9%) > Serine > Leucine > Threonine. The most abundant amino acid in the fruiting bodies of the Baegokjam, Chilbojam and Hachojam infected with P. tenuipes was arginine, while Yangwonjam was Glycine. The most abundant fatty acid in P. tenuipes was Oleic acid on a dry weight basis. The unsaturated fatty acids such as Oleic acid, Linoleic acid and Linolenic acid accounted for more than 78% of the total fatty acids.

Inhibitory Effect of the Culture Broth of Cordyceps longissima and C. scarabaeicola on Nitric Oxide Production (제주긴뿌리동충하초와 풍뎅이동충하초 배양액의 Nitric Oxide 생성 저해)

  • During search for novel bioactive materials from natural resources with the potential as health food and alternative medicine, the culture broth of Cordyceps longissima (CL) J106, J144 and C. scarabaeicola (CS) J94, J123 were prepared, and their effect on cytotoxicity and nitric oxide (NO) production in RAW 264.7 cells were investigated. Whereas the culture broth of CL J144 and CS J123 had cytotoxicity in RAW 264.7 cells, that of CL J106 and CS J94 did not. The culture broth of CL J106 and CS J94 suppressed NO production in RAW 264.7 cells activated with lipopolysaccharide (LPS) at a dose-dependent manner. These results suggest that culture broth, a by-product of Cordyceps, may have active compounds with anti-inflammatory effect. In addition, it appears that their biological activity is dependent on the strains in spite of the same species.

  • Feeding effects of Cordyceps militaris extract powder in rats were as follows. Triglyceride content was appeared a superior decreasing effect, total cholesterol and phospholipid content were showed relatively low decreasing effect. At this time, 3%(w/w) level of Cordyceps militaris extract powder contents was most effective to decrease these lipids content. As increasing the treating amount of Cordyceps militaris extract powder, the level of plasma glucose was decreased. But these decreasing ratio was not proportioned to the increased treating amount directly. The change of insulin level on plasma, pancreas and femur was proportioned to the plasma glucose concentration also. The Treating amount of Cordyceps militaris extract powder was very close to the decreased ratio of insulin level.

  • Many Cordyceps spp. were collected for excellent strains selection at near Wanju-kun from May to October, 2000. They were identified. As the resulting, They were 2 genera and 4 species. 6 strains were got through 4 species culture. 6 strains are Cordyceps militaris f. albino, C, gracilaides, C. militaris and Isaria japoica. Among them, 4 species used for induction of artifitial fruiting body. 3 strains of them were selected for excellent strains. excellent strains are Cordyceps militaris and Isaria japoica.

Tumor Growth Inhibitory and Immunomodulatory Activities of Cordyceps Militaris Water Extracts in ICR Mice Bearing Sarcoma-180 Solid Tumor (누에번데기 및 누에애벌레 밀리타리스동충하초(Cordyceps militaris) 열수추출물의 투여가 고형암이 유발된 마우스의 종양성장 억제 및 면역기능에 미치는 영향)

  • Hot water-extracts prepared from Cordyceps militaris of silkworm pupa (CMP) or Cordyceps militaris of silkworm larva (CML) were tested for tumor growth inhibitory and immunomodulatory activities in ICR mice bearing sarcoma-180 cells solid tumor, and compared with those of the known compound, cordycepin, found in Cordyceps militaris. Mice were subcutaneously injected with sarcoma-180 cells, and i.p. injected with either saline (Control), 50 mg/kg or 100 mg/kg of CMP (CMP50 or CMP100, respectively), or CML (CML50 or CML100, respectively), or 1 or 2 mg/kg of cordycepin (C1 or C2, respectively) for 10 days. Mice injected with CMP50 or CMP100 showed a 47.3% or 57.6% inhibition in the solid tumor growth (P<0.05), while those injected with CML50 or CML100 exhibited a 35.5% or 37.1% reduction (p<0.05) in solid tumor size compared to the value for control mice treated with saline. Animals injected with corcycepin showed a 26∼30% inhibition in the solid tumor growth (P<0.05). Mice bearing solid tumor and injected with CMP or CML showed a significantly increased thymus weight (38∼44% increase), lymphocyte percentages of CD4+ T-cell, CD8+ T-cell, and NK-cell (63∼110% increase) in the spleen, and interleukin-2 excretion (33∼51% increase) by the isolated splenocytes compared to those in control mice (p<0.05). These results indicate that the anti-tumor activity of hot water extracts of Cordyceps militaris, raised on both silkworm pupa and silkworm larva, appears to be associated with their immunomodulatory activity, and these activities found in Cordyceps militaris are superior to those for the single compound, cordycepin.

Anti-wrinkle Effect of Mycelial Culture Broth of Paecilomyces japonica in the Mixture of Cucumber and Grape Extracts (포도와 오이즙액을 이용한 동충하초(Paecilomyces japonica) 균사체 배양액의 피부 주름개선효과)

  • The possibility of usage as cosmetic resource of the mycelial culture broth of P. japonica in the mixture of cucumber and grape extracts was investigated. In the effect of collagen synthesis promotion in human fibroblast cells, the culture broth of P. japonica of 0.1, 0.5 and 1.0% concentration increased the amount of collagen synthesis than that of control cells. The culture broth increased the SOD activity in the concentration dependant manner of 0.01% to 1.0% in the antioxydation activity test. About 90% of superoxide radical was eliminated by 0.5% concentration of the culture supernatant in the antioxydation test. Anticoagulant quercetin in the course of mycelial growth in the mixture of cucumber and grape extract was accumulated to 15 folds than that of pre-culture. In the skin safety test of the culture broth, there is no any skin damage signal in the tested 30 people. Taken together, we concluded that the culture broth of P. japonica in the mixture of grape and cucumber extracts can be used as a cosmetic resource.

Effects of Medicinal Plant Water Extracts on Expression of Anti-diabetic Enzymes mRNA (약용식물 물 추출물이 항당뇨 효소의 유전자 발현에 미치는 영향)

  • This study evaluated the anti-diabetic effects of medicinal plant water extracts on expression of hepatic glucokinase (GCK), pyruvate dehydrogenase (PDH) and acetyl-CoA carboxylase (ACC) mRNA. GCK, PDH and ACC mRNA expression levels were measured by RT-PCR. The medicinal plants used in our study were Cordyceps militaris (CM), Perilla sikokiana (PS), Salvia miltiorrhiza Bunge (SMB), Panax notoginseng (PN) and Angelica utilis Makino (AUM). We found that GCK mRNA expression was increased to about 181% at the 250 ppm of CM water extract. Furthermore, we also found that CM and AUM water extracts stimulated PDH mRNA expression level related to glucose metabolism, however, PS, SMB and PN did not stimulate PDH mRNA expression as expected. Expression of ACC mRNA was also significantly higher in both CM and AUM water extracts. Overall, the results of our study suggest that CM and AUM water extracts stimulate expression of hepatic GCK, PDH and ACC mRNA.

Studies on Distribution and Utilization of Cordyceps militaris and C. nutans (동충하초속균의 분포 및 Cordyceps militaris와 C. nutans의 이용에 관한 연구)

  • The genus Cordyceps known as an insect parasite forms a sclerotium in insect bodies and then produces perithecia on the single or multiple stromata produced from sclerotium. Collected Cordyceps were identified into 5 species: Cordyceps militaris, C. nutans, Cordyceps sphecocephala, Isaria japonica, and Torrubiella sp. The fruit bodies of Cordyceps in petri-dish cover were fixed by tape and put the lid on water agar plates to isolate these collected Cordyceps. The germinated spores were transferred from water agar to Potato dextrose agar(PDA) after six hours. Mycelial growth of C. nutans and C. militaris was the most successful on Hamada media and was also good on Complete media and PDA. Mannose as a carbon source was good for two species and Glutamic acid as a nitrogen source was satisfactory to C. militaris and Asparagine gave a good result to C. nutans. C. militaris and C. nutans showed similar mycelial growth rate on the media that contained thiamine-HCI, biotine or nicotinic acid as a vitamine. When conidia of C. nutans were inoculated to insects, mortality was high in Artogeia napi L, Hemiptera, Plutella xylostella and 50% in Orthoptera, 12% in Acantholyda posticalise M, but not Agelastica coerulea B. in Aphididae, C. nutans was collected from only Hemiptera in nature, but killing effect on other insects was proved. Mycelial growth and fruit-body formation were good on the media that consist of rice powder 5g, wheat flour 5g, water 100ml, but formed fruit-body was not complete stromata but a mass of conidia according to results of observing microscope.
