Analysis of Principal Stresses of O-Ring under Uniform Deformation and Internal Pressure by Stress Freezing Method (응력동결법에 의한 고압기밀용 오링의 주응력 해석)

  • In this research, stress components and principal stresses of O-ring under internal pressure and under uniform squeeze rate were obtained from the stress freezing method of photoelastic experiment and photoelastic experimental Hybrid method for 3-dimensional problems. The obtaining processes of those were introduced. It was certified that the processes of those are effective for the 3-dimensional stress analysis of structures. Stress freezing method, the obtaining processes of those and photoelastic experimental hybrid method were effectively applied to the stress analysis of O-ring made from rubber that under uniform deformation and internal pressure. Stress components and principal stress of Oring under uniform squeeze rate and under internal pressure were analyzed.

  • 본 연구에서는 유동과 냉각수를 이용하는 아이슬러리 제조관정을 관찰하고, 이 시스템의 가장 큰 문제점인 관내동결 특성을 파악하였다. 시험수로는 수도수(city water)를 이용하였다. 시스템을 순환하는 시험수가 제빙관내에서 $0^{\circ}C$ 이하의 과냉각 조건이 형성되어 제빙관을 빠져 나오면서 축냉조에 설치된 충격판상에서 과냉각 해소가 일어났다. 이때 유동과냉각수는 내부에너지를 방출하면서 그 일부가 얼음 입자로 상변화되었고, 나머지는 실험장치를 계속순환하였다. 제빙관에서 시험수와 브라인의 열교환 방식을 대향류와 평행류 두 조건에서 시험수의 과냉각도는 Reynolds 수가 증가함에 따라 감소하는 경향을 보였고, 이들 결과 값으로부터 각각의 관계식을 유도하였다. 또한 브라인의 냉각속도가 0.29~0.53[$^{\circ}C$/min] 범위에서 시험수의 과냉각도는 브라인 냉각속도에 종속되지 않았다.

Cryopreservation of Human Multi-Pronuclear (PN) Zygote by Ultra-Rapid Freezing (인간 다-전핵기 (>2PN) 수정란의 초급속 동결에 관한 연구)

  • The objective of this study was to test whether the developmental capacity of human multi-pronuclear (PN) zygotes after ultra-rapid freezing using EM grid can be maintained. For this experiment, multi-PN zygotes which produced in human IVF program were used as an alternatives of normal 2PN zygotes, and they were separated into 3PN or $\geq4PN$ zygotes to compare their in vitro development and cryoinjury according to PN number. As freezing solution, EFS30 which consisted of 30% ethylene glycol, 18% bcoll, 0.5 M sucrose and 10% FBS added D-PBS was used. The result obtained in this experiment was summarized as follows; When the multi..PN zygotes were ultrarapidly frozen and thawed, the high mean percentages (85.5%) were survived. Also when the cleavage rates between control and freezing group were compared with PN number, there were not significantly different in each group (3PN; 81.3% & 85.4% and $\geq4PN$; 90.0% & 95.7%). When the in vitro development rates after thawing were examined, freezing 3PN group (22.0%) was not differed to control 3PN group (38.5%), although the development result of freezing $\geq4PN$ group (45%) was significantly lower than that of control $\geq4PN$ group (44.4%) (p<0.05). These results demonstrate that developmental capacity of human zygote can be obtained by ultra-rapid freezing method using EM grid and EFS30.

An Performance Evaluation of the Post-installed Anchor System According to the Freezing and Thawing of Concrete and the Corrosion of Anchor (콘크리트의 동결융해와 앵커의 부식에 따른 후 설치 앵커시스템의 성능평가)

  • In this study, performance of the post-installed anchor system was evaluated with reduced strength of concrete and anchor. One of the post-installed anchors was selected to performance evaluation. Concrete strength was reduced by freeze-thawing test, and the post-installed anchor strength was reduced by corrosion test. The post-installed Anchor was installed in concrete of freezing and thawing and original concrete, and corroded anchor was installed in original concrete only. Anchor diameter and installation depth of the anchor were the variable for each specimen. Performance of post-installed anchor system of each specimen was evaluated by pullout test. Anchor diameter and installation depth of anchor, it may affect the performance of the post-installed anchor system from the experimental test results. Fracture mode of each post-installed anchor system had occurred differently depending on the durability of concrete and anchor. The anchor pullout strength from the experimental test results was used in order to compare with the results of CCD (Concrete Capacity Design) method, and CCD equation was modified. Modified equation was able to predict the anchor pullout strength of post-installed anchor system in Original concrete and freezing and thawing of concrete.

Development Process for Decreasing Bitterness of Doraji (Platycodon grandiflorum) (도라지 쓴맛 개선을 위한 공정개발 연구)

  • This study was conducted to evaluate the improvement effects of bitter taste of doraji (Platycodon grandiflorum) with different freezing temperature conditions ($-5^{\circ}C$, $-10^{\circ}C$, $-20^{\circ}C$, and $-70^{\circ}C$). After freezing for 4 h, frozen doraji was thawed and dehydrated. Doraji frozen at $-20^{\circ}C$ showed higher contents of crude fat, crude protein, crude ash, crude fiber, and nitrogen free extract. Moisture, crude saponin, and total polyphenol contents of doraji frozen at $-5^{\circ}C$ were higher than in other samples. Weight loss of doraji was significantly different as freezing temperature decreased and the process repeated. Doraji frozen at $-5^{\circ}C$ showed higher contents of saponin and phenol than the control. Hardness of doraji increased as freezing temperature decreased. According to the preference test, Doraji frozen at $-5^{\circ}C$ showed higher preference and less bitter taste intensity than other samples. These results mean that doraji frozen at $-5^{\circ}C$ with less bitter taste could be utilized widely as a food material.

The Effect of Modified Cryopreservation Method on Viability of Frozen-thawed Blastodermal Cells on the Korean Native Chicken(Ogolgye Breed) (한국재래닭(오골계)종 배반엽세포에 있어서 동결 방법의 개선이 융해 후 생존율에 미치는 영향)

  • For reconstituting genetic resource(Korean Native Chicken: KNC) with grem-line chimeric chicken made with cryopreserved biastdermal cells, the experiments were carried out to optimize cryopreservating conditions. Stage X biastdemal cells were collected from KNC embryos and dissociated. Cells were susupended in medium containing cyopretectant and fetal bovine serum(FBS), and distributed into plastic ampules. Cell susupensions were seeded to induce ice formation at $-7^{\circ}C$ to $-35^{\circ}C$ at in the experiments, the effect of modification of dissociation way, concentration of FBS and cell density on the vaibility of frezen-thawed cells were investigated by trypan blue exclusion. Then change the way of cell dissociation from pipetting to short time vortexing, viability of frozen-thawed cell tended to be increaced from 29 % to 52 %. Increase concentraition of FBS in frozen medium from 20 % to 80 % made viability of thawed cell from 28 % to 35 %. The viability of thawed cells were 33.9% frozen at 2 embryos/0.5 ml, and 43.6 % frozen at 20 embryos/0.5 ml. Furthermore, combination of three modifications make big improvement. The viability of frozen-thawed cell was 60 % for combinated method, and 41 % for general method. This result means the advance to practical cryoreservation of blastdermal cell of the KNC(Ogolgye breed).

Effect of Glycerol and Ethylene Glycol on Post-Thawed Sperm Function in Jeju Horse (제주마의 동결정액 제조에 있어 Glycerol과 EthyleneGlycol이 동결 융해 후 정자의 기능에 미치는 영향)

  • Cryopreservation induces sublethal damage to the spermatozoa, which leads to their reduced fertile life. This study was designed to determine effect of glycerol and ethylene glycol as cryoprotectant in extender on improve the freezability of Jeju horse semen. The semen was cryopreserved with glucose-EDTA extender containing each 5% glycerol, 5% ethylene glycol, 8% glycerol or 8% ethylene glycol, respectively. Post-thawed sperm were evaluated motility, viability, Membrane integrity and acrosome integrity. Post-thawed sperm motility were not significantly differences among treatments. However, sperm viability were significantly higher (p<0.05) in 8% glycerol ($39.85%{\pm}11.41$) than in 5% glycerol treatment ($18.08%{\pm}1.61$). In membrane integrity, swelling sperm ratio was significantly higher (p<0.05) in 8% glycerol ($34.12%{\pm}11.02$) than other treatments. In the percentage of capacitated sperm assessed by CTC staining, F pattern was significantly higher in 8% ethylene glycol than 5% glycerol and 5% ethylene glycol (p<0.05). B pattern ratio was significantly increased in 5% ethylene glycol compared with 8% glcerol and 8% ethylene glycol (p<0.05). Moreover, 8% ethylene glycol treatment was significantly decreased AR pattern ratio compared with other treatments (p<0.05). It is concluded that treatment of 8% glycerol was improved the sperm viability and 8% ethylene glycol was improved the sperm ascrosome integrity after thawing. However, they were not significantly difference between 8% glycerol and 8% ethylene glycol on post-thawed sperm viability. Therefore, 8% ethylene glycol was more effective sperm cryoprotectant than 8% glycerol in Jeju Horse.

  • This study focuses on figuring out the possibility of cosmetics raw materials, especially the A. manihot jinhuakui, The viscosity of the frozen-dried extracts were all increased according to the added concentration, and the high viscosity of the WF extracted with water-based alkaline solvent was confirmed. We used snail mucus to compare the viscosity of the A. manihot jinhuakui. We generated the emulsions of experimental groups with 10% of freezing and drying A. manihot jinhuakui and control group emulsions with 10% of freezing and drying snail mucus. By the results, it shows that the WF experimental group had the highest incremental viscosity rates as 129,200 cPs. In the elastic changes and moisture measurement of the skin, the A. manihot jinhuakui extracts growth rate was the highest more than snail mucus. It demonstrated the possibility of cosmetics raw materials in A. manihot jinhuakui, which takes into account the properties of natural products.

  • Frost heaving and thawing settlement cause unexpected stress around buried pipelines, which results in deformation and permanent demage. A large scale laboratory test has been performed to observe deformation, stress, and temperature of a buried pipe during atmospheric temperature changes. From the experimental results, the stress concentrated around the buried pipe is inevitable and deformation is caused by the frost heaving. Even though backfill materials are sandy soils which are normally assumed to be non frost susceptible, it is revealed that frost demage can happen due to drainage condition, the level of ground water table, and water content.

  • In seasonal frozen areas which have a temperature difference in the winter and spring season like south korea, if stiffness reduction by repeated freezing and thawing occurs to slopes adjacent to private facilities or mountain slopes, safety factor is insufficient to design criteria and landslide could be occurs due to rainfall or snowfall. It can lead to large damage of human life and property. In this study, in order to examine the safety changes of mountain slopes by repeated freezing and thawing, soil samples series of SP and SM by USCS distributed in surface soil of mountain slopes were collected for specimens. Through the direct shear test, the characterestics of frozen soil shear strength were analyzed and by utilizing numerical methods, chracteristics of strength reduction of weathered granite soil according to repeated action of freezing and thawing, changes in the stability of the slopes when applying freezing and thawing of the soil samples were examined. As a result, the maximum shear stress decreased approximately 10%, and slope stability analysis confirmed that required safety factor is less than compare with the non-frozen samples.
