Toxic effects of Aroclor 1016 and bisphenol A on marine green algae Tetraselmis suecica, diatom Ditylum brightwellii and dinoflagellate Prorocentrum minimum (해양 녹조류 Tetraselmis suecica, 규조류 Ditylum brightwellii, 와편모조류 Prorocentrum minimum에 대한 Aroclor 1016과 비스페놀 A의 독성 효과)

  • Microalgae are the potential bioindicators of environmental changes, for the environmental risk assessment as well as to set limits for toxic chemical release in the aquatic environment. Here, we evaluated the effects of two endocrine disrupting chemicals (EDCs), namely bisphenol A (BPA) and Aroclor 1016, on the green algae Tetraselmis suecica, diatom Ditylum brightwellii, and dinoflagellate Prorocentrum minimum. Each species showed wide different sensitivity ranges when exposed to these two EDCs; the 72 h effective concentration ($EC_{50}$) for these test species showed that Aroclor 1016 was more toxic than BPA. $EC_{50}$ values for the diatom D. birghtwellii were calculated at 0.037 mg/L for BPA and 0.002 mg/L for Aroclor 1016, representing it was the most sensitive when compared to the other species. In addition, these results suggest that these EDC discharge beyond these concentrations into the aquatic environments may cause harmful effect to these marine species.

Development of Ecotoxicological Standard Methods using Early Life Stage of Marine Rotifer Brachionus plicatilis and Benthic Copepod Tigriopus japonicus (윤충류 Brachionus plicatilis 및 저서 요각류 Tigriopus japonicus의 초기 생활사를 이용한 해양생태독성시험 방법에 관한 연구)

  • Marine ecotoxicological standard method was applicated using marine rotifer and benthic copepod as primary consumer of marine ecosystem. Marine rotifer, Brachionus plicatilis and benthic copepod, Tigriopus japonicus were designated as standard test species with the endpoints of 24hr neonate mortality (24hr $LC_{50}$) and 48hr population growth (48hr $EC_{50}$) for rotifer, and 48hr nauplius mortality (48hr $LC_{50}$) for benthic copepod. Tests method was referred to those of ASTM (American Society for Testing and Materials) with the replacement of test species which are widely distributed in Korean waters. The two species showed a wide tolerance on salinity ($5{\sim}35\;psu$) and can be easily cultured in small space even they were not as sensitive as in the mortality test using the nauplius of marine invertebrates. However, these species revealed the significant "concentrationresponse relationship" tested with ocean disposal wastes, and reproducibility using cadmium as standard reference material between laboratories. Accordingly, these two species have good potential as test species for marine ecotoxicological test species. Also, we strongly recommend that at least one of these species be included in the test species of "battery test" in marine bioassay.

Effect of Cyclohexnae on the Lung Toxicity in Rats (Cyclohexane에 의한 흰쥐의 폐독성)

  • In order to search the target organ of cylclohexane toxicity, the rats were intraperitoneally treated with cyclohexane (1.56 g/kg of body wt.) four times every other day. In the increasing rate of organ weight per body weight (%) in cyclohexane-treated animals, the lung was highest among the liver, spleen, small intestine, stomach, heart and kidney. And in the decreasing rate of glucose-6-phosphatase (G-6-Pase) activity in each organ, that of lung was also highest among all organs. Lung MDA content was significantly increased (p<0.05) by the cyclohexane treatment. On the other hand, microsomal aniline hydroxylase activity in lung tissue both of control and cyclohexane-treated rats was greatly low as could be scarcely measured, but that in liver possessing high activity was significantly increased (p<0.05) in cyclohexane-treated rats compared with control. Alcohol dehydrogenase activity in lung was markedly higher than that of liver and the latter was significantly (p<0.05) increased by the cyclohexane treatment. In conclusion, cyclohexane treatment to the rats showed mainly lung toxicity and it may be responsible for cyclohexanon, cyclohexane metabolite, distributed from liver.

Follow-up Study of Children with Anthracycline Cardiotoxicity (소아 Anthracycline 심독성의 추적 관찰)

  • Purpose : We studied the relationship between anthracycline cumulative dose and anthracycline cardiotoxicity in childhood cancer and followed up 40 children with anthracycline cardiotoxicity. Methods : A retrospective study was performed in 154 children who received anthracycline chemotherapy between January 1995 to December 2000. Cardiotoxicity was defined when the left ventricular fractional shortening(FS) was below 26%; it was divided into two groups, mild and severe cardiotoxicity, according to the FS. We followed up survivors with cardiotoxicity, and checked their present cardiac function by physical activity, echocardiography, electrocardiography(EKG) and chest X-ray. Results : Of the 154 children treated with anthracyclines, forty(26.0%) were diagnosed as cardiotoxicity. The incidence of cardiotoxicity increased in exponential fashion with increases in the cumulative dose of anthracyclines. There was minimal increase of incidence until a dose of $300mg/m^2$ after which the incidence increased rapidly. After mean $3.8{\pm}1.8year$ follow-up of 23 survivors with cardiotoxicity, FS increased significantly. EKG and chest X-rays were not helpful for the diagnosis of cardiotoxicity because of their low sensitivity and specificity. Conclusion : Although convenient, non-invasive and inexpensive, EKG and chest X-rays were not helpful for the follow-up of anthracycline cardiotoxicity. Almost all survivors with anthracycline cardiotoxicity have improved in both physical activity and echocardiographic findings after discontinuation of anthracyclines.

Research Trends for Soil-Related Algal Toxicity (토양 관련 조류독성 연구동향)

  • Soil ecological risk assessment requires terrestrial toxicity data based on trophic levels including plants, earthworms, nematodes, and springtails. To expand the trophic levels, it is needed to consider primary producer algae, nearly distributed in terrestrial environment, as representative terrestrial test species. In this study, we collected research cases focused on soil-related test species and exposure media from SCI papers, and analyzed exposure media, test species, test chemicals, and other test methods, for reviewing research trends of soil-related algal toxicity. Up to now, in the soil-related algal toxicity, test species were 8 cases (Pseudokirchneriella subcapitata, Chlorella vulgaris, Scenedesmus bijugatus, Chlorococcum infusionum, Scenedesmus subspicatus, Nostoc linckia, Synechococcus elongatus, and Chlorococcum sp.) and endpoints were cell count or photosynthetic pigment content. Also, 5 of exposure media were liquid medium, soil extracts, porewater, agar medium, and soil. Most of papers used algae isolated from natural soils or soil extracts. There were only one case for assessing algal toxicity in soil medium. More researches regarding algal toxicity in soil environments need to be conducted consistently.

Improvement of Performance Test Standards for Marine Pollution Prevention Materials and Chemicals (for Eco-toxicity Test) (해양오염방제 자재·약제의 성능시험기준 개선방안에 관한 연구(생태독성시험 항목))

  • This study suggests ways to improve the standard test method and judgment criterion for the "Eco-toxicity Test" based on the rules and regulations provided in 'performance and qualifying test standards for marine pollution prevention materials and chemicals' in the Republic of Korea. Compared with the relevant references of other countries, this study attempted to find the limitations in the existing standards. As for the growth inhibition test of algae using Skeletonema costatum as an indicator, applying comparative analysis to measure specific growth rates, together with statistical analysis, instead of applying current methods, judged by the appearance of colors from the culture medium was suggested. Considering the exponential growth phase of the test species, the test duration was suggested to be reduced to less than four days. For the test with fish as an indicator, resetting the appropriate body size was suggested to, show consistent sensitivity against chenicals applied during testing. Furthermore, it is necessary to consider the criteria needs, which should be established in reasonable and objective ways. For the testing species, marine rotifer, Brachionus plicatilis could be a replacement for Artemia sp., and a bivalve for fish in the test. To improve the performance effectiveness of the "Eco-toxicity test", it is worth considering the designation of accredited testing institutes, by placing it in the same loop. Thus it is also expected to have a reliable management system in place, having the capacity of cost calculation properly and adjusting testing items if required.

Establishment of Marine Ecotoxicological Standard Method for Larval Fish Survival Test (어류 자어의 사망률을 이용한 해양생태독성시험 방법에 관한 연구)

  • Marine ecotoxicological standard method using fish larvae was established with the standard test species of Oryzias latipes(Japanese Medaka) and Paralichthys olivaceus(flounder) and with the 7 day $LC_{50}$ as endpoint. Test method referred to the USEPA(1994) with the replacement of test species found in the Korean water. Standard test species were selected in terms of the species supply and ecological importance in Korean waters. Japanese medaka can be reared with small tanks in the lab and has wide tolerance on salinity, and flounder eggs can be easily obtained from commercial fish hatcheries. General conditions for larval fish toxicity test are as follows. The possible salinity ranges for toxicity test were $0{\sim}35\;psu$ for medaka and >20 psu for flounder. Test type was designated as static non-renewal test if the dissolved oxygen in the test chamber does not fall below 4.0 mg/L. Ages of test species were selected as 7 days after hatched for medaka(about 5 mm TL) and 25 days for flounder(about 10 mm TL) because of the low natural mortality after these periods. Test can be accepted when the survival rates are over 80% in control. Also, species sensitivity on standard reference materials(copper, cadmium or zinc) must be provided with the toxicity test results.

Application of the Ecotoxicological Standard Method using Population Growth Inhibition of Marine Phytoplankton (식물플랑크톤의 개체군성장저해율을 이용한 해양생태독성 시험방법에 관한 연구)

  • A series of experiments were conducted to establish a marine ecotoxicological standard method using marine primary producers, Skeletonema costatum, Heterosigma akashiwo, Prorocentrum micans, Isochrysis galbana, Tetraselmis suecica as candidate test species. Marine diatom, S. costatum was selected as standard test species in terms of the ecological roles and international uses as test species. Test methods and test acceptability criteria are as follows; $72{\sim}96$ hr population growth inhibition $EC_{50}$ as endpoint. Static non-renewal method, and population growth rates over 0.04/hr in control as test acceptability criterium. S. costatum is widely distributed in the world ocean and used for standard species of marine toxicity test method by ISO (International Standardization Organization). Possible salinity ranges for this test method are $20{\sim}35\;psu$, and reproducibility and interlaboratory test results were consistent through the calibration tests. Sensitivity of the test method was comparable or better than other toxicity tests such as rotifer neonate mortality, bioluminescent bacterial inhibition, seaweed sporulation and sea urchin fertilization tests.

Comparative toxicity of some pesticides on reproduction of Korean native freshwater Cladocerans, Moina macrocopa and Daphnia sp. (한국산 물벼룩 Moina macrocopa와 Daphnia sp.에 대한 수종 농약의 번식독성 비교)

  • The reproduction toxicity of several pesticides on two Korean water flea was investigated to develop a new standard species used for ecological risk assessment of pesticide. Moina macrocopa and Daphnia sp. were exposed to 4 different types of pesticides over 10 and 21 days, respectively. No-ohserved effect concentration (NOEC) for synthetic pyrethroid, fenpropathrin on Moina macrocopa and Daphnia sp. were 0.17 and $0.06\;{\mu}g\;L^{-1}$, respectively. Diazinon, carbofuran and myclobutanil were in the order of their reproduction toxicity to cladocerans tested. There were large differences between Moina macrocopa and Daphnia sp. in their susceptibility to fenpropathrin, diazinon and carbofuran except myclobutanil. Daphnia sp. is more sensitive than M. macrocopa to pesticides tested. Therefore Daphnia sp. may be a good surrogate species to assess the reproduction effect of pesticides on aquatic invertebrates. M. macrocopa also be a good surrogate species because it is one of the most abundant cladocera in agricultural environment, especially rice paddy in Korea. In addition to it's ecological importance of wide spread distribution, it has also economical importance to make possible to shorten period for reproduction test using M. macrocopa due to it's short life cycle.

Marine Ecotoxicological Evaluation on HNS Spill Accident : Nitric Acid Spill Case Study (HNS 유출사고가 해양생물에 미치는 생물독성 영향평가 : HNO3 유출사고 대상)

  • This study intends to evaluate the effect of nitric acid($HNO_3$) spill accidents on the marine ecosystem, while $HNO_3$ is known as one of the typical HNS. For this purpose, we performed (1) the growth inhibition test by using phytoplankton(Skeletonema costatum), (2) acute and chronic toxicity test by using invertebrate(Brachionus plicatilis and Monocorphium acherusicum), (3) fish(Cyprinodon variegatus) and (4) bacteria(Vibrio fischeri). In these tests, we observed the (1) pH changes induced by the nitric acid spill and (2) changes in nitrate($NO_3$) concentration disassociated from nitric acid after the accident, respectively. The toxicity test result on pH changes induced by $HNO_3$ shows that the no observed effect concentration(NOEC), lowest observed effect concentration(LOEC) and 50 % effect concentration($72h-EC_{50}$) values of M. acherusicum are pH 7 (0.3 mM), pH 5(1.1 mM) and pH 5.2(1.4 mM), respectively, indicating that M. acherusicum is the most sensitive species. The chronic toxicity test (population growth rate test) on $NO_3{^-}$ of B. plicatilis show that the NOEC, LOEC and $96h-EC_{50}$ are 5.9 mM, 11.8 mM and 32.6 mM, respectively, indicating that B. plicatilis is the most sensitive species. In conclusion, toxic effecst on the marine organism caused by the nitric acid spill accident is determined to be so slightly except for the most adjacent area of the ship in pH scale and such concentration of nitrate, to the extent of directly influencing the survival and reproduction of the marine organism, is determined practically not to be applicable in the typical accidents in the sea.
