Anti-diabetic Effects of Isaria tenuipes in OLETF Rats as an Animal Model of Diabetes Mellitus Type II (제 2형 당뇨모델 OLETF 랫드에서 동충하초의 항당뇨 효과)

  • We evaluated the anti-diabetic effects of Isaria tenuipes in diabetes mellitus type 2. For the experiments, the diabetic animal model OLETF rats were divided to 4 groups: Isaria tenuipes was administered mixed with the high fat diet 45% at dose levels of 0.0%, 0.1%, 1.0%, and 5.0% for 4 weeks. All animals have free access to water and high fat diet 45%. The diabetic clinical markers, including clinical signs, body weight and food intake, organ weights, blood glucose level, insulin level and HOMA-IR index, oral glucose tolerance test, GLUT4 mRNA and protein were measured at a time. After administration for 4 weeks, the blood glucose levels, insulin levels and HOMA-IR index of test groups were decreased compared with control group in dose-dependent manner. The body weight and diet consumptions were reduced in control group at 4 weeks. The treatments of Isaria tenuipes also showed high expression of GLUT4 mRNA and protein in the muscle of OLETF rats. The results suggest that Isaria tenuipes has anti-hyperglycemic effect attenuating blood glucose in the animal model of type 2 diabetes and might be useful as a functional diet for human diabetic diseases.

Preparation of fermented vinegars using mushroom mycelial cultures and characterization of their physicochemical properties (버섯 균사체를 이용한 발효 식초 제조 및 이화학적 특성)

  • This study was conducted to prepare healthy vinegars using the mycelia of various mushrooms (Cordyceps militaris, Phellinus baumi, Pleurotus cornucopiae, and Ganoderma lucidum). Analyses of the bioactive components of these vinegars revealed that those prepared using mushroom mycelial cultures on fermented brown rice had increased bioactive component levels compared to those cultured on unfermented brown rice. The highest beta-glucan contents (78.7 mg/g) were found in P. baumi cultures and 0.34mg/g of cordycepin was detected in C. militaris cultures. Regarding the chemical properties of these mushroom mycelia vinegars (MMVs), those prepared using C. militaris and P. cornucopiae cultures showed higher acidity and reduced sugar content. Sensory assessments of C. militaris and P. cornucopiae MMVs yielded scores of 7.3 and 6.6, respectively. Analyses of bioactive components revealed that C. militaris MMV contained high levels of polyphenols and 786 mg/L of cordycepin. P. cornucopiae MMV contained the highest amount of beta-glucan (20.9 mg/g).

Effects of Paecilomyces tenuipes Water Extract on the Alcohol Metabolism of Rats (눈꽃동충하초 물추출물이 흰쥐의 알코올대사에 미치는 영향)

  • The effect of Paecilomyces tenuipes water extract (PTWE) on the alcohol metabolism was examined on rats. PTWE of 0, 30, 100 mg/kg body weight was administrated orally to the rats 30 min before oral treatment of 3 g/kg body weight of alcohol. Blood alcohol and acetaldehyde concentrations were measured 0.5, 1, 3, 6, 9 hr after alcohol treatment. Hepatic alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ADH), microsomal ethanol oxidizing system (MEOS) activities were measured 9 hr after alcohol treatment. There were no differences in blood alcohol concentrations and area under the curve (AUC) of alcohol. PTWE decreased acetaldehyde concentration and there were significant differences after 6 hr in 30 mg/kg PTWE and after 3 and 9 hr in 100 mg/kg PTWE, respectively. In particular, 100 mg/kg PTWE decreased AUC of acetaldehyde by 44%. However, there were no changes in the hepatic ADH, ALDH, and MEOS activities.

Biosynthesis of Organic Germanium Using Cordyceps militaris (동충하초를 이용한 유기게르마늄의 생산)

  • In the present study, an attempt has been made to produce a high quality medicinal mushroom Cordycops militaris supplemented with organic Ge. Cordycops militaris was cultivated in SDAY liquid medium containing yeast extract 10 g, peptone 10 g, glucose 40 g per liter and chrysalis media supplemented with inorganic Ge at 100, 500, 1000, and 5000 ppm concentrations. The greatest organic Ge production of 442.4 ppm/g and 284 ppm/g were observed in SDAY liquid and Chrysalis media cultures supplemented with 100 ppm inorganic Ge respectively. Similarly, 4,509.7 ppm/g and 1,058 ppm/g of organic Ge were obtained from liquid and chrysalis media cultures supplemented with 5,000 ppm and 1,000 ppm inorganic Ge, respectively. In addition, higher concentration of organic Ge was obtained in mycelia than fruiting bodies. These results indicate that the concentration of organic Ge increase with decreasing inorganic Ge concentration in the medium. This is the first report on production of high valuable Cordyceps militaris contained with organic Ge.

Anti-oxidant and Anti-proliferative Effects of Water Extract Mixture of Cordyceps Militaris and Allium Tuberosum (동충하초 및 부추 혼합 물추출물의 항산화 및 암세포 증식억제 효과)

  • The present study was performed to evaluate the anti-oxidant and anti-proliferating activity of the water extract mixture of Cordyceps militaris (CM) and Allium tuberosum (AT). The water extract mixture rate of CM and AT was optimized by means of a sensory evaluation test. The optimized mixture rate were decided at 70% of CM, 30% of AT, and 10% of apple concentrate as an additive. The values of total acidity, pH, sugar contents, and turbidity of the water extract mixture were 0.1%, 4.28, 9.10 °Brix, and 1.48 respectively. The water extract mixture had effective DPPH radical scavenging activity, reducing power effect, and ABTS radical activity. DPPH radical activities of the water extract and the water extract mixture were 43.2% and 51.7% respectively; their reducing power (OD700) was 1.14 and 1.43 respectively; and ABTS.+ radical activities were 47.1% and 62.2% respectively. Also, the water extract mixture showed a higher anti-proliferating effect than the AT extract on human prostate cancer cells. These results provided experimental evidence that the water extract mixture of CM and AT is a better source of anti-oxidant and anti-cancer ingredients than a single extract of CM. In conclusion, the water extract mixture of CM and AT will be beneficial in development of a functional drink.

버섯을 이용한 젤리 제조 및 품질특성에 관한 연구

  • 버섯은 특유의 향과 풍미 뿐 아니라 단백질, 다당류, 비타민, 무기질 등을 고루 함유란 저칼로리 영양식품으로서 가치가 높고, 최근에는 생체방어, 항상성 유지, 질병의 회복뿐만 아니라 암, 뇌졸중, 심장병 등의 성인병에 대한 예방과 개선효과가 있는 것으로 알려져 기능성 식품소재로서 활용가치가 높아지고 있다. 그러나 버섯류는 대부분 생체 또는 건조품으로 소비되고 있으며 재배기술이 개선되어 점진적으로 생산량이 증가되나. 수요가 이를 따르지 못해 계절적 공급과잉으로 가격파동이 우려된다. 따라서 출하조절과 버섯 수요를 확대를 위하여 영지, 표고, 눈꽃동충하초 그리고 번데기동충하초를 이용한 버섯젤리의 제조와 제품의 색도, 물성 및 기호도를 조사 비교하였다. 버섯 젤리 제조를 위한 추출액의 적정혼합비율을 선발한 결과, 영지버섯 젤리는 버섯추출액 85%, 대추추출액 10%, 황기추출액 5%를 혼합했을 때, 표고버섯 젤리는 버섯추출액 80%, 대추추출액 10%, 감초추출액 5%, 오미자추출액 5%를 혼합했을 때, 눈꽃동충하초와 번데기동충하초 젤리는 버섯추출액 85%, 대추추출액 10%, 감초추출액 5%를 혼합했을 때 가장 우수하였다. 젤화제 종류별로 버섯 추출액에 대한 응고 효과는 모든 버섯에 대해 carrageenan이 가장 효과적이었다. 버섯 젤리의 색도는 carrageenan 첨가량에 따라 큰 차이가 없었고, hardness, gumminess, chewiness는 증가하는 경향이었다. 관능은 젤화가 완전히 이루어지면서 hardness가 낮은 carrageenan 0.6% 첨가가 가장 우수하였다.

Effect of Paecilomyces tenuipes extract on angiogenesis in prostate cancer cells (눈꽃동충하초 추출물이 전립선 암 세포 내 혈관신생인자 발현에 미치는 영향)

  • In this study, the inhibitory effect of Paecilomyces tenuipes extract on PSA and angiogenesis-related factor expression levels were investigated in human prostate cancer cells, LNCaP. P. tenuipes extract significantly inhibited PSA expression in a dose-dependent manner. We also investigated the inhibitory effect of P. tenuipes extract on the expression of angiogenesis-related genes including VEGF, MMP-2, MMP-9, TIMP-1, and TIMP-2. P. tenuipes extract significantly down-regulated the expression of MMP-2 and MMP-9 in a dose-dependent manner. On the contrary, P. tenuipes increased the expression of TIMP-1 and TIMP-2. Our findings indicate that P. tenuipes exhibits an inhibitory effect on angiogenesis in human prostate cancer cells.

Pathomorphological Peculiarity of Dometic Silkworm, Bombyx mori. Infected with Cordyceps, Paecilomyces tenuipes (동충하초 감염 누에의 병리형태학적 특성)

  • This study was carried out to investigate infection process, symptoms and main component of the domestic silkworm, Bombyx mori. larvae and pupa infected with the entomopathogenic fungus Cordyceps, Paecillomyces tenuipes. The Cordyceps, Paecillomyces tenuipes, was highly infectious to the silkworms. A pathogenicities of Cordyceps, Paecillomyces tenuipes, may be highly virulent because of the low resistance or high susceptibility of the silkworms. The silkworm larva infected with Cordyceps formed phialospores on the phialides at the imperfect stage of the genus Cordyceps, But silkworm pupa infected with Cordyceps formed ascospores in the asci at the perfection stage of the genus Cordyceps. The results of analysis of health silkworm pupa and silkworm pupa infected with Cordyceps were obtained that amino acid, fatty acid, and nucleoside were very different.

Instability in in vitro fruiting of Cordyceps militaris (In vitro에서 큰번데기 동충하초 자실체형성의 불안정성)

  • Traditionally, Cordyceps species have been used as a part of herbal medicine in Oriental countries, including Korea for internal health, vigor and to cure different diseases related to heart, lung etc. In recent years, research on artificial fruiting of some species of the genus Cordyceps including C. militaris has been carried out extensively because of their medicinal value. Instability observed in the in vitro fruiting of C. militaris is reported in the present study.
