Preparation of Seasoned and Semi-Dried Right-Eyed Flounder and Quality of Its Product during Storage (참가자미 조미 반건조제품의 제조와 저장 중 품질안정성)

  • A study was conducted to investigate the possibility of processing seasoned and semi-dried right-eyed flounder (Pseudopleuronects herzensteini) and to examine its quality during storage at $\pm$$3^{\circ}C$ by analyzing the factors such as moisture content, pH, volatile basic nitrogen (VBN), viable cell count and peroxide value (POV). For processing of the seasoned and semi-dried product, optimum conditions of drying temperature and drying time were $20^{\circ}C$ and 4 hrs, respectively, under 50$\pm$5$\%$ of relative humidity and 2 m/sec of air blast speed. The shelf-lives of the products by vacuum-packaging method in nylon/polyethylene/linear low density polyethylene (0.015/0.045/0.040 mm) laminated film bag were 35 and 16 days during storage at $-3^{\circ}C$ and $3^{\circ}C$, respectively.

  • An attempt was made in this study to investigate the possibility of processing seasoned and semi-dried horse mackerel (Tracharus japonnicus) and to examine its keeping quality, by analyzing the factors such as water content, pH, volatile basic nitrogen, viable cell count, peroxide value, color value and sensory evaluation. For processing of the seasoned and semi-dried product, optimum conditions of drying temperature and drying time were $20^{\circ}C$ and 4 hr, respectively, under $50{\pm}5%$ of relative humidity and 2 m/sec of air blast speed. The shelf-life of the products, vacuum-packaging method in nylon/polyethylene/linear low density polyethylene $(0.015/0.045/0.04\;{\mu}m)$ film bag, was 42 days during storage at $-3{\pm}0.5^{\circ}C$, while that of unpackaged one was only 28 days.

Relationship between Quality of Frozen Surimi and Jelly Strength of Kamaboko (동결 surimi의 품질과 어묵 젤리 강도의 관계)

  • To investigate the desirable index for evaluation of frozen surimi's grades, the relationship between indices for grading of surimi, such as ATPase activity $(Ca^{2+}-,\;Mg^{2+}-\;and\;EDTA-)4, solubility, viscosity and K-value of frozen surimi and jelly strength of kamaboko was studied. The myofibrillar $Ca^{2+}-ATPase$ activity and solubility from frozen surimi of grades SA, FA, A, RA and B gave values of $1.184\pm0.2,\;0.956\pm0.14,\;0.766\pm/0.07,\;0.453\pm0.07\;and\;0.227\pm0.08$(umoles Pi/min/mg) for $Ca^{2+}-$ATPase activity and $93.19\pm5,\;84.62\pm4,\;70.63\pm5,\;41.21\pm4\;and\;32.82\pm4(\%)$ for solubility, respectively. Therefore, the myofibrillar $Ca^{2+}-$ATPase activity and solubility of surimi were closely related to the jelly strength of kamaboko from same material, as the correlation coefficient were 0.9584 and 0.9849, respectively. K-value, the index of freshness, was related to the jelly strength of frozen surimi as the correlation coefficient 0.9053 and shown as SA $15.67\pm1.4,\;FA\;14.94\pm 3,\;A\;28.00\pm5,\;RA\;32.16\pm3\;and \;B\;48.68\pm 5(\%)$. $Mg^{2+}-$ and EDTA-ATPase activity and viscosity were not related to the jelly strength. The $Ca^{2+}-$ATPase activity and solubility were found to be useful index for evaluating the quality of frozen surimi.

Seismic Wave Velocity Characteristics of the Mudeungsan Tuff under the Influence of Freeze-Thaw (동결-융해에 따른 무등산 응회암의 탄성파 속도 특성)

  • We analyzed the changes in the properties of the Mudeungsan tuff by conducting an artificial weathering experiment based on the climatic conditions of Mudeungsan National Park, to evaluate the long-term stability of the columnar jointing in the tuff. The climate of Mudeungsan National Park over 20 years suggests the temperature conditions for freeze-thaw are -20 to 30℃. The change in tuff properties due to weathering were estimated by measuring the elastic wave velocity, which was measured after every 40 freeze-thaw cycles. Based on the origin of the Mudeungsan tuff and fracture distribution in the tuff, the elastic wave velocity in samples from 24 locations was measured at regular intervals in the axial and radial directions. The axial elastic wave velocity of the Mudeungsan tuff is 5,187~5,367 m/s, and the radial elastic wave velocity is 4,001~5,290 m/s. As a result of 200 freeze-thaw cycles, the axial elastic wave velocity decreased by 5.53% for sample MT-1, 4.89% for MT-2, and 5.36% for MT-3. The radial elastic wave velocity decreased by 20.00% for MT-1, 17.02% for MT-2, and 19.84% for MT-3. The decrease in elastic wave velocity due to the freeze-thaw cycles is greater for low values of elastic wave velocity. For the axial elastic wave velocity, the weathering is accelerated after 120 cycles and, for the radial elastic wave velocity, weathering actively progresses from the start of the freeze-thaw cycles. In summary, for a low elastic wave velocity, experimental weathering results in a large decrease in elastic wave velocity. In addition, the Mudeungsan tuff and its columnar joints have a distinct anisotropy.

Studies on the Cryopreservation and Survival after Thawing of Hamster Embryos (Hamster 수정란(受精卵)의 동결보존(凍結保存) 및 생존성(生存性)에 관한 연구(硏究))

  • This experiment was carried out to determine the optimum freezing and thawing rates of the hamster embryos. The female hamsters were induced to superovulate by intraperitoneal injections of 30 i.u. PMSG and mated with males of the same strain of 4 days the PMSG injection. They were killed and embryos were flushed from the oviduct and uterine horn on 3 days after mating. Embryos were flushed with a modified Dulbecco's phosphate-buffered saline and equilibrated with 1.5 M-dimethylsulphoxide by a 3-step procedure. The freezing rates of the samples were $1^{\circ}C/min$ from room temperature to $-6^{\circ}C$ and the samples were seeded at $-6^{\circ}C$. After being held for 3 min at the seeding temperature, the rates were $0.3^{\circ}C/min$ from $-6^{\circ}C$ to $-35^{\circ}C$. From $-35^{\circ}C$ to $-70^{\circ}C$, the rates were divided into $0.1^{\circ}C/min$, $1^{\circ}C/min$ and $10^{\circ}C/min$, respectively. At $-70^{\circ}C$ the samples were plunged directly into liquid nitrogen. The samples were thawed at $4^{\circ}C/min$ and $12^{\circ}C/min$ from $-196^{\circ}C$ to $37^{\circ}C$, and for 2 min in $37^{\circ}C$ water bath, respectively. The average numbers of ovulation points and embryos recovered were 35.1 and 27.0 appearing 77.0% recovery rates. Eight cell embryos in the embryos recovered were 24.8. The survival rates of embryos according to the freezing rates were 55.5~67.7% at $0.1^{\circ}C/min$, 58.8~64.9% at $1^{\circ}C/min$ and 40.5~44.7% at $10^{\circ}C/min$, respectively. The survival rates at $10^{\circ}C/min$ were significantly low. The survival rates of embryos according to the thawing rates were 53.5% at $4^{\circ}C/min$, 53.7% at $12^{\circ}C/min$ and 59.1% in $37^{\circ}C$ water bath. The survival rates, in $37^{\circ}C$ water bath were slightly higher, but we did not find any differences among them. In conclusion, the best freezing rates of hamster embryos were $1^{\circ}C/min$ from the room temperature to $-6^{\circ}C/min$, $0.3^{\circ}C/min$ from $-6^{\circ}C/min$ to $-35^{\circ}C$ and $-0.1^{\circ}C/min$ or $1^{\circ}C/min$ from $-35^{\circ}C$ to $-70^{\circ}C$. The hamster embryos thawed for 2 min in $37^{\circ}C$ water bath showed the best survival rates.

  • For the use of antarctic krill as a fond protein source its compositional characteristics were investigated as the first part of the work includes other subjects such as processing of drill paste, concentrates, and fermented or seasoned product. In general composition of fresh frozen and preboiled frozen krill on board, the contents of crude fat and free amino nitrogen were higher in the former than in the latter which contained a high amount of ash. VBN was rather high as much as 37.6 and $26.4\;mg\%$ in both fresh frozen and preboiled krill. The pH of drill homogenates was 7.1 to 7.2 in both cases. Such a low pH might be attributed to a long term storage and temperature fluctuations during frequent transshipping. The amino acid competition of fresh frozen krill meat showed relatively high amount of glutamic acid, aspartic acid, lysine, proline, and leucine while methionine, histidine, serine, tyrosine, and phenylalanine were lower. Among the essential amino acids lysine and leucine were higher and methionine was lower. In tile composition of free amino acid proline, lysing, arginine, and alanine were higher comparatively to the contents of histidine, aspartic acid, serine, and threonine. It is noteworthy for nutritional qualification that tile essential amino acids particularly as lysine were abundant similarly to that of fishes. Heavy metal contents of krill meat 0.039 to 0.048 ppm as Hg, 0.06 to 0.11 ppm as Pb, less than 0.32 ppm as Zn, 0.008 to 0.012 ppm as Cd, 0.61 to 0.68 ppm as Fe, 0.87 to 1.37 ppm as Cu, and nondetective as Cr. A high Cu content seems to be resulted by tile blood pigment of crustacea. The ratio,1 of edible portion to non-edible portion were 37:63 in fresh frozen and 42:58 in preboiled frozen krill respectively. Release of drip after thawing was more in fresh frozen than in preboiled frozen drill marking $36\%$ and $24\%$ of both respectively.

Freezing Time Prediction of Foods by Multiple Regression Analysis (다중회귀분석에 의한 식품의 동결시간 예측)

  • To develop simple and accurate analytical method for freezing time prediction of beef and tylose under various freezing conditions, freezing time (Y) was regressed against the reciprocal $(X_3)$ of difference of initial freezing point and freezing medium temperature, reciprocal $(X_4)$ of surface heat transfer coefficient, the initial temperature $(X_1)$ and thickness $(X_2)$ of samples which should cover most situations arising in frozen food industry. As results of the multiple regression analysis, equations were obtained as follows. $Y_{tylose}=3.45X_1+7642.84X_2+4642.67X_3+2946.89X_4-431.33\;(R^2=0.9568)$ and $Y_{beef}=0.68X_1+7568.98X_2+2430.78X_3+3293.26X_4-299.00\;(R^2=0.9897)$. These equations offered better results than Plank, Nagaoka and Pham's models, shown in satisfactory agreement with models of Cleland & Earle and Hung & Thompson when were compared to previous models, and the accuracy of its was very high as average absolute difference of about 10% in the difference between the fitted and experimental results. Also, thermal diffusivities of beef and tylose were measured as $4.43{\times}10^{-4}m^2/hr$ and $4.39{\times}10^{-4}m^2/hr$ at $6{\sim}7^{\circ}C$, $2.42{\times}10^{-3}m^2/hr$ and $3.32{\times}10^{-3}m^2/hr$ at $-10{\sim}-12^{\circ}C$. Initial freezing points of beef and tylose were $-1.2^{\circ}C\;and\;-0.6^{\circ}C$, respectively. Surface heat transfer coefficients were estimated $20.57\;W/m^2^{\circ}C$ with no-packing, $16.11\;W/m^2^{\circ}C$ with wrap packing and $13.07\;W/m^2^{\circ}C$ with Al-foil packing, and the cooling rate of immersion freezing method was about 10 times faster than that of air blast freezing method.

Autograft Versus Allograft Bone-Patellar Tendon-Bone in Anterior Cruciate Ligament Reconstruction - A Comparison Of Mid-Term Follow-Up Results - (자가 및 동종 골-슬개건-골을 이용한 전방 십자 인대 재건술 -중기 추시 결과의 비교-)

  • Purpose: To compare the mid-term follow-up results of anterior cruciate ligament(ACL) reconstruction with the bone-patellar tendon- bone(BTB) autograft to those with the BTB allograft. Materials and Methods: Retrospective study was performed in 59 cases with BTB autograft and 42 cases with BTB allograft. Evaluations include Lysholm score, 2000 IKDC subjective knee score, Shelbourne patello-femoral pain score , Lachman test, pivot shift test, KT-1000 arthrometer test and 2000 IKDC knee examination. Results: There were no significant statistic differences between two groups in Lysholm score and 2000 IKDC subjective knee score of more than 70 (p<0.05). Five cases(8.5%) showed the patello-femoral pain score less than 80 according to Shelboume with autograft group and two cases(4.8%) with allograft group (p<0.05). Lachman test, pivot shift test and KT-1000 arthrometer test showed no significant statistic differences between two groups(P<0.05). Fifty-four cases(91.5%) were normal or nearly normal according to the 2000 IKDC knee examination with autograft group and thirty-eight cases(90.4%) with allograft group(p<0.05).Conclusion: BTB allograft as well as BTB autograft is considered to be an acceptable choice for ACL reconstruction.

Cold hardiness of Spodoptera litura Fabricius (Lepidoptera: Noctuidae) (담배거세미나방(Spodoptera litura Fabricius)의 내한성)

  • Supercooling points (SCP) and cold tolerance of the tobacco cutworm, Spodol~rera litura Fabricius. were measured in response to subzero temperatures. SCPs were varied among developmental stages. Eggs showed the lowest SCP (-27$^{\circ}$C). Pupae and adults had the intermediate SCP(- 18$^{\circ}$C). The SCPs (- I0 to - 16$^{\circ}$C) of larvae increased with their ages. Lethal low temperature of each stage was higher than its SCP. Preexposure of eggs and larvae to a sublethal low temperature increased their survival capacities under lethal low temperatures. The sublethal temperature also induced the fifth instar larvae to increase hemolymph osmolality and to produce cold-induced proteins of apparent molecular weights of 20 and 27 kD. These results indicate that this species is classified into a freeze-susceptible insect.

Development of Superovulation Method in Korean Native Goats (한국 재래산양의 과배란 처치 방법 개선)

  • 한국 재래산양 체내수정란 생산에 대한 발정동기화 및 과배란 유도방법과 회수된 수정란의 동결 융해 후 생존율을 조사하였다. 발정동기화를 위해 CIDR+FSH 및 CIDR+PMSG의 방법을 이용한 결과, 배란점 및 회수된 수정란의 수는 CIDR+FSH 처리구에서 16.3개 및 9.4개, CIDR+PMSG 처리구에서 16.4개 및 8.7개를 나타내어 두 처리구간에 유의적 차이는 인정되지 않았다. 회수된 수정란을 형태학적으로 평가한 결과 CIDR+FSH 처리구에서 Gade A, B, C 및 D는 75.8%, 15.2%, 4.5% 및 4.5%를 나타낸 반면 CIDR+PMSG 처리구에서는 52.5%, 16.4%, 16.4% 및 14.8%였으며, 이식 가능한 수정란 (Grade A, B) 수는 CIDR+FSH 처리구가 유의적(P<0.05)으로 높게 나타났다. 회수된 수정란의 완만 동결 융해 후 생존성은 CIDR+FSH 처리구에서 73.3%, CIDR+PMSG 처리구에서 63.3%이었으며, 두 군간의 유의적차이는 인정되지 않았다. 따라서 본 결과는 한국 재래산양 체내수정란의 생산과 회수된 수정란의 보존을 위해서 CIDR+FSH로 발정동기화 시키는 것이 효과적이었다.
