Interrelationship of Matrix Metalloproteinase and $TNF-{\alpha}$ in Human Gingiva with Chronic Periodontitis associated to Type 2 Diabetes Mellitus (단순 만성 치주염 환자 및 2형 당뇨병을 가진 만성 치주염 환자의 치은조직에서 Matrix Metalloproteinase와 $TNF-{\alpha}$ 의 발현 양상 비교)

  • 치주질환의 병원균은 세포벽의 항원에 의하여 조직내 존재하는 mononuclear phagocytes가 활성화되어 cytokine들이 생성됨 으로써 치주 결체조직의 파괴를 진행시킨다. 이런 관련된 cytokine들은 순차적으로 상주하는 치은세포 및 대식세포가 Matrix metalloproteinase 합성을 하도록 유도하여 조직파괴를 시작한다. 이들 Matrix metalloproteinase중 MMP-2, MMP-9 (Gelatinase A,B)는 type IV collagen 및 변성된 interstitial collagen을 파괴하며 치주환자의 치은 열구액, 치은조직, 타액 네에서 높게 보고 되어왔다. 당뇨병은 치주질환의 위험요소중 하나로 달뇨 환자에서는 치주질환의 유병율이 일반인에 비해 높고 치주질환의 중증도도 더 심하여 진행도 빠르다고 알려져 있다. 그 병리 기전 중 하나로는 당뇨 환자에서는 치은 열구액 내 중성구 유래의 Matrix metalloproteinase의 활성 증가 및$TNF-{\alpha}$ 의 활성 증가가 추정되고 있다. 본 실험에서는 제2형 당뇨병 환자와 비당뇨 환자들에서 만성 치주염 부위의 치은 및 건강한 치은에서 염증매개체 중 하나인 MMP-2, MMP-9 및 $TNF-{\alpha}$ 의 발현에 대해 상호 비교 분석함으로서 염증, 혈당이 미치는 영향을 밝히고 제 2형 당뇨병 환자에서 심한 치주조직 파괴의 기전을 연구하고자 하였다. 경북대학교병원 치주과 내원환자 중 제2형 당뇨병 환자와 비당뇨 환자들 및 치주질환이 없는 건강인 대조군을 대상으로 여러 가지 환자요소, 임상 치주상태를 기록하고, 전신적으로 건강한 환자의 건강한 부위(n=8,Group 1), 전신적으로 건강한 환자으 만성 치주염 부위(n=8, Group 2), 제2형 당뇨병 환자의 만성 치주염 부위 (n=8,Group 3)에서 각각 변연치은을 채득하고 액화질소에 급속 동결하였다. Western blotting을 이용하여 각 조직 내 MMP-2, MMP-9 및 $TNF-{\alpha}$ 의 발현을 관찰, densitometer를 이용하여 상대적 발현을 정량, 각 조직의${\beta}-actin$을 이용하여 표준화하여 실험군과 대조군들의 평균치를 비교하였다. 비당뇨 환자들의 만성 치주염 부위 및 제2형 당뇨병 환자의 만성 치주염 부위에서 모두 건강 대조군에 비해 MMP-2와 MMP-9 의 발현이 증가되었다. 또한 MMP-2와 MMP-9는 2형 당뇨 환자의 만성 치주염 부위가 비당뇨 환자의 만성 치주염 부위보다 증가된 발현양상을 보였으며, $TNF-{\alpha}$ 발현 비교시 각 군간 유의성 있는 변화는 없었으나 2형 당뇨환자군에서 MMP-2 및 MMP-9의 증가와 함께 다소 증가 양상을 보였다. 결론적으로 본 실험에서 MMP-2 및 MMP-9의 증가가 만성 치주염 및 2형 당뇨 환자에서의 만성치주염에서 비당뇨환자 보다 MMP-2, MMP-9의 증가양상을 보여 주었으며 $TNF-{\alpha}$ 가 2형 당뇨환자의 만성치주염 진행과정에 기여인자로써 역할을 하는 것으로 생각된다.

Mucilage Separation of Korean Yam Using Microparticulation/Air Classification Process (초미세분쇄/공기분급을 이용한 마의 점질물 분리)

  • To separate and concentrate mucilage from yam(Dioscorea batatas DECNE), yam was dried, microparticulated using impact mill and air-classified at different air classifying wheel speed(ACWS) in classifier. As ACWS increased from 5,000 rpm to 22,500 rpm, the contents of dietary fiber, protein and lipid of air classified microparticles(ACM) increased remarkably. Especially the ACM with ACWS over 15,000 rpm showed 36.41% dietary fiber and 16.66% protein. The dietary fiber and protein components were concentrated to $2.5{\sim}9.0$ times as compared with whole yam powder. Concomitantly the non-fibrous carbohydrate decreased from 88.31% to 16.84. The damaged starch(%), WSI and WAI of ACM of ACWS over 15,000 rpm were $1.5{\sim}3.0$ times higher than those of ACM under ACWS 15,000 rpm. The apparent viscosity of ACM was 0.0800 Pa s over ACWS 15,000 rpm and 0.0080 Pa s under ACWS 15,000 rpm. Judging from viscosity of ACM, the mucilage component of yam was concentrated to 10 times. In conclusion, the optimum process to separate and concentrate the mucilage from yam consisted of the microparticulation to $5{\sim}30{\mu}m$ and the air-classification at ACWS over 15,000 rpm.

Time-related Histopathologic Changes of Fresh Frozen Vascular Xenograft in Pig-to-goat Model (돼지-염소 모델에서 신선 동결된 이종 경동맥 이식편의 시간 경과에 따른 조직병리학적 변화)

  • Background: As determined from the recent investigations of discordant cardiac xenotransplantation, hyperacute rejection occurs mainly at the endothelial cells in donor microvascular systems, but this does not occur at cardiac valve leaflets or at medium-to-large caliber vessels. On the basis of this background, this study was performed to look into the biocompatibility for transplantation of a middle or large diameter xenogenic blood vessel by conducting xenogenic arterial transplantation with the carotid artery in a pig-to-goat model. Material and Method: The experimental group was composed of 10 pairs of pig-to-goat combinations. They were divided into each period of 1 week, and 1, 3, 6 and 12 months. Four carotid artery grafts obtained through collection of the bilateral carotid arteries from two pigs were preserved at $-70^{\circ}C$ without other treatment, and then they were transplanted into the bilateral carotid arteries of two goats. Doppler ultrasonography was done on a periodic basis after transplantation to evaluate the patency of the grafted blood vessel. At the ends of a predetermined period, the grafts were explanted from the goats and they underwent gross examination. Hematoxylin-eosin and Masson's trichrome staining were conducted. In addition, in order to examine the immunological rejection of the grafted xenogenic blood vessel, immunohistochemical staining was conducted with T-lymphocyte indicator and von Willebrand factor. Result: Two goats at the each one-week period and the one-year period died during the experimental period because of a reason unrelated to the experimental procedure, and the remaining 8 goats survived until the end of each experiment period. On Doppler ultrasonography, unilateral carotid artery occlusion was found in a goat, whose period was specified as 3 months, among the 8 survived goats. However, the vascular patency was maintained well and there was no graft that formed aneurysms in the other goats. On gross examination, the region of vascular anastomosis was preserved well, and calcification of the grafted blood vessel was not shown. Histologically, the endothelial cells of the graft disappeared one week after transplantation, and then there was progressive spread of the recipients' endothelial cells from the anastomotic site. The reendothelialization occurred over the whole graft at one month after transplantation. The neointimal thickening and adventitial inflammation became severe by 3 months after transplantation, but this lessened at 6 months and 12 months, respectively. The rate of CD3 positive cells was very low among the infiltrated inflammatory cells. Conclusion: The fresh-frozen xenogenic artery kept its patency without being greatly influenced by xenogenic immune reaction.

Quantitative Analysis of 1-deoxynojirimycin in Mulberry Leaves (뽕나무 유전자원의 1-deoxynojirimycin 함량 변이)

  • Countries of the world are competing for the collection and utilization of genetic resources, which is a resource of value-added industrialization. We are building database of silkworm and mulberry resources with morphological, genetic characteristics data etc. Moreover, recent bioactive compound has been receiving increasing attention. So we analyzed 1-deoxynojirimycin(1-DNJ) content as a this item in addition to the basic information using 363 strains(varieties) of mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for 1-DNJ test. As a result, 1-DNJ mean content of 363 mulberry strains was $0.176{\pm}0.077%$, and the coefficient of variation (CV) was 43.5%. The variation between the strains was greatly severe. Among the tested strains, 'C1D89/29' was showed the highest content of 0.47%, whereas 'Pumbo 24' and 'Turkey E' were showed the lowest content of 0.05% respectively. The content of 1-DNJ of 16 mulberry varieties for silkworm rearing were compared. Average concentration was $0.17{\pm}0.04%$, and the coefficient of variation (CV) was 22.8%. Variation among cultivars was not significant. Finally we selected 1-DNJ high-containing 11 strains. They are as follows. 'C1D89/29', 'Hiihak', 'Jeokasibmunja', 'Gweonchil', 'Botongsibmunja', 'Jeokchuk', 'bulguksang', 'Geunsookgojo', 'Busanggeum', 'Guksang 20' and 'Taekwang' which are more than twice of the overall average content.

  • We analyzed rutin content using mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for rutin test. As a result, mean content of mulberry strains was $0.52{\pm}0.25%$, and the coefficient of variation (CV) was 47.9%. The variation between the strains was greatly severe. Among the tested strains, 'Buyoungsang' was showed the highest content of 1.37%, whereas 'Cheongeunsipmunja' and 'Mujeonsipmunja' were showed the lowest content of 0.06% respectively. The content of rutin of 16 mulberry varieties for silkworm rearing were compared. 'Cheongolppong' was showed the highest content of 0.69%, whereas 'Cheongilppong' was showed the lowest content of 0.14 %. Finally we selected rutin high-containing 9 strains. They are as follows. 'Buyoungsang', 'Yeulbon', 'Dangsang 5', 'A8', 'Aja', 'Seongsu 5', 'Sawonppong 12', 'Hwanyoupjosaengnosang' and 'Hwansipjosaeng' which are more than twice of the overall average content.

Shampoo Characteristics of Botanical Extract Containing Green Tea, Saururus chinensis and Prunus padus (녹차, 삼백초, 귀룽나무 등을 포함한 식물성 추출물의 샴푸로서의 특성)

  • In this research, botanical extracts containing green tea, Saururus chinensis and Prunus padus were tested to see possibility as shampoo. Leaves of Green tea and Saururus chinensis were extracted with hot water. Prunus padus bark extract was applied reverse osmosis and ultrasonic extraction. When interfacial tension was measured among shampoo including botanical extracts, Sodium dodecylsulfate(SDS) and Quillaja Bark Saponin(QBS), that of shampoo was lower than that of SDS and QBS at lower concentration, however, it showed similar interfacial tension at 100% concentration. Shampoo showed moderate antimicrobial activity in Staphylococcus aureus and Candida albicans. Botanical extract did not indicate cell toxicity up to $350{\mu}g/ml$ concentration in MTT assay. Shampoo containing botanical extract was stable for 3 months, however, it showed considerable variation in pH and viscosity. In conclusion, shampoo containing botanical extract shows strong possibility for natural shampoo if the formulation is modified.

Detailed Deterioration Diagnosis and Analysis of Site Environment for the Taean Dongmunri Maaesamjonbulipsang (Rock-carved Standing Triad Buddha Statue), Korea (태안 동문리 마애삼존불입상의 정밀 훼손도 진단과 위치환경 분석)

  • This study focus on the surface deterioration diagnosis based on the material characteristics of Taean Dongmunri Maaesamjonbulipsang(rock-carved standing triad Buddha Statue) and the weathering environment analysis. Rock materials of the triad Buddha is coarse-grained biotite granite which is composed mainly of quartz, K-feldspar, plagioclase, biotite and muscovite. The triad Buddha was serious surface irregularity(13.5%), exfoliation(12.3%) and discoloration(46.6%), and the physical characteristics using ultrasonic velocity appeared fourth grade (0.59, highly weathered). Korean style wooden shelter of the triad Buddha(closed type) maintained high humidity environment (daily mean; 86.6%) than exterior environment, and appeared the possibility of deterioration by freezing-thawing because the temperature showed below zero temperatures in winter. The shelter was changed from closed-type to open-type to relieve the moisture problem recently. But the moisture problem is yet to be solved because surface water flowing along northern wayside is flowed into triad Buddha. Therefore, environmental control will need to stop inflow of water into triad Buddha.

Deterioration and Microclimate in the Shelter for the Gaetaesajiseokbulibsang (Standing Triad Buddha Statues in Gaetaesaji Temple Site), Nonsan, Korea (논산 개태사지석불입상의 손상도와 보호각 내부의 미기후 환경)

  • The Gaetaesajiseokbulibsang (Treasure No. 219) consists of light gray and coarse to medium-grained granodiorite with feldspar phenocrysts in part. Magnetic susceptibility of the rock material was measured as 12.06(${\times}10^{-3}$ SI unit), being different from the granite($0.19{\times}10^{-3}$ SI unit) in the Mt. Cheonho. This indicates the raw material has been supplied from the outside. As a result of deterioration assessment, exfoliation of the Right Buddha, cement and dust of the Main Buddha were estimated as 35.2%, 21.1% and 25.0%. The ultrasonic velocity was measured as 2850.2m/s(Main Buddha), 2648.4m/s(Left Buddha) and 2644.5m/s(Right Buddha). The compressive strength calculated from the velocity showed low in the Right Buddha among three and the all pedestal parts which corresponds to the result of deterioration assessment. The indoor mean temperature and relative humidity of the shelter were $13.7^{\circ}C$ and 79.0%. It is evaluated that the indoor microclimate was stable and the shelter functioned to reduce climatic fluctuation of the outdoor. However, water condensation was occurred on the surface of the pedestal part during spring and summer, and freezing in winter season might also be done. These factors were probable to be a main cause of the surface exfoliation of the Triad Buddha Statues. Therefore, dehumidification and heating system in the shelter should be applied to prevent further deterioration.

Comparative Study of Prolonged Preservation Methods in Rabbit Heart for Transplantation (이식을 위한 가토심장의 장기 보존방법에 관한 비교 연구)

  • The successful cardiac transplantation depends partly on the donor heart preservation by a solution that will ensure recovery of myocardial function. The purpose of this study was to perform the evaluation of various preservation solutions and to accumulate the data on the requisites for ideal preservation solution. The experimental setup was the constant pressure Langendorffs perfusion system. Isolated rabbit hearts were perfused for 20minutes with unarm Krebs-Henseleit solution, stored for 4 hours in cold preservation solution after cardioplegia, and then were reperfused for 20minutes. The 4 experimental groups were prepared Hartmann's solution group (group 1, control), modified Euro-collins solution group(group II. MEC), modified University of Wisconsin group (group n, MUW), and CK solution(made by the author) group (group W, CK). The parameters for assessing the preservation ability were levels of enzymes in freezed myocardial tissues (lactate, creatine kinase-MB and adenosine deaminase), coronary flow. left ventricular developing pressure and dpldt. In conclusion, the ability of preservation for isolated rabbit heart was excellent in CK solution and modified University of Wisconsin solution, and poor in modified Euro-collins solution, compared with Hartmann solution. CK solution has low potassium concentrations(34.2mEq/L) and includes various substrates to be salutary on myocardial preservation. This fact may indicates the necessity of further refinements in selection or composition of electrolytes and substrates.

  • Primary malignant neoplasm of the pericardium is very rare, Neoplastic involvement of the pericardium may result in rapidly developing hemorrhagic effusion. A 30-year-old male who occasionally suffered from chest tightness was referred to our hospital under the diagnosis of unstable angina. He presented with acute chest pain and severe dyspnea that had developed one day previously. The diagnostic investigations such as echocardiography, chest CT and magnetic resonance image suggested cardiac tamponade that was caused by rupture of the pericardial teratoma. An operation to remove the tumor and effusion was performed. The pericardial mass was completely excised, and the result of the frozen biopsy favored malignancy. The final pathologic report was malignant fibrosarcoma of the pericardium and no malignant cells were found on the cytology of the pericardial effusion. The patient had a smooth postoperative course and was referred to another hospital for additional radiation therapy. We report here on this case of cardiac tamponade that was caused by primary pericardial fibrosarcoma, and this required urgent diagnosis and surgical management.
